Piemonte is widely acknowledged as a remarkable Pliocene palaeontological spot, due to its potential for the observation and study of marine and terrestrial fossils in abundant outcrops. Our observations on about 80 palaeobotanical localities, representing both terrestrial and marine sedimentary deposits, show that some of them could easily provide more significant materials than those available today. About 30 plant-bearing Pliocene sites were selected for morphological analyses on plant fossils, mainly leaf remains. The main features of each assemblage, including the systematic placement of about 400 plant samples within 52 taxa, are indexed and described in this paper, by combining analysis of palaeontological collections and field observations. The results obtained so far encourage further studies in specific areas of palaeobotanical interest, whose results could easily provide major palaeoenvironmental insights. Finally, we suggest considering more carefully the Piemonte palaeobotanical sites in the frame of geoconservation strategies and we point out the relevance of ex-situ protection of endangered plant fossil samples (collections) in order to assure their availability for future studies. We evaluate the maxcut of the two loops sunrise amplitude with three different masses by direct use of the loop momenta in the Minkoskean (as opposed to the usual Euclidean) continuous dimension regularisation, obtaining in that way six related but different functions expressed in the form of one dimensional finite integrals. We then consider the 4th order homogeneous equation valid for the maxcut, and show that for arbitrary dimension d the six functions do satisfy the equation separately. Landslides represent a serious worldwide hazard, especially in Italy, where exposure to hydrogeological risk is very high; for this reason, a landslide quantitative risk assessment (QRA) is crucial for risk management and for planning mitigation measures. In this study, we present and describe a novel methodological approach of QRA for slow-moving landslides, aiming at national replicability. This procedure has been applied at the basin scale in the Arno River basin (9100 km2, Central Italy), where most landslides are slow-moving. QRA is based on the application of the equation risk = hazard (H) × vulnerability (V) × exposure (E) and on the use of open data with uniform characteristics at the national scale. The study area was divided into a grid with a 1 km2 cell size, and for each cell, the parameters necessary for the risk assessment were calculated. The obtained results show that the total risk of the study area amounts to approximately 7 billion €. The proposed methodology presents several novelties in the risk assessment for the regional/national scale of the analysis, mainly concerning the identification of the datasets and the development of new methodologies that could be applicable over such large areas. Wild and cultivated olives in the Western Cape of South Africa are direct or indirect hosts to a high diversity of Braconidae and Chalcidoidea wasps. Olive-associated Braconidae are known to parasitise the larvae of Bactrocera oleae(Rossi), and probably also Bactrocera biguttula (Bezzi). The lifestyle of olive-associated Chalcidoidea is not fully understood, and may include phytophagous, parasitoid and hyperparasitoid species. Some chalcids could represent olive seed wasps (OSW), a generic term that designates the seed feeders responsible for losses in commercial olive production. In order to obtain direct DNA-based evidence for the lifestyle of four putative OSW – Eupelmus spermophilus Silvestri, Eurytoma oleae Silvestri, Eurytoma varicolor Silvestri, Sycophila aethiopica (Silvestri); and the parasitoid Neochrysocharis formosus (Westwood) – we developed a multiplex PCR assay comprising species-specific primers for each species. The assay was used to survey larvae collected from the seeds of wild and cultivated olives. Eupelmus spermophilus was the most abundant species in wild and cultivated olives, and may represent a threat to the local olive industry. Eurytoma oleae was found only within the seeds of wild olives, and may also represent an OSW. We found direct evidence supporting the potential agricultural relevance of S. aethiopica and E. varicolor as parasitoids of E. spermophilus. Background: BRCA1:c.5017_5019del (p.His1673del) is a founder variant relatively frequent in Northern Italy. Despite previous suggestion of pathogenicity, variant classification in public databases is still conflicting, needing additional evidence. Methods: Maximum likelihood penetrance of breast/ovarian and other cancer types was estimated using full pedigree data from 53 informative Italian families. The effect of the variant on BRCA1‐ABRAXAS1 interaction was assessed using a GFP‐fragment reassembly‐based PPI assay. Results were combined with additional data from multiple sources to classify the variant according to ACMG/AMP classification rules specified for BRCA1/2. Results: Variant‐carriers displayed increased risk for ovarian cancer (HR = 33.0, 95% CI = 7.0–155.0; cumulative risk at age 70 = 27.6%, 95% CI = 12.6–40.0%) but not for breast cancer (HR = 0.7, 95% CI = 0.2–2.2). An increased risk of uterine cancer (HR = 8.0, 95% CI = 1.03–61.6) emerged, warranting further evaluation. Likelihood‐ratio in favor of pathogenicity was 98898642.82 under assumption of standard BRCA1 breast and ovarian penetrance, and 104240832.84 after excluding breast cancer diagnoses (based on penetrance results). Functional analysis demonstrated that the variant abrogates the BRCA1‐ABRAXAS1 binding, supporting the PS3 code assignment within the ACMG/AMP rule‐based model. Collectively, these findings allowed to classify the variant as pathogenic. Conclusion: Pathogenicity of BRCA1:c.5017_5019del(p.His1673del) has been confirmed; however, breast cancer risk in Italian families is not increased, unlike in families from other countries and in carriers of most BRCA1 pathogenic variants. To evaluate whether EBV-DNA can be used as a diagnostic and follow-up parameter for nasopharyngeal tumors in a non-endemic population. The study was carried out in a university hospital. A retrospective study was conducted on 40 paraffin samples of histological preparations. EB-DNA was detected by real-time PCR technique. A prospective study was also conducted on a group of 30 patients who underwent nasopharyngeal biopsy for