Automating workplaces that results in a loss of jobs has become a topic of public debate and study but has paid little attention to differential impacts on age groups. This paper applies OECD data on the risk of jobs being automated to Australian longitudinal survey data following 15 years old students over a ten-year period as they enter the job market. Whilst in the early stages of their working lives, these young Australians often worked in jobs projected to be at high risk of automation, they gradually moved into jobs and occupations projected to be less exposed to automation. Young Australians were also less likely to work in jobs at risk of automation than the average Australian. Besides socio-demographic factors, including educational attainment but also domestic wealth, the incidence and kind of computer use at home were associated with working in jobs at a higher or lower risk of automation, as were young workers' type of employment contract and whether young employees changed employers. Whilst suggestions of large-scale job losses owing to automation may be exaggerated, the socio-economic divide that extends to exposure to at risk occupations ought to be a matter of policy concern. Objective: It has recently been discussed whether hearing screening and hearing threshold assessment can accurately be completed using automated ASSR methods for children with auditory neuropathy spectrum disorder (ANSD). Possible causes for the claimed potential failures were investigated here. Design: The study is based on the analysis of stored ASSR raw data. Study sample: This study reviewed raw ASSR data from 274 patients with a total of 5809 individual recordings. Results: Cochlear microphonics (CM) were found in 18 of the 274 patient records. Four of these 18 were obtained from patients with ANSD. One patient with ANSD without click auditory brainstem responses up to 100 dBnHL demonstrated clear ASSR responses from 65 dBnHL upwards. Where click stimulation suggests an auditory nerve defect, narrow-band chirps were shown to evoke ASSR in certain patients. CMs are elicited by narrow-band chirps in the same way as by broadband stimuli. CM residuals as well as a presumed enlarged wave I with absent neural responses, always accompanied by CM, were found as possible causes of misinterpretation at high stimulus levels. Column liquid chromatography plays a vital role in the downstream processing of biopharmaceuticals, where the goal is to extract and purify a target protein from a mixture. In this work, we examine a real‐world ion exchange chromatography process where electrostatic interactions between proteins and the chromatographic medium are the predominant forces being exploited for the separation of the participating species. More specifically, we successfully separate a recombinant dimeric fragment of the von Willebrand factor, used for the half‐life prolongation of certain proteins, from human serum albumin. Compared to the originally conducted experiments, the optimized experiment achieves a higher product purity in less time and with lower consumption of buffer salts, hence being favorable both in economical and ecological terms. Introduction: Bortezomib (BTZ) is a selective and reversible proteasome inhibitor and first line treatment for multiple myeloma (MM). One of the side effects is BTZ-induced peripheral neuropathy (BIPN). Until now there is no biomarker which can predict this side effect and its severity. Neurofilament light chain (NfL) is a neuron specific cytoskeletal protein, of which higher levels can be detected in peripheral blood in case of axon damage. In this study, we aimed to evaluate the relationship between NfL serum levels and characteristics of BIPN. Methods: We performed a first interim analysis of a monocentric, non-randomized, observational clinical trial including 70 patients (DRKS00025422) diagnosed with MM in the inclusion period of June 2021 until March 2022. Two groups of patients—one with ongoing BTZ treatment at the time of recruiting, and one with BTZ treatment in the past—were compared to controls. NfL in serum was analyzed via the ELLA™ device. Results: Both patients with previous and ongoing BTZ treatment had higher serum NfL levels than controls, and patients with ongoing BTZ treatment had higher NfL levels than patients with BTZ treatment in the past. Serum NfL levels correlated with electrophysiological measures of axonal damage in the group with ongoing BTZ treatment. Skilled migration is an important strategy in developed economies seeking to address skills shortages and population ageing. Research on the labour market outcomes of skilled migrants tends to focus on employers' devaluation of skills without considering the role of immigration policy in the migration process. Moreover, there is little understanding of whether efforts to meet employer demands for local qualifications improve labour market outcomes. Drawing on a study on skilled migrants sponsored under the State‐Specific and Regional Migration Scheme in the regional state of South Australia, we explore the shaping of skills and skills recognition in the migration journey, particularly migrants' strategy of reskilling in response to employer demands for local qualifications. Our logistic regressions on the association between the acquisition of Australian qualifications and labour market outcomes reveal only marginal returns to these efforts. Artificial Intelligence (AI) is taking centre stage in economic growth and business operations alike. Public discourse about the practical and ethical implications of AI has mainly focussed on the societal level. There is an emerging knowledge base on AI risks to human rights around data security and privacy concerns. A separate strand of work has highlighted the stresses of working in the gig economy. This prevailing focus on human rights and gig impacts has been at the expense of a closer look at how AI may be reshaping traditional workplace relations and, more specifically, workplace health and safety. To address this gap, we outline a conceptual model for developing an AI Work Health and Safety (WHS) Scorecard as a tool to assess and manage the potential risks and hazards to workers resulting from AI use in a workplace. A qualitative, practice-led research study of AI adopters was used to generate and test a novel list of potential AI risks to worker health and safety. Risks were identified after cross-referencing Australian AI Ethics Principles and Principles of Good Work Design with AI ideation, design and implementation stages captured by the AI Canvas, a framework otherwise used for assessing the commercial potential of AI to a business. The unique contribution of this research is the development of a novel matrix itemising currently known or anticipated risks to the WHS and ethical aspects at each AI adoption stage. Purpose: The Vibrant Soundbridge (VSB) active middle ear implant can be used to treat adults and children with hearing loss. Intraoperative measurements based on auditory brainstem response (ABR) allow the determination of the implant's performance. Two methods for stimulus presentation during ABR measurements via VSB will be compared. Method: ABRs were measured in patients implanted with a VSB using a standard ABR system. The stimulus was transmitted directly to the VSB using a new transmission unit (AcoustiAP, MED-EL) and for comparison via a standard audio processor coupled to a wireless streaming device. The study included 10 subjects with mild-to-profound sensorineural, conductive, or mixed hearing loss. Results: As opposed to the wireless method, the AcoustiAP transmits the ABR stimulus without additional signal processing. Therefore, only minor distortions of the transmitted signal are to be expected. There was no significant difference between the performance of the two methods (mean wave V amplitudes 313 [75.2] vs. 301 [88.2] nVpp). In addition, the new system allowed for a more stable connection to the implant system. Conclusion: The AcoustiAP is a valuable tool for performing ABR measurements in patients during VSB implantation using standard equipment, as it allows for easier and more controlled measurements of the coupling efficiency of the implant. Intraocular metastases from the lung are a common