The standard Kalman filter and most of its enhancements are typically designed based on the criterion that minimizes the mean squared error, with little discussion of multiple criteria in the positioning and navigation fields. Therefore, a novel data fusion method that takes into account weighted multiple criteria is proposed in this paper, implementing a filtering algorithm based on integrated criteria with different weights determined by a weight adjustment factor. The proposed algorithm and conventional filtering algorithms were utilized for data fusion in GNSS/INS integration. Experiments were conducted using actual data collected from an urban environment. Comparative analysis revealed that, when utilizing the proposed algorithm, the precision of the position, velocity, and attitude of the tested land vehicle could be improved by approximately 24%, 48%, and 35%, respectively. Furthermore, a series of filtering algorithms with different weight adjustment factors was performed to test their influence on the filtering. Simple Summary: Prostate cancer is the second most common cancer among males and is a significant cause of morbidity and mortality. The chemokine C-C motif ligand 2 (CCL2) and its receptor C-C motif chemokine receptor 2 (CCR2) are expressed in prostate cancer but their prognostic value is unclear. This study investigated CCL2 and CCR2 as prognostic biomarkers in locally advanced prostate cancer using the large TROG 03.04 RADAR clinical trial cohort. The results demonstrate that despite being expressed in prostate tumours, CCR2 and CCL2 have no prognostic value in the disease. This study investigated the prognostic value of the chemokine C-C motif ligand 2 (CCL2) and its receptor C-C motif chemokine receptor 2 (CCR2) expression in locally advanced prostate cancer treated with radiotherapy and androgen deprivation using the 10-year outcome data from the TROG 03.04 RADAR clinical trial. CCL2 and CCR2 protein expression in prostate cancer biopsies at the time of diagnosis were quantified by immunohistochemistry and digital quantification. CCR2 protein expression was detected in prostate cancer cells and was associated with prostate-specific antigen serum concentration (p = 0.045). However, neither CCL2 nor CCR2 tissue expression could predict prostate cancer progression, or other clinicopathological parameters including perineural invasion and patient outcome. In serum samples, CCL2 concentration at the time of diagnosis, as assayed by enzyme-linked immunosorbent assay, was significantly higher in patients with prostate cancer compared with benign prostatic hyperplasia (median difference 0.22 ng/mL, 95% CI, 0.17–0.30) (p < 0.0001) and normal controls (median difference 0.13 ng/mL, 95% CI, 0.13–0.17) (p < 0.0001). However, circulating CCL2 was not statistically significant as a predictor of disease progression and patient outcome. In conclusion, this study shows that although CCL2 and CCR2 are expressed in prostate cancer, with an increased level of CCL2 in the serum, neither CCL2 nor CCR2 expression has a clinical prognostic value in locally advanced prostate cancer. Heartbeat star systems have been reported to exhibit two distinct different orbital dynamic evolution processes: apsidal precession (e.g., KIC 4544587) and orbital decay (e.g., KIC 3766353). While experiencing similar dynamic tidal interactions, these binary systems display different dynamical behaviors, which is a puzzling phenomenon. In this work, we deduced a theoretical relation between the timescale of stellar pulsation P pul and orbital periods P orb of heartbeat stars based on the resonance criteria representing the orbital local low-energy configuration. The theoretical relation shows that when the ratio of P orb to P pul is an integer, the specific orbital period is captured in the resonance state, resulting in resonance locking. The resonance criteria are verified by periodograms of the pulsations and orbits of the two systems KIC 4544587 and KIC 3766353 from observations. KIC 4544587 is an apsidal precession heartbeat star with eight observed resonant frequencies available from observations and has an almost integer ratio of P pul / P orb = 67.968 . On the contrary, KIC 3766353 is undergoing the process of orbital shrinkage with only three weak pulsation–orbital resonance frequencies available and shows a non-integer ratio of P pul / P orb = 83.163 . Given the results, the theoretical relation is a potential proxy to distinguish between apsidal precession and orbital decay binary systems. Furthermore, we predict that the orbital period of KIC 3766353 will be reduced to 2.492 days, at which time it will be transformed into apsidal precession. [ABSTRACT FROM AUTHOR]