suspected nasopharyngeal carcinoma (NPC) by comparing EBV-DNA concentrations between the histological specimen and the serum. Quantification of genomic copies of EBV-DNA in serum and detection of anti-EBV antibodies was performed. In both groups the presence of high viral load of EBV-DNA was found in nonkeratinizing squamous cell carcinomas, in three cases of lymphepitomyoma and in 4 out of 6 cases of non-differentiated non-carcinoma lymph node metastases. squamous keratinizing cells. In all cases of NPC, an antibody pattern typical of reactivations (IgGVCA+, IgG-EA+, IgG-EBNA+) and IgA-EA-D, frequently positive in cases of NPC, has been highlighted. A good correlation between the high EBV-DNA charges and the histological diagnosis was highlighted. Wild and cultivated olives harbor and share a diversity of insects, some of which are considered agricultural pests, such as the olive fruit fly. The assemblage of olive-associated parasitoids and seed wasps is rich and specialized in sub-Saharan Africa, with native species possibly coevolving with their hosts. Although historical entomological surveys reported on the diversity of olive wasp species in the Western Cape Province of South Africa, no comprehensive study has been performed in the region in the molecular era. In this study, a dual approach combining morphological and DNA-based methods was used for the identification of adult specimens reared from olive fruits. Four species of Braconidae and six species of Chalcidoidea were identified, and DNA barcoding methodologies were used to investigate conspecificity among individuals, based on randomly selected representative specimens. Morphological identifications were congruent with DNA data, as NJ and ML trees correctly placed the sequences for each species either at the genus or species level, depending on the available taxa coverage, and genetic distances strongly supported conspecificity. No clear evidence of cryptic diversity was found. Overall seed infestation and parasitism rates were higher in wild olives compared to cultivated olives, and highest for Eupelmus spermophilus and Utetes africanus. Laboratory trials were performed to evaluate the action of spinosad and mineral oil on eggs of Grapholita funebrana Treitschke, the key pest in plum orchards. Fruits of cultivars Angeleno, President and Stanley were used in the trials. The first set of tests was carried out by introducing two mated females of G. funebrana into a cage together with fruits of a single cultivar. The second set of trials tested the three cultivars simultaneously. In all trials, one third of the fruits of each cultivar was treated with mineral oil, another third with spinosad and the final third was left untreated. Treatments were carried out before introducing mated females into the cages. The number of fruits with eggs, the number of eggs laid on each fruit and the number of hatched eggs were recorded. The number of eggs per fruit recorded in all trials was the same, indicating that plum moth females, after choosing the fruits for oviposition on the basis of the cultivar or the applied product, tend to use all suitable fruits in the same way. Oviposition in terms of number of fruits was significantly lower in mineral oil and spinosad treatments in all trials, compared to the control. Differences between the two products were found in trials carried out with Angeleno alone and with the three cultivars together. The percentage of egg hatching on the fruits was always significantly lower with spinosad treatment compared to the mineral-oil treatment and the control. In the trials carried out using the three cultivars, the number of infested fruits was significantly higher for Angeleno and no differences in the hatching percentages were found among the cultivars. The ovicidal and antiovideponent activities of commercial products containing mineral oils or spinosad could represent an interesting tool to reduce G. funebrana damage, both in organic and conventional plum orchards. The red gum lerp psyllid, Glycaspis brimblecombei Moore (Hemiptera, Aphalaridae), is an Australian native sapsucking insect pest of eucalypts that has been first reported for the West Palaearctic Region in 2008 and, in 2010, it has been found also in Italy. Subsequently its primary parasitoid, Psyllaephagus bliteus Riek (Hymenoptera: Encyrtidae), was also detected within the main European and North African infested areas, where no release of the parasitoid was ever performed. This study, carried out in 30 Eucalyptus camaldulensis plantations located along the coast, on the hills and the mountains in Mediterranean climatic areas of Sicily (Italy), aimed to determine the influence of environmental parameters on the incidence of both, the psyllid infestation level and the parasitization activity. P. bliteus reached highest average levels in summer samplings and resulted widespread in Sicily at all detected altitudes without statistically significant differences. P. bliteus parasitization is the main factor lowering G. brimblecombei infestation; this result, together with the accidental and contemporaneous arrival of the host and its parasitoid, could explain the absence of high damage level on eucalypts in Sicily. The most significant metric factors positively influencing G. brimblecombei infestation are the percentage of daily hours above 80% of relative humidity and the average maximum temperature, obviously related to other, but less significant climatic factors. The altitude affects both infestation and parasitization, but single sites could explain significantly more, so that the local conditions where the samplings were carried out have to be considered as the main responsibles for the variability in the obtained results. In any sampled Sicilian site, from sea level to 540 m a.s.l., both the psyllid and its parasitoids show a good adaptation to climatic conditions, confirming that areas fitting for E. camaldulensis growth fit also for P. bliteus activity, and proving that Mediterranean climate, differently from some inland areas of California, does not obstacle its parasitic activity. This paper includes a critical review of the existing literature on the use of satellite SAR imagery for subsidence analysis. Land subsidence, related to multiple natural and human-induced processes, is observed globally in an increasing number of areas. Potentially leading to severe impacts on economics and the environment, subsidence