Gun and street group violence remains a serious problem in cities across the United States and the focused deterrence strategy has been a widely applied law enforcement intervention to reduce it. Although two meta-analytical studies concluded that the intervention had a significant effect on violence, questions remain about how violence changes across space and time during and after the intervention. This study applies novel geospatial analyses to assess spatiotemporal changes in gun violence before, during, and after the implementation of Philadelphia Focused Deterrence. Emerging hot spot analysis employing Space-Time cubes of ten annual time bins (2009–2018) at the Thiessen polygon level was used to detect and categorize patterns. The analyses revealed a non-significant decreasing trend across the ten-year period. Furthermore, there were ninety-three statistically significant hot spots categorized into four hot spot patterns: fourteen new hot spots; twenty-three consecutive; one persistent; and fifty-three sporadic. There was no evidence showing statistically significant hot spots for the "diminishing" pattern. Knowledge of these patterns that emerge across micro-locations can be used by law enforcement practitioners to complement data-driven problem solving and fine tune these strategies and other place-based programming. Policymakers can use findings to prioritize resources when developing complementary prevention and intervention efforts by tailoring those efforts to the different emergent patterns. Many inequities exist in serving and supporting Black survivors of violent crime. A key question in reducing inequities in care after victimization is whether police first responders and other formal system providers identify the victim as an "offender" and/or someone who is "undeserving" of supports. These labels and associated biases can directly reduce access to supports through a variety of mechanisms that include police withholding information about one's rights as a victim, among other direct and indirect barriers to social and health services. Unaddressed financial, mental, and physical health consequences of victimization contribute to poorer health outcomes later in life. This paper seeks to bring together the extant research on help-seeking, discrimination in criminal legal system functioning, and barriers to victim services by synthesizing these discrete threads into a theoretically and empirically informed conceptual model that captures the range of factors that shape Black Americans' decision to report their victimization to the police and subsequent help-seeking. Qualitative and quantitative data from a purposive sample of 91 Black victims of community violence is used to ground the developing model. The conceptual model can help lay the foundation for research that seeks to remedy the marked mismatch between the prevalence of violent victimization and help-seeking among Black Americans. It is widely assumed crime and related concerns, including neighborhood incivilities and fear of crime, are barriers to physical activity (PA). Past studies reveal mixed evidence. Studies of impacts for crime-protective factors are less common but have similarly mixed results. This paper evaluates a comprehensive transdisciplinary conceptual framework of cross-sectional associations between crime-related perceptions and reported minutes/week of recreational walking inside and outside one's home neighborhood. Safe and Fit Environments Study (SAFE) recruited and surveyed 2302 participants from adolescents to older adults from four U.S. metropolitan areas. A zero-inflated model estimated two components of each outcome: whether the respondent walked, and minutes/week walked. Correlates of recreational walking were location-specific, differing based on walking location. Fear of crime, risk evaluation, victimization, and incivilities were not consistently associated with walking for recreation inside one's neighborhood. People with crime concerns about their own neighborhoods, however, more commonly walked for recreation outside their neighborhoods. Protective crime-related perceptions that seldom have been studied in relation to PA, such as street efficacy (i.e., the perceived ability to avoid and manage danger), were strongly associated with recreational walking in both locations, indicating the additional heuristic value of the SAFE conceptual framework. This study examines changes in gun violence at the census tract level in Philadelphia, PA before and after the onset of the COVID-19 pandemic. Piecewise generalized linear mixed effects models are used to test the relative impacts of social-structural and demographic factors, police activity, the presence of and proximity to drug markets, and physical incivilities on shooting changes between 2017 and June, 2021. Model results revealed that neighborhood structural characteristics like concentrated disadvantage and racial makeup, as well as proximity to drug markets and police activity were associated with higher shooting rates. Neighborhood drug market activity and police activity significantly predicted changes in shooting rates over time after the onset of COVID-19. This work demonstrates the importance of understanding whether there are unique factors that impact the susceptibility to exogenous shocks like the COVID-19 pandemic. The increasing risk of being in a neighborhood with an active drug market during the pandemic suggests