occurrence and an important differential for cytopathologists reading fine needle aspiration biopsies (FNAB). It is a particularly challenging diagnosis when the patient has had no previous diagnosis of lung cancer. We present two cases of intraocular metastases from lung primaries, and we discuss the clinical, radiological, and cytopathological features that help differentiate intraocular metastases of lung primary from other intraocular tumours, in the setting of FNAB. We also discuss the importance of recognising the spectrum of FNAB cases that can be seen specific to an institution, which may vary according to different patient populations. A thorough metastatic workup and ancillary testing, such as IHC or molecular genetics, ensures an accurate diagnosis. The Carina® implant system is a fully implantable active middle ear implant, which can be coupled to various structures in the middle ear, depending on the nature of the hearing loss and the middle ear anatomy. Currently, there is only one method for determining the coupling efficiency of the actuator of this implant system, and this is limited to incus coupling. The proposed method is based on the intraoperative recording and evaluation of auditory brainstem responses (ABRs) while directly stimulating the hearing system via the actuator. The acoustic stimulation was achieved using an optimised broadband chirp stimulus (CE-Chirp®). This study included 10 subjects having moderate to severe sensorineural or mixed hearing loss. The intraoperative ABR measurements show, that it is possible to derive reliable AEP thresholds via the actuator of the implant. The ABR thresholds correlate well with preoperative BC PTA4 thresholds (r = 0.87) while the postoperative OC-direct thresholds (in situ audiogram via the implant) correlate with r = 0.77. The results demonstrated that the direct actuator stimulation allow for reliable intraoperative ABR measurements and that the proposed method can be used to estimate the coupling efficiency of the actuator and facilitate its positioning. Objective: To produce sound-evoked cervical vestibular evoked myogenic potentials (cVEMPs). These are widely used for the diagnosis of peripheral vestibulopathies and disturbances of the sacculocollic reflexes. The main, sensitive, frequency for eliciting cVEMPs has been shown to be around 500 Hz. Design: Narrow-band chirps in different frequency ranges (middle frequencies of 0.5, 1, 2 and 4 kHz) will be used for sequential and quasi-simultaneous stimulation of the cervical vestibular organ. As benchmarks, peak-to-peak p13 and n23 amplitudes were analysed. Study samples: The pilot study used five normal-hearing subjects (mean age 38.6 years) none of whom have balance disorders. Results: From 40 measurements, p13 and n23 could be identified in a total of 34 detected cVEMPs. As expected, amplitudes were highest at 500 Hz, followed by 1, 2 and 4 kHz. Using the quasi-simultaneous stimulation method led to a slight reduction in cVEMP amplitudes, but these remained in the same order according to magnitude: 0.5, 1, 2 and 4 kHz. Conclusion: Sequential and quasi-simultaneous narrow-band chirps are effective for evoking cVEMPs. High amplitudes contribute to fast identification across an extended frequency range (0.5-4 kHz) and also contribute to a reduction in examination time and overall sound exposure. Abstract We compare the cost of maintaining a proposed subsidy for New York's three upstate nuclear power plants with the cost of replacing the plants with renewable technologies from 2016 to 2050. Keeping nuclear operating with subsidy until 2050 is the most expensive option, costing $32.4 billion (2014 USD) over that period in the base business as usual case. The least expensive option is to shut down nuclear today and replace it with onshore wind, saving $7.9 billion. All analyzed renewable scenarios lead to 20.1 to 27.4 Mt CO 2 greater life-cycle emission reductions. In addition, re-investing the cost savings of the renewable scenarios into additional onshore wind increase CO 2 savings up to 32.5 Mt. Graphical abstract Costs and life-cycle CO 2 emissions. Highlights • A novel storage expansion planning tool designs a 100% renewable power system. • Modeling energy autonomy and power reserves strongly impacts storage investments. • The resulting total storage energy/power capacities are up to 3.2/1.6 times larger. • Both the total installed capacities (of storage) and the technology mix are affected. Abstract Energy storage systems (ESS) are a structural solution for the integration of renewable energy systems. To plan the optimal combination of ESS, storage expansion planning approaches are commonly used. They tend to focus on balancing the energy fluctuations from renewable technologies but are usually blind to the need for specific additional services required for dealing with forecast errors. Hence, they underestimate the real operating costs of the future power system and lead to suboptimal investment recommendations. In response, we propose a multi-service storage expansion approach. A linear programming optimization is developed, LEELO, to find the optimal investments in a 100% renewable system (based on solar photovoltaic and wind power) deciding on renewable generators and storage systems. In our formulation, we explicitly model the provisioning of power reserves and energy autonomy as additional services. A case study applies our model to Chile considering four regions and the (existing) hydropower park, for a complete year with an hourly resolution. We systematically assess how our novel multi-service planning differs from conventional energy-based planning in terms of total costs, operation, and investment decisions (with a focus on ESS). Considering power reserves and energy autonomy reveals on average 20% higher costs that otherwise would not be captured in the expansion planning process. Regarding operation, ESS show only slight differences in the two planning models. All ESS participate in the provision of energy. As might be expected, batteries are the main provider of (short-term) power reserves, assisted by pumped-hydro, whereas hydrogen storage is responsible for providing (long-term) energy autonomy. However, the storage investment decisions differ significantly between both models. In our multi-service model, the attained power capacities and energy capacities are up to 1.6 and 3.2 times larger, respectively than in conventional planning. The resulting storage mix changes even more strongly: a general shift towards hydrogen systems is observed. Mainly batteries are substituted, while pumped-hydro capacities stay relatively constant. The trend of the above results is consistent for various scenarios of wind and photovoltaic generation and for sensitivities of service parameters. Electrical energy storage (EES) is a promising flexibility source for prospective low-carbon energy systems. In the last couple of years, many studies for EES capacity planning have been produced. However, these resulted in a very broad range of power and energy capacity requirements for storage, making it difficult for policymakers to identify clear storage planning recommendations. Therefore, we studied 17 recent storage expansion studies pertinent to the U.S., Europe, and Germany. We then systemized the storage requirement per variable renewable energy (VRE) share and generation technology. Our synthesis reveals that with increasing VRE shares, the EES power capacity increases linearly; and the energy capacity, exponentially. Further, by analyzing the outliers, the EES energy requirements can be at least halved. It becomes clear that grids dominated by photovoltaic energy call for more EES, while large shares of wind rely more on transmission capacity. Taking into account the energy mix clarifies—to a large degree—the apparent conflict of the storage requirements between the existing studies. Finally, there might exist a negative bias towards storage because transmission costs are frequently optimistic (by neglecting execution delays and social opposition) and storage can cope with uncertainties, but these issues are rarely acknowledged in the planning process. Young people’s transition from education into the labour market are diverse: sometimes direct from school