Background: The long-term outcomes (≥ 2 years of follow-up) of volar locked plate (VLP) fixation versus closed reduction and casting (CRC) for the treatment of displaced distal radial fractures (DRFs) remain unclear. This study aimed to conduct a meta-analysis comparing the long-term clinical outcomes of VLP and CRC in elderly patients (aged ≥ 60 years). Methods: A comprehensive search of PubMed, Web of Science, and Cochrane Library was performed to identify studies comparing the long-term outcomes of VLP and CRC for DRFs. Only randomized controlled trials (RCTs) with a mean follow-up duration of at least 2 years and participants aged 60 years or older were included. The risk of bias in the included studies was assessed. The primary outcome measure was the Patient-Rated Wrist Evaluation (PRWE). Secondary outcomes included the Disabilities of the Arm, Shoulder, and Hand (DASH) questionnaire, EuroQol 5 Dimension 5-level score (EQ-5D-5 L), grip strength, and incidence of reoperation. Results: Four RCTs, comprising 423 patients, were included. The meta-analysis revealed that VLP fixation was associated with significantly improved outcomes compared to CRC in terms of PRWE score (pooled mean difference: -6.21; 95% CI: -10.28 to -2.15; p = 0.003), DASH score (pooled mean difference: -8.18; 95% CI: -13.35 to -3.01; p = 0.002), and grip strength (pooled mean difference: -6.63; 95% CI: 0.25 to 13.01; p = 0.04). There were no significant differences in EQ-5D-5 L score (95% CI: -0.08 to 0.05; p = 0.74) or incidence of reoperation (RR = 0.56; 95% CI: 0.22 to 1.42; p = 0.22). ABSTRACT Nickel‐aluminum bronze (NAB) is extensively utilized across various industries, particularly marine applications due to its exceptional corrosion resistance. The mechanical properties of NAB are highly dynamic, and influenced by factors such as manufacturing processes, operating environments, and microstructure. This variability introduces complexities and challenges in addressing fatigue, the most common failure mode in metals. This review offers a thorough examination of the fatigue properties of NAB in both their conventionally fabricated and additively manufactured (AM) forms. This review explains the mechanisms that modulate fatigue in NAB by analyzing the existing literature and identifying critical factors such as microstructure, defects, and processing parameters. The goal is to improve the comprehension and dependability of NAB by contrasting the fatigue resistance of cast and AM NAB. The ultimate objective of this comprehensive examination is to predict and reduce component failures, thereby extending the service life and performance of NAB components in demanding environments. PLACK syndrome (OMIM 616295) is a rare genodermatosis associated with peeling skin, leukonychia, acral punctate keratosis, cheilitis, and knuckle pads and is caused by loss‐of‐function mutations in the CAST gene, which encodes calpastatin, a calcium‐dependent protease. This case report highlights a case of PLACK syndrome presenting with the unique findings of striate hyperkeratosis on the palms as well as life‐threatening cardiomyopathy. Background: Various treatment approaches for individuals with ankle sprains can reduce treatment costs and enhance recovery. This study aimed to compare the efficacy of spring ankle braces with splints and casts in treating ankle sprains. Materials and methods: This cross-sectional study involved 60 patients diagnosed with ankle sprains at the orthopedic clinic of Imam Khomeini Hospital in Jiroft in 2022. Following diagnosis confirmation through additional examinations and imaging, patients with ankle sprains not requiring surgery were selected and placed in two groups: one treated with spring ankle braces and the other with splints or casts. Both groups underwent a 4-week treatment regimen, comprising 30 individuals each. Data were collected and analyzed using SPSS version 26. Results: The average age of patients was 32.5 ± 13.4 years. Of the ankle sprain patients, 56.7% were male. Patients reported the highest satisfaction levels with the plaster cast treatment method. A statistically significant relationship was found between patient satisfaction and the treatment methods of spring ankle braces and plaster casting (P < 0.05). Patients treated with plaster casts reported the lowest pain levels, with a significant relationship between pain levels and the two treatment methods (P < 0.05). Range of motion results were similar for both treatment methods, while the cast treatment showed the highest incidence of skin complications. A significant relationship was observed between spring ankle braces and plaster casts regarding skin complications (P < 0.05). Conclusion: Treating ankle sprains with plaster casts leads to higher satisfaction and lower pain levels compared to using spring ankle braces. [ABSTRACT FROM AUTHOR]

A meta-analysis including all relevant randomized controlled trials was conducted to compare soft bandage, splint and cast as the treatment of torus fracture. PubMed, Scopus, and Web of Science databases were searched in January 2023. Two comparisons were made: (1) splint versus cast, and (2) bandage versus rigid immobilization (i.e. splint or cast). Main outcomes were pain, clinical healing of the fracture and return to activities. Secondary outcomes were adverse events (skin issues, problems with cast/splint/bandage) and patient/parental satisfaction. Seven studies with 1550 patients were included. Splint was associated with higher pain scores at 3 days compared to cast (Mean difference [MD] 1.00, CI 0.06–1.94) and at 1 week (MD 1.46, CI 0.84–2.08, moderate-certainty evidence), but faster return to activities (at 3 weeks RR 1.77, CI 1.09–2.88, at 4 weeks RR 1.44, CI 1.11–1.82, moderate-certainty evidence). All torus fractures heal clinically within 3–4 weeks (low-certainty evidence). Bandage may lead to slightly higher pain score (MD 0.35, CI 0.04–0.66, moderate-certainty evidence) at first day after treatment compared to rigid immobilization, but no evidence of a difference was found in later time points. The prevalence of skeletal muscle atrophy, caused by disease and aging, is rising as life expectancy increases. Exercise is the most effective treatment option; however, it is often impractical for individuals suffering from disease or bedridden. The formulation of non-exercise-based interventions is necessary. This study assessed the impact of acupuncture (AC), electro-acupuncture (EA), and electrical stimulation (ES) on muscle mass and contractile properties in a model of casting-induced muscle atrophy. Sprague-Dawley rats (n = 40) were assigned to five groups: control (CON), cast (CT), cast receiving AC (CT-AC), cast receiving EA (CT-EA), and cast receiving ES (CT-ES) (n = 8 each). Treatments were 15 min and three times/week for 14 days. Contractile properties and protein markers of atrophy and inflammation were measured. Casting decreased muscle mass and fiber cross-sectional area, but AC, EA, and ES attenuated cast-induced muscle atrophy. All treatments increased peak twitch tension compared to CT. CT increased the protein levels of MAFbx and MuRF1, while AC, EA, and ES mitigated the elevation of these proteins. Our results indicate that acupuncture, electro-acupuncture, and electrical stimulation show promise as therapeutic strategies to counteract skeletal muscle loss and dysfunction resulting from disuse atrophy caused by injury, disease, and aging. Genome editing in non-model bacteria is important to understand gene-to-function links that may differ from those of model microorganisms. Although species of the Burkholderia cepacia complex (Bcc) have great biotechnological capacities, the limited genetic tools available to understand and mitigate their pathogenic potential hamper their utilization in industrial applications. To broaden the genetic tools available for Bcc species, we developed RhaCAST, a targeted DNA insertion platform based on a CRISPR-associated transposase driven by a rhamnose-inducible promoter. We demonstrated the utility of the system for targeted insertional mutagenesis in the Bcc strains B. cenocepacia K56-2 and