Simple Summary: SMARCA4-mutated tumors are associated with poor prognosis. However, it is not clear if male and female patients have the same or different prognosis and if additional common mutations within the tumor play a role in prognosis. We have found that male patients had substantially worse prognosis than female patients whose tumor carried a SMARCA4 mutation. In addition, different co-existing mutations including the mutation of TP53, KRAS, CDKN2A, STK11, and Keap1 were associated with differential prognosis. Our study provides helpful insight for prognostic stratification in clinical practice and for understanding the pathogenic mechanism of this unique subtype of malignancy. Background: Whether sex and co-mutations impact prognosis of patients with SMARCA4-mutated (mutSMARCA4) malignancies is not clear. Methods: This cohort included patients from Northern California Kaiser Permanente with next-generation sequencing (NGS) performed from August 2020 to October 2022. We used Cox regression modeling to examine the association between sex and overall survival (OS), adjusting for demographics, performance status, Charlson comorbidity index, receipt of treatment, tumor mutation burden (TMB), and TP53, KRAS, CDKN2A, STK11, and Keap1 co-mutations. Results: Out of 9221 cases with NGS performed, 125 cases (1.4%) had a mutSMARCA4. The most common malignancies with a mutSMARCA4 were non-small cell lung cancer (NSCLC, 35.2%), esophageal and stomach adenocarcinoma (12.8%), and cancer of unknown primary (11.2%). The most common co-mutations were p53 (mutp53, 59.2%), KRAS (mutKRAS, 28.8%), CDKN2A (mutCDKN2A, 31.2%), STK11 (mutSTK11, 12.8%), and Keap1 (mutKeap1, 8.8%) mutations. Male patients had substantially worse OS than female patients both among the entire mutSMARCA4 cohort (HR = 1.71, [95% CI 0.92–3.18]) with a median OS of 3.0 versus 43.3 months (p < 0.001), and among the NSCLC subgroup (HR = 14.2, [95% CI 2.76–73.4]) with a median OS of 2.75 months versus un-estimable (p = 0.02). Among all patients with mutSMARCA4, mutp53 versus wtp53 (HR = 2.12, [95% CI 1.04–4.29]) and mutSTK11 versus wtSTK11 (HR = 2.59, [95% CI 0.87–7.73]) were associated with worse OS. Among the NSCLC subgroup, mutp53 versus wtp53 (HR = 0.35, [0.06–1.97]) and mutKRAS versus wtKRAS (HR = 0.04, [0.003-.45]) were associated with better OS, while mutCDKN2A versus wtCDKN2A (HR = 5.04, [1.12–22.32]), mutSTK11 versus wtSTK11 (HR = 13.10, [95% CI 1.16–148.26]), and mutKeap1 versus wtKeap1 (HR = 5.06, [95% CI 0.89–26.61}) were associated with worse OS. Conclusion: In our cohort of patients with mutSMARCA4, males had substantially worse prognosis than females, while mutTP53, mutKRAS, mutCDKN2A, mutSTK11 and mutKeap1were differentially associated with prognosis among all patients and among the NSCLC subgroup. Our results, if confirmed, could suggest potentially unidentified mechanisms that underly this sex and co-mutation-dependent prognostic disparity among patients whose tumor bears a mutSMARCA4. As a typical application of geodesy, the GNSS/INS (Global Navigation Satellite System and Inertial Navigation System) integrated navigation technique was developed and has been applied for decades. For the integrated systems with multiple sensors, data fusion is one of the key problems. As a well-known data fusion algorithm, the Kalman filter can provide optimal estimates with known parameters of the models and noises. In the literature, however, the data fusion algorithm of the GNSS/INS integrated navigation and positioning systems is performed under a certain norm, and performance of the conventional filtering algorithms are improved only under this fixed and limited frame. The mixed norm-based data fusion algorithm is rarely discussed. In this paper, a mixed norm-based data fusion algorithm is proposed, and the hypothesis test statistics are constructed and adopted based on the chi-square distribution. Using the land vehicle data collected through the multi-GNSS and the IMU (Inertial Measurement Unit), the proposed algorithm is tested and compared with the conventional filtering algorithms. Results show that the influences of the outlying measurements and the uncertain noises are weakened with the proposed data fusion algorithm, and the precision of the estimates is further improved. Tunicate nanocellulose with its unique properties, such as excellent mechanical strength, high crystallinity, and good biodegradability, has potential to be used for the preparation of light management film with tunable transmittance and haze. Herein, we prepared a whole tunicate cellulose film with tunable haze levels, by mixing tunicate microfibrillated cellulose (MFC) and tunicate cellulose nanofibrils (CNF). Then, the obtained whole tunicate cellulose film with updated light management was used to modify the organic solar cell (OSC) substrate, aiming to improve the light utilization efficiency of OSC. Results showed that the dosage of MFC based on the weight of CNF was an important factor to adjust the haze and light transmittance of the prepared cellulose film. When the dosage of MFC was 3 wt.