to work or inactivity, sometimes characterised by stop-starts and combinations of different activities. This paper explores the association between transition pathways of Australian youth and their outcomes, in terms of earnings, perceptions of employment opportunities and debt. It builds on analysis by Fry and Boulton [2013.Prevalence of Transition Pathways in Australia. Canberra: Productivity Commission Staff Working Paper] who identified five pathways most typically taken by 15–24 year old Australians over a 10 year period, including quasi linear transitions from education to work with and without study, combinations of work and study, multiple churning between labour market statuses, and transitions into prolonged periods of inactivity. The present study adds four more years of panel data that have since become available, and compares the labour market outcomes of the different pathways at age 29–38. Earnings and employment perception converged over time, but debt did not. Socio-demographics were most strongly associated with outcomes, but earnings were also greater for churners and for young people in extended education increasingly combined with work. Transitions that combined learning and work seemingly strategically or ‘flexibly’ appeared most rewarding in the medium term, although neither compensated for socio-economic difference. Expansion planning models are often used to support investment decisions in the power sector. Towards the massive insertion of renewable energy sources, expansion planning of energy storage systems (SEP – Storage Expansion Planning) is becoming more popular. However, to date, there is no clear overview of the available SEP models in the literature. To shed light on the existing approaches, this review paper presents a broad classification of SEP, which is used to analyze a database of about 90 publications to identify trends and challenges. The trends we found are that while SEP was born more than four decades ago, only in the last five years increasing research efforts were put into the topic. The planning has evolved from adequacy criteria to broader targets, such as direct costs, mitigation of CO 2 emissions, and renewable integration. The modeling of the network, power system, energy storage systems (ESS), and time resolution are becoming more detailed. Uncertainty is often considered and the solution methods are still very diverse. As outstanding challenges, we found that (1) the large diversity of ESS, in contrast to conventional generation technologies, and (2) the complex lifetime and efficiency functions need to be addressed in the models. (3) Only a high temporal and spatial resolution will allow for dimensioning the challenge of integrating renewables and the role of ESS. (4) Although the value of ESS lies beyond shifting energy in time, current SEP is mostly blind to other system services. (5) Today, many flexibility options are available, but they are often assessed separately. In the same line, although cross-sectorial (power, heat, transport, water) SEP is becoming more frequent, there are many open tasks towards an integrated coordination. The planning of future energy systems will be multi-sectorial and multi-objective, consider the multi-services of ESS, and will inherently require interdisciplinary efforts. [ABSTRACT FROM AUTHOR]

Objective: Active middle ear implants are widely used to treat adults and children with sensorineural, conductive, or mixed hearing loss. Currently, there is no adequate method to determine the performance of active middle ear implant systems. Design: The proposed method is based on measuring the auditory brainstem response while stimulating the hearing system via the active middle ear implant (Vibrant SoundbridgeTM, VSB; MEDEL, Austria). The acoustic stimulation was achieved via an optimised chirp stimulus (CE-Chirp), implemented in the Eclipse system (Interacoustics, Denmark). To compensate for the frequency-specific delays in the VSB system, the underlying model function of the CE-Chirp was adjusted accordingly (VSB-CE-Chirp). Study samples: The study includes 12 subjects having mild to profound sensorineural, conductive or mixed hearing loss. Results: The use of an optimised VSB-CE-Chirp instead of the CE-Chirp increases significantly the ABR wave V amplitudes (1.63 times) and so also increases their identifiability (by 15.2%). On average, wave V could be identified at a 7.5 dB lower stimulation level. Conclusion: The constructed VSB-CE-Chirp stimulus, after it had been transmitted through the VSB system, follows well the shape of the original CE-Chirp. Preliminary measurements in VSB patients demonstrated a significantly improved ABR amplitude with the VSB-CE-Chirp. [ABSTRACT FROM AUTHOR]

Debates about the Global Financial Crisis of 2007 have pointed at institutional and individual-behavioural factors as its causes. Using the British Household Panel Survey, this article highlights marked differences in perceptions of societal and economic fairness among financial services employees in investment or management positions in the United Kingdom and the general working population at the brink of the Global Financial Crisis. Panel data analysis suggests that financial services and occupations did not necessarily attract employees with pro-market attitudes, but that employment in these institutions and occupations made it more likely that employees came to display these perceptions, contributing to the construction of a distinct attitudinal profile of finance employees. Flexibility requirements in prospective energy systems will increase to balance intermittent electricity generation from renewable energies. One option to tackle this problem is electricity storage. Its demand quantification often relies on optimization models for thermal and renewable dispatch and capacity expansion. Within these tools, power plant modeling is typically based on simplified linear programming merit order dispatch (LP) or mixed-integer unit-commitment with economic dispatch (MILP). While the latter is able to capture techno-economic characteristics to a large extent (e.g. ramping or start-up costs) and allows on/off decision of generator units, LP is a simplified method, but superior in computational effort. We present an assessment of how storage expansion is affected by the method of power plant modeling and apply a cost minimizing optimization model, comparing LP with MILP. Moreover, we evaluate the influence of wind and photovoltaic generation shares and vary the granularity of the power plant mix within MILP. The results show that LP underestimates storage demand, as it neglects technical restrictions which affect operating costs, leading to an unrealistically flexible thermal power plant dispatch. Contrarily, storage expansion is higher in MILP. The deviation between both approaches however becomes less pronounced if the share of renewable generation increases. [ABSTRACT FROM AUTHOR]

Asthma is the most common airway chronic disease with treatments aimed mainly to control the symptoms. Adrenergic receptor agonists, corticosteroids and anti-leukotrienes have been used for decades, and the development of more targeted asthma treatments, known as biological therapies, were only recently established. However, due to the complexity of asthma and the limited efficacy as well as the side effects of available treatments, there is an urgent need for a new generation of asthma therapies. The anti-inflammatory and bronchodilatory effects of prostaglandin E2 in asthma are promising, yet complicated by undesirable side effects, such as cough and airway irritation. In this review, we summarize the most important literature on the role of all four E prostanoid (EP) receptors on the lung's immune and structural cells to further dissect the relevance of EP2/EP4 receptors as potential targets for future asthma therapy. As the ophthalmology accreditation system undergoes major changes, training programs must evaluate residents in the 6 core competencies, including appropriately communicating bad news. Although the literature is replete with recommendations for breaking bad news across various non-ophthalmology specialties, no formal training programs exist for ophthalmology. There are many valuable lessons to be learned from