efforts related to disrupting drug organizations, or otherwise curbing violence stemming from drug markets, may go a long way towards quelling citywide increases in gun violence. This study examines whether victims of violent street crimes who are known to the police as past offenders, when compared with victims with no arrest history, have different outcomes related to receipt of victim and health-related services, while taking into consideration whether or not police responded to the victimization incident. The sample is comprised of 103 men and women between the age of 18 and 40 living in one Mid-Atlantic city who were victims of street violence within the year before study recruitment. Logistic regression was used to assess the impact of police response to the victimization incident on receipt of victim services, and receipt of victim services on engagement with counseling and mental health services. The results show that prior arrests were not associated with receipt of services. However, having police officers respond to the victimization was associated with higher odds of receiving victim services, and in turn, victim services were associated with receiving mental health treatment. Police response appears to set victims on a path to accessing services. Although the number of arrests was not associated with service receipt, a small percentage of victims who did not receive services stated they were reluctant to cooperate with the police, thus limiting their opportunity for victim services. Because most victims who did not access victim services did not know that they existed, policies that promote more knowledge of and initial engagement with victim services could improve access to needed health and mental health services. In this article, we explore some aspects of Fourier analysis on fuzzy sets. In particular, we give a complete characterization of the Fourier coefficients of fuzzy subsets of the unit circle. The main result hinges on a theorem by the Italian mathematician Aldo Ghizzetti. This work was part of a broader project jointly with Professor Vittorio Cafagna, who, sadly, passed away unexpectedly in 2007. The number of jurisdictions implementing focused deterrence strategies targeted at gangs continues unabated. Although recent research suggests positive impacts of the strategy on reductions in gun violence, little is known about the particular mechanisms operating behind the strategy. This article provides a descriptive analysis of the law enforcement activities or levers undertaken after enforcement operations in Philadelphia as a part of the focused deterrence strategy. The article quantifies the execution of levers related to arrest, case processing, and probation sanctioning during enforcement activities after shootings. The results show that Philadelphia achieved success in implementing the enforcement levers as intended, and there was little evidence that arrest practices were overly aggressive. The authors suggest that future evaluations seek to carefully document the wide array of levers used in concert with an assessment of community understanding of and reactions to the strategy as well as an examination of reactions of group members targeted. In line with emerging work on the role of fatherhood in prisoner reentry, this study directs attention to the financial obligations that connect fathers to their families in reentry. Specifically, the study provides a descriptive picture of soon-to-be-released male state prisoners with child support obligations using a multi-state, longitudinal dataset, and examines whether characteristics of incarcerated men with child support orders and associated debt are significantly different from incarcerated males without child support orders. Whether males attached to comprehensive reentry programs received more services related to their debt obligation after prison release compared with non-program participants is also analyzed. Findings show that although prisoners identify having substantial needs vis-à-vis child support obligations, few reported receiving assistance related to these needs upon release. However, reentry program participants received significantly more child support–related services than non-program participants. The purpose of this article is to situate knowledge on the pushes and pulls of disengagement from gangs within the inventory of gang intervention programs. Drawing on developmental and life course criminological theory and three major, multi-site studies on gang disengagement, we examined the self-reported pushes and pulls that led gang members to reduce their gang embeddedness and move toward disengagement more effectively. We found that (1) multiple rather than single factors for leaving gangs were most common, (2) push factors exceeded pull factors in prevalence and frequency, and (3) motivations for disengagement may be age-graded, and appear to increase in complexity with age. We complemented the multi-site findings by examining prominent programmatic efforts to reduce or prevent gang involvement that have the most explicit theory of change related to the pushes and pulls in gang disengagement: focused deterrence, hospital-based interventions, jobs programs, and relationship-based interventions that have street-outreach, therapeutic, family, and fatherhood areas of focus. Programs that address individuals disillusioned with their gang and simultaneously offer sustained opportunities