X-linked dystonia parkinsonism (XDP) is a neurogenetic combined movement disorder involving both parkinsonism and dystonia. Complex, overlapping phenotypes result in difficulties in clinical rating scale assessment. We performed wearable sensor-based analyses in XDP participants to quantitatively characterize disease phenomenology as a potential clinical trial endpoint. Wearable sensor data was collected from 10 symptomatic XDP patients and 3 healthy controls during a standardized examination. Disease severity was assessed with the Unified Parkinson's Disease Rating Scale Part 3 (MDS-UPDRS) and Burke-Fahn-Marsden dystonia scale (BFM). We collected sensor data during the performance of specific MDS-UPDRS/BFM upper- and lower-limb motor tasks, and derived data features suitable to estimate clinical scores using machine learning (ML). XDP patients were at varying stages of disease and clinical severity. ML-based algorithms estimated MDS-UPDRS scores (parkinsonism) and dystonia-specific data features with a high degree of accuracy. Gait spatio-temporal parameters had high discriminatory power in differentiating XDP patients with different MDS-UPDRS scores from controls, XDP freezing of gait, and dystonic/non-dystonic gait. These analyses suggest the feasibility of using wearable sensor data for deriving reliable clinical score estimates associated with both parkinsonian and dystonic features in a complex, combined movement disorder and the utility of motion sensors in quantifying clinical examination. We present linear viscoelastic data with anionically synthesized and critically fractionated polybutadiene (rich in vinyl content) rings having about Z = 22 entanglements. These rings are experimentally as pure as currently possible. They exhibit a power-law stress relaxation G(t) that is well-described by the state-of-the-art fractal loopy globule (FLG) model (power-law exponent of − 3/7). Previously reported data with polystyrene rings, prepared by anionic synthesis in dilute solution and purified by liquid chromatography at the critical condition, having Z = 14 entanglements, showed a power-law G(t) as well. Recent developments with different synthetic methods yielding not so well-characterized rings with a very large number of entanglements (up to 300), suggest that a rubbery plateau emerges in the linear viscoelastic response for Z > 15. Our work confirms the power-law G(t) with the FLG exponent with another chemistry and contributes to the current discussion about different regimes of rheological behavior, indicating that a possible deviation from the power-law FLG type of behavior toward rubbery plateau may occur for Z > 22. To fully capture the experimental G(t) data, the FLG model is complemented by two additional relaxation modes which are attributed to ring-ring (RR) and ring-linear (RL) threading, in accordance with recent reports in the literature. The faster RR mode likely reflects a new mechanism of stress relaxation not described by FLG, and the slower RL mode is attributed to synthetic and material handling imperfections (for example, due to thermal treatment). However, it does not change the punchline of the work: no rubbery plateau for entangled rings with up to 22 entanglements. For many years, wire-guided localization represented the gold standard method for the detection of non-palpable breast lesions, despite the acknowledged limitations. LOCalizer™ is the most recent and promising radiofrequency identification system, characterized by the capability of measuring the distance from the tagged lesion in millimeters and the association of each marker with a unique code. A 45-year-old woman with two lesions suspected of cancer in the same breast was referred to our department. She underwent double localization with LOCalizer™ (Hologic, Santa Carla, CA, USA) of the lumps into opposite quadrants. We performed a mini-invasive and oncologically safe surgical excision through a tunneling approach after a peri-areolar incision. The surgical technique realized with Localizer™ could be an effective method for locating multiple non-palpable breast lesions and is able to offer excellent oncological and cosmetic results, as also confirmed by several reports in the literature. Acute generalized exanthematous pustulosis (AGEP) is a rare, acute, severe cutaneous adverse reaction mainly attributed to drugs, although other triggers, including infections, vaccinations, ingestion of various substances, and spider bites, have also been described. AGEP is characterized by the development of edema and erythema followed by the eruption of multiple punctate, non-follicular, sterile pustules and subsequent desquamation. AGEP typically has a rapid onset and prompt resolution within a few weeks. The differential diagnoses for AGEP are broad and include infectious, inflammatory, and drug-induced etiologies. Diagnosis of AGEP depends on both clinical and histologic criteria, as cases of overlap with other disease processes have been reported. Management includes removal of the offending drug or treatment of the underlying cause, if necessary, and supportive care, as AGEP is a self-limited disease. Lymphoid organs are unusual multicellular tissues: they are densely packed, but the lymphocytes trafficking through them are actively moving. We hypothesize that the intriguing ability of lymphocytes to avoid jamming and clogging is in part attributable to the dynamic shape changes that cells undergo when they move. In this work, we test this hypothesis by investigating an idealized system, namely, the flow of self-propelled, oscillating particles passing through a narrow constriction in two dimensions (2D), using numerical simulations. We found that deformation allows particles with these properties to flow through a narrow constriction in conditions when non-deformable particles would not be able to do so. Such a flowing state requires the amplitude and frequency of oscillations to exceed threshold values. Moreover, a resonance leading to the maximum flow rate was found when the oscillation frequency matched the natural frequency of the particle related to its elastic stiffness. To our knowledge, this phenomenon has not been described previously. Our findings could have important implications for understanding and controlling flow in a variety of systems in addition to lymphoid organs, such as granular flows subjected to vibration. This study reports a set of primeval marine incursions identified in two drill cores, 1PS-06-CE, and 1PS-10-CE, which recovered the Barbalha Formation, Araripe Basin, Brazil. Based on a multi-proxy approach involving stratigraphy, microbiofacies, ichnofossils, and microfossils, three short-lived marine incursions were identified, designated Araripe Marine Incursions (AMI) 1–3. AMI-1 and AMI-2, which occur within the shales of the Batateira Beds (lower part of the Barbalha Formation), were identified by the occurrence of benthonic foraminifera, calcareous nannofossils, dinocysts, and a mass mortality event of non-marine ostracods. AMI-3 was recognized in the upper part of the Barbalha Formation, based on the occurrence of ichnofossils and planktonic foraminifera. The observation of the planktonic foraminifera genus Leupoldina for the first time in the basin indicates early Aptian/early late Aptian age for these deposits, and the first opportunity of correlation with global foraminifera biozonation. Our findings have implications for the breakup of the Gondwana Supercontinent, as these incursions represent the earliest marine-derived flooding events in the inland basins of northeastern Brazil. The aim of this paper is to provide an analytical tool, which might improve models in which the particle-in-a-box approach has been applied and that may be also used when the thin disk approximation could not be longer appropriate. The dispersion velocity is the root-mean-square planetesimal, asteroid, or Kuiper belt