Accurate forecasting of wind speed holds significant importance for the economic and social development of humanity. However, existing numerical weather predictions have certain inaccuracies due to various reasons. Therefore, it is highly necessary to perform statistical post-processing on forecasted results. However, traditional linear statistical post-processing methods possess inherent limitations. Hence, in this study, we employed two deep learning methods, namely the convolutional neural network (CNN) and the U-Net neural network, to calibrate the forecast of the Global Ensemble Forecast System (GEFS) in predicting 10-m surface wind speed in Southwest China with a forecast lead time of one to seven days. Two traditional linear statistical post-processing methods, the decaying average method (DAM) and unary linear regression (ULR), are conducted in parallel for comparison. Results show that original GEFS forecasts yield poorer wind speed forecasting performance in the western and eastern Sichuan provinces, the eastern Yunnan province, and within the Guizhou province. All four methods provided certain correction effects on the GEFS wind speed forecasts in the study area, with U-Net demonstrating the best correction performance. After correction using the U-Net, for a 1-day forecast lead time, the proportion of the 10-m U-component of wind with errors less than 0.5 m/s has increased by 46% compared to GEFS. Similarly, for the 10-m V-component of wind, the proportion of errors less than 0.5 m/s has increased by 50% compared to GEFS. Furthermore, we employed the mean square error-based error decomposition method to further diagnose the sources of forecast errors for different prediction models and reveal their calibration capabilities for different error sources. The results indicate that DAM and ULR perform best in correcting the Bias2, while the correction effects of all methods were variable for the distribution with the forecast lead time. More and more researches have been carried out recently on Weigh-In-Motion (WIM) technology for solving the traffic safety problems caused by overload. In this article, we aim to study the measurement accuracy of the WIM system. Based on the electromechanical theory and elastic half-space method, we establish a theoretical model of multi-layer structure to investigate the correlation between the output voltage of the piezoelectric sensor and the applied force. In addition, we performed cyclic and moving load experiments to verify the accuracy of the analytical calculations. The load magnitude identified by this theoretical model matched the experiments very well, which shows that this model is effective for the WIM system. In addition, we proved that the load frequency is an important factor affecting the measurement accuracy of the sensor, which further enables us to design more suitable sensors for certain use scenarios. The grouting quality of tendon ducts is very important for post-tensioning technology in order to protect the prestressing reinforcement from environmental corrosion and to make a smooth stress distribution. Unfortunately, various grouting defects occur in practice, and there is no efficient method to evaluate grouting compactness yet. In this study, a method based on wavelet packet transform (WPT) and Bayes classifier was proposed to evaluate grouting conditions using stress waves generated and received by piezoelectric transducers. Six typical grouting conditions with both partial grouting and cavity defects of different dimensions were experimentally investigated. The WPT was applied to explore the energy of received stress waves at multi-scales. After that, the Bayes classifier was employed to identify the grouting conditions, by taking the traditionally used total energy and the proposed energy vector of WPT components as input, respectively. The experimental results demonstrated that the Bayes classifier input with the energy vector could identify different grouting conditions more accurately. Since the first approval of the anti-CD3 recombinant monoclonal antibody (mAb), muromonab-CD3, a mouse antibody for the prevention of transplant rejection, by the US Food and Drug Administration (FDA) in 1986, mAb therapeutics have become increasingly important to medical care. A wealth of information about mAbs regarding their structure, stability, post-translation modifications, and the relationship between modification and function has been reported. Yet, substantial resources are still required throughout development and commercialization to have appropriate control strategies to maintain consistent product quality, safety, and efficacy. A typical feature of mAbs is charge heterogeneity, which stems from a variety of modifications, including modifications that are common to many mAbs or unique to a specific molecule or process. Charge heterogeneity is highly sensitive to process changes and thus a good indicator of a robust process. It is a high-risk quality attribute that could potentially fail the specification and comparability