to develop and engage in prosocial networks are most likely to enjoy success. Recently released prisoners in the United States are increasingly facing the burden of financial debt associated with correctional supervision, yet little research has pursued how—theoretically or empirically—the burden of debt might affect life after prison. To address this gap, we employ life course and strain perspectives and path analysis to examine the impact of child support debt on employment and recidivism, using longitudinal data from an evaluation of a prisoner reentry program known as the Serious and Violent Offender Reentry Initiative. Results indicate that having more debt has no effect on recidivism; however, more debt was significantly associated with a decrease in later legitimate employment. Implications for community reintegration and justice processing are discussed within the framework of past and emerging work on legal financial obligations, employment, and desistance from crime after prison. Blastic plasmacytoid dendritic cell neoplasm ( BPDCN) is a rare haematological malignancy derived from the precursors of plamacytoid dendritic cells, with an aggressive clinical course and high frequency of cutaneous and bone marrow involvement. Neoplastic cells express CD4, CD43 (also termed SPN), CD45 RA and CD56 (also termed NCAM1), as well as the plasmacytoid dendritic cell-associated antigens CD123 (also termed IL3 RA), BDCA-2 (also termed CD303, CLEC4E) TCL1 and CTLA1 (also termed GZMB). The median survival is only a few months as the tumour exhibits a progressive course despite initial response to chemotherapy. The best modality of treatment remains to be defined. Generally, patients receive acute leukaemia-like induction, according to acute myeloid leukaemia ( AML)-type or acute lymphoid leukaemia ( ALL)-type regimens. The frequent neuromeningeal involvement indicates systematic pre-emptive intrathecal chemotherapy in addition to intensive chemotherapy. Allogeneic haematopoietic stem cell transplantation ( HSCT), particularly when performed in first remission, may improve the survival. Preliminary data suggest a potential role for immunomodulatory agents and novel targeted drugs. Herein epidemiology, clinical manifestations, diagnosis and management of BPDCN will be presented. In detail, this review focuses on the therapeutic aspects of BPDCN, proposing a treatment algorithm for the management of the disease, including induction chemotherapy, allogeneic HSCT and intrathecal prophylaxis at different steps of treatment, according to compliance, biological and clinical characteristics of patients. The association of formaldehyde dehydrogenase from Pseudomonas putida (FalDH) with two different magnetic nanoparticles led to distinct enzyme responses depending upon the chemical modification process applied. The magnetite nanoparticles coated with aminopropyltriethoxysilane (MagNP-APTS) exhibited a lower activity and stability, while magnetite coated with a silica shell and APTS (MagNP@SiO2APTS) afforded excellent results, improving the stability and performance of FalDH in relation to the free enzyme. Such differences were ascribed to unfavorable conformational changes in the MagNP-APTS–FalDH system, as indicated by the kinetics and Raman spectroscopy data, not discarding the possible interference from the exposed Fe(ii)/(iii) ions. The MagNP@SiO2APTS–FalDH catalyst could be successfully recycled by using an external magnet, keeping its highest performance close to 100%, during the first four cycles, and decaying slightly up to 70% after the 10th cycle. Background: Today, the ability of deoxyribonucleic acid (DNA) evidence to place persons at crime scenes with near certainty is broadly accepted by criminal investigators, courts, policy makers, and the public. However, the public safety benefits of investments in DNA databases are largely unknown and research attempting to quantify these benefits is only gradually emerging. Given the inherent difficulty in randomly assigning offenders to treatment and comparison groups for the purpose of inferring specific deterrence and probative effects (PREs) of DNA databases, this study developed an alternate strategy for extracting these effects from transactional data. Research Design: Reoffending patterns of a large cohort of offenders released from the Florida Department of Corrections custody between 1996 and 2004 were analyzed across a range of criminal offense categories. First, an identification strategy using multiple clock models was developed that linked the two simultaneous effects of DNA databases to different clocks measuring the same events. Then, a semiparametric approach was developed for estimating the models. Results: The estimation models yielded mixed results. Small deterrent effects—2-3% reductions in recidivism risk attributable to deterrence—were found only for robbery and burglary. However, strong PREs—20-30% increase in recidivism risk attributable to PREs—were uncovered for most offense categories. A binuclear cobalt complex, [Co2BPP], derived from the ( R, R)-Trost-bis-ProPhenol (BPP) ligand, has been investigated as a catalyst for CO2 uptake and its electrochemical reduction. The complex exhibits labile acetate ligands that can be readily exchanged with CO2, leading to pronounced changes in its vibrational spectra, with νC=O peaks observed at 1700 and 1596 cm-1. The electrochemical reduction of the CoIII centers at -0.5 