object velocity with respect to the local mean circular orbit. This velocity is a function of the object orbital eccentricity and inclination. We calculate a general expression of the dispersion velocity for the planar case in which the object's orbit has no inclination with respect to the local mean circular orbit and for the spatial case in which it has an inclined orbit. Our general expression of the square of the dispersion velocity may be expanded around any value of e for the planar and spatial cases, being in space an exact solution of the orbital inclination i. We expanded our expression around e = 0 with i = e / 2 to study solid accretion rates and collision probabilities. We find that in the whole range of eccentricities and inclinations, our results are lower than solid accretion rates and collision probabilities computed by using the expressions of the dispersion velocity usually adopted in the literature. We apply our expressions of the square of the dispersion velocity expanded around e=0 and up to sixth order in e in our numerical model of planetary formation with planetesimal fragmentation and in our model of the collisional frequency on large asteroids. Our formalism, although generally giving lower values than previous approximations, validates the formerly used estimates for the applications presented here. We report a measurement of the equation of state of superfluid 4 He ( T ∼ 1 K) at negative pressure. A stimulated Brillouin gain spectrometer, used together with an optical interferometer, allows us to probe simultaneously both the compressibility and the density of acoustically driven metastable states of the liquid. Objective: There are few real-world setting studies focused on apremilast effectiveness (i.e., retention rate) in psoriatic arthritis (PsA). The main aim of this retrospective observational study is the assessment of apremilast 3-year retention rate in real-world PsA patients. Moreover, the secondary objective is to report the reasons of apremilast discontinuation and the factors related to treatment persistence. Methods: In fifteen Italian rheumatological referral centers, all PsA consecutive patients who received apremilast were enrolled. Anamnestic data, treatment history, and PsA disease activity (DAPSA) at baseline were recorded. The Kaplan–Meier curve and the Cox analysis computed the apremilast retention rate and treatment persistence-related risk factors. A p-value < 0.05 was considered statistically significant. Results: The 356 enrolled patients (median age 60 [interquartile range IQR 52–67] yrs; male prevalence 42.7%) median observation period was 17 [IQR 7–34] months (7218 patients-months). The apremilast retention rate at 12, 24, and 36 months was, respectively, 85.6%, 73.6%, and 61.8%. The main discontinuation reasons were secondary inefficacy (34% of interruptions), gastro-intestinal intolerance (24%), and primary inefficacy (19%). Age and oligo-articular phenotype were related to treatment persistence (respectively hazard ratio 0.98 IQR 0.96–0.99; p = 0.048 and 0.54 IQR 0.31–0.95; p = 0.03). Conclusion: Almost three-fifths of PsA patients receiving apremilast were still in treatment after 3 years. This study confirmed its effectiveness and safety profile. Apremilast appears as a good treatment choice in all oligo-articular PsA patients and in those ones burdened by relevant comorbidities. The transitioning of nanotechnology from laboratory to industrial-scale manufacturing poses various challenges in nanoparticle realization. From this perspective, beside the conventional synthetic procedure, based on the seed-mediated growth approach, a reshaping thermal strategy has been investigated to improve the control on gold nanorods aspect ratio, with the aim to point out a potential and encouraging way to better manage the scalability and reproducibility of nanoparticles. For this purpose, nanorods covered with CTAB and nanorods enclosed within a silica shell of tuned thickness have been synthesized and submitted to a post-thermal treatment at various temperatures, up to 300 °C for CTAB recovered gold nanorods (AuNR@CTAB), and up to 500 °C for silica-shell embedded gold nanorods (AuNR@SiO2). For AuNR@CTAB, through accurate temperature control, the longitudinal plasmonic band can be moved very close to the transversal one upon slight reduction of their length. Instead, for AuNR@SiO2, owing to the fully inorganic shell, a higher temperature of treatment can be reached leading to the possibility of reshaping the nanorods into spheres without the observation of any by-products. Preoperative planning in primary hip arthroplasty is an essential step that guides the surgeon in restoring the anatomy and biomechanics of the joint. This study aims to evaluate the accuracy of the 2D digital planning, considering cup sizing, stem sizing, and limb length discrepancy. Additionally, we conducted a multivariable analysis of demographic data and comorbidities to find factors influencing preoperative planning.This retrospective study analyzed the planning accuracy in 800 consecutive uncemented primary total hip arthroplasty. We compared the preoperatively planned total hip arthroplasty with postoperative results regarding the planned component size, the implanted size, and the lower limb length restoration. Therefore, we investigated factors influencing planning accuracy: overweight and obesity, sex, age, past medical history, comorbidities, and implant design. All the surgeries were performed in the posterolateral approach by one expert surgeon who did the preoperative planning. The preoperative planning was determined to be (a) exact if the planned and the implanted components were the same size and (b) accurate if exact ± one size. The restoration of postoperative limb length discrepancy was classified into three groups: ± 3 mm, ± 5 mm, and ± 10 mm. This assessment was performed through a digital method 2D based on a standard hip X-ray.This court of 800 implants showed that planning was exact in 60% of the cups and 44% of the stems and was accurate in 94% of the cups and 80% of the stems. The postoperative limb length discrepancy was ± 3 mm in 91% and ± 5 mm in 97%.This study showed preoperative 2D digital planning great precision and reliability, and we demonstrated that it was accurate in 94% of the cups and 80% of the stems. Internalizing problems (e.g., depression, anxiety) and substance use are common among young people and often co-occur. However, youths face myriad barriers to access needed treatment, and existing evidence-based interventions tend to focus on internalizing problems or substance use, rather than both simultaneously. Brief interventions that target both problems may, therefore, be an efficient and accessible resource for alleviating youth difficulties; however, this possibility has been insufficiently evaluated. This systematic review evaluated the intervention characteristics and quality of six studies spanning 2015 to 2019 that examined intervention effects on internalizing and substance use outcomes. Based on independent calculations and author reports (respectively), 3–4 interventions significantly reduced youth internalizing symptoms; 3–5 reduced youth substance use; and 2–3 reduced symptoms in both domains. All six interventions identified substance use as a primary target. Four interventions were administered by interventionists to youths in inpatient, outpatient, primary care, or school settings. The remaining two studies delivered content through voicemail messages or an online design. Interventions ranged from ~ 15 to 240 min. Results highlight the sparsity and heterogeneity of youth-focused brief interventions that have evaluated program effects on both internalizing problems and substance use outcomes, suggesting a clear need for integrated supports that are also designed for accessibility. Future investigations of brief youth-focused interventions