our colleagues regarding this important skill. We examine the historic basis for breaking bad news, explore current recommendations among other specialties, and then evaluate a pilot study in breaking bad news for ophthalmology residents. The results of this study are limited by a small number of residents at a single academic center. Future studies from multiple training programs should be conducted to further evaluate the need and efficacy of formal communication skills training in this area, as well as the generalizability of our pilot training program. This paper documents evidence of a ‘normalisation’ of the risk of homelessness experienced by apprentices and trainees in Australia. It draws on a bespoke survey of past and current learners in its Southern state, group and one-to-one interviews with apprentices and trainees, and a review of a government support programme, conducted in 2012 and 2013. The research produced evidence of apprentices and trainees experiencing homelessness in the course of their studies, but also applying preventative strategies to avoid or lessen its impact. At the same time, the study encountered behaviours and judgements reflecting a generalised acceptance among learners and training providers of this risk, which has been reinforced by the marketisation of training provisions in Australia. Air conducted vestibular evoked myogenic potentials (VEMP) can be elicited by various low frequency and intense sound stimuli, mainly clicks or short tone bursts (STB). Chirp stimuli are increasingly used in diagnostic audiological evaluations as an effective means to obtain acoustically evoked responses in narrowed or extended frequency ranges. We hypothesized in this study that band limited chirp stimulation, which covers the main sensitivity range of sound sensitive otolithic afferents (around 500 Hz), might be useful for application in cervical and ocular VEMP to air conduction. For this purpose we designed a chirp stimulus ranging 250-1000 Hz (up chirp). The chirp stimulus was delivered with a stimulus intensity of 100 dB nHL in normal subjects ( n = 10) and patients with otolith involvement (vestibular neuritis) ( n = 6). Amplitudes of the designed chirp ('CW-VEMP-chirp, 250-1000 Hz') were compared with amplitudes of VEMPs evoked by click stimuli (0.1 ms) and a short tone burst (STB, 1-2-1, 8 ms, 500 Hz). CVEMPs and oVEMPs were detectable in 9 of 10 normal individuals. Statistical evaluation in healthy patients revealed significantly larger cVEMP and oVEMP amplitudes for CW-VEMP-chirp (250-1000 Hz) stimuli. CVEMP amplitudes evoked by CW-VEMP-chirp (250-1000 Hz) showed a high stability in comparison with click and STB stimulation. CW-VEMP-chirp (250-1000 Hz) showed abnormal cVEMP and oVEMP amplitudes in patients with vestibular neuritis, with the same properties as click and STB stimulated VEMPs. We conclude that the designed CW-VEMP-chirp (250-1000 Hz) is an effective stimulus which can be further used in VEMP diagnostic. Since a chirp stimulus can be easily varied in its properties, in particular with regard to frequency, this might be a promising tool for further investigations. [ABSTRACT FROM AUTHOR]

A new National Disability Insurance Scheme is being trialled in Australia, following criticism of the fragmented and inequitable nature of existing disability supports (e.g. in the 2009 ‘Shut Out’ report by the National People with Disabilities and Carer Council) and reform recommendations made by the Australian Government's Productivity Commission in 2011. The Insurance Scheme distinguishes between people living with disability who will be eligible for different types of supports: either mainly information about services provided in the community or direct supports and self-managed funding. Analysis of the categories highlights differences in socio-demographics, unmet need for help, and social and labour market inclusion. Unmet need for help was disproportionately prevalent among people with disability when compared to people not living with disability. A higher level of educational capital among people with most severe or profound disability, however, contributed to reduced levels of reported unmet need for some. Overlapping associations between disability, social characteristics and inclusion suggest that nuances in support needs be observed and interventions included that also support people with less severe disability. Objective: A common testing approach for automated ABR detection is to use a sequential test strategy. Repeated testing increases the error probability for a falsely detected response (Type I error rate). To compensate for this effect, the statistically critical test value must be increased with each test step. The aim of the study was to improve response detection by a reduction of the number of test steps using a progressively increasing test step of iteration, defined here as step width. Design: A progressively increasing test step width was tested with and without the table-related testing (adjusting the critical test value to each test step) proposed by Stürzebecher & Cebulla (2013). For this study the same data pool was used. Study sample: The investigation was performed on raw EEG data collected during routine clinical measurement of frequency-specific ASSR for hearing threshold assessment. Results: The reduction of the test step number combined with a progressive test step width led to a significantly improved response detection. In combination with table-related testing a slight but not significant improvement compared to table-related testing alone was revealed. Training and education are acknowledged routes into employment, but they also entail risks of contemporaneous financial loss, and economic and social insecurity. This paper explores the specific risk of homelessness among apprentices and trainees, drawing on a survey conducted in South Australia in 2013. Housing has been largely overlooked by studies of the wellbeing of apprentices and trainees, and by explorations of the drivers of attrition rates that continue to plague Australia's training schemes. The data examined here reveal the high proportion of income that trainees spent on their housing; home moves motivated by the desire to reduce rental or mortgage payments; and a small proportion of learners who experienced periods of homelessness. Closer statistical analysis reveals that apprentices and trainees with past experiences of homelessness were disproportionately likely to be pursuing courses in retail and personal services, or in transport. They were also likely to be receiving Youth Allowance or AUSTUDY payments. We recommend better recording of apprentices' and trainees' housing situations and greater use of administrative data to improve our understanding and reduce the incidence of homelessness among this population [ABSTRACT FROM AUTHOR]

Background: To evaluate the prevalence of BAP1 germline mutations in a series of young patients with uveal melanoma (UM), diagnosed before age 30. Materials and Methods: The study was carried out on 14 young uveal melanoma patients (average age 21.4 years, range 3 months to 29 years). Germline DNA was extracted from peripheral blood. BAP1 sequencing was carried out using direct sequencing of all exons and adjacent intronic sequences. We also tested for germline mutations in additional melanoma-associated candidate genes CDKN2A and CDK4 (exon 4). Results: We identified one patient with a pathogenic mutation (c. 1717delC, p.L573fs*3) in BAP1. This patient was diagnosed with UM at age 18 years and had a family history of a father with UM and a paternal grandfather with cancer of unknown origin. One additional patient had an intronic variant of uncertain significance (c.123-48T > G) in BAP1 while the remaining 12 patients had no alteration. None of the patients had CDKN2A or CDK4 (Exon 4) mutations. Family history was positive for a number of additional malignancies in this series, in particular for cutaneous melanoma, prostate, breast and colon cancers. There were no families with a history of mesothelioma or renal cell carcinoma. Conclusions: This study suggests that a small subset of patients with early onset UM has germline mutation in BAP1. While young patients with UM should be screened for germline BAP1 