V was significantly enhanced in the presence of CO2, evidencing a chemical reaction coupled to an electrode process. Instead of yielding CO or HCOOH as observed in most artificial systems, formaldehyde was identified as the major product below -1.0 V, showing that CO2 can undergo four-electron reduction by the [Co2BPP] electrocatalyst. BACKGROUND This study integrated criminological and public health perspectives to examine the influence of bullying victimization and the school environment on physical activity (PA). METHODS We used a weighted sample of 7786 US middle school students surveyed as part of the Health Behavior in School-Aged Children study to conduct a multilevel analysis of bullying victimization as a predictor of self-reported PA and number of days attending physical education ( PE) classes. Hierarchical models assessed the contribution of school climate and anti-crime programs. RESULTS Results indicated that bullying victimization was associated with fewer days in PE and lower odds of reporting at least 60 minutes of PA more than once a week. Although one of the school crime prevention policies examined was associated with more days in PE, the school-level factors did not account for a large portion of the variance in the 2-level models for either days in PE or PA. CONCLUSIONS The results suggest that reduced levels of PA can be added to the growing list of health issues associated with bullying victimization and provide support for the importance of bullying prevention efforts in conjunction with health promotion programs targeted to middle school students. A combined interpolation – three‐dimensional level set method (3D LSM)‐based segmentation process is presented. The performance in terms of accuracy of the 3D LSM in the segmentation of throat regions from highly anisotropic magnetic resonance imaging (MRI) volumes, with and without an interpolation step is evaluated. Qualitative and quantitative results from real MRI data suggest that performing interpolation, to reconstruct isotropic MRI volumes, prior to 3D LSM improves the accuracy of the segmentation results, compared to interpolation post 3D LSM and no interpolation at all. Zygomycosis is an invasive infection that can occur particularly in patients with haematological malignancy. The causative fungi are members of the order Mucorales, and individual species within this group require a high level of laboratory skill to be identified. Zygomycosis can present as rhinocerebral, pulmonary, or disseminated disease, with a rapid clinical course. The optimal management of these cases requires early diagnosis, aggressive antifungal therapy and, when possible, surgical debridement. Founded on clinical experience, but without the benefit of comparative studies, liposomal amphotericin B has become the therapeutic agent of choice. Posaconazole is an orally administered triazole with a demonstrated in vitro and in vivo activity against most Zygomycetes that is comparable to that of amphotericin B. Studies on salvage therapy with posaconazole have yielded promising results, and successful case reports are also available. As an adjuvant approach, iron chelation with deferasirox has shown promising results, although clinical experience is still limited. [ABSTRACT FROM AUTHOR]

Within the realm of active living in urban neighborhoods in the United States, only a few studies have addressed the factors that promote or inhibit active living among residents in public housing. This paper examines the environmental and interpersonal factors associated with active living and health in public housing. We specifically examine the environmental predictors of fear of crime, and whether fear is related to moderate physical activity, and in turn, health and obesity. The analysis drew upon data from a sample of 328 African-American residents living in two severely distressed public housing developments in 2007. Structural equation modeling was used to calculate a path model of direct and indirect effects. Perceptions of violence were positively associated with fear, and both fear and physical activity predicted health. However, neither fear nor physical activity was associated with obesity. Background: Although a number of studies have tested ecologic models that postulate relationships among social networks, the built environment, and active living, few neighborhood-based studies have considered the role of crime and violence. This study investigates the degree to which individual-level demographic characteristics and neighborhood-level physical and social characteristics are associated with increased fear of crime. Methods: Data were analyzed in 2007 from a 2005 survey of 901 randomly selected individuals living in 55 neighborhoods in Washington DC. Multilevel ordered logit regression was used to examine associations between individual-level and neighborhood-level characteristics and how often fear of crime prevents a respondent from walking outdoors. Results: Age and female gender were associated with an increase in fear; the percentage of a resident’s life spent in the same neighborhood was associated with a decrease in fear. Results of cross-level interactions showed that at the neighborhood level, women were more fearful than men in neighborhoods without violence, but that the difference in fear between men and women shrinks as neighborhood violence increases. Collective efficacy was found to increase fear among black respondents and had no effect on fear among nonblack respondents. Conclusions: If the study of neighborhoods and active living is to progress and contribute to both etiologic understanding and policy formation, it is essential that theoretical and empirical models consider the impact of violence and fear on walking. Efforts to increase active living in urban neighborhoods that do not account for the impact of crime and fear may fall short of their intended outcomes. [Copyright &y& Elsevier]