has attracted growing scientific attention and, over the last decades, new tools and methods have been developed for accurately measuring the spatial and temporal evolution of surface deformations associated with subsidence phenomena. The collection of the existing scientific literature on the satellite InSAR for subsidence analysis was conducted in January 2022 exploiting the WoS's freely accessible web search engine. An extensive database of 1059 scientific contributions was compiled, covering the period 1997–2021. The content of each record in the literature database has been critically examined to collect and store information regarding the study area location, microwave band adopted, satellite used, processing approach, subsidence cause, application type, field evidence and strategies to validate and compare InSAR data. Analysis of temporal distribution revealed a substantial growth in scientific production and an increasing interest of geoscientists, with a mean value of 21 articles per year from 1997 to 2014, rising to about 100 articles per year between 2015 and 2021. All continents include at least a study area, with Asia and Europe having the largest number of case studies, with 586 and 281 analyses in their territory, respectively, and revealing a clear geographical bias in subsidence study locations. Graphical visualizations and syntheses of current applications are presented. The large availability of different acquisition bands, the increasing imaging capabilities, refinement of processing approaches, and growing expertise in data interpretation allowed InSAR data to be used at different scales of analysis, for different purposes and subsidence types, in a wide range of physiographic settings. This review highlights that satellite InSAR has moved from being a niche topic to an operative tool with a major role in subsidence studies. Despite more than 25 years of progress and advancements, technical and operational challenges remain to be faced. The identification of founder mutations in cancer predisposing genes is important to improve risk assessment in geographically defined populations, since it may provide specific targets resulting in cost-effective genetic testing. Here, we report the characterization of the BRCA1 c.190T>C (p.Cys64Arg) mutation, mapped to the RING-finger domain coding region, that we detected in 43 hereditary breast/ovarian cancer (HBOC) families, for the large part originating from the province of Bergamo (Northern Italy). Haplotype analysis was performed in 21 families, and led to the identification of a shared haplotype extending over three BRCA1-associated marker loci (0.4 cM). Using the DMLE+2.2 software program and regional population demographic data, we were able to estimate the age of the mutation to vary between 3,100 and 3,350 years old. Functional characterization of the mutation was carried out at both transcript and protein level. Reverse transcriptase-PCR analysis on lymphoblastoid cells revealed expression of full length mRNA from the mutant allele. A green fluorescent protein (GFP)-fragment reassembly assay showed that the p.Cys64Arg substitution prevents the binding of the BRCA1 protein to the interacting protein BARD1, in a similar way as proven deleterious mutations in the RING-domain. Overall, 55 of 83 (66%) female mutation carriers had a diagnosis of breast and/or ovarian cancer. Our observations indicate that the BRCA1 c.190T>C is a pathogenic founder mutation present in the Italian population. Further analyses will evaluate whether screening for this mutation can be suggested as an effective strategy for the rapid identification of at-risk individuals in the Bergamo area. The susceptibility of olive cultivars to the olive fruit fly, Bactrocera oleae ( Rossi) ( Diptera: Tephritidae), has seldom been studied. This article examines factors associated with olive fruit fly infestation of 16 commonly planted Sicilian olive cultivars. Total infestation data were simultaneously correlated with categorical and quantitative factors using ordinal logistic regression. When all factors were included in the analysis, year, sampling date, cultivar, and fruit color were highly significant, but the quantitative factors fruit volume, fruit elongation, and fruit hardness were not. When the analysis was repeated excluding cultivar, all quantitative factors were significant, and elongation and volume were highly significant. Spherical, large, and hard fruit seemed to be preferred by B. oleae over fruit that are elongate, small, and soft. Therefore, fruit color, elongation, volume, and hardness provide useful information regarding the susceptibility of cultivars. In both organic and conventional olive cultivation, information about olive cultivar susceptibility to olive fruit fly will help orchard managers to produce quality oil and table olives while reducing treatments for olive fruit fly control. The parasitoid complex of the western conifer seed bug, Leptoglossus occidentalis, was studied in British Columbia and California on Pinus monticola and P. contorta var. latifolia. Three egg-parasitoid species were identified: Gryon pennsylvanicum (Hymenoptera: Platygastridae), Ooencyrtus johnsoni (Hymenoptera: Encyrtidae) and Anastatus pearsalli (Hymenoptera: Eupelmidae). Leptoglossus occidentalis represents a new host record for O. johnsoni, and California is a new distribution record for G. pennsylvanicum. Gryon pennsylvanicum was the dominant species during the entire oviposition period of the bug, reaching peak parasitization levels in July (25%). Ooencyrtus johnsoni and A. pearsalli emerged only from eggs collected on Pinus monticola. The parasitization level by G. pennsylvanicum recorded on the two different Pinaceae hosts did not show any statistically significant difference. No tachinid eggs or exit holes by tachinid larvae were observed in any adult L. occidentalis collected in either British Columbia or California. The data collected encouraged the introduction of G. pennsylvanicum in Italy where L. occidentalis has become a serious pest. Abstract: Spread of the exotic parasitoid Closterocerus chamaeleon (Girault) and its parasitism on the Eucalyptus gall wasp Ophelimus maskelli (Ashmead) (Hymenoptera, Eulophidae) were studied in Sicily after C. chamaeleon introduction in May 2006. Parasitoid spread was evaluated by sampling sites at increasing distances from the five release sites. C. chamaeleon quickly established