The article examines the new political-ideological face of sugarcane plantation owners in the Northeastern sugarcane region, encompassing the three main producing states of the region: Alagoas, Pernambuco, and Paraíba. The central argument is that this traditional segment of the ruling classes has undergone significant changes in the past two decades, resulting in an expansion of its internal composition and its accommodation to the dynamics of agribusiness. Consequently, they have crafted a new rhetoric of legitimation, grounded in the defense of sugarcane monoculture as an agro-social business, drawing on concepts such as sustainability and socio-environmental responsibility. The limitations and contradictions of this discourse are scrutinized in the context of class conflicts and the economic and ideological dimensions contained therein, which explain why sugarcane plantation owners, through their representative bodies, seek social legitimacy for the full exercise of their activities. The water influence on electrochemical behaviour of Ag+ ions in urea and choline chloride mixture was investigated by cyclic voltammetry technique, while the molecular insights about the investigated systems were obtained from molecular dynamic (MD) simulation. The water content was variated from 0 up to 10% (v/v). Cyclic voltammetry technique showed that the peak potential for Ag+/Ag redox couples shifted in direction to more positive potentials with the gradual increase of water content in solution, indicating that the addition of water electrocatalyses the kinetics of the reduction of Ag+ ions. The MD simulations demonstrated that water molecules do not interact strongly with Ag+ ions but induce a small reduction in the number of urea molecules around of the ion and that the water molecules adjust to free spaces in the mixture. The excess level of carbon dioxide in the atmosphere has contributed a lot to global warming, occasioning several damages to the planet. Therefore, it is urgent to find ways to capture this gas. Then, the present work analyzed the temperature effect in CO 2 absorption through deep eutectic solvents (DESs) based on urea and choline chloride using an in silico approach. The Molecular Dynamics (MD) simulations indicated that the increased temperature reduced the interaction potential of carbon dioxide molecules with the DESs components, indicating that the absorption process is more favorable at 303 K. On the other hand, the Noncovalent Interactions (NCI) simulations suggest that the increased temperature reduced the strong attractions and increased repulsive interactions between the carbon dioxide molecules with the solvent analyzed. Therefore, both in silico approaches suggest that the carbon dioxide absorption is more indicated at 303 K. Molecules presenting reversible photoreaction are highly interesting because, in principle, can be used as photocatalysts in electron transfer reactions for the designing of molecular switches. Herein we present the results obtained for dipyridoquinoxalinophenazine (dpqpz), a non-photoluminescent product of a condensation reaction between 1,10-phenanthroline-5,6-dione and 2,3-diaminophenazine. After 2-electron/2-proton electrochemical or chemical reduction, the product dihydro-dpqpz exhibits red emission with maximum at 610 nm under excitation at 530 nm. Spectroelectrochemical studies showed this is a reversible process wherein the dpqpz molecule is recovered upon air contact. Theoretical calculations showed a new allowed electronic transition emerges after 2-electron reduction making the photoreduction possible. The emission efficiency of dihydro-dpqpz showed to be dependent on the solvent nature with the polarity being the dominant effect. The strong emission suppression in protic solvents (quantum yields of 0.02 and 0.15 in ethanol and acetonitrile, respectively) was ascribed to the influence of hydrogen bonds. The reversible redox-responsive fluorescence emission property of dihydro-dpqpz in the absence of a sacrificial donor and under visible light irradiation is an essential feature that qualifies its application as a feasible switch material. Introduction: Major depressive disorder (MDD), an incapacitating mental disorder, is characterized by episodes of at least 2 weeks of apparent changes in