Burkholderia multivorans ATCC17616. We showed that the RhaCAST system can be used for loss- and gain-of-function applications. Importantly, the selection marker could be excised and reused to allow iterative genetic manipulation. The RhaCAST system is faster, easier, and more adaptable than previous insertional mutagenesis tools available for Bcc species and may be used to disrupt pathogenicity elements and insert relevant genetic modules, enabling Bcc biotechnological applications. Background: The aim of this study was to compare the effects of four different immobilization methods [single sugar tong splint (SSTS), double sugar tong splint (DSTS), short arm cast (SAC), and long arm cast (LAC)] commonly used for restricting forearm rotation in the upper extremity. Methods: Forty healthy volunteers were included in the study. Dominant extremities were used for measurements. Basal pronation and supination of the forearm were measured with a custom-made goniometer, and the total rotation arc was calculated without any immobilization. Next, the measurements were repeated with the SAC, LAC, SSTS and DSTS. Each measurement was compared to the baseline value, and the percentage of rotation restriction was calculated. Results: The most superior restriction rates were observed for the LAC (p = 0.00). No statistically significant difference was detected between the SSTS and DSTS in terms of the restriction of supination, pronation or the rotation arc (p values, 1.00, 0.18, and 0.50, respectively). Statistically significant differences were not detected between the SAC and the SSTS in any of the three parameters (p values, 0.25; 1.00; 1.00, respectively). When the SAC and DSTS were compared, while there was no significant difference between the two methods in pronation (p = 0.50), a statistically significant difference was detected in supination (p = 0.01) and in the total rotation arc (p = 0.03). Conclusion: The LAC provides superior results in restricting forearm rotation. The SAC and SSTS had similar effects on forearm rotation. (1) Background: Dislocations of the trapeziometacarpal joint (TMC) are uncommon in children and adolescents. Only a few isolated cases are reported in the literature. Therapeutic guidance is minimal and inconclusive. (2) Methods: The authors present four patients treated for this unusual lesion. We evaluated the evolution according to treatment, age, patient activity, and quickDASH. Despite the clear limitation of the small number of patients, it is relevant to try to better understand this lesion and its evolution. A systematic review of the literature was also conducted. (3) Results: This is the largest published series of TMC dislocations in children and adolescents. Patients included a 12-year-old girl treated conservatively with a poor quickDASH; a 9-year-old girl treated surgically with the Eaton–Littler technique for a new dislocation with a partially modified quickDASH; a 13-year-old boy with two necessary closed reductions for a new dislocation and a very good final quickDASH; and a 12-year-old boy treated with closed reduction and percutaneous fixation with excellent final results with quickDASH. (4) Conclusions: In the absence of scientific evidence, conservative treatment and ligament reconstruction did not provide good functionality. In contrast, closed reduction with percutaneous fixation provided excellent results. Purpose: Different studies have shown that weightbearing is safe in stable transsyndesmotic, isolated lateral simple ankle fractures. Despite this evidence, AO guidelines still recommend immobilization with above-the-knee cast for 4–6 weeks for these fractures. The objective of this study was to compare the outcomes of mobilization and weightbearing to those of immobilization and non-weightbearing in patients with stable transsyndesmotic, lateral isolated simple ankle fractures. Methods: Fifty patients were randomly assigned to permissive weightbearing in a walking boot or non-weightbearing immobilization using a below-the-knee cast. Primary outcome was ankle functionality as scored by the Olerud-Molander Ankle Score (OMAS). Secondary outcomes were radiological displacement of fracture, range of motion (ROM), calf circumference, and RAND 36-item health survey. Patients were in follow-up for 24 months. Results: Ankle functionality after six and twelve weeks was significantly higher for the intervention group, with respectively 30 points (p = 0.001) and 10 points (p = 0.015) of difference. ROM improved significantly in the intervention group after six weeks. All fractures showed radiological progression of fracture healing. RAND 36-item showed differences in both physical (60.3 vs. 46.3, p = 0.017) and mental (78.5 vs. 58.2, p = 0.034) components in favor of the intervention group. In 16% of patients who initially showed stable fractures on radiographic imaging, joint dislocation was identified on weightbearing radiographs prior to randomization, leading to exclusion. The purpose of the article is to study the director's technique of destroying the stereotypical perception of a comic actor in a dramatic role and to prove that this is a way to create a counterpoint in the acting ensemble. Research methodology. The following scientific research methods were used: analysis (to process films, theoretical works, articles by film critics and journalists), comparison (to identify the features of stylistic decisions in films), induction (to derive general principles from specific examples in the work of Jim Carey), generalisation (to write conclusions). Scientific novelty. For the first time, the possibilities of using a comedic actor in film dramas are analysed, the semantic and stylistic effect arising from such a directorial decision is investigated. The emotional impact on the viewer's perception is determined, the principles of casting for supporting roles to create a counterpoint in the acting ensemble are derived. Conclusions. A comic actor in a dramatic role has a significant impact on the style of the film. Such a directorial decision can lead to a duality of perception of the character; the author, imposing tragic elements of the newly created image on the previous experience of contemplating the performer's comic roles, creates a problem of "trust" in the actor for the audience. The introduction of the factor of unrealism allows for a natural combination of stylised and realistic acting. This combination creates a counterpoint in the acting ensemble and charges the conflict with the very coexistence of the actors. The combination of the comic and the acutely dramatic works to increase tension and create additional conflicts. This can be used as the following directorial tools: creating emotional amplification and empathy; softening the perception of a complex topic; creating a sense of background for a character; recognising another layer of meaning in a familiar style of acting. The article presents the results of testing the flow curves of cast magnesium alloy MgAl9Zn1. Experimental tests were carried out on a deformation dilatometer in a compression test of cylindrical specimens heated to 380 °C, 400 °C, 420 °C at strain rates of 0,1 s-1, 1 s-1, 10 s-1. The experimental curves were described by functions used in programs for simulating metal forming processes based on the finite element method. The Generalized Reduced Gradient optimization method (GRDM) implemented in Microsoft Excel was used to determine the function coefficients. Based on the results obtained, the best functions characterizing the flow curves were selected for use in describing the material models used in numerical simulations of cast magnesium alloy MgAl9Zn1. The Horseshoe Canyon Formation has provided a wealth of vertebrate skeletal fossils over the past 100 years of collection. Vertebrate trace fossils such as footprints and trackways, however, have been markedly underrepresented. Trackways can provide a wealth of information regarding behaviour and soft tissues reflective of autopodial anatomy. Discovery of several giant, high-fidelity ichnites in series from the Morrin Member represents the first definitive dinosaur trackway reported