%, the haze of the obtained film increased 74.2% compared to the pure CNF film (39.2%). Moreover, the optimized tunicate cellulose film exhibited excellent mechanical properties (e.g., tensile strength of 168 MPa, toughness of 5.7 MJ/m3) and high thermal stability, which will be beneficial to the workability and durability of OSC. More interestingly, we applied the obtained whole tunicate cellulose film with a high haze (68.3%) and high light transmittance (85.0%) as an additional layer to be adhered to the glass substrate of OSC, and a notable improvement (6.5%) of the power conversion efficiency was achieved. With the use of biodegradable tunicate cellulose, this work provides a simple strategy to enhance light management of the transparent substrate of OSC for improving power conversion efficiency. Objectives: This study aimed to investigate factors associated with sleep quality in healthcare students and to determine whether depressive and anxiety symptoms may explain some of the associations between sleep quality and self-rated health. Study design: This is a cross-sectional study at wave one. Methods: A total of 637 healthcare students were recruited via a stratified random sampling method in Hangzhou, China. The Sleep Quality Questionnaire (SQQ) and the four-item Patient Health Questionnaire (PHQ-4) were used to assess sleep quality, depressive, and anxiety symptoms, respectively. Self-rated health was assessed via a self-developed questionnaire of both physical and psychological health. Structural equation modeling was used to examine the direct and indirect effects of sleep quality on self-rated health through depressive and anxiety symptoms. Results: Students engaged in part-time employment (p = 0.022), who had poor perceived employment prospects (p = 0.009), and who did not participate in recreational sports (p = 0.008) had worse sleep quality. Structural equation modeling revealed a significant total effect of sleep quality on self-rated health (b = 0.592, p < 0.001), a significant direct effect of both sleep quality and depressive and anxiety symptoms on self-rated health (b = 0.277, 95% CI: 0.032–0.522), and a significant indirect effect of sleep quality on self-rated health through depressive and anxiety symptoms (b = 0.315, 95% CI: 0.174–0.457). Conclusions: Depressive and anxiety symptoms partially explain the association between sleep quality and self-rated health. As an optimal estimation method, the Kalman filter is the most frequently-used data fusion strategy in the field of dynamic navigation and positioning. Nevertheless, the abnormal model errors seriously degrade performance of the conventional Kalman filter. The adaptive Kalman filter was put forward to control the influences of model errors. However, the adaptive Kalman filter based on the predicted residuals (innovation vector) requires reliable observation information, and its performance is significantly affected by outliers in the measurements. In this paper, a novel adaptively-robust strategy based on the Mahalanobis distance is proposed to weaken the effects of abnormal model deviations and outliers in the measurements. In the proposed scheme, the judging index is defined based on the Mahalanobis distance, and the adaptively-robust filtering is performed when the observations are reliable, otherwise, the robust filtering is performed based on the robust estimation method. Various experiments with the actual data of GPS/INS integrated navigation systems are implemented to examine validity of the proposed scheme. Results show that both the influences of model deviations and outliers are weakened effectively by using the proposed adaptive robust filtering scheme. Recent theory suggests that both the orienting response and arousal inhibition play roles in the effect of the concealed information test (CIT). However, the neural signatures associated with these two processes remain unclear. To address this issue, participants were motivated to either conceal or reveal crime-related stimulus during CIT while EEG was recorded. By using a temporal principal component analysis, we found that crime-related stimuli produced a larger early P3 than crime-irrelevant stimuli in both the conceal condition and reveal condition. This result suggests that this early P3 reflects an orienting response. In addition, we found that crime-related stimuli elicited a larger frontal negative slow wave than crime-irrelevant stimuli in the conceal condition but not the reveal condition, which suggests that the frontal negative slow wave reflects the arousal inhibition process. Although the 3D structures of active and inactive cannabinoid receptors type 2 (CB2) are available, neither the X-ray crystal nor the cryo-EM structure of CB2-orthosteric