should assess program effects on both internalizing and substance use outcomes; examine mechanisms driving the varied efficacy of identified interventions; and create, refine, and test interventions with potential to address co-occurring internalizing problems and substance use in young people. REF-FTP78 is a class IIb medical device present on the market with different trade names and developed for the treatment of gastroesophageal reflux disease (GERD). This medical device is based on polysaccharides from Aloe Barbadensis and fucoidans from brown seaweeds, such as Undaria pinnatifida and Fucus vesiculosus, and aims to exert a protective effect on the esophageal mucosa against the noxious components of refluxate. The present study reports on the efficacy of REF-FTP78 devoting a particular attention to the barrier effect and wound healing properties, combined with antioxidant and anti-inflammatory activities. Film-forming properties and barrier effect were investigated on in vitro reconstructed human esophageal epithelium, through TEER measurement and evaluation of caffeine and Lucifer yellow permeability, and in an ex vivo swine model of esophageal mucosa damage. Antioxidant and anti-inflammatory properties were evaluated in terms of scavenging activity towards DPPH, ABTS and NO radicals and a wound healing assay was carried out to study the influence of the product on cell migration. The obtained results highlighted a significant barrier effect, with a reduction in caffeine penetration equal to 65.3%, the ability to both repair and prevent the damage caused by an acid insult, confirmed by a good transepithelial resistance for the tissue treated with the tested item, and the capacity to promote wound healing. Furthermore, the tested product showed good antioxidant and anti-inflammatory properties in the performed radical scavenging assays. AI systems often need to deal with inconsistent information. For this reason, since the early 2000s, some AI researchers have developed ways to measure the amount of inconsistency in a knowledge base. By now there is a substantial amount of research about various aspects of inconsistency measuring. The problem is that most of this work applies only to knowledge bases formulated as sets of formulas in propositional logic. Hence this work is not really applicable to the way that information is actually stored. The purpose of this paper is to extend inconsistency measuring to real world information. We first define the concept of general information space which encompasses various types of databases and scenarios in AI systems. Then, we show how to transform any general information space to an inconsistency equivalent propositional knowledge base, and finally apply propositional inconsistency measures to find the inconsistency of the general information space. Our method allows for the direct comparison of the inconsistency of different information spaces, even though the data is presented in different ways. We demonstrate the transformation on four general information spaces: a relational database, a graph database, a spatio-temporal database, and a Blocks world scenario, where we apply several inconsistency measures after performing the transformation. Then we review so-called rationality postulates that have been developed for propositional knowledge bases as a way to judge the intuitive properties of these measures. We show that although general information spaces may be nonmonotonic, there is a way to transform the postulates so they can be applied to general information spaces and we show which of the measures satisfy which of the postulates. Hydrocephalus is the most common neurosurgical disorder in children, and cerebrospinal fluid (CSF) diversion with shunt placement is the most commonly performed pediatric neurosurgical procedure. CT is frequently used to evaluate children with suspected CSF shunt malfunction to assess change in ventricular size. Moreover, careful review of the CT images is important to confirm the integrity of the imaged portions of the shunt system. Subtle shunt disruptions can be missed on multiplanar two-dimensional (2-D) CT images, especially when the disruption lies in the plane of imaging. The use of volume-rendered CT images enables radiologists to view the extracranial shunt tubing within the field of view as a three-dimensional (3-D) object. This allows for a rapid and intuitive method of assessing the integrity of the extracranial shunt tubing. The purpose of this pictorial essay is to discuss how volume-rendered CT images can be generated to evaluate CSF shunts in the pediatric population and to provide several examples of their utility in diagnosing shunt disruption. Long-term exposure to air pollution has been associated with the development of some inflammatory processes related to skin. The goal of modern medicine is the development of new products with antiflammatory action deriving from natural sources to improve environmental and economic sustainability. In this study, two different humic acids (HA) were isolated from from lignite (HA-LIG) and composted artichoke wastes (HA-CYN) and characterized by infrared spectrometry, NMR spectroscopy, thermochemolysis-GC/MS, and high-performance size-exclusion chromatography (HPSEC), while their antiflammatory activity was evaluated on HaCaT cells. Spectroscopic results showing the predominance of apolar aliphatic and aromatic components in HA-LIG, whereas HA-CYN revealed a presence of polysaccharides and polyphenolic lignin residues. The HA application on human keratinocyte pre-treated with Urban Dust revealed a general increase of viability suggesting a protective effect of humic matter due to the content of aromatic, phenolic and lignin components. Conversely, the gene expression of IL-6 and IL-1β cytokines indicated a significant decrease after application of HA-LIG, thus exhibiting a greater antiflammatory power than HA-CYN. The specific combination of HA protective hydrophobic components, viable conformational arrangements, and content of bioactive molecules, suggests an innovative applicability of humic matter in dermatology as skin protectors from environmental irritants and as antiflammatory agents. We consider spin systems in the d-dimensional lattice Z d satisfying the so-called strong spatial mixing condition. We show that the relative entropy functional of the corresponding Gibbs measure satisfies a family of inequalities which control the entropy on a given region V ⊂ Z d in terms of a weighted sum of the entropies on blocks A ⊂ V when each A is given an arbitrary nonnegative weight α A . These inequalities generalize the well known logarithmic Sobolev inequality for the Glauber dynamics. Can machine vision step beyond the ocularcentric metaphysics of the Western gaze and the reproduction of racial capital? Paul Virilio argued that machine vision requires no perceptual response or recognition of the world. The computer's series of coded impulses mediate the real autonomously from physical and energetic analogies. This article calls "negative optics" this inhuman mode of machine vision that withdraws from the ocularcentric rules of transparency. Not only negative optics offers an internal critique of ocular metaphysics, but it also defies the equation of value between 0 and 1 s that sustains the universal law of capital. The equation of value needs the nullification of the racialized and gendered body to meet the function of surrogate machines feeding the neo-liberal human subject. Here, however, the negative optics of machine vision withdraws matter from the equation insofar as the AI mismatches of concepts and objects requires the formation of socio-technical assemblages amongst surrogates of all kinds. The intricacies of this surrogacy are discussed in connection to artist Elisa Giardina's video installation entitled, The Cleaning of Emotional Data (2019). This video presents