Objective: This study examined intermediate outcomes of a faith-based prisoner reentry program by assessing how client spirituality related to client- and program-level characteristics, investigating differences between completers and terminators, and examining how religious preference, religiosity/spirituality, religious salience, and incarceration's impact on spirituality influenced program completion, satisfaction, and perceived progress. Method: The study employed independent t tests, chi-square tests, and regression models to examine the influence of predictor variables. Results: Religious preference was positively associated with progress and satisfaction. In addition, individuals reporting decreases in spirituality during prison were 34 times as likely to drop out of the program compared to those who had more positive in-prison spiritual changes. Salience, general religiosity/spirituality, and spiritual experiences did not predict outcomes. Implications of the findings are discussed. [ABSTRACT FROM AUTHOR]

Abstract: Prisoner reentry constitutes one of the central criminal justice challenges confronting U.S. society. Coinciding with this emerging social problem has been increased policymaker interest in faith-based programs to improve outcomes for vulnerable populations, including released prisoners. Critical questions about the nature and effects of faith-based reentry programs remain largely unaddressed, however: (1) What is a “faith-based” program? (2) How does or could such a program reduce recidivism and improve other behavioral outcomes among released offenders? (3) What is the evidence concerning the impacts of faith-based reentry programs? (4) What are critical implementation issues that may affect the operations and impacts of such programs? This article examines each of these questions and identifies critical conceptual, theoretical, and research gaps in the literature. It highlights that the term “faith-based” is used inconsistently, that the precise causal relationship, if any, between various measures of faith and crime remains in question, and that few rigorous evaluations of faith-based reentry programs exist. Despite a small but growing literature on gang disengagement and desistance, little is known about how social networks and changes in networks correspond to self-reported changes in street gang membership over time. The current study describes the personal or "ego" network composition of 228 street gang members in two east coast cities in the United States. The study highlights changes in personal network composition associated with changes in gang membership over two waves of survey data, describing notable differences between those who reported leaving their gang and fully disengaging from their gang associates, and those who reported leaving but still participate and hang out with their gang friends. Results show some positive changes (i.e., reductions) in criminal behavior and many changes toward an increase in prosocial relationships for those who fully disengaged from their street gang, versus limited changes in both criminal behavior and network composition over time for those who reported leaving but remained engaged with their gang. The findings suggest that gang intervention programs that increase access to or support building prosocial relationships may assist the gang disengagement process and ultimately buoy desistance from crime. Research has found that drug markets tend to cluster in space, potentially because of the profit that can be made when customers are drawn to areas with multiple suppliers. But few studies have examined how these clusters of drug markets—which have been termed "agglomeration economies"—may be related to accidental overdose deaths, and in particular, the spatial distribution of mortality from overdose. Focusing on a large neighborhood in Philadelphia, Pennsylvania, known for its open-air drug markets, this study examines whether deaths from accidental drug overdose are clustered around street corners controlled by drug trafficking gangs. This study incorporates theoretically-informed social and physical environmental characteristics of street corner units into the models predicting overdose deaths. Given a number of environmental changes relevant to drug use locations was taking place in the focal neighborhood during the analysis period, the authors first employ a novel concentration metric—the Rare Event Concentration Coefficient—to assess clustering of overdose deaths annually between 2015 and 2019. The results of these models reveal that overdose deaths became less clustered over time and that the density was considerably lower after 2017. Hence, the predictive models in this study are focused on the two-year period between 2018 and 2019. Results from spatial econometric regression models find strong support for the association between corner drug markets and accidental overdose deaths. In addition, a number of sociostructural