required for batch disposition. Failure to meet product specifications or comparability can substantially affect clinical development timelines. To mitigate these risks, the general rule is to maintain a comparable charge profile when process changes are inevitably introduced during development and even after commercialization. Otherwise, new peaks or varied levels of acidic and basic species must be justified based on scientific knowledge and clinical experience for a specific molecule. Oryza officinalis Wall ex Watt is one of the most important wild relatives of cultivated rice and exhibits high resistance to many diseases. It has been used as a source of genes for introgression into cultivated rice. However, there are limited genomic resources and little genetic information publicly reported for this species. To better understand the pathways and factors involved in disease resistance and accelerating the process of rice breeding, we carried out a de novo transcriptome sequencing of O. officinalis. In this research, 137,229 contigs were obtained ranging from 200 to 19,214 bp with an N50 of 2331 bp through de novo assembly of leaves, stems and roots in O. officinalis using an Illumina HiSeq 2000 platform. Based on sequence similarity searches against a non-redundant protein database, a total of 88,249 contigs were annotated with gene descriptions and 75,589 transcripts were further assigned to GO terms. Candidate genes for plant-pathogen interaction and plant hormones regulation pathways involved in disease-resistance were identified. Further analyses of gene expression profiles showed that the majority of genes related to disease resistance were all expressed in the three tissues. In addition, there are two kinds of rice bacterial blight-resistant genes in O. officinalis, including two Xa1 genes and three Xa26 genes. All 2 Xa1 genes showed the highest expression level in stem, whereas one of Xa26 was expressed dominantly in leaf and other 2 Xa26 genes displayed low expression level in all three tissues. This transcriptomic database provides an opportunity for identifying the genes involved in disease-resistance and will provide a basis for studying functional genomics of O. officinalis and genetic improvement of cultivated rice in the future. Porosity defects are still a challenging issue in the fusion welding of molybdenum and its alloys due to the pre-existing interior defects associated with the powder metallurgy process. Fiber laser welding of end plug and cladding tube made of nanostructured high-strength molybdenum (NS-Mo) alloy was performed in this work with an emphasis on the role of welding heat input. The distribution and morphology of porosity defects in the welded joints were examined by computed tomography (CT) and scanning electron microscopy (SEM). Preliminary results showed that laser welding of NS-Mo under low heat input significantly reduced the porosity defects in the fusion zone. The results of computed tomography (CT) showed that when the welding heat input decreased from 3600 J/cm (i.e., 1200 W, 0.2 m/min) to 250 J/cm (i.e., 2500 W, 6 m/min), the porosity ratio of the NS-Mo joints declined from 10.7% to 2.1%. Notable porosity defects under high heat input were related to the instability of the keyhole, expansion and the merging of bubbles in the molten pool, among which the instability of the keyhole played the dominant role. The very long chain fatty acid elongase (ELOVL) plays an important role in the synthesis of long-chain polyunsaturated fatty acids (LCPUFA). Previous studies suggest that chicken could be an alternate source of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). In this study, we detected that ELOVL5, which plays a key role in the biosynthesis of omega-3 (n-3) and omega-6 (n-6) polyunsaturated fatty acids (PUFA), was highly expressed in the liver of laying hens and increased rapidly after sexual maturity. Bioinformatic analysis revealed ELOVL fatty acid elongase 5 (ELOVL5) gene as a putative target of miR-218-5p, miR-19a-3p, miR-19b-3p, miR-30a-5p, miR-30b-5p, and miR-30e-5p. We demonstrated estrogen downregulated microRNA (miRNA), and that ELOVL5 is a direct target of miR-218-5p, which was located in intron 14 of the Slit guidance ligand 2 (SLIT2) gene and co-expressed with the host gene. Overall, estrogen enhanced hepatic synthesis of LCPUFA by functioning as a negative regulator of miRNA thereby augmenting the expression of these miRNA target genes, especially ELOVL5, which plays a key role in the biosynthesis of n-3 and n-6 LCPUFA. This study provides a novel model for the use of estrogen in the poultry industry as an inducer of ELOVL5 expression to enhance hepatic n-3 and n-6 LCPUFA synthesis at the post-transcriptional level. Nonylphenol (NP) is an ultimate degradation product of nonylphenol polyethoxylates (NPE) that is primarily used in cleaning and industrial processes. Its widespread use has led to the wide existence of NP in various environmental matrices, such as water, sediment, air and soil. NP can be decreased by