mood, cognition, and neurovegetative functions. Many neuroimaging studies using magnetic resonance imaging (MRI) have examined morphometric changes in patients with MDD, but the results are not conclusive. This study aims to review the literature and perform a meta-analysis on hippocampal volume (HcV) in patients with MDD. Methods: Studies on HcV in patients with MDD diagnosis were identified from major databases (MEDLINE, EMBASE, The Cochrane Library, Scopus, PsycINFO, and SciELO) using the search terms depression, major depressive disorder, MDD, unipolar, magnetic resonance imaging, MRI, and hippocampus. Results: A meta-analysis of 29 studies fulfilling specific criteria was performed. The sample included 1327 patients and 1004 healthy participants. The studies were highly heterogeneous with respect to age, sex, age of onset, and average illness duration. However, the pooled effect size of depression was significant in both hippocampi. MDD was associated with right (-0.43; 95% confidence interval [95%CI] -0.66 to -0.21) and left (-0.40; 95%CI -0.66 to -0.15) hippocampal atrophy. Conclusions: MDD seems to be associated with global HcV atrophy. Larger longitudinal follow-up studies designed to analyze the influence of sociodemographic variables on this relationship are required to yield better evidence about this topic. • The physicochemical properties of DES 1ChCl:2G were minimally affected with the addition of AgNO 3 due to the low concentration of 0.05 mol⋅L−1. • The reduction process of Ag+/Ag that have peak potentials around −0.05 V. • Ag+ ions strongly interact with the chloride anion in DES. The thermodynamic and electrochemical properties of the silver ions (Ag+) dissolved in choline chloride (ChCl) and glycerol (G) mixture at eutectic composition were investigated. In addition, a computational calculation was carried out using Molecular Dynamic (MD) and Density Functional Theory (DFT) methods to understand the prevailing interactions in the investigated system. Specific mass, speed of sound, viscosity, and refractive index were measured at temperatures between 293 and 343 K and 101.3 kPa. The electrochemical behavior of Ag+ ions was analyzed by cyclic voltammetry in the range of 313 and 343 K. Silver coatings were successfully electrodeposited on the gold (Au) surface without adding additives to the electroplating solution. Scanning Electron Microscopy (SEM) analyzed the surface morphologies of the electrodeposits. The computational calculation indicated that adding silver nitrate to the eutectic solvent provided a predominance of repulsive forces for 1ChCl:2G and had negligible influence on sound velocity and refractive index. Early embryonic mortality caused by maternal-fetal recognition failure in the three weeks after fertilization represents a major cause of reproductive inefficiency in the cattle industry. Modifying the amounts and ratios of prostaglandin (PG) F 2α and PGE 2 can benefit the establishment of pregnancy in cattle. Adding conjugated linoleic acid (CLA) to endometrial and fetal cells culture affects PG synthesis, but its effect on bovine trophoblast cells (CT-1) is unknown. The aim of this study was to determine the effects of CLA (a mixture of cis - and trans -9, 11- and −10,12-octadecadienoic acids) on PGE 2 and PGF 2α synthesis and the expression of transcripts involved with maternal-fetal recognition of bovine trophectoderm. Cultures of CT-1 were exposed to CLA for 24, 48 and 72 h. Transcript abundance was determined by qRT-PCR and hormone profiles were quantified by ELISA. The PGE 2 and PGF 2α concentrations were reduced in the culture medium of CLA-exposed CT-1 compared to that of unexposed cells. Furthermore, CLA supplementation increased the PGE 2 :PGF 2α ratio in CT-1 and had a quadratic effect (P < 0.05) on the relative expression of MMP9, PTGES2, and PTGER4. The relative expression levels of PTGER4 were reduced (P < 0.05) in CT-1 cultured with 100 μM CLA than in the unsupplemented and 10 μM-CLA groups. Treatment of CT-1 with CLA decreased PGE 2 and PGF 2α synthesis but a biphasic effect of CLA was observed on the PGE 2 :PGF 2α ratio and relative abundance of transcripts with 10 μM CLA providing maximal improvements in each endpoint. An alternative for replacing conventional organic solvents and ionic liquids, the Deep