and spread; within 5months, it caused 62% parasitism at release sites and 38% parasitism at sites 2km from release sites. One year after its introduction (spring 2007), C. chamaeleon was detected more than 50km distant from release sites. By winter 2007–08, the parasitoid was recovered throughout Sicily and in many surrounding islets, with parasitism rates always >65% and usually at 100%. The dispersal rate was 0.15km/month in the 2months after release. It increased to 0.6km/month in the next 3months, reaching 7.5km/month 5–7months after release and 20km/month in the following 11months, when the entire island was colonized. The pattern of spread followed the spread latency model of biological invasions; the spread latency period was very short because of the parasitoid’s biological characteristics and a favourable environment. Parasitism trends differed between suburban and afforested sites, showing a longer spread latency period in the afforested sites due to the greater extent of potentially colonisable area. The parasitoid’s use of continuous and long-distance dispersal mechanisms enabled it to rapidly colonize even very distant regions and enhances its effectiveness as a biological control agent. A systematic survey of soil CO2 concentrations was carried out on the flanks of Somma–Vesuvius volcano in order to constrain possible pathways responsible of carbon dioxide diffuse degassing taking place during the present state of quiescence. Measurements were performed at 1162 sites in late winter–spring 2000, highlighting that soil CO2 concentrations range from 50 to 10500 ppmV. A statistical analysis was developed in order to define the threshold value of anomaly and separate the biogenic CO2 component, produced by soil respiration, from the inorganic component of deep provenance. A computer routine was also elaborated to interpret the grid of CO2 anomalous concentration values and define the actual location, orientation and length of degassing structures. The results obtained by this procedure reveal a main control of the regional stress field on the patterns of gas migration. The identified degassing lineaments are typically oriented along the Apenninic (NW–SE) and anti-Apenninic (NE–SW) trends, which are known to govern the past geological and structural evolution of the Campanian Plain and present seismicity and deformation pattern of Mount Vesuvius. A main degassing area was recognized on the eastern and southern flanks of the volcano, which likely relates to the geometry of the underlying carbonate basement, reaching its top (500 m depth) in this sector of the volcano. The analytical performance of the new Aspergillus Galactomannan Ag VirCLIA® mono-test (Vircell S.L.) was compared to the Platelia™ Aspergillus Ag ELISA assay (Bio-Rad). Prospective serum and bronchoalveolar lavage (BAL) samples from patients at risk of invasive aspergillosis (IA) were tested using both the Aspergillus Galactomannan Ag VirCLIA® mono-test and the Platelia™ Aspergillus Ag ELISA assay. Concordance, sensitivity, specificity, and positive and negative predictive values were calculated using the manufacturer-recommended cutoff levels. Receiver operating characteristic (ROC) analysis and the Youden index were performed to determine the optimal cutoff. A total of 187 serum samples and 73 BAL samples were analyzed with both assays. The concordance between the Aspergillus Galactomannan Ag VirCLIA® mono-test and the Platelia™ Aspergillus Ag ELISA assay was 87.8%, with a Cohen's kappa of 0.75. The sensitivity and specificity of the Aspergillus Galactomannan Ag VirCLIA® mono-test were 78.6% and 96.2%, respectively, with positive and negative predictive values of 94.8% and 83.3%. The ROC curve for the Aspergillus Galactomannan Ag VirCLIA® mono-test demonstrated an area under the curve (AUC) of 0.87, and the Youden index at the manufacturer's established cutoff was 0.73. This new Aspergillus Galactomannan Ag VirCLIA® mono-test exhibited adequate analytical and clinical performance, showing good correlation with the Platelia™ Aspergillus Ag ELISA assay. The single-sample, semi-automated test is user-friendly, allowing small laboratories to perform the test on demand without the need for batch evaluations, providing a useful solution for timely diagnostic support for clinicians. Simple Summary: Parasitoid wasps in the family Braconidae are generally highly specialized and can be used as agents for biological control of arthropod pests. Psyttalia concolor, Psyttalia humilis and Psyttalia lounsburyi parasitize the larvae of the olive fruit fly (Bactrocera oleae), the most damaging pest of cultivated olives in the world. Psyttalia concolor is native to the Mediterranean, and P. humilis and P. lounsburyi are native to sub-Saharan Africa. Despite their potential for agricultural pest control, these species have been poorly characterized at the genetic level. We sequenced the mitochondrial genome of the three species and compared its organization with other Braconidae. Psyttalia had a unique gene rearrangement involving the positions of transfer RNA genes. We also present a phylogenetic reconstruction of the Braconidae and confirm the phylogenetic placement of Psyttalia in the subfamily Opiinae. The family Braconidae consists mostly of specialized parasitoids, some of which hold potential in biocontrol of agricultural pests. Psyttalia concolor, Psyttalia humilis and Psyttalia lounsburyi are parasitoids associated with Bactrocera oleae, a major pest of cultivated olives. The native range of Psyttalia concolor is the Mediterranean, and P. humilis and P. lounsburyi are native to sub-Saharan Africa. This study reports the mitochondrial genomes of the three species, thus laying the foundation for mitogenomic analyses in the genus Psyttalia. Comparative mitogenomics within Braconidae showed a novel gene arrangement in Psyttalia in involving translocation and inversion of transfer RNA genes. The placement of Psyttalia in the subfamily Opiinae was well-supported, and the divergence between Psyttalia and its closest relative (Diachasmimorpha longicaudata) was at ~55 MYA [95% highest posterior density (HPD): 34–83 MYA]. Psyttalia lounsburyi occupied the most basal position among the three Psyttalia, having diverged from the other two species ~11 MYA (95% HPD: 6–17 MYA). Psyttalia concolor and P. humilis were recovered as sister species diverged at ~2 MYA (95% HPD: 1.1–3.6 MYA). This phylogeny combining