from the Horseshoe Canyon Formation. The trackway is composed of three footprint casts in series, alongside 22 associated ichnites. The ichnites were formed in soft sediment molds along a non-uniform walking surface. The deep substrates allowed for the preservation of both the plantar surface and dorsal components of the trackmakers' feet. Some ichnites show soft tissue morphologies like interdigital webbing, toe pads, and skin impressions. Presence of these features and small kidney-shaped forelimb prints allow referral to Hadrosauropodus langstoni. The preservation of the trackway demonstrates asymmetries useful for comparison of the relief and impression surfaces of prints using both discrete and morphometric characteristics, improving confidence in identifying partial or isolated Hadrosauropodus prints of various modes of formation. Background: Pediatric femoral shaft fractures are one of the common fractures that we encounter in the orthopedics outpatient department. These fractures have various treatment options ranging from traction and early hip spica casting to elastic titanium intramedullary nails [titanium elastic nailing system (TENS)] and reamed intramedullary nails. In this study, we are evaluating the functional outcome and results following the use of flexible intramedullary nails and conservative management for femoral shaft fractures in children. Methods: This was a prospective observational study which was carried out in a tertiary care hospital from October 2015 to May 2017 for two groups of children who sustained traumatic femoral shaft fractures. A total of 40 patients were taken into consideration with 20 patients each in two groups. Results: The male-to-female ratio was 2.6:1 in our study population. The overall post-operative complication rate was more in the TENS group. Besides, we found that the TENS group of patients had a less limb length discrepancy (LLD) (P = 0.004) and a lower degree of angular deformity (P = 0.36). Conclusion: TENS demonstrated advantages including sufficient axial stability, early mobilization, and more rapid return to function than spica cast treatment; however, it had the drawback of possible infection and pin site irritation. Overall, TENS offers a minimally invasive method for treatment of the femoral shaft fractures in children which stands superior to traditional spica casting. [ABSTRACT FROM AUTHOR]

Earthworms have a prominent role in supporting soil functioning and thus in providing key services to humanity. Their beneficial role relates to effects on soil structure, carbon and nutrient cycling as well as the soil microbial community. Optimizing the role of earthworms in agricultural systems is therefore crucial for maintaining or improving soil quality and supporting a more sustainable, circular agriculture. Here, we summarize established knowledge on the role of earthworms in agronomy; present novel insights from the past decades; and identify key knowledge gaps to be addressed in the future to fully benefit from earthworms in our agricultural soils. We start by discussing how earthworms affect basic soil processes through their effects on soil structure, microbial communities and biogeochemical cycles. Further, we show how as a result of these changes, earthworms indirectly affect plant growth, the soil greenhouse gas balance and play a role in remediation of contaminated arable soils. We further address one of the paradoxes of earthworm ecology: that they are often not present in the soils where they are most needed. We subsequently discuss potential solutions to this paradox. Finally, we identify 10 key questions that need to be addressed in the near future. In our view, recognizing that earthworms are not a stand-alone solution to improving the sustainability of cropping systems, but an essential piece of the puzzle is crucial for optimizing the benefits they offer in agronomic systems. Objective: Plastic bronchitis (PB) is a rare disease characterized by obstruction of the airway by fibrinous mucus plugs. The etiology can be idiopathic or secondary to systematic diseases such as congenital heart diseases. Definitive diagnosis is made by pathological examination of the sputum or bronchial sample taken by bronchoscopy. In this study, the clinical status and treatment status of patients with PB were evaluated. Materials and Methods: Medical records of the patients diagnosed as PB were reviewed retrospectively. Age, gender, clinical symptoms, radiology, bronchoscopic findings, and pathology results were documented. Results: Six patients with PB were included in this study (female:male, 2:4). The median age of the diagnosis was 45 months. The most common symptoms are persistent wet cough and shortness of breath. The duration of symptoms ranged from 30 to 90 days. Atelectasis was the most common radiological finding. Diagnosis was made with pathological examination of the mucus in all patients. All of the patients were treated with bronchoscopic removal of the mucus, and 4 patients required oral prednisolone therapy. Symptoms and radiological findings resolved completely in all patients. Conclusion: Although PB is a rare disease, it should be kept in mind in relation to patients with persistent radiological and clinical respiratory symptoms. [ABSTRACT FROM AUTHOR]

Objectives: Distal forearm fractures are the most common pediatric fractures. This study aimed to investigate the effectiveness of below-elbow cast treatment for displaced distal forearm fractures in children compared to above-elbow cast through meta-analysis of randomized controlled trials. Methods: Several databases from January 1, 2000 until October 1, 2021 were searched for randomized controlled trials that assessed below versus above-elbow cast treatment of displaced distal forearm fractures in pediatric patients. The main meta-analysis comparison was based on the relative risk of loss of fracture reduction between children undergoing below versus above-elbow cast treatment. Other outcome measures including re-manipulation and cast-related complications were also investigated. Results: Nine studies were eligible of the 156 articles identified, with a total of 1049 children. Analysis was undertaken for all included studies with a sensitivity analysis conducted for studies with high quality. In the sensitivity analysis, the relative risks of loss of fracture reduction (relative risk = 0.6, 95% confidence interval = 0.38, 0.96) and re-manipulation (relative risk = 0.3, 95% confidence interval = 0.19, 0.48) between the below and above-elbow cast groups were in favor of below-elbow cast and statistically significant. Cast-related complications were in favor of below-elbow cast but did not attain statistical significance (relative risk = 0.45, 95% confidence interval = 0.05, 3.99). Loss of fracture reduction was noted in 28.9% of patients treated with above-elbow cast and 21.5% in below-elbow cast. Re-manipulation was attempted in 48.1% versus 53.8% of children who lost fracture reduction in the below-elbow cast and above-elbow cast groups, respectively. Conclusion: Below-elbow cast treatment was favored, with statistical significance, in terms of loss of fracture reduction and re-manipulation, and was not associated with a higher risk of cast-related complications. The accumulative evidence currently does not support above-elbow cast treatment and below-elbow cast treatment should be the mainstay for displaced distal forearm fractures in children. The cytomatrix at the active zone-associated structural protein (CAST) and its homologue, named ELKS, being rich in glutamate (E), leucine (L), lysine (K), and serine (S), belong to a family of proteins that organize presynaptic active zones at nerve terminals. These proteins interact with other active zone proteins, including RIMs, Munc13s, Bassoon, and the β subunit of Ca2+ channels, and have various roles in neurotransmitter release. A previous study showed that depletion of CAST/ELKS in the retina causes morphological changes and functional impairment of this structure. In this study, we investigated the roles of CAST and ELKS in ectopic synapse localization. We found that the