mutations, our results suggest that there is a need to identify other candidate genes which are responsible for UM in young patients. Background:The phosphoinositide-3 kinase (PI3K) pathway is an attractive therapeutic target. However, difficulty in predicting therapeutic response limits the clinical implementation of PI3K inhibitors. This study evaluates the utility of clinically relevant magnetic resonance imaging (MRI) biomarkers for noninvasively assessing the in vivo response to the dual PI3K/mTOR inhibitor BEZ235 in two ovarian cancer models with differential PI3K pathway activity.Methods:The PI3K signalling activity of TOV-21G and TOV-112D human ovarian cancer cells was investigated in vitro. Cellular and vascular response of the xenografts to BEZ235 treatment (65 mg kg−1, 3 days) was assessed in vivo using diffusion-weighted (DW) and dynamic contrast-enhanced (DCE)-MRI. Micro-computed tomography was performed to investigate changes in vascular morphology.Results:The TOV-21G cells showed higher PI3K signalling activity than TOV-112D cells in vitro and in vivo. Treated TOV-21G xenografts decreased in volume and DW-MRI revealed an increased water diffusivity that was not found in TOV-112D xenografts. Treatment-induced improvement in vascular functionality was detected with DCE-MRI in both models. Changes in vascular morphology were not found.Conclusions:Our results suggest that DW- and DCE-MRI can detect cellular and vascular response to PI3K/mTOR inhibition in vivo. This paper draws on an analysis of one of the UK's most prominent longitudinal datasets, the British Household Panel Survey. The panel was used to study and track the realisation of job aspirations of young people born in the early 1980s and entering the workforce in the 2000s. The study compared the aspirations of those growing up in socio-economically disadvantaged areas with those growing up in non-disadvantaged areas. The analysis confirmed strong differences in the occupational preferences and choices of young people in these two types of areas. Young people in disadvantaged areas more often sought manual occupations, often following their parents' example. They were also more likely to become unemployed, reflecting economic tertiarisation and decreasing availability of manual jobs in those areas. However, the study also found evidence of young people from disadvantaged areas using repeated changes in jobs to achieve employment in higher-level occupations. The research demonstrated that growing up in disadvantaged areas did not prevent the proactive construction of career biographies per se, but it required overcoming more barriers to do so. Objectives: The aim of this study is to evaluate the sensitivity of our hearing screening program. The evaluation was done using a questionnaire for parents of children who participated in the NHS for a targeted time frame of two years. A survey was accomplished to identify children who passed our screening protocol in the newborn period, but who were later identified to have hearing loss. Methods: For the survey a one-year cohort was established in 2008 which included 500 children who received a hearing screening at our Center with the ABR newborn screener, MB11 BERAphone®, two years before. Two hearing questionnaires were chosen for the survey. The LittlEARS questionnaire (MED-EL Medical Electronics GmbH) for investigating the hearing behavior of the children during the first two years of life and a second, custom-developed questionnaire (WÃ?rzburger questionnaire) investigating some aspects which are not included in the LittlEARS tool, such as speech/language development, general development as well as pathological factors that might eventually lead to a temporary hearing loss. Results: Analysis of the WÃ?rzburger questionnaires revealed normal speech development for 92.9% of the children. For 4.7% male and 2.4% female children delayed speech development was reported. Although twice as many males were found, the statistical comparison showed no significant difference according to gender. The results of the LittlEARS questionnaire are identical to those of the WÃ?rzburger questionnaire in 98.3% of the investigated cases and in 1.7% of the cases slightly different results but on borderline: The LittlEAR scores showed normal auditory development for the childrens’ age but the WÃ?rzburger questionnaire results showed delayed speech development. Conclusions: Based on the follow-up analysis and the results from the two questionnaires, no permanent hearing loss was found in any child two years after they passed the newborn hearing screening. We determined the phase diagram of magnesium oxide with finite-temperature density functional theory molecular dynamics simulations up to temperatures and pressures as relevant for the deep interior of super-Earths and in rocky cores of giant planets such as Jupiter. The equation of state data, the Hugoniot, and a ramp compression curve are computed and compared to earlier results from diamond anvil cell and (decaying) shock wave experiments. In addition, the dynamical electrical conductivity and the reflectivity along the experimental Hugoniot curve are calculated in order to characterize electronic structure changes under compression. The structural properties of MgO are identified using pair correlation functions and self-diffusion coefficients. The solid-solid coexistence line is calculated by comparing the free enthalpies of the Bl and the B2 phase. The free energy of the solid phases is determined via thermodynamic relations using the ab initio simulation results and phonon calculations in the harmonic approximation. Our results indicate that the solid B2 phase of MgO does not occur in the interior of the Earth but may play an important role in super-Earths and in rocky planetary cores. [ABSTRACT FROM AUTHOR]

Abstract: Objectives: There has been a rapid growth in using chirp stimuli for evoking auditory brainstem response (ABR). This study aims to investigate the waveform morphology, latency, and amplitude values in newborns and compare them with those evoked by the click in order to determine whether chirp-evoked responses are easier to detect at near-threshold values, as has been proposed, and to assess the usefulness of the chirp stimulus in improving ABR detection thresholds in this population. Methods: Chirp- and click-evoked ABR were measured in 96 normal-hearing newborns (52 males and 44 females) using the MB11 BERAphone®. Only children who passed the hearing screening and were not older than 5 days were included in this study. Two stimulation levels were used: 40dB nHL and 60dB nHL. The stimulation rate was 20.3s–1. The recorded ABR were analyzed based on visual identification of waves I, III, and V, and their amplitude and latency values. Results: Chirp-evoked responses showed clearly larger wave amplitudes compared to those evoked by click stimulus. The gain in amplitude for wave V was 77% for 60dB nHL and 100% for 40dB nHL, for wave III the gain was 22% at 60dB nHL and 46% at 40dB nHL and for wave I the gain was 64% and 100%, respectively. Generally, the gain in amplitude associated with the chirp was greater at the lower stimulus level of 40dB nHL. The wave's latencies were shorter in response to the chirp stimulus. Interpeak intervals differed between click-evoked and chirp-evoked ABR, slightly in the I–III interval and significantly in the I–V interval. Conclusions: Chirp-evoked potentials are distinguished from click-evoked potentials by their significantly larger ABR amplitudes. Therefore they are expected to yield more reliable results and shorter measurement times. It should be noted that, for diagnostic purposes, the typical peak latencies of chirp-evoked ABR are clearly different to those of clicks. [Copyright &y& Elsevier]

The article discusses the study that analyzed proximal impact of being a member of a gang on offending, victimization and attitudinal and experiential outcomes, theorized to be connected to relations between gang membership and offending, particularly in a British sample of young people. Using data from Offending Crime and Justice Survey, the authors found gang onset to have influence on offending, antisocial behavior, drug use, commitment to deviant peers and neutralization techniques.