new sequences and a set of 31 other cyclostomes and non-cyclostomes highlights the importance of a comprehensive taxonomic coverage of Braconidae mitogenomes to overcome the lack of robustness in the placement of several subfamilies. Genetic testing for BRCA1 and BRCA2 genes has led to the identification of many unique variants of uncertain significance (VUS). Multifactorial likelihood models that predict the odds ratio for VUS in favor or against cancer causality, have been developed, but their use is conditioned by the amount of necessary data, which are difficult to obtain if a variant is rare. As an alternative, variants mapping to the coding regions can be examined using in vitro functional assays. BRCA1 and BRCA2 proteins promote genome protection by interacting with different proteins. In this study, we assessed the functional effect of two sets of variants in BRCA genes by exploiting the green fluorescent protein (GFP)-reassembly in vitro assay, which was set-up to test the BRCA1/BARD1, BRCA1/UbcH5a, and BRCA2/DSS1 interactions. Based on the findings observed for the validation panels of previously classified variants, BRCA1/UbcH5a and BRCA2/DSS1 binding assays showed 100% sensitivity and specificity in identifying pathogenic and non-pathogenic variants. While the actual efficiency of these assays in assessing the clinical significance of BRCA VUS has to be verified using larger validation panels, our results suggest that the GFP-reassembly assay is a robust method to identify variants affecting normal protein functioning and contributes to the classification of VUS. The Mediterranean fruit fly Ceratitis capitata is a major pest of fruit orchards worldwide. In order to develop control methods based on natural products, particularly required in organic farming, a laboratory study was carried out to evaluate the response of adults to leaf extracts from four plant species: Ruta graveolens, Eriobotrya japonica, Rubus ulmifolius, and Ficus carica. Air-dried leaf powders were subjected to an ethanolic cold extraction. Chemical composition was assessed by GC/ MS analysis. Extracts were tested in laboratory bioassays to assess the electroantennographic (EAG) response, attraction/ repellence toward ovipositing females, and induced mortality toward adults. Among the extracts tested, only R. graveolens elicited significant differences compared to the control in all the bioassays. This extract, whose main volatile compounds were 2-nonanone and 2-undecanone (45% of the total), recorded the greatest EAG response, and stimulated a positive oviposition response compared to the control. Ruta graveolens extract elicited a significant insecticidal activity, confirmed to be dose-dependent. LD50 and LD90 concentrations were 3.64 and 6.01% (v/v), respectively. To our knowledge, this is the first study reporting a single plant extract, from R. graveolens, acting as attractant and oviposition stimulant, and at the same time having insecticidal properties toward C. capitata adults. A review is reported in the present paper on invasive alien insects introduced in Sicily on Eucalyptus trees, together with unpublished results from recent surveys. As to the latter ones, observations were conducted especially on Thaumastocoris peregrinus (Carpintero & Dellapé) (Hemiptera, Thaumastocoridae), the most recently introduced species. Overall, eight alien insect pests have been accidentally introduced in the island on Eucalyptus, belonging to the orders Hemiptera (Aphalaridae, 2 spp.; Thaumastocoridae, 1 sp.), Coleoptera (Cerambycidae, 2 spp.; Curculionidae, 1 sp.) and Hymenoptera (Eulophidae, 2 spp.). Two encyrtid parasitoids, Avetianella longoi Siscaro and Psyllaephagus bliteus Riek, obtained from Phoracantha spp. and Glycaspis brimblecombei Moore respectively, and Closterocerus chamaeleon (Girault) (Hymenoptera, Eulophidae) attacking Ophelimus maskelli (Ashmead), are also reported. Simple Summary: Anchonocranus oleae is a southern African weevil that feeds on the seeds of the African Wild Olive, a close relative of the European cultivated olive tree. The species is known to occur in the Western Cape of South Africa, the main region of olive production in Southern Africa. We generated reference DNA barcodes and the complete mitogenome of A. oleae as part of our ongoing genetic cataloguing of insects associated with wild and cultivated olives in South Africa. The phylogenetic position of A. oleae in the family Curculionidae was inferred to be in the Curculioninae, Conoderinae, Cossoninae, Molytinae, and Scolytinae (CCCMS) clade but could not be precisely determined due to the paucity of genetic data for adequate taxonomic context, highlighting the need for further coverage of related tribes and genera. Nevertheless, the data generated in this study contribute to the enrichment of baseline information on olive-associated insects, in general, and on the genus Anchonocranus, in particular. Anchonocranus oleae Marshall (Coleoptera: Curculionidae) is a seed-feeding weevil native to southern Africa; its larvae are known to develop in the fruits of the African Wild Olive and, more rarely, cultivated olives. The species has been mainly found in the Western Cape province of South Africa, but it has remained in relative obscurity because it does not seem to represent a current threat to commercial olive production. As part of an ongoing effort to produce baseline genetic data for olive-associated entomofauna in South Africa, we generated reference DNA barcodes for A. oleae collected from wild and cultivated olives and sequenced its mitogenome for assessment of the phylogenetic position of the species in the family Curculionidae. The mitochondrial phylogeny estimate indicated that A. oleae shares a common ancestor with Elaidobius (tribe Derelomini), but a definite and close relationship to this tribe and the precise tribal placement of A. oleae in the subfamily Curculioninae could not be inferred due to the lack of representative mitogenomes of other relevant curculionine tribes and genera. Several unclassified variants (UVs) have been identified in splicing regions of disease-associated genes and their characterization as pathogenic mutations or benign polymorphisms is crucial for the understanding of their role in disease development. In this study, 24 UVs located at BRCA1 and BRCA2 splice sites were characterized by transcripts analysis. These results were used to evaluate the ability of