factors, such as concentrated disadvantage, and physical environmental factors, particularly blighted housing, are associated with a higher rate of overdose deaths. Implications from this study highlight the need for efforts that strategically coordinate law enforcement, social service provision and reductions in housing blight targeted to particular geographies. Copper nitrite reductase mimetics were synthesized using three new tridentate ligands sharing the same N,N,N motif of coordination. The ligands were based on L -proline modifications, attaching a pyridine and a triazole to the pyrrolidine ring, and differ by a pendant group (R = phenyl, n-butyl and n-propan-1-ol). All complexes coordinate nitrite, as evidenced by cyclic voltammetry, UV-Vis, FTIR and electron paramagnetic resonance (EPR) spectroscopies. The coordination mode of nitrite was assigned by FTIR and EPR as κ2O chelate mode. Upon acidification, EPR experiments indicated a shift from chelate to monodentate κO mode, and 15N NMR experiments of a Zn2+ analogue, suggested that the related Cu(II) nitrous acid complex may be reasonably stable in solution, but in equilibrium with free HONO under non catalytic conditions. Reduction of nitrite to NO was performed both chemically and electrocatalytically, observing the highest catalytic activities for the complex with n-propan-1-ol as pendant group. These results support the hypothesis that a hydrogen bond moiety in the secondary coordination sphere may aid the protonation step. [ABSTRACT FROM AUTHOR]

Urease is an enzyme containing a dinuclear nickel active center responsible for the hydrolysis of urea into carbon dioxide and ammonia. Interestingly, inorganic models of urease are unable to mimic its mechanism despite their similarities to the enzyme active site. The reason behind the discrepancy in urea decomposition mechanisms between inorganic models and urease is still unknown. To evaluate this factor, we synthesized two bis-nickel complexes, [Ni2L(OAc)] (1) and [Ni2L(Cl)(Et3N)2] (2), based on the Trost bis-Pro-Phenol ligand (L) and encompassing different ligand labilities with coordination geometries similar to the active site of jack bean urease. Both mimetic complexes produced ammonia from urea, (1) and (2), were ten- and four-fold slower than urease, respectively. The presence and importance of several reaction intermediates were evaluated both experimentally and theoretically, indicating the aquo intermediate as a key intermediate, coordinating urea in an outer-sphere manner. Both complexes produced isocyanate, revealing an activated water molecule acting as a base. In addition, the reaction with different substrates indicated the biomimetic complexes were able to hydrolyze isocyanate. Thus, our results indicate that the formation of an outer-sphere complex in the urease analogues might be the reason urease performs a different mechanism. [ABSTRACT FROM AUTHOR]

Abstract: Purpose: The purpose was to examine how fear of crime, crime victimization, and perceived level of community incivilities are related to physical activity participation and outdoor recreation among Latino adolescents. Method: The study utilized a mixed methods approach that included 25 qualitative interviews and 390 school-based surveys collected from youth across three schools in Little Village, Chicago, Illinois. Results: Results showed that Latino adolescents who expressed greater fear of crime also engaged in less physical activity and outdoor recreation. There was no association between crime victimization and physical activity and outdoor recreation. Those who perceived greater levels of community incivilities also engaged in less outdoor recreation, but perception of incivilities had no significant association with physical activity levels. Interview data revealed most of the children believed crime was a serious problem in their neighborhood and it impacted their ability to be physically active and play outside. Conclusions: Fear of crime was related to lower physical activity and outdoor recreation. It is imperative that communities provide safe environments for children to be active. Increasing police and adult presence in parks and school grounds is recommended. Moreover, efforts must be made to reduce the gang problems in Latino communities. [Copyright &y& Elsevier]

Focused deterrence is a gang violence reduction strategy that relies on a unique mix of strong enforcement messages from law enforcement and judicial officials coupled with the promise of additional services. At the heart of the intervention is a coordinated effort to communicate the costs and consequences of gun violence to identified gang members during face-to-face meetings and additional community messaging. In Philadelphia, focused deterrence was implemented between 2013 and 2016, and although an impact evaluation showed a significant decrease in shootings in targeted areas relative to matched comparison neighborhoods, the effect on targeted gangs was not universal, with some exhibiting no change or an increase in gun-related activity. Here, we employ data on group-level social media usage and content to examine the correlations with gun violence. We find that several factors, including the nature of social media activity by the gang (e.g., extent of activity and who is engaging), are associated with increases in the average rate of gang-attributable shootings during the evaluation period, while