biodegradation through the action of microorganisms under aerobic or anaerobic conditions. Half-lives of biodegradation ranged from a few days to almost one hundred days. The degradation rate for NP was influenced by temperature, pH and additions of yeast extracts, surfactants, aluminum sulfate, acetate, pyruvate, lactate, manganese dioxide, ferric chloride, sodium chloride, hydrogen peroxide, heavy metals, and phthalic acid esters. Although NP is present at low concentrations in the environment, as an endocrine disruptor the risks of long-term exposure to low concentrations remain largely unknown. This paper reviews the occurrence of NP in the environment and its aerobic and anaerobic biodegradation in natural environments and sewage treatment plants, which is essential for assessing the potential risk associated with low level exposure to NP and other endocrine disruptors. Simple Summary: In this study, we analyzed the microRNA (miRNA) sequencing libraries of the hen ovary at four different developmental stages (15, 20, 30, and 68 W) and found 209 differently expressed (DE) miRNAs in the six comparisons of the four stages, which might be involved in ovarian growth and functions. In particular, five key miRNAs (gga-miR-2954, gga-miR-6634-5p, gga-miR-449b-5p, gga-miR-449c-3p, and gga-miR449c-5p) targeting 23 predicted genes were obtained in the interaction network and functional enrichment analysis through a combination of DE miRNAs and differentially expressed genes (DEGs), which might be involved in cell differentiation and proliferation, steroid hormone biosynthesis, and angiogenesis in ovarian development. Our findings can contribute to a better understanding of the role of functional miRNAs in ovarian development and the ovulatory cycle during the four developmental stages of hens. It is well-known that multiple functional miRNAs are found in mammals' ovaries, which are linked not only to ovarian development, but also to maturation and apoptosis. However, there is still a lack of knowledge regarding the role of miRNAs in the hen ovary. In the present study, we analyzed the miRNA sequencing libraries of ovaries at the four different developmental stages of hens (15, 20, 30, and 68 W) and a total of 677 known miRNAs and 61 novel miRNAs were identified. In total, 209 of them were differently expressed miRNAs (DE miRNAs) obtained from comparisons of the four stages, including 84 upregulated and 125 downregulated DE miRNAs. Furthermore, the five key DE miRNAs gga-miR-2954, gga-miR-6634-5p, gga-miR-449b-5p, gga-miR-449c-3p, and gga-miR449c-5p were screened using an analysis of the miRNA-mRNA interaction network and functional enrichment annotated in seven significantly enriched pathways, such as endocytosis, lysine degradation, the biosynthesis of amino acids, and the MAPK signaling pathway, which may primarily participate in cell differentiation and proliferation, steroid hormone biosynthesis, and angiogenesis by targeting the related genes. For instance, gga-miR-449 family members were predicted to target 15 genes, including TGFB1, TPM1, TPM3, and CAMKB2, which were reported to regulate follicular growth, selection, and the ovulatory cycle. Taken together, our results illustrate the ovarian miRNA profiles of the four classic developmental stages of hens and highlight the significant role of miRNAs in ovarian development and functions. Few studies have been conducted regarding the biological function and regulation role of gga-miR-221-5p in the liver. We compared the conservation of miR-221-5p among species and investigated the expression pattern of gga-miR-221-5p, validating the direct target genes of gga-miR-221-5p by dual luciferase reporter assay, the biological function of gga-miR-221-5p in the liver was studied by gga-miR-221-5p overexpression and inhibition. Furthermore, we explored the regulation of gga-miR-221-5p and its target genes by treatment with estrogen and estrogen antagonists in vivo and in vitro. The results showed that miR-221-5p was highly conserved among species, expressed in all tested tissues and significantly downregulated in peak-laying hen liver compared to pre-laying hen liver. Gga-miR-221-5p could directly target the expression of elongase of very long chain fatty acids 6 (ELOVL6) and squalene epoxidase (SQLE) genes to affect triglyceride and total cholesterol content in the liver. 