nine bioinformatics programs in predicting genetic variants causing aberrant splicing (spliceogenic variants) and the nature of aberrant transcripts. Eleven variants in BRCA1 and 8 in BRCA2, including 8 not previously characterized at transcript level, were ascertained to affect mRNA splicing. Of these, 16 led to the synthesis of aberrant transcripts containing premature termination codons (PTCs), 2 to the up-regulation of naturally occurring alternative transcripts containing PTCs, and one to an in-frame deletion within the region coding for the DNA binding domain of BRCA2, causing the loss of the ability to bind the partner protein DSS1 and ssDNA. For each computational program, we evaluated the rate of non-informative analyses, i.e. those that did not recognize the natural splice sites in the wild-type sequence, and the rate of false positive predictions, i.e., variants incorrectly classified as spliceogenic, as a measure of their specificity, under conditions setting sensitivity of predictions to 100%. The programs that performed better were Human Splicing Finder and Automated Splice Site Analyses, both exhibiting 100% informativeness and specificity. For 10 mutations the activation of cryptic splice sites was observed, but we were unable to derive simple criteria to select, among the different cryptic sites predicted by the bioinformatics analyses, those actually used. Consistent with previous reports, our study provides evidences that in silico tools can be used for selecting splice site variants for in vitro analyses. Leptoglossus occidentalis (Heteroptera, Coreidae), a Nearctic species, was accidentally introduced into Northern Italy in the late 1990s, from where it has spread throughout Europe. The bug causes abortion of immature cones of Pinus pinea L., with economic impact on the pine-nut industry. As part of a pest control research program, the egg parasitoid Gryon pennsylvanicum Ashmead (Hymenoptera, Platygastridae) was collected from British Columbia, Canada, and legally introduced to a quarantine climatic chamber in Florence, Italy. The egg parasitoid will be tested against native non-target heteropterans, an environmental impact assessment will be conducted, and a mass rearing method will be developed if appropriate. The objectives of this study were the specific taxonomic confirmation of the main olive seed wasp (OSW) attacking commercial olives in the Western Cape, to investigate monitoring methods and seasonal occurrence of OSW, to determine the potential economic damage of infestations, and to ascertain the geographic distribution of OSW in the regions where olives are cultivated in the Western Cape. Morphological and molecular methods were used to identify all the species obtained from cultivated olives at two trial sites near Stellenbosch and Agter-Paarl. Eupelmus spermophilus Silvestri (Hymenoptera: Chalcidoidea) was by far the most frequent and widespread olive seed wasp. Monitoring with yellow sticky traps over three seasons indicated OSW presence in orchards when olives were small enough for OSW to oviposit in, but traps were not reliable indicators of OSW infestation levels. Prematurely dropped olives, collected from February until harvest over three seasons, and olives picked at harvest were examined for OSW infestation. Total yield for each data tree was recorded at harvest, and yield loss due to OSW infestation calculated. Economically significant yield losses occurred sporadically. Fruits collected from both cultivated and wild olive trees indicated that OSW occurs in most olive-growing regions of the Western Cape. OSW does not pose a significant threat to the local olive industry as a whole and cannot be considered a key pest of olives, but during some seasons it can affect production yields where wild olive trees are located near commercial olive groves. Accurate identification of parasitoids is crucial for biological control of the invasive brown marmorated stink bug, Halyomrpha halys (Stål). A recent work by Talamas et al. (2017) revised the Palearctic fauna of Trissolcus Ashmead, egg-parasitoids of stink bugs, and treated numerous species as junior synonyms of T. semistriatus (Nees von Esenbeck). In the present paper, we provide a detailed taxonomic history and treatment of T. semistriatus and the species treated as its synonyms by Talamas et al. (2017) based on examination of primary types, molecular analyses and mating experiments. Trissolcus semistriatus, T. belenus (Walker), T. colemani (Crawford), and T. manteroi (Kieffer) are here recognized as valid and a key to species is provided. Abstract Bactrocera biguttula is an African olive fruit fly that does not attack cultivated olives but rather develops in the fruits of wild species of Olea and Noronhia. The complete mitochondrial genome of an individual specimen was characterized in comparison to other Bactrocera. The phylogenetic relationships of B. biguttula with other Dacini were investigated, with special focus on B. oleae , an agricultural pest known to attack cultivated and wild olives. The sequence had a total length of 15,829 bp, and included the typical features of insect mitogenomes, similarly to the other Bactrocera analysed. Start codons included ATG, ATC, ATT, and TCG (in COI). The majority of stop codons (TAA) were fully encoded, whereas in some cases only TA or T were present. The complete sequence was biased towards A + T, with a positive AT-skew and a negative GC-skew. The predicted cloverleaf structure of tRNA Ser1 showed absence of the DHU arm, a common feature in insects and other Metazoans. Phylogenetic reconstruction showed that B. biguttula and B. oleae are sister species, having diverged from a common ancestor < 10 Myr ago. This result warrants future genomic comparisons between these two closely related species for investigating the specific adaptations to the different hosts. [ABSTRACT FROM AUTHOR]