Objective: Automated auditory response detection is always performed by applying an appropriate statistical test to a sample of stimulus-related epochs of the raw EEG. The often-used sequential test strategy saves time, but the multiple testing increases the probability of falsely detected responses. Therefore, the critical test value must guarantee the specified error probability for the maximum test step number. However, response detection at all lower test step numbers is disadvantaged. We propose calculating the critical test values for each test step number, which correspond exactly to the given error probability. Design: The critical test values for each test step were calculated by the method described by Stürzebecher et al (2005). A table with the test values was implemented with customized software of the Eclipse ASSR system® (Interacoustics, Denmark). Study sample: Table-related testing was performed on a sample of raw EEG data collected during the routine clinical measurement of frequency-specific auditory steady-state responses (ASSR) for hearing threshold assessment. Results: The new test strategy leads to a significantly increased detection rate and a significantly shorter detection time. Objective: The present paper reports the results of a study on a group of newborns whose chirp-evoked auditory brainstem responses (ABR) were recorded at different repetition rates. The study was aimed at finding an optimum repetition rate for the chirp stimulus that facilitates a short response detection time in the frequency domain. Design: Chirp-evoked ABR were recorded at six different stimulation rates between 20/s and 100/s. The stimulation level was 35 dB nHL. The mean harmonic related SNR was calculated, and the response detection time and rate were assessed using a detection algorithm. Study sample: A group of 80 sleeping and 27 awake newborns with normal hearing were included. Results: The highest mean harmonic-related SNR in both groups was found at 60/s. The signal-to-noise ratio (SNR) was significantly smaller for the awake newborns. A significantly shorter detection time can be achieved for both newborn groups at 60/s compared to the other rates tested. Response detection in the group of awake newborns profited more from the change to 60/s compared to the sleeping newborns. Conclusions: It can be expected that with this optimized stimulus paradigm a shorter test time and a higher specificity of the hearing screening can be reached. [ABSTRACT FROM AUTHOR]

Objectives: The diagnosis of Auditory Neuropathy Spectrum Disorder (ANSD) is still challenging: because of the fluctuating hearing thresholds, presence of OAEs and very poor or absent Click-ABR-thresholds. Several studies demonstrated good correlations between ASSR-thresholds and pure-tone-thresholds as well as Click-ABR-thresholds in cochlear hearing loss. However, this is ambiguous in patients with ANSD, because there are often mismatches between mostly better ASSR-thresholds and worse, if at all detectable Click-ABR-thresholds. Aim: The aim of the present study is to evaluate, if an ANSD may be overseen due to the presence of normal ASSR-thresholds and if ASSR-thresholds correlate with pure-tone-thresholds or with the thresholds of cochlear microphonics (CM) in ANSD. Material and methods: 28 patients (4 adults, 24 children) with ANSD were examined. 6 of whom suffered from an aplasia- or hypoplasia of the cochlear nerve. A pure-tone or behavioral audiometry, Click-ABR and ASSR were performed in all patients. 21 patients showed CM in Click-ABR (n=16) or in electrocochleography (EcochG) (n=11). Results: None of the patients with ANSD showed completely normal ASSR-thresholds. The ASSR-thresholds were usually better than expected from Click-ABR results. No correlation of the single frequencies of the pure-tone-thresholds and the frequency specific ASSRthresholds could be demonstrated. However, there were only little differences in the mean or median in all frequencies (mean --1.25--3.5 dB, median --5--2.5dB). There was no correlation between ASSR- and Click-ABR-thresholds. But a correlation between ASSR-thresholds especially at 2 and 4 kHz and Click evoked CM-thresholds in the EcochG was detected (r=0.49--0.69). Conclusions: At present there are only few studies concerning the value of ASSR in the diagnosis of ANSD. None of the patients in the present study showed normal ASSR-thresholds, which implies that an ANSD is not likely to be overseen due to the ASSR results. The ASSR by itself is not sufficient for the diagnosis of ANSD, because it's thresholds are usually better than Click-ABR-thresholds. The analysis of two British cohort studies (of people born in 1958 or 1970) and one British panel study (of people born in the early 1980s) tracked the educational, employment and marital preferences of three generations of young people between ages 16 and 23/26. It found a steady decline in young people achieving their ambitions. Supporting evidence from in-depth interviews with parents and their children suggested that the perceived need, ability and opportunity to disconnect from tradition and to engage in autonomous decision-making had become the main drivers of aspirations. Although this autonomy was greater for current than previous generations, it remained socially inequitable, with parents and their children accepting the widening gap that separates past and present transitions to adulthood. Biological drugs intended for multi-dose application require the presence of antimicrobial preservatives to avoid microbial growth. As the presence of certain preservatives has been reported to increase protein and peptide particle formation, it is essential to choose a preservative compatible with the active pharmaceutical ingredient in addition to its preservation function. Thus, this review describes the current status of the use of antimicrobial preservatives in biologic formulations considering (i) appropriate preservatives for protein and peptide formulations, (ii) their physico-chemical properties, (iii) their in-/compatibilities with other excipients or packaging material, and (iv) their interactions with the biological compound. Background: Development of retinal detachment models in small animals can be difficult and expensive. Here we create and characterize a novel, cone-rich retinal detachment (RD) model in the chick. Methodology/Principal Findings: Retinal detachments were created in chicks between postnatal days 7 and 21 by subretinal injections of either saline (SA) or hyaluronic acid (HA). Injections were performed through a dilated pupil with observation via surgical microscope, using the fellow eye as a control. Immunohistochemical analyses were performed at days 1, 3, 7, 10 and 14 after retinal detachment to evaluate the cellular responses of photoreceptors, Müller glia, microglia and nonastrocytic inner retinal glia (NIRG). Cell proliferation was detected with bromodeoxyuridine (BrdU)-incorporation and by the expression of proliferating cell nuclear antigen (PCNA). Cell death was detected with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL). As in mammalian models of RD, there is shortening of photoreceptor outer segments and mis-trafficking of photoreceptor opsins in areas of RD. Photoreceptor cell death was maximal 1 day after RD, but continued until 14 days after RD. Müller glia up-regulated glial fibriliary acidic protein (GFAP), proliferated, showed interkinetic nuclear migration, and migrated to the subretinal space in areas of detachment. Microglia became reactive; they up-regulated CD45, acquired amoeboid morphology, and migrated toward outer retina in areas of RD. Reactive NIRG cells accumulated in detached areas. Conclusions/Significance: Subretinal injections of SA or HA in the chick eye successfully produced retinal detachments and cellular responses similar to those seen in standard mammalian models. Abstract: The aim of the study was to investigate the molecular genetics of uveal melanoma (UM) metastases and correlate it with disease progression. Twelve pathologically confirmed UM metastases from 11 patients were included. Molecular genetic alterations in chromosomes 