Eutectic Solvents (DES) emerged, a class of compounds with essential properties in the industry. These solvents have numerous advantages regarding ionic liquids, such as low price, biodegradability, and low toxicity. Furthermore, the DES has many applications in science, for example, organic synthesis, electrodeposition of metals, catalytic process. In this work, the behavior of Sn2+ and In3+ ions were analyzed in the solvents 1ChCl:2EG (chloride choline and ethylene glycol, DES) and water through computational simulations by molecular dynamics (MD) and Quantum Theory of Atoms in Molecules (QTAIM) calculations. The results showed that the Sn2+ and In3+ ions strongly interact with the chloride anion in DES. Furthermore, only the Sn2+ ion showed interaction with the ethylene glycol molecules in DES, which was not observed for In3+, which led to no interaction with EG. In contrast, the most likely interaction is between cations and oxygen from water in a water solvent. The analysis of Bond Critical Points (BCPs) showed that the strength of these interactions follows the following sequence: In-Ow > In-Cl > Sn-Ow > Sn-Cl. The behavior of both ions in the metallic mixture was invariant in DES and in water when compared to the same isolated ions. Deep eutectic solvents (DESs) have many advantages, making them a promising alternative in replacing ionic liquids and organic solvents. Besides, DESs have received much prominence due to their diverse applications: Electrodeposition of metals, organic synthesis, gas adsorption, and biodiesel production. Therefore, this work analyzed the effect of the temperature increase (298 K–353 K) on the behavior of the Co2+ ions in three eutectic solvents through electrochemical techniques and computational simulations. From the electrochemical analysis realized, the increase in temperature caused a reduction in specific mass and an increase in the diffusion coefficient. Besides, the activation energy values were of 15.3, 29.9, and 55.2 kJ mol−1 for 1ChCl:2 EG, 1ChCl:2U, and 1ChCl:2G, respectively. The computational simulations indicate that the increased temperature effect caused the replacement of HBD molecules by anions chloride around Co2+ ions for the SDW1 and SDW3 systems between the temperatures of 298 K–353 K, except for the SDW2 system that the replaced occurred in the interval of 313 K–353 K. Besides, the increase of temperature occasioned the increase of strength for Co–Cl interaction and weakened the interactions between the Co2+ ions with the oxygen of HBD molecules. Chagas disease (CD) is a tropical disease caused by the parasite Trypanosoma cruzi, transmitted by the barber insect. Currently, there are approximately 7 million infected people in the world, and it is estimated that 70 million people could contract this disease. The anacardic acid (AA) showed effectiveness in in silico and in vitro tests. The antichagasic potential of five sulfonamide molecules, derived from anacardic acid, was evaluated from a molecular approach based on the density functional theory (DFT), molecular dynamics (MD), and molecular docking (docking) calculations. Methyl 2-methoxy-6- (8- (methylsulfonamide) octyl) benzoate (SA1); 2-methoxy-6- (8- (phenylsulfonamide) octyl) benzoate (SA2); methyl 2-methoxy-6- (8- (2methylphenyl sulfonamide) octyl) benzoate (SA3); methyl 2-methoxy-6- (8-(methylphenylsulfonamide)octyl)benzoate (SA4); methyl2-(8-(2,5-dimethylphenylsulfonamide)octyl)-6-methoxybenzoate (SA5) were the investigated molecules. The DFT calculations were performed using the B3LYP/6-311+G (d, p) level of theory. The global and local reactivity data showed that SA1 shows the highest molecular reactivity, while SA2 is the most stable derivative. In addition, the structures of investigated molecules were confirmed by the linear correlations higher than 0.98 displayed between the experimental and calculated spectroscopic data (IR and NMR). Molecular docking of the molecules showed a greater prominence for the SA1, SA2, and SA4 molecules in the results of distances of ligand-cruzain. Photophysical studies of 4′-methyl-2,2′-bipyridine derivatized with the polycyclic aromatic hydrocarbons (PAHs) anthracene (mbpy-anth), naphthalene (mbpy-naph), and pyrene (mbpy-pyr) were acquired in different solvents. While the absorption spectra were