the background to address a surrogate human–machine alliance that steps beyond the human–machine equation of value. By starting from the negativity of the image, the racialized and gendered conditions of techno-social labor under artificial intelligence capitalism show that the equation of value maintains the condition of the zero of blackness, which like matter without form, has no value (On matter beyond the equation of value 2017). Knee osteoarthritis (KOA) is a common degenerative disease in which several treatments and treatment associations have been investigated. This review analyzed the efficacy of the association of photobiomodulation therapy (PBMT) and exercises for people with KOA in randomized controlled clinical trials. PubMed, Scopus, and EMBASE databases were searched using the following terms: "knee osteoarthritis," "laser," "low-level laser," "photobiomodulation," "phototherapy," and "exercise." Seven RCT studies involving humans that examined PBMT treatment in association with were found. Most studies used mainly near-infrared PBMT irradiation, with a fluence ranging from 610 mJ/cm2 to 200 J/cm2, 23.55 J to 2400 J total energy per knee, and number of treatment sessions from 10 to 24. In addition, all the protocols included exercises to increase lower limb muscle strength that were performed alone or in association with other types of exercises. However, only 2 studies, considered as a high quality, showed the additional effect of PBMT (lower doses) on an exercise program (involving warming-up, motor learning, balance coordination and strengthening exercises, and stretching) for improvement of pain and functional capacity in people with KOA. This review demonstrates that there is a controversy on the effects PBMT associated with exercises for pain and functional capacity improvement for people with KOA, because there is a heterogeneity between studies in related to PBMT parameters, as dose, number of therapy sessions and the type of PBMT (either LLLT and HILT), and the exercise protocols proposed. During pregnancy, supply of nutrients and exposure of the mother to environmental factors can influence fetus phenotype, possibly modifying growth of fetal tissues and organs. Few studies inconsistently reported that fetuses exposed to an insufficient energy supply, as those born small for gestational age, may have a reduced volume of uterus and ovaries. A retrospective analysis was performed on ultrasound data performed between 2012 and 2018 in 69 young premenarchal girls, 5 to 9 years of age, attending our endocrine–gynecologic clinic for a suspect of early puberty. Length of pregnancy and birthweight was also retrieved. When corrected for age, and presence of ovarian follicles, ovarian volume was positively (R2 = 0.210; p = 0.001) related to percentiles of birthweight (beta coefficient 0.012; 95% CI, 0.002–0.021). Similarly, uterine volume was positively (R2 = 0.237; p = 0.005) related to percentiles of birthweight (beta coefficient 0.067; 95% CI, 0.021–0.114). Ovarian (p = 0.034) and uterine (p = 0.014) volume was higher in the upper 3rd distribution of birthweight percentiles. In conclusion, development of ovarian and uterine volume increases progressively with the increase of birthweight percentiles. Objectives: The aim of the study was to investigate whether the osteoinductive properties of bone-conditioned medium (BCM) harvested from cortical bone chips within a clinically relevant short-term period can enhance the biologic characteristics of deproteinized bovine bone mineral (DBBM) in vitro. Materials and methods: To assess the biofunctionalization of DBBM, the adhesive, proliferative, and differentiation properties of mesenchymal stromal ST2, pre-osteoblastic MC3T3-E1, and primary bone-derived cells grown on BCM-coated DBBM were examined by crystal violet staining of adherent cells, BrdU ELISA, and qRT-PCR, respectively. Results: BCM extracted within 20 min or 24 h in either Ringer's solution (BCM-RS) or RS mixed with autologous serum (BCM-RS + S) increased the adhesive properties of all three cell types seeded on DBBM. The 20-min BCM-RS preparation appeared more potent than the 24-h preparation. BCM-RS made within 20 min or 24 h had strong pro-proliferative effects on all cell types grown on DBBM. RS + S alone exhibited a considerable pro-proliferative effect, suggesting an impact of the serum on cellular growth. DBBM coated with BCM-RS or BCM-RS + S, made within 20 min or 24 h each, caused a significant induction of osteogenic differentiation marker expression with a higher potency of the BCM-RS + S. Finally, a strong additive effect of fresh bone chips combined with BCM-coated DBBM on the osteogenic differentiation of the three cell types was observed. Conclusions: Altogether, the data strongly support the biofunctionalization of DBBM with BCM extracted within a clinically relevant time window of 20 min. Congenital encephalocele is a very rare entity, with herniation of normal brain or gliotic tissue through a defect in the skull. The objective is to present a newborn child diagnosed with transethmoidal encephaloceles at birth. She developed respiratory problems, feeding difficulties, and failure to thrive since the first days of life and so required early surgery at her 33th day of life, through an endoscopic nasal approach. Technical difficulties encountered, complications, and management are discussed. To the best of our knowledge, this is the first report of endoscopic treatment of transethmoidal encephalocele in a newborn. Lin28 is an evolutionary conserved RNA-binding protein that plays important roles during embryonic development and tumorigenesis. It regulates gene expression through two different post-transcriptional mechanisms. The first one is based on the regulation of miRNA biogenesis, in particular that of the let-7 family, whose expression is suppressed by Lin28. Thus, loss of Lin28 leads to the upregulation of mRNAs that are targets of let-7 species. The second mechanism is based on the direct interaction of Lin28 with a large number of mRNAs, which results in the regulation of their translation. This second mechanism remains poorly understood. To address this issue, we purified high molecular weight complexes containing Lin28a in mouse embryonic stem cells (ESCs). Numerous proteins, co-purified with Lin28a, were identified by proteomic procedures and tested for their possible role in Lin28a-dependent regulation of the mRNA encoding DNA methyltransferase 3a (Dnmt3a). The results show that Lin28a activity is dependent on many proteins, including three helicases and four RNA-binding proteins. The suppression of four of these proteins, namely Ddx3x, Hnrnph1, Hnrnpu or Syncrip, interferes with the binding of Lin28a to the Dnmt3a mRNA, thus suggesting that they are part of an oligomeric ribonucleoprotein complex that is necessary for Lin28a activity. Filter-feeding organisms accumulate xenobiotics and other substances in their tissues. They can be useful as sentinel organisms in biomonitoring of the marine compartment. Bivalve cellular immunity is ensured by phagocytosis and cytotoxic reactions carried out by hemocytes in a network with humoral responses. These can be affected by chemical contaminants in water that can be immunosuppressors also at a low concentration increasing the sensibility to pathogens. This work is an attempt to individuate cellular markers for pollution detection, investigating the effect of methylmercury (CH3HgCl) at different concentrations on the activity and hemocyte morphology of the Mediterranean mussel, Mytilus galloprovincialis. We assessed the effect of three sub-lethal concentrations of the organometal on the cellular morphology, the efficacy of phagocytosis toward yeast cells, the alteration of the lysosomal membrane and the ability to release cytotoxic molecules. The results provide information on the alteration of hemocyte viability, modification