content-specific variables (e.g., direct threats towards rivals and law enforcement) were not associated with increases in shootings. PlsX is the first enzyme in the pathway that produces phosphatidic acid in Gram-positive bacteria. It makes acylphosphate from acyl-acyl carrier protein (acyl-ACP) and is also involved in coordinating phospholipid and fatty acid biosyntheses. PlsX is a peripheral membrane enzyme in Bacillus subtilis, but how it associates with the membrane remains largely unknown. In the present study, using fluorescence microscopy, liposome sedimentation, differential scanning calorimetry, and acyltransferase assays, we determined that PlsX binds directly to lipid bilayers and identified its membrane anchoring moiety, consisting of a hydrophobic loop located at the tip of two amphipathic dimerization helices. To establish the role of the membrane association of PlsX in acylphosphate synthesis and in the flux through the phosphatidic acid pathway, we then created mutations and gene fusions that prevent PlsX's interaction with the membrane. Interestingly, phospholipid synthesis was severely hampered in cells in which PlsX was detached from the membrane, and results from metabolic labeling indicated that these cells accumulated free fatty acids. Because the same mutations did not affect PlsX transacylase activity, we conclude that membrane association is required for the proper delivery of PlsX's product to PlsY, the next enzyme in the phosphatidic acid pathway. We conclude that PlsX plays a dual role in phospholipid synthesis, acting both as a catalyst and as a chaperone protein that mediates substrate channeling into the pathway. [ABSTRACT FROM AUTHOR]

Lipase B from Candida antarctica (CAL-B) is largely employed as a biocatalyst for hydrolysis, esterification, and transesterification reactions. CAL-B is a good model enzyme to study factors affecting the enzymatic structure, activity and/or stability after an immobilization process. In this study, we analyzed the immobilization of CAL-B enzyme on different magnetic nanoparticles, synthesized by the coprecipitation method inside inverse micelles made of zwitterionic surfactants, with distinct carbon chain length: 4 (ImS4), 10 (ImS10) and 18 (ImS18) carbons. Magnetic nanoparticles ImS4 and ImS10 were shown to cross-link to CAL-B enzyme via a Michael-type addition, whereas particles with ImS18 were bond via pyridine formation after glutaraldehyde cross-coupling. Interestingly, the Michael-type cross-linking generated less stable immobilized CAL-B, revealing the influence of a cross-linking mode on the resulting biocatalyst behavior. Curiously, a direct correlation between nanoparticle agglomerate sizes and CAL-B enzyme reuse stability was observed. Moreover, free CAL-B enzyme was not able to catalyze transesterification due to the high methanol concentration; however, the immobilized CAL-B enzyme reached yields from 79.7 to 90% at the same conditions. In addition, the transesterification of lipids isolated from oleaginous yeasts achieved 89% yield, which confirmed the potential of immobilized CAL-B enzyme in microbial production of biodiesel. Aims To carry out statistical validation of a newly developed magnetic resonance imaging (MRI) auto-contouring software tool for gross tumour volume (GTV) delineation in head and neck tumours to assist in radiotherapy planning. Materials and methods Axial MRI baseline scans were obtained for 10 oropharyngeal and laryngeal cancer patients. GTV was present on 102 axial slices and auto-contoured using the modified fuzzy c-means clustering integrated with the level set method (FCLSM). Peer-reviewed (C-gold) manual contours were used as the reference standard to validate auto-contoured GTVs (C-auto) and mean manual contours (C-manual) from two expert clinicians (C1 and C2). Multiple geometric metrics, including the Dice similarity coefficient (DSC), were used for quantitative validation. A DSC ≥ 0.7 was deemed acceptable. Inter- and intra-variabilities among the manual contours were also validated. The two-dimensional contours were then reconstructed in three dimensions for GTV volume calculation, comparison and three-dimensional visualisation. Results The mean DSC between C-gold and C-auto was 0.79. The mean DSC between C-gold and C-manual was 0.79 and that between C1 and C2 was 0.80. The average time for GTV auto-contouring per patient was 8 min (range 6–13 min; mean 45 s per axial slice) compared with 15 min (range 6–23 min; mean 88 s per axial slice) for C1. The average volume concordance between C-gold and C-auto volumes was 86.51% compared with 74.16% between C-gold and C-manual. The average volume concordance between C1 and C2 volumes was 86.82%. Conclusions This newly designed MRI-based auto-contouring software tool shows initial acceptable results in GTV delineation of oropharyngeal and laryngeal tumours using FCLSM. Purpose: The purpose of this study was to examine the association between adolescent and parental perceptions of neighborhood safety and adolescents' physical activity in multiple locations and to investigate the moderating effect of sex within this association.Method: This cross-sectional study was conducted with 928 adolescents aged 12 to 16 years old and 1 of their parents. Adolescents and parents reported their perceptions of neighborhood safety (traffic safety, pedestrian safety, crime safety, and stranger danger safety). Adolescents reported how often they