3 (including the BAP1 region), 8q, 6p, and 1p were investigated by microsatellite genotyping. Mutations in codon 209 of GNAQ and GNA11 genes were studied by restriction-fragment length polymorphism (RFLP). We identified monosomy of chromosome 3 in tumors from four patients with an average survival of 5 months (range 1–8 months) from time of diagnosis of metastatic disease. In contrast, tumors with either disomy or partial chromosome 3 alterations showed significantly slower metastatic disease progression with an average survival of 69 months (range 40–123 months, p = 0.003). Alterations in chromosomal arms 1p, 6p, and 8q and mutations in either GNAQ or GNA11 showed no association with disease progression. Prominent mononuclear inflammatory infiltrate was observed in tumors from patients with slowly progressive disease. In conclusion, in UM metastases, monosomy 3 is associated with highly aggressive, rapidly progressive disease while disomy or partial change of 3 and prominent mononuclear inflammatory infiltrate in the tumor is associated with better prognosis. Abstract: Objective: At our center, the Maico MB11 BERAphone® device is used for newborn hearing screening based on Auditory Brainstem Responses (ABR). In 2006, an optimized chirp stimulus was implemented in the device to increase the reliability and quality of the screening method. In 2002, an automated response detection algorithm had been implemented. This study analyzes the screening results using the MB11 BERAphone® device with the implemented chirp stimulus and automated response detection method. Methods: The data presented were collected in the well-baby nursery as part of the newborn hearing screening program following a two stage screening protocol. To focus the study on the typical routine screening, data from at-risk babies were not included. Overall, data from 6866 babies (3604 males and 3262 females) screened from March 2006 to April 2011 were analyzed in this study. Results: Out of the 6866 babies screened, 6607 passed bilaterally prior to hospital discharge (defined as 1st stage in this hearing screening program). Therefore, the pre-discharge pass rate of the hearing screening with the MB11 BERAphone® device was 96.2%. The resulting referral rate was 3.8%. The median test time per ear (excluding time for preparation and data reporting) was 28s with a range of 15–112s (5–95th percentile). The number of infants referred for 2nd stage, post-discharge re-screening was 259. Of this group, 71 passed bilaterally and 188 failed the re-screening in one or both ears. Therefore, including both the pre-discharge and post-discharge screening results, the bilateral pass rate was 97.3% and 2.7% were referred for diagnostic evaluation. Diagnostic testing was performed on all of the 188 infants who were referred. Results showed that 47 of these babies had hearing loss. This equates to a positive predictive value for a refer result of 25%. The observed prevalence of hearing impairment in our population was 0.684%. Diagnostic results for 141 of the referred newborns proved that they had normal hearing. That is, 141 out of 6866 newborns had a false-positive result with the MB11 BERAphone® screening. Therefore, the device had a specificity of 97.9%. During the time period of this study, no baby who passed the hearing screening was later found to have hearing impairment, suggesting a sensitivity of 100%. However, due to the limited number of newborns and the short time period after screening for the more recently screened babies, a reliable estimate of the sensitivity cannot be yet made from our program statistics. The theoretical sensitivity of the MB 11 device is 99.9%. Conclusions: The Maico MB11 BERAphone® is a reliable device for auditory brainstem response newborn hearing screening and it provides results within a very short time. Due to the implemented method for automatic detection of ABR, the use of the device does not require an experienced examiner, allowing it to be used by trained technicians in locations outside the department of audiology such as the well-baby nursery. Abstract: In the following study we investigated the utility of molecular genetic testing of the DNA extracted from routinely stained and processed smears from uveal melanoma (UM). Smears from five uveal melanoma cell lines and 12 primary tumors were prepared and stained with Papanicolaou and Romanowsky stains. Genotyping was carried out utilizing 14 microsatellite markers on chromosomes 3, 6 and 8. Mutational screening for alterations in GNAQ and GNA11 genes was carried out by restriction fragment length polymorphism. The results were compared to those obtained through direct sequencing of frozen tumor tissues. High quality DNA was extracted from the stained slides with no difference in the efficiency of DNA extraction between the two staining techniques. The extracted DNA was of adequate quality for genotyping and mutational screening. DNA extracted from approximately 200 tumor cells is sufficient for reproducible testing of allelic imbalances and for studying the common somatic mutations in GNAQ and GNA11 genes. In conclusion, we presented the feasibility of utilizing routinely stained cytology smears from UM for molecular genetic testing. The DNA obtained is of sufficient quality to carry out genotyping for markers on chromosome 3, 6 and 8, as well as screening for somatic mutations in GNAQ and GNA11 genes. [Copyright &y& Elsevier]

In the risk society thesis, most notably forwarded by Ulrich Beck and Anthony Giddens, the labour market plays a key role in individualization processes. While for previous generations, family and personal networks, and also government institutions, were important in providing access to and mobility within the labour market, cohorts entering the labour market since the 1970s and onwards are perceived to be living in a modern ‘risk regime’, requiring each individual to make choices and decisions in relation to a market that no longer accommodates employment based on kinship and friendship. Based on data from 58 qualitative interviews with parents and their adult children, this article examines more closely these purported changes. The study’s main observation is that important changes towards increased perceived individualization have taken place from one generation to the next. While affirming the disjuncture posited by Beck between a ‘collectivized past’ and an ‘individualized present’, this study’s empirical evidence from two generations of individuals indicates that the disjuncture is muddier and more complex than previously understood. Background: A cochlear delay model has previously been proposed for the construction of a chirp stimulus in order to compensate for the temporal dispersion in the auditory periphery. The large intersubject variability in the model data suggests that a chirp constructed from the average model data will not be able to compensate equally well for the temporal dispersion in all normal-hearing subjects. For the recording of the auditory brain stem response (ABR), it has been suggested that the most efficient chirp for generating the largest response amplitude changes in duration with level, indicating that the delay model exhibits a latency change with frequency, which becomes larger at lower levels. Purpose: To investigate in normal-hearing subjects how the ABR varies in response to five different chirps and to study how the efficiency of each chirp changes with stimulus level. Research Design: A click and five chirps of different durations and constructed from the proposed delay model were designed with identical amplitude spectra. The six stimuli were used to record the ABR from 50 normal-hearing test subjects using a quasi-simultaneous stimulation technique at 50 and 30 dB nHL. The ABR recordings were evaluated by the peak-to-trough amplitude and the peak latency. Results: For the test group the following level effect was demonstrated: at 50 dB nHL the largest response amplitude was provided by a shorter chirp, whereas at 30 dB nHL the largest response amplitude was provided by a longer chirp. There is, however, large variability as to which of the five chirps