of the morphological and cytoskeletal features and besides the cellular spreading, intrinsic ability of motile cells was used as a complementary investigation method. Exposure to the contaminant affected the percentage of phagocytosis and the phagocytosis index. Moreover, morphological and cytoskeleton alteration, caused by the pollutant, leads to reduced ability to incorporate the target and adhere to the substrate and the low ability of cells to retain neutral red could depend on the effects of methylmercury on membrane permeability. These results reinforce the use of the Mediterranean mussel as model for the evaluation of environmental quality in aquatic ecosystems integrating the novel information about hemocyte functions and morphology sensibility to organic mercury. Purpose: The use of cyanoacrylate (CA)-based tissue adhesives for mesh fixation in abdominal hernia repair is increasing due to the fast action and bond strength of these glues. The aim of the present study was to assess tissue changes induced by different CA glues used for mesh fixation in an animal model.Methods: Parietal defects were induced in the abdominal wall of 60 rats and repaired by polyvinylidene fluoride (PVDF) mesh fixation using different CA glues. At 1, 7, 15, and 30 days post-surgery, macroscopic and histopathological studies were performed to evaluate mesh adhesion, the presence of complications and the tissue response.Results: All meshes were successfully fixed without signs of inflammatory reaction, displacement or detachment. In areas where CA adhesives were applied, the acute tissue response was limited and transient. At 7 days post-surgery, collagen fibril production around prosthetic materials was observed, and collagen maturation was achieved at 30 days post-surgery. Good mesh incorporation was detected with all three glues, but the application of Glubran-2 was associated with an early macrophagic response and the early production and maturation of collagen fibrils.Conclusions: Our study confirmed that CA tissue adhesives induced the good incorporation of prosthetic mesh within host tissue with a low incidence of complications and reduced acute tissue reaction. At 30 days post-surgery no signs of mesh disinsertion or migration were observed, the prosthetic mesh adhesion was due to the presence of a dense mature connective tissue rich in type I collagen fibres. Vagus nerve stimulation (VNS) is a surgical treatment available for patients affected by generalized refractory epilepsy. The authors report the case of a 15-year-old girl affected by CHD2-related myoclonic encephalopathy and BLM haploinsufficiency due to a deletion of 15q25.3q26.2 region, who suffered from secondary hemophagocytic lymphohistiocytosis (SHLH) after a VNS wound infection. SHLH has sporadically been described in epileptic patients. Based on indirect evidence that shows immune dysregulation in patients with CHD2 mutations and BLM mutations, we hypothesize that the genetic background of this patient may have played a critical role in the development of the syndrome. The photocatalytic partial oxidation of tyrosol to hydroxytyrosol, the most powerful and expensive natural antioxidant currently known, has been studied in irradiated aqueous suspensions of TiO2 as the photocatalyst. Three TiO2 samples, two commercial (Merck and P25® Evonik) and a home prepared one, have been used bare or surface modified. The influence of surface hydroxylation, surface fluorination, and silver nanoparticles decoration on the selectivity towards hydroxytyrosol has been investigated. The higher the hydroxyl group density, the lower the selectivity. On the other hand, fluorination improved the selectivity towards hydroxytyrosol for all of the samples. In particular, selectivity values up to 10%, i.e. ca. 10 times higher than those obtained with bare sample, were obtained in the presence of fluorinated P25 Evonik TiO2. It is proposed that the higher production of hydroxyl radicals and the lower hydrophilicity of the surface of the fluorinated sample favour hydroxylation of tyrosol and desorption of hydroxytyrosol, respectively, thus hindering the overoxidation of tyrosol. The presence of silver on fluorinated TiO2 was detrimental and it did not significantly improve the selectivity even on bare TiO2. Even if the selectivity hereby reported is relatively low, results show that the surface modification of the photocatalyst and the fluidodynamic optimization of the reacting system could make viable this interesting photocatalytic route of synthesis, which can be used also for the enrichment of some commercial polyphenolic solutions in the very valuable hydroxytyrosol. Introduction and hypothesis: This study compared anatomical and clinical outcomes of traditional McCall culdoplasty versus a modified McCall technique with double ligament suspension (DLS). Methods: This retrospective study presents outcomes of 68 patients who underwent vaginal hysterectomy and vaginal suspension for apical prolapse ≥ stage II according to the POP-Q score system, at, between January 2016 and February 2018. In 34 women vaginal cuff suspension was obtained with traditional McCall culdoplasty (McCall group), while in 34 women we performed a modified McCall, which consists of a double ligament suspension (DLS group), suspending the vaginal cuff to uterosacral ligaments and also to adnexal peduncles. Primary outcome was prolapse recurrence ≥ stage II according to the POP-Q system. Fisher's, Mann-Whitney U and Student's t tests were used for statistical analysis. Results: There were no statistical differences among patients' preoperative characteristics, operative time, blood loss or postoperative complications. Follow-up mean duration was 23.2 ± 6.7 and 22.4 ± 8.7 months in the McCall and DLS group, respectively. Prolapse recurrence occurred in 11 (32.3%) women in the McCall group versus 2 (5.9%) women in the DLS group (p < 0.05): among them, 2 patients (5.9%) in the McCall group and 1 (2.9%) in the DLS group required further treatment. Total vaginal length was 6.1 ± 0.9 cm in the McCall group versus 6.9 ± 0.7 cm in the DLS group (p < 0.001). No statistical difference in quality of life assessment was observed. Conclusions: DLS group patients had better anatomical outcomes and lower recurrence rates than McCall group patients, without increasing operative time or complications. Filling a polymer with halloysite nanotubes is considered a promising strategy to generate nanocomposites with tailored physicochemical properties. We have focused our attention on pluronic block copolymer/halloysite nanocomposites prepared by melt blending. The effect of composition on thermal stability and polymer crystallinity was investigated by thermogravimetry and differential scanning calorimetry. Electron microscopy was used to monitor the nanoparticle distribution in the polymeric matrix. The pluronic thermal stability is reduced by the clay nanoparticles. Concerning the polymer crystallinity, it is slightly decreased even if the melting temperature is lowered by halloysite. Furthermore, waterlogged archeological wood samples are consolidated using the nanotubes/pluronic nanocomposite, and the penetration of the nanocomposites into the lignin channels is confirmed by measurements based on X-ray computed microtomography. This study investigated the application of a novel hybrid artificial intelligence (AI)-based classifier for aiding prediction of the prognosis in patients with chronic hepatitis. Nineteen biomarkers on 155 patients with hepatitis from the University California Irvine Machine Learning repository were used as input data. Weights derived by applying the geometric margin maximisation criterion of a Lagrangian support vector machine (LSVM) were used for selecting the features associated with the highest relative importance towards the required classification, i.e. to predict whether a patient with