The pre-release risk assessment, a recommended practice in biological control programmes, was carried out before introducing Closterocerus chamaeleon (Girault), a eulophid parasitoid of the eucalypt gall maker Ophelimus maskelli (Ashmead) (Hymenoptera, Eulophidae), in Sicily, Italy. We evaluated its host specificity in laboratory small arena no-choice tests on six non-target hosts, using O. maskelli as a control species. The non-target species fit with at least one of the following criteria: a) common geographical origin; b) ecological or behavioural affinities with the target host; c) concealed habit of the preimaginal stages; d) taxonomic affinity; e) taxonomic affinity with hosts of congeneric parasitoids of the candidate agent; and f) presence in Italy/Sicily. Closterocerus chamaeleon performed an oviposition sequence of 10 recognisable behaviours, always leading to parasitization when exposed to O. maskelli. The same behaviour sequence was carried out on the gall maker Leptocybe invasa Fisher et La Salle, the only non-target species living on Eucalyptus, but just until Step 6, i.e. until ovipositor extraction and insertion, and never until egg-laying. All of the other non-target hosts were ignored. Furthermore, the adult parasitoid longevity was assessed in three rearing conditions: 1) no food/no parasitisation permitted; 2) food/no parasitisation permitted; and 3) food provided after parasitisation. Adult wasp longevity was significantly higher in treatment 2 (21.1 days ± 4.1 SE) than in treatment 1 (3.6 days ± 0.5 SE) or 3 (3.8 days ± 0.6 SE). Leptoglossus occidentalis Heidemann ( Heteroptera: Coreidae) is a North American conifer seed pest that was accidentally introduced to Europe. In the Mediterranean area, it threatens the production of Pinus pinea Linnaeus seeds. The egg-parasitoid Gryon pennsylvanicum ( Ashmead) ( Hymenoptera: Platygastridae), the main natural enemy in the native range of L. occidentalis, was imported from British Columbia to Italy. Pre-release risk assessments were made under quarantine conditions by no-choice tests conducted with naïve and experienced G. pennsylvanicum offering single eggs of target and non-target species for varying exposure times (1, 4, 48 h). G. pennsylvanicum successfully parasitized from 75% to 100% of the target host eggs. Only one female specimen of the egg-parasitoid emerged from a non-target egg ( Gonocerus juniperi Herrich- Schaeffer, Heteroptera: Coreidae). Two dead female specimens were found, one inside an egg of Coreus marginatus ( Linnaeus) ( Heteroptera: Coreidae) and one in an egg of Camptopus lateralis ( Germar) ( Heteroptera: Alydidae). All three cases occurred at the longest oviposition exposure time. In the last few years, several studies have focused on the interpretation of unclassified variants (UVs) of BRCA1 and BRCA2 genes. Analysis of UVs through a unique approach is not sufficient to understand their role in the development of tumors. Thus, it is clear that assembling results from different sources (genetic and epidemiological data, histopathological features, and in vitro and in silico analyses) represents a powerful way to classify such variants. Building reliable integrated models for UV classification requires the joining of many working groups to collaborative consortia, allowing data exchange and improvements of methods. This will lead to improvement in the predictivity of gene testing in BRCA1 and BRCA2 and, consequently, to an increase in the number of families that can be correctly classified as linked or unlinked to these genes, allowing more accurate genetic counseling and clinical management. Summary: A key issue in oral HPV infection is whether it can be associated with a genital HPV infection, or whether it can be considered as an independent event. This analysis evaluated the frequency and type-concordance of oral HPV infection in women with cervical HPV infection by means of: (i) a cross-sectional study on a sample (n =98) of Italian women; and (ii) a literature-based metanalysis, including the experimental study the subject of this Paper and nine other published studies (n =1017), which also examined the influence of oral sampling procedure (oral brushing vs oral rinse) and HIV status on oral HPV detection. The prevalence of oral HPV infection in the Italian study was 14.3% (95% CI: 7.4–21.2); the prevalence of type-concordance was 21.4% (95% CI: 0.0–43.6) and it was only marginally significant (P =0.05). The prevalence of oral HPV infection in the metanalysis was estimated as 18.1% (95% CI: 10.3–25.9); the prevalence of type-concordance was 27.0% (95% CI: 12.3–41.7), and it was statistically significant (P =0.002). The metanalysis also showed that the oral sampling procedure was not a determinant of HPV detection; however, HIV status increased the likelihood of oral HPV infection (HIV-positive vs negative: 27.2%; 95% CI: 22.1–32.2 vs 15.5%; 95% CI: 6.9–24.2) and type-concordance (HIV-positive vs negative: 46.8%; 95% CI: 34.7–58.9 vs 15.6%; 95% CI: 0.8–30.4). Oral HPV infection and type-concordance in women with cervical HPV infection are more prevalent than could be expected by chance; this finding is consistent with the notion of a degree of dependence of the oral site on the cervical site. We determined the prevalence of human papillomavirus (HPV) cervical infection and HPV genotypes among 115 women immigrating to Sicily (Italy), with regard to abnormal cytology and socio-behavioral characteristics in a cross-sectional, observational study. Information was collected with the help of cultural mediators/translators. HPV-DNA was assayed by the INNOLiPA HPV assay and a nested PCR/sequencing method. Sixty (52.2%) women came from sub-Saharan Africa and 55 (47.8%) from Eastern Europe. HPV infection was found in 55 (47.8%) women. The most frequent types were the oncogenic types HPV-16 (7.8%), HPV-18 and 51 (6.0% each), HPV-52 (5.2%), 31, 53, and 68 (4.3% each). Twenty-seven (23.5%) women had cytological abnormalities associated with HPV infection (p=0.04). Being single (OR = 2.98; 95%CI: 1.30-6.84) and parity (OR = 0.29; 95%CI: 0.12-0.65) were consistent predictors of HPV infection. Only 21 (18.2%) women returned to collect the results of their Pap and HPV tests. The high prevalence of HPV infection and oncogenic types among immigrant women make them a priority group for cervical cancer screening. Linguistically and culturally appropriate prevention efforts are needed to sensitize immigrant women regarding HPV-related issues and to conduct vaccine strategies for cervical cancer prevention. Abstract: The solitary parasitoid Eretmocerus mundus Mercet (Hymenoptera: Aphelinidae) is one of the key biological control agents of the whitefly Bemisia argentifolii Bellows & Perring, also known as B. tabaci (Gennadius) B-biotype (Homoptera: Aleyrodidae). We present new information on its biology, and particularly about larval elimination during the first instar in cases of superparasitism. For the first time, physical elimination