involvement of these proteins in ribbon synapse distribution is complex. Unexpectedly, CAST and ELKS, in photoreceptors or in horizontal cells, did not play a major role in ribbon synapse ectopic localization. However, depletion of CAST and ELKS in the mature retina resulted in degeneration of the photoreceptors. These findings suggest that CAST and ELKS play critical roles in maintaining neural signal transduction in the retina, but the regulation of photoreceptor triad synapse distribution is not solely dependent on their actions within photoreceptors and horizontal cells. Air transport is considered to be the safest means of transport. However, if an accident occurs, it often ends in catastrophe. Thus, significant efforts have been paid to sustain successful operations in aviation. Several studies have been carried out to understand the underlying reasons for accidents. This study used Fault Tree Analysis (FTA), Failure Mode and Effects Analysis (FMEA) and Causal Analysis based on Systems Theory (CAST) methods to analyse Tenerife aircraft accident and to compare the findings of different methods. The findings showed that while all three methods provided some overlapping findings, the CAST method led to the identification of all causes that were identified by other methods. Considering the nature of the causal factors, FMEA provided more causal factors that are related to organisation and technology than FTA. Introduction: Computed tomography can be used to screen and diagnose the presence of drug-filled packets in the body. We present an unusual computed tomography finding in a suspected body packer. Case summary: A traveller suspected of being a body packer was brought to the emergency department complaining of low back pain. On examination, there was an abdominal cast circumferentially encasing his abdomen, which he alleged was part of the management of his lower back pain in his own country. Images: A plain computed tomogram of the abdomen and pelvis revealed that the cast was made of materials with a nodular appearance and heterogeneous density, which was different from that of a plaster of Paris cast. An investigation later confirmed the cast contained cocaine. Conclusion: Our case demonstrates how a body packer can traffic drugs outside their body for an alleged medical reason. Background: The current study aims to investigate the clinical efficacy of closed reduction and cast immobilization for patients with developmental dysplasia of the hip (DDH) who failed Pavlik harness treatment. Methods: Patients with DDH who underwent cast immobilization under general anaesthesia after the failure of the Pavlik harness or Tübingen brace treatment between January 2015 and December 2020 were retrospectively recruited. General information, including Graf classification of initial diagnosis, initial treatment, age of cast immobilization, IHDI classification, AI index, avascular necrosis (AVN), and residual dysplasia, was collected. The incidence of AVN and late acetabular dysplasia (LACD) was also estimated. Moreover, factors related to AVN and LACD were investigated by multiple logistic regression analysis. Results: Thirty‐four patients (47 hips) were finally included in the current study. Of these patients, 31 hips (66.0%) were successfully treated with closed reduction and cast immobilization. Open reduction was successfully performed in 16 hips (34.0%). Till the latest follow‐up, LACD and AVN were found in 13 (27.7%) and 10 (21.3%) hips, respectively. In the open reduction group, type III and IV of the IHDI classification and type IV of the Ultrasound Graf classification were significantly higher when compared with the closed reduction group. Multiple logistic regression showed that failure of closed reduction was related to the initial types of the Ultrasound Graf and IHDI classifications. Conclusions: Although the success rate of closed reduction after early harness failure in DDH is only 66%, we still advocate closed reduction as a first‐line treatment for children who have failed sling treatment. Studies have reported a high percentage of ankle fracture dislocations with secondary loss of reduction during primary treatment with a splint or cast. This study aimed to assess the rate of secondary loss of reduction in unimalleolar ankle fracture dislocations treated primarily with a cast or external fixator, identify the potential influence of fracture morphology, and investigate the potential implications. Unimalleolar ankle fracture dislocations with and without posterior malleolar fracture between 2011 and 2020 were included. Patients were categorized into two groups, depending on the method of temporary treatment. Fracture morphology, time to definitive surgery, and soft-tissue complications were compared. Of 102 patients, loss of reduction tended to occur more often in the cast group (17.3%) than in the external fixator group (6.0%). The presence of a posterior malleolar fracture did not have a significant influence on loss of reduction in cast immobilization; however, the fragment proved to be significantly bigger in cases with loss of reduction. No statistically significant differences in soft tissue complications or time to definitive surgery were found. Surgeons should consider the application of interval external fixation in the primary treatment of unimalleolar ankle fracture dislocations with additional posterior malleolar fractures. A complete ostrich femur from the Late Pleistocene deposits of the Upper Cave at Zhoukoudian (China) was referred by Shaw to Struthio anderssoni in the 1930s, but its present whereabouts are unknown. A good quality plaster cast of the missing specimen has been found in the collections of the Natural History Museum (London). This cast provides interesting information about the morphology of this large ostrich femur, which had previously been only summarily described and not illustrated. Although smaller than the femora of the Early Pleistocene giant ostrich Pachystruthio, the robust femur from Zhoukoudian shows morphological similarities with them, and it is suggested that 'Struthio' anderssoni should be placed in the genus Pachystruthio. The best choice of orthosis in the treatment of acute Achilles tendon rupture is still under debate. To investigate if choice of orthosis in the first 3 weeks of treatment affected patient reported outcome (Achilles tendon Total Rupture Score (ATRS)), tendon elongation (Achilles Tendon Resting Angle (ATRA) and Heel Rise Height (HRH)) and re-rupture. Registry study in the Danish Achilles tendon Database. Patients treated with cast and patients treated with walker in the first 3 weeks of treatment were compared using a linear mixed-effects model adjusted for potential confounders. 1304 patients were included in the study. No clinically relevant difference was found: Adjusted mean difference (using walker the whole period as reference)(95% CI) ATRS after 1 year = 0.1(−3.0; 4.1), ATRS after 6 months = 2.0(−4.5; 5.8), ATRS after 2 years = 3.0(−0.7; 7.0), HRH difference = 0.6(−6.6; 8.2), ATRA difference = 0.03°(−1.5; 1.6), re-rupture(odds ratio) = 0.812(0.4; 1.61). Bi-films are double-layered oxide films entrapped in the casting parts. Characterization of such nano-sized defects has always been an engrossing issue. The oxide/Metal/Oxide (OMO) sandwich method provides a physical simulation of oxide layers with areas thin enough to be studied by High-Resolution Transmission Electron Microscopy (HRTEM). The effect of Be on the oxide composition on the surface of Al–5Mg melt was studied with the addition of 2000 ppm Be via OMO technique. The average thickness of the oxide films was measured to be 51.4 and 10 nm for Al–5Mg and Al–5Mg–Be samples, respectively. After EDS and XRD Investigations, the study was carried on with HRTEM equipped with Selective Area Electron Diffraction (SAED). OMO sandwich samples were susceptible to providing areas thin enough to be studied by HRTEM, with no need for sample preparation. While thermodynamics predicts the formation of MgO, BeO, MgAl2O4, BeAl2O4, and Al2O3 oxides, SAED patterns only confirmed the formation of polycrystalline MgO and BeO layer in