hepatitis would have survived or died. Thus, the 19 initial features were reduced to the 16 most important prognostic factors and were fed into various AI-based classifiers. Results indicated an overall classification accuracy and area under the receiver operating characteristic curve of 100% for the proposed hybrid algorithm, the LSVM multilayer perceptron (MLP), thus demonstrating its potential for aiding prediction of prognosis in patients with hepatitis in a clinical setting. Among painful disorders, migraine is distinguishable by its chronic pathology and episodic clinical manifestation. Only a small percentage of patients with migraine progress to a chronic form of migraine. Both peripheral and central portions of the trigeminal system are involved in the pathophysiology of migraine pain, as they are involved in the processes of peripheral and central sensitization, alongside various subcortical and cortical brain structures. This review focuses on clinical, neurophysiological, and neuroimaging data underscoring cortical pain processing in migraine. Data obtained from quantitative sensory testings are inconclusive and support the involvement of the peripheral portion of the trigeminovascular system as indirect evidence of peripheral sensitization, solely during the headache phase. The assessment of subjective pain intensity in response to several painful modalities has not been conclusive for the clear state of central sensitization in between migraine attacks but for the subclinical allodynia state that defines the boundary between behavioural responses and an irritable nervous state. Modulation of the brainstem and midbrain pain pathways, in conjunction with the thalamic and thalamocortical pathways, may be critical for the initiation and maintenance of migraine attacks. Several studies using different neuroimaging techniques have demonstrated that brains experiencing migraine undergo plastic changes in both microstructure and macrostructure and in the functioning of cortical networks, which may manifest early in the life of a patient with migraine. The hydrolysis reactions of triazolo–triazole derivatives, which are characterized by the presence of substituents of different electronic character (electron donor or acceptor) on the bicycle, in 0.5 mol·dm−3 NaCl as ionic medium, at 25 °C, have been investigated. Acid–base titrations, volumetric or coulometric, have been performed. The pH range investigated is 0.3–12. UV–vis spectra recorded at various pH values showed that the substitution pattern at the bicycle strongly affects the absorption and emission properties of the triazoles. Current/voltage curves recorded at various pH values also indicate that only a cationic monoprotonated species undergoes irreversible reduction at potential values spanning the range − 0.9 V to − 1.3 V (vs. Ag/AgCl), depending on the pH and on the substituent's nature. Particularly, on the fused-ring N-rich bicycle, with pentafluorophenyl as electron withdrawing group, reduction takes place at a potential of 0.05 V higher than the analogue with aminophenyl electron donor group. The mononuclear complexes formation between Cu(II) and a triazolo–triazole compound has been also highlighted by UV–vis spectra and current/voltage curves recorded at 0.3 < pH < 6. Tapirs are perissodactyl ungulates of the genus Tapirus. The family Tapiridae was more diverse in the past. Genus Tapirus include five living species: T. indicus, T. pinchaque, T. bairdii, T. terrestris, and T. kabomani. Despite all the information available about tapirs, evolutionary relationships among species within the genus are still uncertain. Recent works suggest that T. terrestris may be a species complex. A better understanding of the evolutionary history of this clade is essential to better support conservation strategies for the species of this genus, which are keys in the dynamics of tropical forests in Southeast Asia and Central and South America. Geometric morphometry has been proved to be a useful tool for the study of morphological evolution in mammals, but studies involving cranial geometric morphometry of tapiroids have never been done. We hereby propose landmarks for the study of tapir cranial shape through 2D geometric morphometric technique, including 20 in lateral cranial view (n = 71), 14 in dorsal cranial view (n = 51), and 21 in ventral cranial view (n = 44), followed by PCA multivariate statistical analysis that ordinated specimens from each of the three data groups along the major axis of shape variation. Lateral and dorsal view landmarks proved to be the most diagnostic for the species studied, providing interesting insights and trends on tapiroid cranial evolution. Ventrally, the species analyzed do not differentiate significantly. Aim: Identify key factors driving legume seedlings performance in saline-sodic soils. Methods: Five plots were established in paired sub-humid steppes with saline-sodic soils dominated by Distichlis spicata or Panicum coloratum. In each plot, Lotus tenuis was sown in autumn and individual plants were collected close to nurse plants (dominant species), and in open areas, at the end of the cold and warm seasons. Carbon and nutrient acquisition (through C and O isotopic composition and N and P content measurements) and plant symbiotic functioning (through N fixation by rhizobia and mycorrhizal colonization measurements) were determined. Results: Biomass of legume grown close to a nurse plant was always higher than in open areas. This increase was higher close to P. coloratum and to D. spicata in the cold and warm seasons, respectively. In both cases, L. tenuis improved its nutrient acquisition and symbiosis functioning. N-acquisition and rhizobia efficiency increased in the most facilitated L. tenuis plants in the cold season while P-acquisition was greater in the warm season. Conclusions: Grass-to-legume facilitation in sub-humid steppes with saline-sodic soils was detected in cold and warm seasons, differing between steppes in relationship with the vegetation growing rate and the establishment and functioning of legume-rhizobia-mycorrhiza symbioses. Differentiated thyroid cancer in children is a rare disease, accounting for only 1.4% of all pediatric malignancies. The diagnosis, biological behavior and treatment of differentiated thyroid cancer in children is different from that in adults. While there are many unresolved issues regarding approaches to management of differentiated thyroid cancer in the pediatric population, there is near universal consensus that treatment of this disease, which includes total thyroidectomy, central lymph node dissection at the time of initial surgery in those with nodal metastases, and the possible use of iodine-131 radiotherapy, is best performed by specialists including high-volume endocrine surgeons and experts with experience in calculating and administering radioactive iodine in children, when deemed appropriate. Aims and methods: This multicenter retrospective study aims to evaluate the correlations between Body Weight Loss (BWL), Body Mass Index (BMI) and clinical outcomes (ORR, PFS, and OS) of advanced gastric cancer (aGC) patients treated with second-line ramucirumab-based therapy in a "real-life" setting. Results: From December 2014 to October 2018, 101 consecutive aGC patients progressed to a first-line chemotherapy were treated with ramucirumab alone (10.9%) or in combination with paclitaxel (89.1%). Median BMI was 21.2 kg/m2 and mBWL since first-line treatment commencement was 4.5%. Among 53 patients who underwent primary tumor resection (PTR), 73.6% experienced BWL, while 26.4% did not experience BWL (p = 0.0429). Patients who underwent PTR had a significantly higher probability of experiencing BWL (yes vs no) [OR = 2.35 (95% CI 1.02–5.42), p = 0.0439]. Among the 89 evaluable patients, ORR was 26.9% (95% CI 17.2–40.1). At a median follow-up of 17.3 months, mPFS was 5.4 