generated the largest response in each individual subject, but at the two levels of stimulation, the best chirps were significantly correlated across the test group. All five chirps generated significantly larger ABRs than the click, but at 30 dB nHL the gain in response amplitude by using the chirps instead of the click was larger than at 50 dB nHL. Conclusions: A chirp that evokes the largest broadband ABRs in normal-hearing subjects changes in duration with level--that is, being relatively short at higher levels (50 dB nHL) and relatively long at lower levels and near the threshold. However, the changes in amplitude in response to chirps of different durations are not very large, and it is therefore uncertain whether the outcome from using such chirps actually would outweigh the instrumental complexity of implementation. Abstract: Spectral domain optical coherence tomography (SD-OCT) was used to image retinal detachments in vivo, in a murine model of retinal detachment (RD). Subretinal injections of hyaluronic acid (Healon) were delivered to the right eye of seventeen 10–20 week-old C57Bl6 mice. Evaluation of the fundus with an operating microscope and fundus photography were performed. In vivo, non-contact, ultra high resolution SD-OCT imaging was performed on day 0, day 1–2, day 5–6 and day 15–16. The retinal morphology at the edge and in the area of maximal RD was evaluated. Eyes were enucleated for histologic analysis. The retinal detachment was confirmed by microscopy in all mice. The extent of the retinal detachment was evaluated by measuring the height of the retinal detachment. The retinal layers, including the photoreceptor layer, were evaluated. Retinal layers appeared indistinct soon after RD (day 1, 5), particularly over areas of maximal detachment. By day 5 and 15 the external limiting membrane was no longer visible and there was increased reflectivity of the photoreceptor layer and undulation of the outer retina in areas of RD on both SD-OCT and histology. The thickness of the outer nuclear layer and photoreceptor outer segments decreased on day 5 and 15. Abstract: Objective: Somatoform disorders (SFD) are defined by symptoms that lack medical explanation. This study examined the type and pattern of patients'' causal attributions using a new semistructured interview technique Methods: The Causal Attributions Interview allows to assess and weigh 15 common explanations of physical symptoms. Attributions given by 79 patients with SFD were compared with those obtained from 187 chronic pain patients. Results: The test–retest reliabilities of the interview-elicited attributions were satisfactory to good. SFD patients attributed most of their symptoms to mental/emotional problems (46.9%) and somatic disease (41.1%), while the pain sample preferred physical overloading/exhaustion (56.1%), daily hastiness/time pressure (41.7%), somatic disease (39.6%), and weather influence (39.0%). On average, SFD patients chose 2.57 and pain patients 3.86 different attributions for each symptom. These numbers were substantially larger than those of initial spontaneous attributions. Correspondence analysis revealed underlying dimensions with three groups labeled “environmental,” somatic,” and “psychological/stress.” While pure environmental attributions were rare (1.1%), somatic factors were chosen for 28.3% of the symptoms, psychological/stress for 22.1%, and the combination of both for 25.6%. Approximately 10% were attributed in a multicausal sense to all three groups. Depression was found to correlate positively with psychological/stress and negatively with somatic attributions. Conclusion: The results do not support the perspective that SFDs generally result from poor acknowledgement of emotional factors. SFD and chronic pain showed distinguishable attributional patterns. AbstractPurpose:The aims of this study are (1) to evaluate the spatial distribution of neovessels and mature vessels in human uveal melanoma tumors and (2) to determine whether vessel maturation is associated with the major indicators for poor prognosis. Methods:Immunohistochemical analyses were performed on human tissue specimens from enucleated eyes (n = 14) to assess total vessels, neovessels, mature vessels, and cell proliferation. Tumor morphology was analyzed by hematoxylin and eosin and modified periodic acid-Schiff (PAS) staining.The spatial distribution of neovessels and mature vessels was analyzed by immunohistochemistry, and correlated with major indicators of poor prognosis (i.e., aggressive PAS patterns, epithelioid cytology, mitotic figures, extraocular extension, anterior tumor location, ciliary body involvement, large tumor size, cell proliferation, and angiogenic activity). Results:Neovesseldensity was greater than mature vessel density in apical (p = 0.17), central (p = 0.036), and peripheral (p = 0.31) regions of the tumors, while mature vessel density was greater than neovessel density in basal areas of the tumor (p = 0.47). This pattern indicated that vessel maturation begins at the base of the tumor and later extends to the peripheral and apical regions. The difference between mature and neovessel densities for the apical (–0.8 ± 1.9) and central areas (–0.8 ± 1.3) of the tumor was significantly higher than the difference obtained for the basal area (0.3 ± 1.6; p = 0.014 and p = 0.012, respectively), indicating a higher density of mature vessels compared to neovessels at the base. Statistical correlations were found between mature vessel density and tumor size (r = 0.48, p = 0.084), cell proliferation (r = 0.62, p = 0.042), and mitotic figures (r = 0.76, p = 0.001). Conclusions:Significant differences exist in the spatial distribution of mature versus neovessels in human uveal melanoma. Vessel maturation is associated with known clinical and pathologic indicators of poor prognosis (e.g., cell proliferation). Antiangiogenic therapy should be considered for the treatment of ocular malignancies; however, the results of this study indicate that blood vessel maturation heterogeneity may limit the efficacy of vessel targeting agents.Copyright © 2009 S. Youth and adolescence are associated with significant changes in a person's life. According to risk theory, the changes experienced by young people today are fundamentally different from those experienced by previous generations and entail a greater degree of uncertainty. In this paper, survey and interview data are used to describe how contemporary risk experiences of young people in Britain differ from those of older generations, highlighting, in particular, their greater frequency and difference in type. Young people are also more likely to be worried about risk, and this remains the case well beyond the years of adolescence. In dealing with these real or anticipated risks, young and adolescent people turn to the traditional sources of family and friends to obtain advice. Using career decisions as a case study, we show that, whereas young people and their parents often share their risk assessments as they take into account each other's decision-making context, understanding is reduced if career paths diverge. Multimodal chromatography is a powerful tool in the downstream processing of biopharmaceuticals. To fully benefit from this technology, an efficient process strategy must be determined beforehand. To facilitate this task, we employ a recent mechanistic model for multimodal chromatography, which takes salt concentration and pH into account, and we present a mathematical framework for the optimization of chromatographic processes. This framework also includes the use of discrete process controls in order to cover a wider range of chromatographic applications. We describe a procedure to numerically solve the resulting nonlinear mixed-integer optimal control problems. We discuss results of computational experiments, covering the cases where one wants to optimize the yield of the product or the batch-cycle time under specified purity requirements. The results indicate that a good separation can be achieved in a two-component system and that both salt concentration and discrete pH play an important role within the purification process.