not affected by the solvent nature, the emission ones showed red shifts with increase polarity consistent with an intramolecular charge-transfer (ICT) character. Theoretical calculations found a dominant "Hole" → "Particle" contribution for the first five transitions involving local excited-state (LE) and ICT transitions, with the latter being dominant at higher wavelengths (S 0 → 1CT). The fluorescence solvatochromic data suggested a hybridized local and charge transfer (HLCT) state for the mbpy-R compounds with the CT contribution prevailing in high polar solvents; a result that was corroborated by the monoexponential decay lifetimes. The Stokes shifts difference between heptane and dimethylsulphoxide, the most and least polar solvents used in this work, was calculated as 1315, 2969, and 808 cm−1 for mbpy-naph, mbpy-anth, and mbpy-pyr, respectively. The increase of the solvent polarity results in a red shift and a broadening of the emission and excitation bands for mbpy-anth and mbpy-naph with the spectra of the latter being the most affected. For mbpy-pyr, on the contrary, the emission spectra were narrower than the excitation ones indicating the L a transitions lie at lower energy than the L b. For this species, the results indicated an effective mixing of LE and CT states in the moderately polar solvents being in accordance with the observed single exponential decay which is likely related to one hybridized state. The rapid and safe removal of critical newborn infants (RNs), such as premature infants and those with complex congenital malformations, born in centers that have insufficient resources for Neonatal Intensive Care Units (ICUs) by means of interhospital transportation is considered one of the recommendations of the Ministry of Health due to its capacity to significantly reduce neonatal mortality from preventable causes. Objective: to characterize neonatal transport through the Aeromedical Service Metódos: This is an integrative review study, with a descriptive and exploratory approach, on the topic: Attendance of Neonatal Patients performed by the Aeromedical Service. In order to perform the search in the Virtual Health Library (VHL), we used combinations of the following keywords, considered descriptors in DeCS: "Neonatology", "Air Rescue" and "Urgency". The terms were cross-referenced as descriptors and also as title and abstract words. The search was performed in June 2019. Results and discussion: The literature the Aeromedical Service is mainly used among high risk NBs, in which there are pre-established indications and with this allows the introduction of better approaches. In which it is necessary to use pre-established materials and protocols, as well as a team prepared to perform care in neonatal patients. Conclusion: In view of this perspective, it is possible to understand that the technological advances aligned to the development of a quality medicine can provide better approaches to neonatal patients and thus establish a better quality of life and decrease of the morbimortality of these patients. PPP-RTK (Precise Point Positioning – Real Time Kinematic) is a positioning method by GNSS (Global Navigation Satellite Systems), based on the SSR (State Space Representation) concept. One challenge for the fast convergence of PPP-RTK solutions in real-time applications is the mitigation of atmospheric effects (troposphere and ionosphere). Thus, using atmospheric models, PPP solutions can achieve accurate positions and shortened initialisation time. Such performance depends on the topology of permanent station networks and atmospheric conditions. The main objective of this contribution is to study the PPP-RTK and the optimised infrastructure in terms of costs and benefits to realise the method using atmospheric corrections. This work was essential to analyze the behavior of Fe2+ and Mn2+ ions in water and deep eutectic solvent (DES) based on choline chloride and ethylene glycol (1ChCl:2EG (choline chloride and ethylene glycol, DES)) to evaluate the electrodeposition of metallic ions. Due to their low toxicity, high solubility, biodegradability and thermal stability, DES have shown promise as alternatives to traditional solvents such as water in various applications, including biological studies. They also dissolve insoluble substances