months (95% CI 3.6–6.8) and mOS was 8.7 months (95% CI 7.3–11.9). In the multivariate analysis, only ECOG-PS and BMI were confirmed independent predictors for shorter PFS [HR = 1.69 (95% CI 1.01–2.82), p = 0.04] [HR = 1.97 (95% CI 1.12–3.46), p = 0.01] and OS [HR = 1.69 (95% CI 1.01–2.83), p = 0.04] [HR = 2.08 (95% CI 1.17–3.70), p = 0.01]. Conclusion: Efficacy of ramucirumab is confirmed in this "real-life" analysis. BWL seems not to have correlations with clinical outcomes in these patients, while BMI and ECOG-PS remain major prognostic factors. In this study, we used long-term passive acoustic monitoring (PAM) to improve knowledge about the use and spatiotemporal distribution of Erignathus barbatus vocalisations. Two autonomous passive acoustic recorders were deployed between August 2014 and July 2015 in the inner and outer parts of the Kongsfjorden (Svalbard Islands, Norway) resulting in 1728 h of data recorded and 17,220 vocalisations detected. We investigated the vocalisation occurrence with respect to the presence of ice in the fjord (sea ice and glacier iceberg) and to the light cycle. The vocalisation rate varied significantly daily and monthly, and a correlation to the light and darkness turnover was shown during the period between the end of February and the first part of April. Bearded seal vocalisations, which started on 12 February and ceased on 22 June, intensified with the ice coverage peak but also continued during ice retreat, coinciding with the mating season. Encephalocele is a rare malformation consisting in herniation of cranial contents through a cranial defect. A transsphenoidal location is uncommon, representing 5% of all basal encephaloceles. The surgical treatment of transsphenoidal encephaloceles is challenging. An optimal approach has not yet been determined, and it varies according to the surgical experience. We report the surgical management of a transsphenoidal encephalocele. The encephalocele and the sellar defect were repaired through a sublabial transsphenoidal microsurgical approach (TSM). Preoperative magnetic resonance imaging (MRI) and computed tomography (CT) scans were crucial for surgical planning. The sublabial transsphenoidal microsurgical approach offered a good and complete exposure of both the sac and the bone defect. Therefore, the congenital defect was successfully repaired with complete resolution of the encephalocele without any surgical or medical complications. Postoperative CT scan and MRI showed the restoration of the bone defect and the recovery of a normal anatomy with herniated structures pushed back into the sella. The described sublabial transsphenoidal microsurgical approach represents a minimally invasive, safe, and effective treatment strategy for transsphenoidal encephalocele. [ABSTRACT FROM AUTHOR]

This article reports on the impact of the Experience Corps® (EC) Baltimore program, an intergenerational, school-based program aimed at improving academic achievement and reducing disruptive school behavior in urban, elementary school students in Kindergarten through third grade (K-3). Teams of adult volunteers aged 60 and older were placed in public schools, serving 15 h or more per week, to perform meaningful and important roles to improve the educational outcomes of children and the health and well-being of volunteers. Findings indicate no significant impact of the EC program on standardized reading or mathematical achievement test scores among children in grades 1-3 exposed to the program. K-1st grade students in EC schools had fewer principal office referrals compared to K-1st grade students in matched control schools during their second year in the EC program; second graders in EC schools had fewer suspensions and expulsions than second graders in non-EC schools during their first year in the EC program. In general, both boys and girls appeared to benefit from the EC program in school behavior. The results suggest that a volunteer engagement program for older adults can be modestly effective for improving selective aspects of classroom behavior among elementary school students in under-resourced, urban schools, but there were no significant improvements in academic achievement. Background: Changes after reimplantation of the autologous bone have been largely described. However, the rate and the extent of resorption in cranial grafts have not been clearly defined. Aim of our study is to evaluate the bone flap resorption (BFR) after cryopreservation.Methods: We retrospectively reviewed 27 patients, aged 18 years or older, subjected to cranioplasty (CP) adopting autologous cryopreserved flap. The BFR was derived from the percentage of decrease in flap volume (BFR%), comparing the first post-operative computed tomography (CT) and the last one available (performed at least 1 year after surgery). We also proposed a semiquantitative scoring system, based on CT, to define a clinically workable BFR classification.Results: After a mean ± SE follow-up of 32.5 ± 2.4 months, the bone flap volume decreased significantly (p < 0.0001). The mean BFR% was 31.7 ± 3.8% and correlated with CT-score (p < 0.001). Three BFR classes were described: mild (14.8% of cases) consisting in minimal bone remodelling, CT-score ≤ 6, mean BFR% = 3.5 ± 0.7%; moderate (51.9% of cases) corresponding to satisfactory cerebral protection, CT-score < 13, mean BFR% = 25.6 ± 2.2%; severe (33.3% of cases) consisting in loss of cerebral protection, CT-score ≥ 13, mean BFR% = 54.2 ± 3.9%. Females had higher BFR% than males (p = 0.022). BFR classes and new reconstructive surgery were not related (p = 0.58).Conclusions: BFR was moderate or severe in 85.2% of re-implanted cryopreserved flaps. The application of photochemistry in larger scales relies on studies in photoreactor engineering. This report investigates the structural parameters of externally irradiated micro-structured photochemical packed bed reactors (PBRs). It reports on the impacts of the size and material of spherical packings on the reactor ability to harness incoming photons. Tubular reactors were built from borosilicate glass tubes of two outer diameters (20 and 30 mm) packed with soda-lime glass spheres of three average diameters (1.0, 3.0 and 6.0 mm), alumina spheres (6.0 mm dia.), and polished iron spheres (5.0 mm dia.). The aqueous-phase oxidation of benzoic acid by oxygen radicals generated by UV-A irradiation of nitrite ions was used as a model homogeneous photochemical reaction. It was observed that increasing the D/dp ratio for the narrower reactors led to improved specific yields (Y˜3.0 mm = 1.05Y˜0), suggesting that intensification is possible in these reactors. Lastly, it was found that glass spheres can be replaced by alumina spheres with considerably little loss in the light-collection efficiency (ca. 20%), indicating an attractive pathway for fixed-bed solar photocatalysis. Aims: To investigate the function of localized retinal areas in highly selected type 1 diabetes mellitus patients (DM1) with no or mild signs of diabetic retinopathy (NO DR and NPDR, respectively) and its correlation with age, diabetes duration and glycemic control.Methods: Multifocal electroretinograms (mfERG) were recorded in 35 eyes of 18 NO DR patients and 38 eyes of 19 NPDR patients. Thirty-one eyes of 17 normal subjects were enrolled as controls. N1-P1 response amplitude densities (RADs) and P1 implicit times (ITs) from isolated (R1: 0°-2.5°, R2: 2.5°-5°, R3: 5°-10°) and combined (R1 + R2, R2 + R3 and R1 + R2 + R3) annular rings and from four retinal sectors (nasal, N; temporal, T; superior, S and inferior, I) with increasing eccentricities up to 10° (S1, S2, S3, S1 + S2, S1 + S2 + S3) were measured. The statistical differences between DM1 groups and controls were tested by ANOVA. The electrophysiological data were correlated with age, duration of diabetes and glycated hemoglobin (HbA1c) level using the Pearson’s test.Results: MfERG RADs, but