Be containing alloy. [Display omitted] Anatomical 3D-printed nasal casts are valuable models to investigate intranasal drug deposition, providing preclinical data that cannot be obtained in animal models. However, these models are limited since they are often derived from a single patient or represent a mean of several groups. The present study aimed to better characterize the anatomical differences of the nasal cavity in a European sub-population and to assess the potential impact of anatomical variations on intranasal deposition by medical devices. Ninety-eight cranial computed tomography scans of patients were selected and analyzed in 2D and 3D conformations. They showed symmetry of cavities and a high level of heterogeneity of measurements, especially volume and area, in the population. Three anatomical groups with distinct nasal geometry were identified and 3D nasal casts of the most representative patient of each group were printed. Fluorescein was administered using three medical devices: a nasal spray, a sonic jet nebulizer and a prototype mesh-nebulizer. The deposition profiles were compared with the Aeronose® as a reference. Our results show that anatomical variations influenced the deposition profiles depending on the device, with a higher variation with spray and the mesh-nebulizer. This work emphasises the importance of anatomical parameters on drug intranasal deposition and the need to evaluate inhaled drugs on different 3D nasal casts reflecting the target population. Earthworms are pivotal in shaping soil ecological processes through their bioturbation activity and organic matter consumption. Earthworm species are known to have different impacts on soil structure, but only a small number of species have been studied so far, and few studies have examined how earthworms simultaneously affect soil functions. Here, we measured the impact of different earthworm species on soil structure (bioturbation function), carbon (C) and nitrogen (N) dynamics and the microbial community (organic matter transformation function), while exploring the links between these functions, across distinct soil compartments (surface casts, below-ground drilosphere, and bulk soil). Six earthworm species (Lumbricus terrestris , Allolobophora chlorotica , Octolasion cyaneum, Octodrilus complanatus , Aporrectodea caliginosa meridionalis and Microscolex dubius) of different ecological categories and functional groups were incubated in soil cores with soil and alfalfa litter for 6 weeks. Our results on the bioturbation function illustrated a great diversity of behaviors and confirmed the relevance of a functional classification based on bioturbation metrics. The main microbial hotspots were surface casts, whose characteristics allowed to distinguish two groups of species. Octod. complanatus, L. terrestris and M. dubius induced high humidity (respectively, +57, +48, +74%), high C (respectively, 19.9, 24.8, 33.2 g kg⁻1 dry soil) and N (respectively, 2, 2.3, 3.2 g kg⁻1 dry soil) content and microbial community selection, promoting C and N mineralization. The three other species had a lower impact. The below-ground drilosphere only showed specific characteristics in the case of L. terrestris. The effects of the studied species on the organic matter transformation function did not align with their bioturbation activities nor with their ecological category. These findings show that the contribution of earthworms to C and N turnover is only partially dependent on their bioturbation effects and suggest the usefulness of developing distinct functional groups based on the specific soil functions under consideration. There are lots of gene editing tools for targeting genome sequences. Some are almost known, and most are a complete mystery and undiscovered. CRISPR/Cas editing tools have brought about a major revolution in medicine. Researchers have shown that CRISPR can modify DNA much more accurately, economically and easily than previous methods. CRISPR has proven itself effective for the deletion, replacement and insertion of DNA fragments into cell types, tissues and organisms. Recently, combining CRISPR/Cas with factors (transcription factors/repressors, exonucleases, endonucleases, transposons, caspase, fluorescent proteins, oxidoreductive enzymes, DNA/RNA polymerases), and elements (aptamers, barcodes, fluorescent probes, Trigger) have provided genome, transcriptome, proteome and epigenome modification. These modules are being investigated for cancer prevention and therapy and this review focuses on such innovative combinations that hopefully will become a clinical reality in the near future. Heterogeneous W-steel joining components will produce brittle intermetallic compounds (IMCs) and significant residual stress in the interface. Adding a Cu interlayer serves as an effective solution. Nevertheless, the strengthening of W-Cu-steel joints is restricted because W-Cu and Cu-steel are members of binary immiscible and finite solid solution systems. Thus, accomplishing interface alloying by overcoming the positive generating energy of insoluble systems and opening up interatomic diffusion channels is a crucial issue to be addressed. In this work, casting and brazing technologies were incorporated into bonding W-Cu-steel to provide a high temperature field, as well as the dissolving and wetting of Cu-based liquid phase to refractory W. It is shown that the superior tensile strength of the W/Cu castings-steel brazed joints (∼264 MPa) was achieved, and the joint survived 1000 cycles of thermal fatigue under 1 MW/m2. To assess the effects of brazing and casting on the W-Cu-steel joint, a detailed analysis was conducted on the mechanism of atomic diffusion in the joint interface. It is considered that in W-Cu joining, casting provided a higher thermodynamic driving force than brazing, thus achieving better interatomic diffusion and a wider microalloying region. Cu-steel joining achieved good alloying and forming dendritic extensions by intergranular diffusion. Based on the process optimization results, the feasibility of preparing the U-shaped first wall (FW) mock-up with W armor using brazing technology was verified. The importance of earthworms for soil functioning is widely accepted and is mainly linked to their burrowing, feeding and casting behaviours. However, our knowledge of earthworm behavior is still limited by the lack of efficient and easy-to-use methods. In this work, we describe and test a new marker of earthworm bioturbation. Dense particles of tungsten, visible in X-ray images, were located at a certain depth in repacked soil cores inoculated by either Lumbricus terrestris or Aporrectodea caliginosa , alone or in combination. This marker was compared to the classical luminophore method (i.e. colored sand particles) generally used in aquatic ecology. The dynamics of both the burrow systems and the casting activities was followed using X-ray tomography every two weeks for a period of 10 weeks. Below ground casting activities could be assessed since parts of the casts excreted by the earthworms were marked by tungsten and appeared white in the images. The time dynamics of both burrow and cast productions were almost linear and significantly influenced by the earthworm species and the presence of litter. As expected, L. terrestris created larger (up to 50 cm3) and deeper burrow systems and made fewer macropores compared to A. caliginosa. Surprisingly, both species were equally influenced by the presence of litter, and this highlighted their behavioral plasticity. For example, A. caliginosa decreased by 32 % the volume of burrows in the presence of litter and the resulting burrow system was significantly shallower which showed that this endogeic species is indeed able to partly feed on surface litter. The addition of TiC nanoparticles induced a refined basket-weave structure, replacing coarse α colonies in cast Ti-2.5Al. Thus, the yield strength and elongation of Ti-2.5Al+TiC are increased by 46.3 % and 57.6 %, respectively. Purpose: Above-knee casts pose a major challenge in the day-to-day activities among walking age clubfoot patients due to complete restriction of