in other types of solvents, working from the purification of chemicals to the manufacture of semiconductor devices. Water, choline and ethylene glycol molecules were optimized by Density Functional Theory (DFT) through B3LYP/6–311 +G(d,p) model implemented with the Gaussian 09 W software. Molecular Dynamics (MD) simulations were performed by GROMACS software 2020.04 in a cubic box of dimensions 8 nm × 8 nm × 8 nm. Quantum Theory of Atoms in Molecules (QTAIM) and analysis of Non-covalent Interactions (NCI) used Multiwfn 3.8 software. The preliminary results of MD simulations showed that gradually adding water increased the interaction between the metal center and water. Fe2+ and Mn2+ ions were placed in water (OW) and 1ClCh:2EG, indicating that the interaction between the cations and the chloride anion in DES was strong, followed by the ethylene glycol molecules (O1, O2). In the presence of water, however, Fe-OW and Mn-OW interactions were more substantial in all systems, either with the ions alone or in a mixture. Electrostatic forces acting on the electrons in the last filled layer of the iron atom provided the formation of shorter and stronger bonds than the Mn2+ ions. When placed in a 1:1 equimolar mixture, they showed little difference from when they were isolated. Thus, the Mn-Cl interaction was more significant in the presence of a eutectic solvent than the Fe-Cl interaction. QTAIM analysis showed for the FeDES and MnDES systems that the Electron Localization Function (ELF) values increased for the Fe-(O1, O2) and Mn-(O1, O2) interactions but decreased for the Fe-Cl and Mn-Cl interactions in the metal mixture. It revealed strong hydrogen bonding interactions between the water molecules, Fe2+ and Mn2+, which may affect their chemical reactivity, as the Fe-Cl interaction in DES was stronger than Fe-Cl in any water. The Laplacians of the electronic densities were positive, and so were the ELF values, indicating a predominance of non-covalent interactions of the van der Waals forces type. Considering the two metal ions alone, in the presence of DES, the interaction strength followed Fe-Cl > Mn-Cl > Fe-(O1, O2) > Mn-(O1, O2). In the mixture, Fe-Mn followed the pattern Mn-Cl > Fe-Cl > Fe-(O1, O2) > Mn-(O1, O2). Thus, comparing the 3 systems, the interaction strength was Fe-Cl > Mn-Cl > Mn-Cl mixture > Fe-Cl mixture e Fe-(O1, O2) mixture > Fe-(O1, O2) > Mn-(O1, O2) mixture > Mn-(O1, O2). The interaction strength for the systems with water was Fe-Cl mixture > Fe-Cl > Mn-Cl > Mn-Cl mixture. However, for the interaction with OW, the observed behaviors were: W300, Mn-OW > Fe-OW mixture > Mn-OW mixture > Fe-OW mixture ; W600, Mn-OW mixture > Fe-OW mixture > Mn-OW > Fe-OW; W900, Mn-OW > Fe-OW > Mn-OW mixture > Fe-OW mixture ; W1200, Mn-OW > Mn-OW mixture > Fe-OW mixture > Fe-OW mixture ; W5580, Mn-OW > Fe-OW > Mn-OW mixture > Fe-OW mixture. The mixtures in the 1:1 molar ratio of the cations presented a little discrepant from their isolated ions, suggesting a new approach to observing the behavior of these systems and the coordination possibilities. INTRODUCTION: Hemophilic arthropathy affects people with hemophilia (PwH) and causes joint dysfunction and disability. Brazil has a unique situation and implemented policies to improve health care for PwH. The aim of this study was to evaluate the Functional Independence Score in Hemophilia (FISH), Hemophilia Joint Health Score (HJHS), and associated factors among adult PwH attending a Hemophilia Comprehensive Care Center in Brazil. METHODS: A post hoc analysis was conducted, including 31 patients who had submitted to a physical evaluation during a previously published cross-sectional study performed from June 2015 to May 2016 at the Brasília Blood Center Foundation, Brazil. RESULTS: The mean age was 30.8±9.4 years, and 80.6% had severe hemophilia. FISH was 27.0±3.8, and HJHS 18.0±10.8. The ankle was the most often affected joint (25/31, 80.6%). There were significant correlations between FISH and HJHS scores and the Hemophilia Quality of Life Questionnaire for Adults. Patients with severe hemophilia (P = 0.029) and PwH aged = 30 years (P = 0.049) had lower FISH scores. Monthly household income > 2 Brazilian minimum wages was independently associated with improved HJHS (P = 0.033). The factors simultaneously