Clinicians rely on occlusal contact detection products to identify high contacts and to equilibrate occlusions. Concerns about these products have stimulated numerous investigations into marking reproducibility, accuracy and interpretation, but none have looked at their effects on the occlusion itself. The aim of this study was to assess whether these products alter the occlusion that they purport to measure by determining whether there are differences in the forces and moments experienced by occluding teeth with and without their presence. A matched pair of IPN Portrait 33° molar denture teeth was placed into occlusion with the mandibular tooth supported by a load sensor and the maxillary tooth mounted onto a vertically sliding assembly with a total weight of 15·1N. The three-dimensional force and moment components on the mandibular tooth were measured when the teeth were in direct crown-crown contact (control) and with the products in place. All six products, (Accufilm I, Accufilm II, Hanel Articulating Silk, Rudischhauser Thick and Thin, and T-scan) showed significant ( P < 0·05) differences in forces and moments from control. Conclusion: A formulation based on sodium hyaluronate (NaHYA) was the most promising candidate vehicle for intra-tympanic drug administration regarding conductive hearing loss, inflammatory reactions, and elimination. Objectives: Recent advances in inner ear research support the idea of using the middle ear cavity for drug administration to target the inner ear. This paper presents rheological and safety assessments of three candidate polymer formulations for intra-tympanic drug administration. Method: The formulations were based on sodium carboxymethyl cellulose (NaCMC), sodium hyaluronate (NaHYA), and poloxamer 407 (POL). Rheological studies were performed with a controlled rate instrument of the couette type. Safety studies were performed in guinea pigs subjected to an intra-tympanic injection of the formulations. Hearing function was explored with ABR before and 1, 2, and 3 weeks after the injection. Elimination of the formulations marked with coal was explored with an endoscopic digital camera 1, 2, and 3 weeks after injection. Middle and inner ear morphology was examined with light microscopy 6 days after injection. Results: The results speak in favor of NaHYA, since it did not cause prolonged hearing threshold elevations. The results of the elimination and morphological investigations support the conclusion of NaHYA being the most promising candidate for intra-tympanic administration. [ABSTRACT FROM AUTHOR]

Paeoniflorin (PF) is a principal bioactive component, which exhibits many pharmacological effects, including protection against ischemic injury. This paper aimed to investigate the protective effect of PF both in vivo and in vitro. Middle cerebral artery occlusion (MCAO) was performed on male Sprague-Dawley (SD) rat for 2 h, and different doses of PF or vehicle were administered 2 h after reperfusion. Rats were sacrificed after 7 days treatment of PF/vehicle. PF treatment for 7 days ameliorated MCAO-induced neurological deficit and decreased the infarct area. Further study demonstrated that PF inhibited the over-activation of astrocytes and apoptosis of neurons, and PF promoted up-regulation of neuronal specific marker neuron-specific nuclear (NeuN) and microtubule-associated protein 2 (MAP-2) in brain. Moreover, NMDA-induced neuron apoptosis was employed. The in vitro study revealed that PF treatment protected against NMDA-induced cell apoptosis and neuronal loss via up-regulation of neuronal specific marker NeuN, MAP-2 and Bcl-2 and the down-regulation Bax. Taken together, the present study demonstrates that PF produces its protective effect by inhibiting the over-activation of astrocytes, apoptosis of neurons and up-regulation of neuronal specific marker NeuN, MAP-2, and B-cell lymphoma-2 (Bcl-2), and down-regulation Bax. In this paper, a novel small-molecular apoptotic PET imaging probe, 18F-ML-8 with a malonate motif structure, is presented and discussed. After study, the small tracer that belongs to a member of ApoSense family is proved to be capable of imaging merely apoptotic regions in the CTX treated tumor-bearing mice. The experimental result is further confirmed by in vitro cell binding assays and TUNEL staining assay. As a result, 18F-ML-8 could be used for noninvasive visualization of apoptosis induced by antitumor chemotherapy. [ABSTRACT FROM AUTHOR]

Background: Alpine ski racing is a popular international winter sport that is complex and challenging from physical, technical, and tactical perspectives. Despite the vast amount of scientific literature focusing on this sport, including topical reviews on physiology, ski-snow friction, and injuries, no review has yet addressed the biomechanics of elite alpine ski racers and which factors influence performance. In World Cup events, winning margins are often mere fractions of a second and biomechanics may well be a determining factor in podium place finishes. Objective: The aim of this paper was to systematically review the scientific literature to identify the biomechanical factors that influence the performance of elite alpine ski racers, with an emphasis on slalom, giant slalom, super-G, and downhill events. Methods: Four electronic databases were searched using relevant medical subject headings and key words, with an additional manual search of reference lists, relevant journals, and key authors in the field. Articles were included if they addressed human biomechanics, elite alpine skiing, and performance. Only original research articles published in peer-reviewed journals and in the English language were reviewed. Articles that focused on skiing disciplines other than the four of primary interest were excluded (e.g., mogul, ski-cross and freestyle skiing). The articles subsequently included for review were quality assessed using a modified version of a validated quality assessment checklist. Data on