knee movement. This current study investigates the effectiveness of below-knee casts compared to above-knee casts for managing walking age clubfoot deformity. Methods: After approval from the institutional ethical committee, we enrolled walking age clubfoot patients for deformity correction through corrective manipulation and casting through below-knee casts over 2 years. The corrective manipulation was performed using the Ponseti technique. The patients were followed for a minimum of 2 years period. To compare the effectiveness of below-knee casts over above-knee casts, we enrolled equal numbers of walking age clubfeet matched for age and gender. We compared the two groups in terms of initial and post-correction Dimeglio scores, individual deformities corrections (maximum ankle dorsiflexion, heel varus correction, foot abduction), failure rates, relapses, and complication rates. Results: 56 patients with 80 clubfeet with a mean follow-up of 39.98 months were considered for the final analysis. There were 29 patients in the below-knee cast group and 27 in the above-knee cast group, with 40 clubfeet each. The patients in the above-knee casting group had significantly better post-correction Dimeglio scores and individual deformity components corrections than the below-knee cast group. There were nil failures in the above-knee casting group and 7.5% in the below-knee cast group. The relapse rate was 15% in both groups, with none requiring any extensive soft tissue procedure. We did not encounter any major complications during the treatment and follow-up. Conclusion: The deformity correction and maintenance are suboptimal with the below-knee casts. Forearm fractures such as distal radius fractures are traditionally treated with a plaster or synthetic cast. Patients commonly report inconvenience of the cast, skin problems, and occasionally radial sensory nerve numbness. A known issue with casting is that the rate of secondary dislocation is high. As an alternative to casts, personalized 3D-printed braces are increasingly used. This review provides an inventory of current developments and experience with 3D-printed forearm braces. Main focus was on the design requirements, materials used, technical requirements, and preclinical and clinical results. Review of 12 studies showed that all printed braces used an open design. Fused Deposition Modelling is most commonly used 3D-printing technique (seven studies) and polylactic acid is the most commonly used material (five studies). Clinical evaluation was done in six studies, mainly involving distal radius fractures, and generally showed a low complication rate and high patient satisfaction with the printed brace. Simple Summary: A bibliometric analysis was carried out to know the evolution of research on genes associated with meat tenderness, of interest for the development of selection programs. Since 1993, studies have been limited to a few researchers in high-income countries due to the costs associated with the techniques. The main findings showed that the scientific production had a discontinuous growth because science experienced a significant change since approximately 2010. Marker-assisted selection was widely used, evaluating mainly CAPN (calpain) and CAST (calpastatin) genes for their contribution to meat tenderness, especially in cattle. However, the effects are small; therefore, genomic selection was implemented by genotyping thousands of single nucleotide polymorphisms (SNPs) for further explanation of genetic variation. The results shown are important for scholars to identify emerging methodologies and gaps in the literature and to know who the prolific authors and institutions in the field for possible collaborations, etc., are. Tenderness is one of the main characteristics of meat because it determines its price and acceptability. This is the first bibliometric study on the trend of research on the role of genes in meat tenderness. A total of 175 original and English-language articles published up to 2021 were retrieved from Scopus. The bibliometric analysis was carried out with VOSviewer (version 1.6.18, Eck and Waltman, Leiden, Netherlands) and complemented with the Analyze search results service from Scopus. Erroneous and duplicate data were eliminated, and incomplete information was added to standardize the results. Scientific production was evaluated by means of quantity, quality and structure indicators. As a first glance, 8.816% of authors have published more than 50% of papers mainly related to genes encoding the calpain (CAPN)-calpastatin (CAST) system and single nucleotide polymorphisms (SNPs). Among other findings, a strong link was found between the contribution of the main countries (led by the United States with) and their institutions (led by the USDA Agricultural Research Service with) to their gross domestic product. Most studies on the topic are published in the Journal of Animal Science, and other journals with high impact according to the number of citations and different metrics. Finally, when evaluating the most cited articles, the occurrence and association of the main keywords, it was confirmed that research is focused on the role of CAPN and CAST genes and of SNPs in beef tenderness. The change in science was emphasized; although marker-assisted selection is still used, genes have an infinitesimal effect on complex traits. Objective: There is limited evidence about the cross-cultural validity of autistic symptoms in schoolaged children in Brazil. We used data from a large school survey to evaluate the factor structure of autism symptoms in community-dwelling children and adolescents. Methods: We translated the Childhood Autism Spectrum Test to Brazilian Portuguese and performed factor analyses to investigate the factor structure of parent-reported autistic symptoms in a large sample (n=8,571) of children/adolescents from a school survey in the metropolitan area of São Paulo. Results: Autistic symptoms were best conceptualized under a correlated-factors model with two factors: one predominantly characterized by social-communication symptoms and the other by symptoms of inflexible/restricted language, behaviors, and interests. Conclusions: These findings provide evidence that the structure of autistic symptoms in Brazil is similar to that described in other countries, indicating the cross-cultural validity of autism in Brazil. Vehicle emission regulations in Europe and many Asian countries include a particle number limit. The number concentration is measured, typically, with condensation particle counters (CPCs). For research purposes, the size distributions provide useful information. Scanning mobility particle sizers (SMPSs) accurately provide the size distribution but are not suitable for transient aerosol. Engine (fast) exhaust particle sizers (EEPSs) cover this gap, but with less accuracy. Fast size distribution instruments are commonly used in the research and development of engines. In the last few years, instrument algorithms have been improved, but studies assessing the improvements are limited, in particular in their lower size range, around 10–20 nm, and for soot-like aerosol. In this paper, we compared the three instruments using salt, silver, diffusion flame soot (CAST), and spark discharge graphite particles. Moreover, vehicle exhaust number concentration measurements with EEPSs over a seven-year period were presented. In terms of particle concentration, EEPS overestimated, on average, 25% compared to CPC, in agreement with previous studies. Its accuracy for mean particle size determination was better than 5 nm compared to SMPS. The agreement between the instruments was satisfactory but the uncertainty increased at low concentrations and larger particle sizes, showing that there is still room for further improvements. Experimental challenges, such as low concentration levels of modern vehicles, losses in the diluters, use of photometric mode by the CPCs, and the material impact, are also discussed. The flowability of Ultra-high molecular weight polyethylene (UHMWPE) is very poor so flow modification is needed to improve its processing efficiency, but flow