associated with better HJHS and FISH were age < 30 years (P = 0.021) and monthly household income < 2 minimum wages (P = 0.013). CONCLUSION: FISH and HJHS showed favorable scores despite being performed in a country with unfavorable socioeconomic conditions. In addition to hemophilia severity and age, monthly household income was independently associated with functional and articular state of PwH. Improving the binding of metal complexes to DNA to boost cancer cell cytotoxicity requires fine tuning of their structural and chemical properties. Copper has been used as a metal center in compounds containing intercalating ligands due to its ability to catalytically generate reactive oxygen species (ROS), such as hydroxyl radicals (OH˙). We envision the synergy of DNA binding and ROS generation in proximity to target DNA as a powerful chemotherapy treatment. Here, we explore the use of [Cu(2CP-Bz-SMe)]2+ (2CP-Bz-SMe = 1,3-bis(1,10-phenanthrolin-2-yloxy)-N-(4-(methylthio)benzylidene)propan-2-amine) for this purpose by characterizing its cytotoxicity, DNA binding, and ability to affect DNA replication through the polymerase chain reaction – PCR and nuclease assays. We determined the binding (Kb) and Stern–Volmer constants (KSV) for complex-DNA association of 5.8 ± 0.14 × 104 and 1.64 (±0.08), respectively, through absorption titration and competitive fluorescence experiments. These values were superior to those of other Cu-complex intercalators. We hypothesize that the distorted trigonal bipyramidal geometry of [Cu(2CP-Bz-SMe)]2+ allows the phenanthroline fragments to be better accommodated into the DNA double helix. Moreover, the aromaticity of these fragments increases the local hydrophobicity thus increasing the affinity for the hydrophobic domains of DNA. Nuclease assays in the presence of common reducing agents ascorbic acid, nicotinamide adenine dinucleotide, and glutathione showed the effective degradation of DNA due to the in situ generation of OH˙. The [Cu(2CP-Bz-SMe)]2+ complex showed cytotoxicity against the following human cancer cells lines A549, MCF-7, MDA-MB-231 and MG-63 with half maximal inhibitory concentration (IC50) values of 4.62 ± 0.48, 5.20 ± 0.76, 5.70 ± 0.42 and 2.88 ± 0.66 μM, respectively. These low values of IC50, which are promising if compared to that of cisplatin, are ascribed to the synergistic effect of ROS generation with the intercalation ability into the DNA minor grooves and blocking DNA replication. aims to identify compliance with a treatment protocol in adult patients with hemophilia and to evaluate the factors associated with and the impact on mortality of noncompliance with a hemophilia treatment protocol. Methods: This was a cross-sectional study that was performed between June 2015 and May 2016, followed by a cohort to evaluate mortality until July 2019 that included all adult patients with hemophilia registered in the Federal District, Brazil. Results: Among 138 patients enrolled in the study, 35 patients were compliant with all items of the treatment protocol (25.4%). Regarding each item, compliance with the medical consultations was 71.0% (98/138); the clotting factor regimen was 65.9% (91/138); and the serological tests were 51.4% (71/138). The mortality was 7.2% (10/138). Noncompliance with any aspect of the protocol was associated with mortality: medical consultations (p< 0.001), clotting factor regimen (p=0.013), and serological tests (p=0.006). All deaths occurred in those who did not comply with the protocol, and the majority were due to bleeding. Patients who were noncompliant with all protocol items showed the highest mortality (50.0%, 5/10). Treatment at the hemophilia treatment center (OR: 2.388; 95% CI: 1.052– 5.418, p=0.037) was positively and independently associated with compliance with the protocol in multivariate analysis. Conclusion: Noncompliance with the treatment protocol was high. Treatment at a hemophilia treatment center was positively and independently associated with compliance with the protocol, which reinforces the importance of comprehensive care by a multidisciplinary team. Objetivo: Avaliar a associação do sódio sérico no momento da admissão e mortalidade em pacientes idosos admitidos em uma unidade de terapia intensiva de hospital público do Distrito Federal (DF). Métodos: Estudo