the study population, design, location, and findings relating biomechanics to performance in alpine ski racers were extracted from each article using a standard data extraction form. Results: A total of 12 articles met the inclusion criteria, were reviewed, and scored an average of 69 ± 13 % (range 40-89 %) upon quality assessment. Five of the studies focused on giant slalom, four on slalom, and three on downhill disciplines, although these latter three articles were also relevant to super-G events. Investigations on speed skiing (i.e., downhill and super-G) primarily examined the effect of aerodynamic drag on performance, whereas the others examined turn characteristics, energetic principles, technical and tactical skills, and individual traits of high-performing skiers. The range of biomechanical factors reported to influence performance included energy dissipation and conservation, aerodynamic drag and frictional forces, ground reaction force, turn radius, and trajectory of the skis and/or centre of mass. The biomechanical differences between turn techniques, inter-dependency of turns, and abilities of individuals were also identified as influential factors in skiing performance. In the case of slalom and giant slalom events, performance could be enhanced by steering the skis in such a manner to reduce the ski-snow friction and thereby energy dissipated. This was accomplished by earlier initiation of turns, longer path length and trajectory, earlier and smoother application of ground reaction forces, and carving (rather than skidding). During speed skiing, minimizing the exposed frontal area and positioning the arms close to the body were shown to reduce the energy loss due to aerodynamic drag and thereby decrease run times. In actual races, a consistently good performance (i.e., fast time) on different sections of the course, terrains, and snow conditions was a characteristic feature of winners during technical events because these skiers could maximize gains from their individual strengths and minimize losses from their respective weaknesses. Limitations: Most of the articles reviewed were limited to investigating a relatively small sample size, which is a usual limitation in research on elite athletes. Of further concern was the low number of females studied, representing less than 4 % of all the subjects examined in the articles reviewed. In addition, although overall run time is the ultimate measure of performance in alpine ski racing, several other measures of instantaneous performance were also employed to compare skiers, including the aerodynamic drag coefficient, velocity, section time, time lost per change in elevation, and mechanical energy behaviours, which makes cross-study inferences problematic. Moreover, most studies examined performance through a limited number of gates (i.e., 2-4 gates), presumably because the most commonly used measurement systems can only capture small volumes on a ski field with a reasonable accuracy for positional data. Whether the biomechanical measures defining high instantaneous performance can be maintained throughout an entire race course remains to be determined for both male and female skiers. Conclusions: Effective alpine skiing performance involves the efficient use of potential energy, the ability to minimize ski-snow friction and aerodynamic drag, maintain high velocities, and choose the optimal trajectory. Individual tactics and techniques should also be considered in both training and competition. To achieve better run times, consistency in performance across numerous sections and varied terrains should be emphasized over excellence in individual sections and specific conditions. This paper quantifies and discusses the three-dimensional kinematic and kinetic characteristics of the female softball swing as performed by fourteen female collegiate amateur subjects. The analyses were performed using a three-dimensional computer model. The model was driven kinematically from subject swings data that were recorded with a multi-camera motion analysis system. Each subject used two distinct bats with significantly different inertial properties. Model output included bat trajectories, subject/bat interaction forces and torques, work, and power. These data formed the basis for a detailed analysis and description of fundamental swing kinematic and kinetic quantities. The analyses revealed that the softball swing is a highly coordinated and individual three-dimensional motion and subject-to-subject variations were significant in all kinematic and kinetic quantities. In addition, the potential effects of bat properties on swing mechanics are discussed. The paths of the hands and the centreof- curvature of the bat relative to the horizontal plane appear to be important trajectory characteristics of the swing. Abstract: Ethnopharmacological relevance: The lateral root of Aconitum has been popularly used in traditional Chinese medicine (TMC) known as Fuzi which is beneficial for the treatment of various diseases, such as rheumatism, painful joints, syncope and bronchial asthma. However, it has a potential carditoxicity with a relatively narrow margin of safety. Aim of the study: This paper was designed to explore the mechanisms of Fuzi's toxicity and find out potential tissue-specific biomarkers of toxic effects. Material and methods: A myocardial lipidomics based on ultraperformance lipid chromatography coupled with quadrupole-time-of-flight mass spectrometry (UHPLC/Q-TOF MS) was developed to compare three cardiac lipid extraction methods and investigate the changes of lipids in mice heart of three different dosage groups. In addition, we concurrently inspected the biochemical parameters in plasma, observed the histology of the heart and recorded the electrocardiogram (ECG). Results: The cardiotoxicity of Fuzi was dose-dependent, and the high-dose group obviously manifested the heart damage in histology and a certain degree of arrhythmia. Significant changes of 14 lipid metabolites which primarily involved in phospholipid metabolism, sphingolipid metabolism, saturated fatty acid oxidation and unsaturated fatty acid peroxidation were identified and considered as the potential biomarkers of Fuzi toxicity. Conclusion: The lipidomics approach is helpful to search potential tissue-specific biomarkers and understand the underlying mechanisms of Fuzi toxicity on the heart. [Copyright &y& Elsevier]