modification could significantly deteriorate its wear resistance. To solve this technical problem, it is necessary to recognize the reasons why flow modifiers deteriorate the wear resistance of UHMWPE. In current research, influence of calcium stearate (CaSt) which is a commonly used flow modifier on the wear resistance of UHMWPE was investigated, the experimental results showed that a small amount of CaSt (0.5wt%) could significantly deteriorate the wear resistance of UHMWPE (11%). Subsequently, the effect of CaSt on the three-phase structure (amorphous, crystalline and interphase) of UHMWPE was investigated to explain why CaSt deteriorates the wear resistance of UHMWPE in terms of structure–property relationship. The study of crystallization and melting behavior indicates that the incorporation of CaSt makes the matrix produce more well-formed crystals thus increasing the hardness of the matrix, and the increase in hardness can alleviate the degree of abrasive wear. The study of the amorphous region found that CaSt has a unique disentanglement effect, which reduces the degree of entanglement of molecular chains of the matrix, and the reduction of the degree of entanglement deteriorates the shear resistance of the matrix, thus aggravating the shear damage of the sample during the wear process. Meanwhile, the disentanglement effect of CaSt can also improve the recrystallization ability of the matrix, reducing the interphase of the matrix thus making the matrix more susceptible to structural damage during the wear process. Furthermore, the incorporation of CaSt significantly reduces the heat resistance of the matrix, which makes the wear resistance of the matrix deteriorate more caused by the friction heat during the wear process. This paper can provide theoretical guidance to solve the problem of severe degradation of wear resistance after flow modification of UHMWPE and provide inspiration for the structural design of other wear-resistant materials. This paper deals with the topics of violence and enmity both in terms of their traditional and cultural characteristics, as well as in light of the transformations undergone between the 16th and 17th centuries following the measures implemented in various European environments to target above all the alarming phenomenon of banditry, and adopted in consideration of the new-found need for safety and tranquillity. Enmity is typical of all human groups everywhere and at any point in time. And since for long it was often closely related to a distinct dimension of violence, its characteristics and changes reveal the symbolic meanings and values that each society ascribes to conflicts and to the methods of their resolution. In fact, the concept of enmity, deeply intertwined with that of revenge, played an essential role within a society in which a peace-centred order was not only pre-eminent but also constituted as the safeguarding of social hierarchies focused on status, kinship, and honour. In the second half of the 16th century, in the wake of widespread social insecurity and the crisis of political polycentrism that had characterised the mediaeval state, the cultural system of enmity lost its traditional stamp and became, often unawares, a means of extreme violence. This passage is clearly noticeable in the ruthless challenge that the central powers mounted against banditry, enacting a legislation that led to a veritable state of emergency, which among other things entailed an explicit mandate for the use of private violence. This paper addresses enmities between burghers and other inhabitants of early modern Ljubljana, documented in town council records from 1521 to 1671. In traditional dispute settlement, the concept of enmity was closely related to violent retribution, which was antithetical to the ideals of urban communities, which since the Middle Ages had been established upon the renunciation of the right to feud. Focused on the conception of enmity in the Carniolan capital following the general prohibition of feuding in 1495, the paper analyses the role, language and emotional dimension of traditional practices of dispute settlement, their intertwinement with court proceedings and coexistence with criminal law. While town council records cannot provide a complete picture of the attitude towards enmity or feud in early modern Ljubljana, as no criminal court records survive, they are an important source for understanding social relations more broadly and, in particular, the manner in which everyday enmities that undermined ideals of neighbourliness were mediated by the secular authorities. The analysis shows that in Ljubljana the traditional conception of enmity as a public expression of hostility and discord - the opposite of the urban values of peace, concord and good neighbourliness - remained in use well into the seventeenth century, despite its normative prohibition, as did the traditional terms and emotions expressing this relationship, especially 'resentment', 'bitterness' and (just) 'anger', while 'rage' seems to have still been used very rarely to justify transgressions of social norms. Although physical violence did not always erupt in the investigated cases, and remained limited when it did, it was a viable threat in enmities, even when they were largely carried out before the town council or court. These were the central fora for both conducting and settling enmities among burghers, as well as other lay non-noble inhabitants of early modern Ljubljana. Underpinned by community mediation in disputes, the burgomaster, town judge and councillors principally acted as mediators and arbiters, attempting to lead conflicting parties to settlement before enmity was exacerbated into violence - in particularly bitter cases by forcing them to settle ex officio under threat of sanctions (heavy fines, prison sentences) or even 'harsher', i.e. corporal punishment. The council and court also retained traditional expressions of renewed friendship and neighbourliness, (mutual) public apologies and handshakes to demonstrate reconciliation between the parties. Consequently, the autonomous town authorities were instrumental for the long perseverance of traditional practices of dispute settlement in Ljubljana, which is comparable to findings elsewhere in Europe. In this study, we synthesized a novel high-Zn 7xxx Al alloy through a comprehensive two-step thermo-mechanical treatment that includes hot extrusion and hot rolling. This approach effectively mitigates casting defects caused by high Zn content and preserves a significant number of dislocations and low-angle grain boundaries. These deformation substructures, coupled with high Zn concentrations, promote the dense formation of η' nanoprecipitates. As a result, this alloy exhibits an exceptional combination of mechanical properties, with an ultimate tensile strength (UTS) of 789 MPa and a tensile elongation of 9.2 %. Earthworm could regulate their body concentration of arsenic via storage or excretion, and the ability of As efflux among different earthworms is not consistent. Here, whole and semi As exposure patterns with 0-10-30-60-100 mg kg−1 exposure concentrations were set to characterize the As efflux in geophagous earthworm, Metaphire guillelmi. Cast As (As–C) and earthworms' antioxidative responses were monitored to explore the efflux mechanisms under 30 mg kg−1 As-spiked soil (As30), besides, As concentration in earthworm tissue after egestion and dissection depurations were compared. In the whole exposure pattern, As concentration in gut content (As-G, 19.2–120.3 mg kg−1) surpassed that in the tissue (As-T, 17.2–53.2 mg kg−1), and they both increased with exposure concentrations. With the prolong time, they firstly increased and kept stable between day 10–15, then As-G increased while As-T decreased between day 15–20. In the semi-exposure pattern, both As-G and As-T decreased when M. guillelmi was transferred to clean soil for 5 days. During the 42-day incubation in As30, the antioxidative responses including reactive oxygen species (ROS), glutathione (GSH) and glutathione-S-transferase (GST) were firstly increased and then decreased, and As–C (13.9–43.9 mg kg−1) kept higher than As-G (14.2–35.1 mg kg−1). Significantly positive correlations were found between As-T and GSH, As–C and GST. Moreover, tissue As after dissection (11.6–22.9 mg kg−1) was obviously lower than that after egestion (11.4–26.4 mg kg−1), but significantly related to ROS and GSH. Taken together, M. guillelmi exhibited excellent capacity of As efflux, and GSH explained tissue As accumulation while GST facilitated the As elimination via cast. Besides, dissection instead of egestion revealed the As efflux in M. guillelmi more accurately. These findings contributed to a better understanding of how geophagous earthworm M. guillelmi regulated tissue As accumulation for As stress tolerance, and recommended an optimal depuration mode to characterize As accumulation. • Mechanism responsible for the anomalously severe HCS of Al-Li alloys were investigated. • Strong strain localization resulting from Li-rich inclusions induces microcrack initiation. • Liquid refilling is impeded by Li-rich inclusions occupying the interdendritic channels. This work reports a new mechanism for the severe hot cracking susceptibility (HCS) of cast Al-Li alloys. In contrast to the narrow vulnerable solidification temperature range increasing cracking resistance, alloys with high Li content or solute content, which are supposed to show lower HCS, instead exhibit anomalously high HCS. Results reveal that this phenomenon arises from a variety of Li-rich inclusions distributed in the interdendritic paths, including Li 2 O, LiAlO 2 , Li 2 CO 3 and LiH. These particles can form spontaneously with O 2 , CO 2 and H 2 O during solidification, deteriorating the interfacial bonding capacity. Meanwhile, the occupation of refilling channels for residual liquid increases the potential risk of microcrack initiation. Background: Pressure offloading treatment is critical for healing diabetes-related foot ulcers (DFU). Yet the 2011 Australian DFU guidelines regarding offloading treatment are outdated. A national expert panel aimed to develop a new Australian guideline on offloading treatment for people with DFU by adapting international guidelines that have been assessed as suitable to adapt to the Australian context. Methods: National Health and Medical Research Council procedures were used to adapt suitable International Working Group on the Diabetic Foot (IWGDF) guidelines to the Australian context. We systematically screened, assessed and judged all IWGDF offloading recommendations using best practice ADAPTE and GRADE frameworks to decide which recommendations should be adopted, adapted or excluded in the Australian context. For each recommendation, we re-evaluated the wording, quality of evidence, strength of recommendation, and provided rationale, justifications and implementation considerations, including for geographically remote and Aboriginal and Torres Strait Islander peoples. This guideline, along with five accompanying Australian DFU guidelines, underwent public consultation, further revision and approval by ten national peak bodies (professional organisations). Results: Of the 13 original IWGDF offloading treatment recommendations, we adopted four and adapted nine. The main reasons for adapting the IWGDF recommendations included differences in quality of evidence ratings and clarification of the intervention(s) and control treatment(s) in the recommendations for the Australian context. For Australians with plantar DFU, we recommend a step-down offloading treatment approach based on their contraindications and tolerance. We strongly recommend non-removable knee-high offloading devices as first-line treatment, removable knee-high offloading devices as second-line, removable ankle-high offloading devices third-line, and medical grade footwear as last-line. We recommend considering using felted foam in combination with the chosen offloading device or footwear to further reduce plantar pressure. If offloading device options fail to heal a person with plantar DFU, we recommend considering various surgical offloading procedures. For people with non-plantar DFU, depending on the type and location of the DFU, we recommend using a removable offloading device, felted foam, toe spacers or orthoses, or medical grade footwear. Background: Gastric cancer (GC) is one of the leading malignant diseases worldwide, especially in Asia. CAST is a potential oncogene in GC carcinogenesis. The character of macrophage infiltration in the GC microenvironment also remains unaddressed. Methods: We first applied machine searching to evaluate gene candidates for GC. CAST expression and pan-cancer surveyance were analyzed using the Human Protein Atlas (HPA) and Gene Expression Profiling Interactive Analysis 2 (GEPIA2) database. The protein–protein interaction (PPI) network was downloaded from STRING. We investigated the impact of CAST on clinical prognosis using a Kaplan–Meier plotter. The correlations between CAST and Lgr5 and macrophage infiltration in GC were determined using TIMER 2.0. Finally, GeneMANIA was also used to evaluate the possible functional linkages between genes. Results: After the machine-assisted search, CAST expression was found to significantly influence the overall survival of GC patients. STRING revealed CAST-related proteomic and transcriptomic associations, mainly concerning the CAPN family. Moreover, CAST significantly impacts the prognosis of GC based on the validation of other datasets. Notably, high CAST expression was correlated with worse overall survival in GC patients (hazard ratio = 1.59; log-rank P = 9.4 × 10−8). CAST and Lgr5 expression were both positively correlated with WNT 2 and WNT 2B. Among the GC patients in several datasets, CAST and macrophage infiltration, evaluated together, showed no obvious association with poor clinical overall survival. Conclusions: CAST plays an important role in the clinical prognosis of GC and is associated with WNT 2/WNT 2B/Lgr5. The use of extrathoracic airway replicas in optimization of drug delivery to the lungs with nebulizers, dry powder inhalers (DPIs) and pressurized metered-dose inhalers (pMDIs) is discussed. Such airway replicas have been useful in evaluating new pulmonary drug delivery platforms mainly based on the comparison of the total lung dose (TLD) and the aerodynamic particle size distribution (APSD) of the aerosol distal to the physical models. The ability of these in vitro methods to replicate in vivo results has allowed advancements in respiratory drug delivery and in the accuracy and utility of in vitro-in vivo correlations (IVIVCs). We used calibrated 2D images uploaded by patients to an online platform to generate a 3D digital model of the limb. This was used to 3D print a splint. This method of 3D printing of splints was used for two patients who were not able to visit the hospital in person due to restrictions placed by the COVID-19 pandemic. Both patients were satisfied with the splint. We feel that this technology could be used to offer additional options to conventional splinting that allows contactless splint fitting. This study aims to identify the association between CAST/MspI and MSTN/HaeIII polymorphism with growth traits in Awassi sheep. A total of 129 (male and female) Awassi sheep were used in the study. Growth traits (Body weight BW, body length BL, chest depth CD, heart girth HG, and withers height WH) were taken from animals at one year old. PCRRFLP analysis was used to detect CAST polymorphism (Exon 1C/1D) and MSTN (Exon 3) genes. Three genotypes (MM, MN and NN) were observed for CAST/MspI polymorphism with allele and genotype frequency 0.78(M) and 0.22(N); 0.70(MM), 0.16(MN), and 0.14(NN). Two genotypes (MM and mm) were found for MSTN/HaeIII polymorphism with allele and genotype frequency as 0.10(M) and 0.90(N); 0.10(MM) and 0.90(mm). The genes were in agreement with Hardy-Weinberg equilibrium (p>0.05). The association analysis showed an association between CAST/MspI polymorphism and BW, BL, CD, and HG (P<0.05). The MM genotypes had the highest BW compared to MN and NN genotypes. At the same time, the MN genotypes had the highest BL, CD, and HG compared to MM and NN genotypes.