not ITs, from all isolated and combined rings and sectors up to 10° of foveal eccentricity were statistically different between DM1 groups compared to controls. No significant differences were found between NO DR and NPDR patients. The mfERG abnormalities of the central retinal areas were correlated significantly with age in both DM1 groups and with diabetes duration mainly in NPDR group.Conclusions: In DM1 patients, localized retinal dysfunction, described by reduced mfERG RAD, can be observed also in the absence of clinical signs of DR and it is related to aging. Soluble CD163, soluble CD14 and cellular HIV-1-DNA levels reflect two different aspects of HIV infection: immune activation and the reservoir of infected cells. The aim of this study was to describe their relationships in a cohort of HIV-HCV co-infected patients successfully treated for both HCV and HIV infections. Fifty-five patients were recruited and studied prior to the start of direct-acting antivirals (DAAs) (T0), at week 12 of DAA treatment (T1) and 24 weeks after T0 (T2). The subjects were classified as having undetectable plasma HIV viraemia (UV) or low-level viraemia (LLV) in the 18 months before T2. Plasma levels of sCD163 and of sCD14 were comparable in patients with UV and in subjects with LVL at T0, T1 and T2. The HIV DNA level was positively correlated with LLV but not with sCD163 and sCD14 levels; these two markers of inflammation were positively correlated (p = 0.017). Soluble CD163 and sCD14 decreased over time from T0 to T2 (p = 0.000 and p = 0.034, respectively). In conclusion, the significant decrease in sCD163 and sCD14 levels in patients cured of HCV infection, regardless of the presence of LLV, suggests a main role for HCV in immune activation in HIV-HCV co-infected patients. Dietary restriction, such as low glycemic index diet (LGID), have been successfully used to treat drug-resistant epilepsy. However, if such diet could also counteract antiepileptogenesis is still unclear. Here, we investigated whether the administration of LGID during the latent pre-epileptic period, prevents or delays the appearance of the overt epileptic phenotype. To this aim, we used the Synapsin II knockout (SynIIKO) mouse, a model of temporal lobe epilepsy in which seizures manifest 2–3 months after birth, offering a temporal window in which LGID may affect epileptogenesis. Pregnant SynIIKO mice were fed with either LGID or standard diet during gestation and lactation. Both diets were maintained in weaned mice up to 5 months of age. LGID delayed the seizure onset and induced a reduction of seizures severity only in female SynIIKO mice. In parallel with the epileptic phenotype, high-density multielectrode array recordings revealed a reduction of frequency, amplitude, duration, velocity of propagation and spread of interictal events by LGID in the hippocampus of SynIIKO females, but not mutant males, confirming the gender-specific effect. ELISA-based analysis revealed that LGID increased cortico-hippocampal allopregnanolone (ALLO) levels only in females, while it was unable to affect ALLO plasma concentrations in either sex. The results indicate that the gender-specific interference of LGID with the epileptogenic process can be ascribed to a gender-specific increase in cortical ALLO, a neurosteroid known to strengthen GABAergic transmission. The production of pasture in Ethiopia was simulated by means of a dynamic model. Most of the country is characterized by a tropical monsoon climate with mild temperatures and precipitation mainly concentrated in the June-September period (main rainy season). The production model is driven by solar radiation and takes into account limitations due to relocation, maintenance respiration, conversion to final dry matter, temperature, water stress, and nutrients availability. The model also considers the senescence of grassland which strongly limits the nutritional value of grasses for livestock. The simulation for the 1982-2009 period, performed on gridded daily time series of rainfall and maximum and minimum temperature with a resolution of 0.5°, provided results comparable with values reported in literature. Yearly mean yield in Ethiopia ranged between 1.8 metric ton per hectare (t ha-1) (2002) and 2.6 t ha−1 (1989) of dry matter with values above 2.5 t ha-1 attained in 1983, 1985, 1989, and 2008. The Ethiopian territory has been subdivided in 1494 cells and a frequency distribution of the per-cell yearly mean pasture production has been obtained. This distribution ranges from 0 to 7 t ha-1 and it shows a right skewed distribution and a modal class between 1.5-2 t ha-1. Simulation carried out on long time series for this peculiar tropical environment give rise to as lot of results relevant by the agroecological point of view on space variability of pasture production, main limiting factors (solar radiation, precipitation, temperature), and relevant meteo-climatic cycles affecting pasture production (seasonal and inter yearly variability, ENSO). H1069Q substitution represents the most frequent mutation of the copper transporter ATP7B causing Wilson disease in Caucasian population. ATP7B localizes to the Golgi complex in hepatocytes but moves in response to copper overload to the endo-lysosomal compartment to support copper excretion via bile canaliculi. In heterologous or hepatoma-derived cell lines, overexpressed ATP7B-H1069Q is strongly retained in the ER and fails to move to the post-Golgi sites, resulting in toxic copper accumulation. However, this pathogenic mechanism has never been tested in patients’ hepatocytes, while animal models recapitulating this form of WD are still lacking. To reach this goal, we have reprogrammed skin fibroblasts of homozygous ATP7B-H1069Q patients into induced pluripotent stem cells and differentiated them into hepatocyte-like cells. Surprisingly, in HLCs we found one third of ATP7B-H1069Q localized in the Golgi complex and able to move to the endo-lysosomal compartment upon copper stimulation. However, despite normal mRNA levels, the expression of the mutant protein was only 20% compared to the control because of endoplasmic reticulum-associated degradation. These results pinpoint rapid degradation as the major cause for loss of ATP7B function in H1069Q patients, and thus as the primary target for designing therapeutic strategies to rescue ATP7B-H1069Q function. This longitudinal study described cellular HIV-DNA changes and their correlation with HIV low-level plasma viremia (LLV) in HIV-HCV co-infected patients on successful antiretroviral and anti-HCV therapy by treatment with direct-acting antivirals (DAA). Thirty-nine patients were examined prior to the start of DAA (T0), after week 12 (T1) and 24 weeks (T2) of anti-HCV therapy. Cellular PBMC HIV-DNA was analysed as an absolute value and as the percentage of increase or decrease from T0 to T2. Patients were classified as having undetectable plasma HIV viraemia (UV) or LLV in the year before the start of anti-HCV treatment and within the T0-T2 study period. Thirty-five patients (89.7%) of the 39 subjects enrolled had the same plasma HIV viraemia control in the year before HCV treatment and in the T0-T2 interval. The HIV-DNA value at T0 and at T2 was higher in patients with LLV than in subjects with UV ( p = 0.015 and p = 0.014, respectively). A similar proportion of patients with LLV and UV experienced an increase or decrease of HIV-DNA from T0 to T2. The percentage increase in HIV-DNA value (262.8%) from T0 to T2 was higher compared to the decrease (43.5%) in patients with UV ( p = 0.012), and it was higher compared to the percentage increase in HIV-DNA value reported in subjects with LLV (262.8 versus 49%, p = 0.026). HIV-HCV co-infected patients experienced a multifaceted perturbation of cellular HIV-DNA levels within a 24-week period during anti-HCV treatment; the extent of the phenomenon was greater in subjects with UV.