were physically active in multiple locations (physical activity in the neighborhood, in parks, and for active transport). Mixed-effects linear regression models were used to investigate these associations while controlling for demographics and the Walkability Index.Results: Parent-perceived crime safety was positively associated with adolescents' physical activity in parks (B = .094, p = .024). Parent-perceived traffic safety was positively associated with adolescents' reported physical activity in the neighborhood (B = .186, p = .014). Adolescents' physical activity for active transport was positively associated with parent-perceived traffic safety (B = .179, p = .001), stranger danger safety (B = .110, p = .013), and crime safety (B = .077, p = .035). There were 2 interactions by sex on the relation between adolescent traffic safety perception and parent pedestrian safety perception in the neighborhood and adolescents' physical activity in parks (i.e., statistically significant only for boys).Conclusions: Parents' perceptions of traffic, stranger danger, and crime safety were all related to adolescents' active transportation. Although Italian mental health (MH) services are community based, user and relative participation in service evaluation lagged behind until lately. We here review three recent studies involving stakeholder participation in service evaluation: two were quantitative studies, one on 204 users in an MH service in Pistoia (Central Italy) and the other on 2259 relatives, conducted with the National Union of Associations for Mental Health. The third (supported by The Centro per il Controllo delle Malattie, the ministerial Center for Disease Control) was a qualitative study in seven MH services, involving users, relatives, and professionals together, which collected interviews from 136 users, 119 relatives, and 79 professionals. In the quantitative studies, positive evaluations outnumbered negative ones. The qualitative study explored negative aspects in greater depth. Common findings were insufficient information, underinvolvement of users-relatives in planning, no choice of clinician, psychiatrist domination, and limited helpfulness of interventions. Coronary heart disease clusters within families, but there may be several reasons for this phenomenon to occur. A possible way to elucidate this is to study biological relatives of affected individuals. The aim of our study was thus to compare a number of clinical, metabolic, clotting and immunologic factors between offspring with paternal history of premature myocardial infarction and controls and to propose a model which could safely allow to identify the high risk subgroup among them. Sixty-nine offspring of both sexes mean age 18.1 years old (cases) and thirty-two frequency matched relative to age and gender controls were studied. Cases compared to controls had significantly increased diastolic blood pressure levels (74.0±9.9 vs. 67.4±8.3 mmHg, P=0.002), leptin plasma levels (11.8±10.8 vs. 6.8±3 ng/ml, P=0.046) and fibrinogen, plasminogen, fibrin degradation products and plasminogen activator inhibitor-1 plasma levels (306.6±52.5 vs. 280.6±28.9 mg%, P=0.03, 97.4±23.5 vs. 83.6±15 mg%, P=0.0007, 292.0±148.5 vs. 219.2±69.4 ng/ml, P=0.036, 14.7±5.3 vs. 8.7±3.1 I.U./ml, P=0.0001, respectively), while cases had significantly decreased HDL-cholesterol serum levels (45.9±12.5 vs. 50.5±8.8 mg%, P=0.03) and protein S plasma levels (89.9±17.5 vs. 101.3±13.7%, P=0.001). Study Objective: To compare total blood product requirements in liver transplantation (LT) assisted by thromboelastography (TEG) or conventional coagulation tests (CCTs).Design: Retrospective observational study.Setting: A tertiary care referral center for LT.Patients: Adult patients undergoing LT from deceased donor.Intervention: Hemostasis was monitored by TEG or CCTs and corresponding transfusion algorithms were adopted.Measurements: Number and types of blood products (red blood cells, RBC; fresh-frozen plasma, FFP; platelets, PLT) transfused from the beginning of surgery until the admission to the intensive care unit.Methods: We compared data retrospectively collected in 226 LTs, grouped according to the type of hemostasis monitoring (90 with TEG and 136 with CCTs, respectively). Confounding variables affecting transfusion needs (recipient age, sex, previous hepatocellular carcinoma surgery, Model for End Stage Liver Disease - MELD, baseline hemoglobin, fibrinogen, creatinine, veno-venous by pass, and trans-jugular intrahepatic portosystemic shunt) were managed by propensity score match (PSM).Main Results: The preliminary analysis showed that patients in the TEG group received fewer total blood products (RBC + FFP + PLT; p = 0.001, FFP (p = 0.001), and RBC (p = 0.001). After PSM, 89 CCT patients were selected and matched to the 90 TEG patients. CCT and TEG matched patients received similar amount of total blood products. In a subgroup of 39 patients in the top MELD quartile (MELD ≥25), the TEG use resulted in lower consumption of FFP units and total blood products. Nevertheless, due to the low number of patients, any meaningful conclusion could be achieved in this subgroup.Conclusions: In our experience, TEG-guided transfusion in LT does not reduce the intraoperative blood product consumption. Further studies are warranted to assess an advantage for TEG in either the entire LT population or the high-MELD subgroup of patients. [ABSTRACT FROM AUTHOR]