17β-estradiol could significantly inhibit the expression of gga-miR-221-5p but promote the expression of ELOVL6 and SQLE genes. In conclusion, the highly conservative gga-miR-221-5p could directly target ELOVL6 and SQLE mRNAs to affect the level of intracellular triglyceride and total cholesterol. Meanwhile, 17β-estradiol could repress the expression of gga-miR-221-5p but increase the expression of ELOVL6 and SQLE, therefore promoting the synthesis of intracellular triglyceride and cholesterol levels in the liver of egg-laying chicken. The fatty acid-binding protein (FABP) gene family, which encodes a group of fatty acid-trafficking molecules that affect cellular functions, has been studied extensively in mammals. However, little is known about the gene structure, expression profile, and regulatory mechanism of the gene family in chickens. In the present study, bioinformatics-based methods were used to identify the family members and investigate their evolutionary history and features of gene structure. Real-time PCR combined with in vivo and in vitro experiments were used to examine the spatiotemporal expression pattern, and explore the regulatory mechanism of FABP genes. The results show that nine members of the FABP gene family, which branched into two clusters and shared a conserved FATTYACIDBP domain, exist in the genome of chickens. Of these, seven FABP genes, including FABP1, FABP3-7, and FABP10 were abundantly expressed in the liver of hens. The expression levels of FABP1, FABP3, and FABP10 were significantly increased, FABP5 and FABP7 were significantly decreased, and FABP4 and FABP6 remained unchanged in hens at the peak laying stage in comparison to those at the pre-laying stage. Transcription of FABP1 and FABP3 were activated by estrogen via estrogen receptor (ER) α, whilst FABP10 was activated by estrogen via ERβ. Meanwhile, the expression of FABP1 was regulated by peroxisome proliferator activated receptor (PPAR) isoforms, of which tested PPARα and PPARβ agonists significantly inhibited the expression of FABP1, while tested PPARγ agonists significantly increased the expression of FABP1, but downregulated it when the concentration of the PPARγ agonist reached 100 nM. The expression of FABP3 was upregulated via tested PPARβ and PPARγ agonists, and the expression of FABP7 was selectively promoted via PPARγ. The expression of FABP10 was activated by all of the three tested PPAR agonists, but the expression of FABP4-6 was not affected by any of the PPAR agonists. In conclusion, members of the FABP gene family in chickens shared similar functional domains, gene structures, and evolutionary histories with mammalian species, but exhibited varying expression profiles and regulatory mechanisms. Accumulating evidence has shown that miR-34a serves as a posttranscriptional regulatory molecule of lipid metabolism in mammals. However, little studies about miR-34a on lipid metabolism in poultry have been reported until now. To gain insight into the biological functions and action mechanisms of miR-34a on hepatic lipid metabolism in poultry, we firstly investigated the expression pattern of miR-34a-5p, a member of miR-34a family, in liver of chicken, and determined its function in hepatocyte lipid metabolism by miR-34a-5p overexpression and inhibition, respectively. We then validated the interaction between miR-34a-5p and its target using dual-luciferase reporter assay, and explored the action mechanism of miR-34a-5p on its target by qPCR and Western blotting. Additionally, we looked into the function of the target gene on hepatocyte lipid metabolism by gain- and loss-of-function experiments. Our results indicated that miR-34a-5p showed a significantly higher expression level in livers in peak-laying hens than that in pre-laying hens. miR-34a-5p could increase the intracellular levels of triglycerides and total cholesterol in hepatocyte. Furthermore, miR-34a-5p functioned by inhibiting the translation of its target gene, long-chain acyl-CoA synthetase 1 (ACSL1), which negatively regulates hepatocyte lipid content. In conclusion, miR-34a-5p could increase intracellular lipid content by reducing the protein level, without influencing mRNA stability of the ACSL1 gene in chickens. [ABSTRACT FROM AUTHOR]

As one of the relatively new titanium (Ti) alloys in the engineering field, β-Ti alloy–Ti55531 has attracted a great deal of attention due to its excellent mechanical properties, while a few research papers on weldability and the post-weld heat treatment (PWHT) process of Ti55531 have been reported. Based on an orthogonal experiment design, the parameters of laser beam welding (LBW) of Ti55531 alloy with a thickness of 2 mm were optimized. Moreover, the influences of welding parameters and PWHT on the microstructures and performance of the laser-welded joint of Ti55531 were analyzed. The results showed that, for microstructures in different zones of as-welded joints of Ti55531: three forms of α phases (i.e., equiaxial αp phase, lamellar αS phase, and αGB phase at grain boundaries) were observed in base metal (BM); in the heat affected zone (HAZ), part of lamellar αS phase had dissolved while equiaxial αp phase had grown; the fusion zone (FZ) mainly consisted of β phase, which presented as coarse columnar crystals. After the PWHT process, the microstructures of the welded joint were changed: in the BM zone, α phase at grain boundary disappeared and lamellar α phase decreased; in the HAZ, the edge of αp phase obviously dissolved; in the FZ, plenty of compact needle-like α phases were observed. The tensile strength of the as-welded joint was about 940 MPa and then increased to 1161 MPa after PWHT, which were 78.4% and 96.8% of that of the original BM respectively.