Abstract: Arctiin, an important lignan compound in Fructus Arctii, has been reported to possess various kinds of bioactivities. Previous studies on the pharmacokinetic of arctiin after oral administration showed that it had a rapid absorption phase followed by a sharp but lasting disappearance. To gain deep insight into the action mechanism of arctiin, the excretion and metabolism of arctiin in vivo should be further studied. In this paper, three metabolites were isolated and identified in rat feces as (−)-enterolactone (M-1), (−)-arctigenin (M-2) and [(2R,3R)-2-(3′-hydroxybenzyl)-3-(3″,4″-dimethoxybenzyl)-butyrolactone] (M-3). Based on the structures of three metabolites, possible metabolic pathways of arctiin in rats are proposed. At the same time, the cumulative excretion rate of arctiin and its metabolites in rat urine and feces were determined, indicating that arctiin was excreted 19.84% in urine and 1.80% in feces, respectively, enterolactone, the most main metabolite, was excreted 35.80% in feces. These results provide very important information for understanding the metabolism and excretion of arctiin in vivo and speculating its action mechanism, they can provide useful information and reference for further metabolic investigations on arctiin in humans. Abstract: It is difficult to develop injectable sustained delivery systems for herbal medicines because of their composition complexity. Encapsulating various compounds with different physiochemical properties and achieving their synchronized and sustained release seem too hard to realize. In this paper, an injectable nanoparticulate system based on an mPEG–PLGA–mPEG (PELGE) platform was prepared for co-encapsulation and sustained release of four active components (ginkgolides A, B, C and bilobalide) in Ginkgo biloba extract. Different carriers were screened by macrophage uptake experiment for their ability to be long-circulation. Drug loaded nanoparticles were prepared with 10% PEG2000 modified PLGA by a co-precipitation method. The encapsulation efficiency of the total ginkgo terpenes (GT) in the optimal formulation was 78.84±2.06% with a loading dose of 11.90±0.31mg/150mg PELGE. The particles exhibited a spherical shape with a mean diameter of 123.3±44.0nm and zeta potential of −30.86±2.49mV. Sustained and synchronized release of the four components from PELGE nanoparticles was observed both in vitro and in vivo, which was mainly contributed to the long circulation of PEGylated nanoparticles and the slow degradation of PLGA. The half-life time of the four terpenoid compounds were also significantly improved by incorporation into PELGE nanoparticles. The results indicate that a PELGE nanoparticle is a promising carrier system for sustained and synchronized release of herbal medicines containing multiple components. [Copyright &y& Elsevier]

Most infants with long-gap esophageal atresia receive an esophageal replacement with tissue from stomach or colon, because the native esophagus is too short for true primary repair. Tissue-engineered esophageal conducts could present an attractive alternative. In this paper, circular decellularized porcine esophageal scaffold tissues were implanted subcutaneously into Sprague-Dawley rats. Depending on scaffold cross-linking with genipin, glutaraldehyde, and carbodiimide (untreated scaffolds : positive control; bovine pericardium : gold standard), the number of infiltrating fibroblasts, lymphocytes, macrophages, giant cells, and capillaries was determined to quantify the host response after 1, 9, and 30 days. Decellularized esophagus scaffolds were shown to maintain native matrix morphology and extracellular matrix composition. Typical inflammatory reactions were observed in all implants; however, the cellular infiltration was reduced in the genipin group. We conclude that genipin is the most efficient and best tolerated crosslinking agent to attenuate inflammation and to improve the integration of esophageal scaffolds into its surrounding tissue after implantation. Purpose and background: The focus was directed to the study of two of the most lignan-rich food sources: sesame and flaxseeds. Recent epidemiological and experimental evidences suggesting that these foods may improve metabolic functions underlying metabolic syndrome (MetS). Methods: To characterize the effect of these oilseeds on metabolic functions, we conducted an experimental study aimed at preventing adiposity and metabolic imbalance in a mouse model of high-fat diet (HFD)-induced MetS. Statistical analysis was performed by two-way analysis of variance test followed by post hoc Bonferroni analysis. Results: We studied the effect of the oilseeds sesame and flaxseed on metabolic parameters in mice on a HFD. When the HFD was integrated with 20 % of sesame or flaxseed flours, the mice showed a decrease in body fat, already at day 15, from time 0. The size of the adipocytes was smaller in epididymal fat, liver steatosis was inhibited, and insulin sensitivity was higher in mice on the supplemented diets. The supplemented diets also resulted in a significant increase in the serum levels of the lignan metabolites enterodiol and enterolactone compared with the controls. The expression of genes associated with the inflammatory response, glucose metabolism, adipose metabolism and nuclear receptor were altered by the oilseed-supplemented diets. Some of the most abundant lignans in these oilseeds were studied in 3T3-L1 preadipocyte cells and were effective in inhibiting adipocyte differentiation at the minimal dose of 1 nM.