of supernumerary larvae was observed, both outside and inside the host. These findings are documented with confocal microscopy images and video recordings. We observed more than 350 B. argentifolii nymphs, parasitized with one, two or more than two larvae. Physical attack took place only when parasitoid eggs were laid in contact with each other. In this case the percentage larval mortality outside the host was significantly higher (45.7%) than in cases of single larvae (12.0%) or larvae hatched under the same host but not in contact with each other (23.3%). These new findings are important in view of a possible horizontal transfer of a parthenogenesis inducing Wolbachia bacterium from a thelytokous to an arrhenotokous strain, with the aim of augmenting the biological control of B. argentifolii. Abstract: Human papillomavirus (HPV) infection is the commonest sexually transmitted infection, and high-risk HPV types are associated with cervical carcinogenesis. This study investigated: the HPV type-specific prevalence in 970 women with an abnormal cytological diagnosis; and the association of HPV infection and cervical disease in a subset of 626 women with a histological diagnosis. HPV-DNA was researched by nested PCR/sequencing and the INNOLiPA HPV Genotyping assay. The data were analysed by the chi-square test (p ≤0.05 significant). Overall, the HPV prevalence was 37.7%; high-risk genotypes were found in 88.5% of women and multiple-type infections in 30.9% of the HPV-positive women. The commonest types were HPV-16 (8.2%), HPV-6 (5.0%), HPV-51 (4.2%) and HPV-53 (3.6%). Among the women with histological diagnosis, HPV was evident in 19.9% of those without lesions, 65.8% of those with low-grade lesions and 100% (p =0.002) of those with high-grade lesions. The commonest types were HPV-16 (in 14.7% low-grade and 42.8% high-grade lesions), HPV-31 (4.7% and 14.3%, respectively) and HPV-33 (2.0% and 14.3%, respectively). Two high-grade lesions contained exclusively one uncommon type, namely, HPV-83 and -85. This study confirmed the high prevalence of HPV infection and high-risk genotypes among women with cervical abnormalities living in Italy. Introduction: Adequate brushing of oral mucosa is important for accurate human papillomavirus (HPV) detection in potentially malignant (oral leukoplakia [OL], oral lichen planus [OLP]) and malignant (oral squamous cell carcinoma [OSCC]) lesions. Since various factors may limit the adequacy of oral brushing and, consequently, the accuracy of HPV detection, modified sampling procedures should be evaluated for their effect on HPV frequency and/or types detected. Aim: To compare the HPV frequency in samples obtained by brushing the lesion site with the frequency in samples obtained by brushing an apparently normal adjacent site. The correlation between HPV frequency and keratinization of the site affected by the lesion, as well as sociodemographic variables (age, sex, smoking and drinking habits), was also examined. Methods: HPV DNA was detected in brushing samples from 50 patients with OL, 49 with OLP, and 17 with OSCC. Polymerase chain reaction (PCR) amplification was performed by MY09/MY11 and GP05+/GP06+ primers; the HPV type was identified by DNA sequencing and a reverse hybridization (line probe) assay. Data were analyzed by the Z test, the Fisher's exact test, the chi-square test, odds ratio (OR), and a logistic regression model. Results: HPV DNA was detected in 22% of samples from lesion sites and in 16% of samples from adjacent sites (p = 0.22) in patients with OL, in 24.5% and 22.4% of samples from lesion and adjacent sites, respectively, in patients with OLP (p = 0.40), and in 35.3% and 41.2% of samples from lesion and adjacent sites, respectively, in patients with OSCC (p = 0.36). Lesions adjacent to HPV-positive normal sites had an increased rate of HPV detection (OR = 30; 95% CI 9.57, 94.1). HPV-18 was the most frequent genotype, followed by HPV-6, -16, -33, and -53. HPV prevalence was reduced in lesions at keratinized sites (14.5%) compared with non-keratinized sites (34.4%; p = 0.007; OR = 0.32; 95% CI 0.13, 0.81). Discussion: In patients with OL, OLP, or OSCC, a high prevalence of HPV infection was shown in apparently normal sites adjacent to lesion sites infected by HPV. The lower HPV frequency in lesions at keratinized sites suggests that HPV detection by lesion brushing is affected by keratinization. The keratinized epithelium may be less susceptible to HPV infection or, alternatively, the highly proliferative activity in non-keratinized sites may predispose to HPV infection. Conclusion: Results from this study indicate that taking samples from normal sites adjacent to oral lesions may be of value in HPV detection, particularly when the lesions are located at keratinized sites. This sampling procedure may allow more accurate diagnosis of HPV infection compared with sampling only the lesion site, and may also represent a reliable method to investigate the biological characteristics of HPV infection and related oral carcinogenesis. The olive fruit fly, Bactrocera oleae, is considered the main olive pest worldwide, and has been the target of biological control programmes through the release of the braconid parasitoid Psyttalia concolor. Laboratory tests were performed to evaluate the influence of distance from the host on parasitisation, placing larvae of the substitute host Ceratitis capitata at seven distances (0, 0.5, 1, 1.5, 2, 2.5, 3 mm) and four different time periods (7, 15, 30, 60 min). Moreover, field collected olives of Ogliarola Barese cultivar infested by B. oleae were exposed to P. concolor females to confirm its ability to parasitise B. oleae in small olives. Psyttalia concolor oviposition was inhibited at 2.5 and 3 mm due to the ovipositor length of the parasitoid females (2.7 mm). Hosts were easily parasitised at distances between 0 and 1.5 mm. The thin fruit pulp (up to 3.5 mm) of field collected olives allowed the parasitisation to occur also in mature fruits. At the best combination distance/time (0 mm, 30 min), tests performed with different larvae/parasitoid female ratio showed an increasing emergence of P. concolor (from 20% to 57%) with larvae/parasitoid ratio increasing from 0.11 to 0.74. The results of the present study might optimise the mass rearing of P. concolor, through a proper setting of its parameters, such as the host/parasitoid ratio, exposure distances, and interaction time.