coorte realizado em pacientes com 65 anos ou mais internados na UTI geral do Hospital Regional de Taguatinga entre agosto/2014 a julho/2016. Hiponatremia foi definida como sódio sério abaixo de 135 mEq/L. Os pacientes foram divididos em dois grupos: com hiponatremia e sem hiponatremia. Resultados: Foram incluídos 113 pacientes. Idade média foi 74±6 anos e o SOFA na admissão na UTI era 10±4. O tempo de internação na UTI apresentou mediana de 14 (IQ25-75%: 2,5-29) dias. Hiponatremia no momento da admissão na UTI foi observada em 40 pacientes (35,3%). A mortalidade em 4 dias foi de 29,2% (N=33) e em 28 dias de 51,3% (N=58). Pacientes com hiponatremia apresentaram maior mortalidade em 4 dias (40,0% vs 21,9%, p=0,02) e 28 dias (60,0% vs 46,6%, p=0,04). Conclusão: Hiponatremia esteve associada a maior mortalidade em 4 e 28 dias em pacientes idosos internados em UTI. [ABSTRACT FROM AUTHOR]

The prevalence of multidrug-resistant (MDR) bacteria and the limited efficacy of current available antibiotics cause every year approximately 700 000 deaths per year. This study aimed to evaluate the anti-inflammatory effect and antibacterial potential of the ibuprofen derivative Methyl 2-(-4-isobutylphenyl)propanoate (MET-IBU). The molecular structure of MET-IBU was confirmed by Nuclear Magnetic Resonance (NMR) and, Attenuated Total Reflectance Fourier Transform Infrared spectroscopy (ATR-FTIR) spectroscopy. Our in vivo study using adult zebrafish model demonstrated that the ibuprofen derivative MET-IBU also possesses anti-inflammatory effect, and in vitro antibacterial activity assays showed that in the association of ampicillin, norfloxacin, and gentamicin with MET-IBU occurred reduction in the minimum inhibitory concentration (MIC) for MDR bacterial strains of Escherichia coli 06 and Staphylococcus aureus 10, indicating a potentiating in the growth inhibition of these pathogenic bacteria. Regarding the strain of Staphylococcus aureus K2068 (overexpressing mepA gene), a potentiation of ethidium bromide was found in the association with MET-IBU, indicating the action of this compound on the efflux pump mechanism present in this strains. This result corroborates the molecular docking study that indicated a high affinity of the MET-IBU with the MepA efflux pump. It was also noticed an antibiotic potentiating activity in the association MET-IBU with norfloxacin against strains of Staphylococcus aureus 1199B (overexpressing norA gene) when compared to the norfloxacin control. This enhanced antibiotic effect of MET-IBU is associated with a second resistance mechanism, which is due to the modification in the topoisomerase enzyme. These results bring attention to the ibuprofen derivative MET-IBU as possible candidate for the development of new options for the treatment of bacterial infections with protective anti-inflammatory action. Guanylate cyclase C (GUCY2C or GC-C) and its ligands, guanylin (GUCA2A or Gn) and uroguanylin (GUCA2B or Ugn), are expressed in intestinal epithelial cells and regulate ion secretion, intestinal barrier function, and epithelial monolayer homeostasis via cGMP-dependent signaling pathways. The aim of this study was to determine whether GC-C and its ligands direct the course of intestinal inflammation. In this article, we show that dextran sodium sulfate (DSS)-induced clinical disease and histological damage to the colonic mucosa were significantly less severe in GC-C-/- mice and moderately reduced in Gn-/- animals. Relative to wild-type controls, GC-C-/- and Gn-/- mice had reduced apoptosis and increased proliferation of intestinal epithelial cells during DSS colitis. Basal and DSS-induced production of resistin-like molecule β (RELMβ) was substantially diminished in GC-C-/- mice. RELMβ is thought to stimulate cytokine production in macrophages in this disease model and, consistent with this, TNF-α and IFN-γ production was minimal in GC-C-/- animals. RELMβ and cytokine levels were similar to wild-type in Gn-/- mice, however. Colonic instillation of recombinant RELMβ by enema into GC-C-/- mice restores sensitivity to DSS-mediated mucosal injury. These findings demonstrate a novel role for GC-C signaling in facilitating mucosal wounding and inflammation, and further suggest that this may be mediated, in part, through control of RELMβ production.