ligand-modulator has been resolved, prohibiting the drug discovery and development of CB2 allosteric modulators (AMs). In the present work, we mainly focused on investigating the potential allosteric binding site(s) of CB2. We applied different algorithms or tools to predict the potential allosteric binding sites of CB2 with the existing agonists. Seven potential allosteric sites can be observed for either CB2-CP55940 or CB2-WIN 55,212-2 complex, among which sites B, C, G and K are supported by the reported 3D structures of Class A GPCRs coupled with AMs. Applying our novel algorithm toolset-MCCS, we docked three known AMs of CB2 including Ec2la (C-2), trans-β-caryophyllene (TBC) and cannabidiol (CBD) to each site for further comparisons and quantified the potential binding residues in each allosteric binding site. Sequentially, we selected the most promising binding pose of C-2 in five allosteric sites to conduct the molecular dynamics (MD) simulations. Based on the results of docking studies and MD simulations, we suggest that site H is the most promising allosteric binding site. We plan to conduct bio-assay validations in the future. Sugar beet cyst nematode (SBCN, Heterodera schachtii) is an important nematode that causes significant yield losses of 25–50% or more in most areas of sugar beet production worldwide. Rapid and accurate identification of this species is essential to support decisions on pest management. However, the difference between H. schachtii and other Heterodera spp. based on morphology is a challenging task. In the present study, a SCAR-PCR assay was developed to identify and differentiate H. schachtii in infected root and soil samples. H. schachtii-species-specific SCAR-PCR primers OPA06-HsF and OPA06-HsR were designed from the randomly amplified polymorphic DNA (RAPD) marker amplified with random primer OPA06. The developed primers specifically amplify a 922-bp fragment from the target populations but did not amplify DNA from non-target cyst nematodes including Heterodera, Globodera, Cactodera, and other related species tested in this study. The sensitivity detection indicated that 5 × 10−4 of a single cyst, 1/320 of a single second-stage juvenile (J2), or 10 pg of genomic DNA could be detected. The assay accurately identifies the different stages of H. schachtii in sugar beet and oilseed rape roots as well as a single J2 in 10 g of soil. Finally, the SCAR-PCR assay detected H. schachtii in seven samples out of the fifteen field samples. The assay will not only be useful for differentiating H. schachtii from mixed populations of Heterodera spp. but also for effective detection of the species directly from infested samples. With the ongoing advancement in oil exploration, microemulsion, as an innovative oil displacement method, has garnered considerable attention owing to its exceptional physicochemical properties in enhancing crude oil recovery. As such, this study initially delineates the fundamental concepts, classifications, formation mechanisms, advantages, and preparation methodologies of microemulsions. Subsequently, it introduces the selection criteria for microemulsion components, followed by an elucidation of the characterization methods for microemulsions based on these criteria. Furthermore, it examines the factors influencing the efficacy of microemulsions in enhancing oil recovery through two distinct methods, along with the effects of various formulation microemulsions under laboratory and oilfield conditions. There is increasing evidence that nerve growth factor (NGF) and its receptors, the neurotrophic receptor tyrosine kinase 1 (NTRK1/TrkA), the common neurotrophin receptor (NGFR/p75NTR) and the membrane receptor sortilin, participate in cancer growth. In melanoma, there have been some reports suggesting that NGF, TrkA and p75NTR are dysregulated, but the expression of the NGF precursor (proNGF) and its membrane receptor sortilin is unknown. In this study, we investigated the expression of NGF, proNGF, TrkA, p75NTR and sortilin by immunohistochemistry in a series of human tissue samples (n = 100), including non-cancerous nevi (n = 20), primary melanomas (n = 40), lymph node metastases (n = 20) and distant metastases (n = 20). Immunostaining was digitally quantified and revealed NGF and proNGF were expressed in all nevi and primary melanomas, and that the level of expression decreased from primary tumors to melanoma metastases (p = 0.0179 and p < 0.0001, respectively). Interestingly, TrkA protein expression was high in nevi and thin primary tumors but was strongly downregulated in thick primary tumors (p < 0.0001) and metastases (p < 0.0001). While p75NTR and sortilin were both expressed in most nevi and melanomas, there was no significant difference in expression between them. Together, these results pointed to a downregulation of NGF/ProNGF and TrkA in melanoma, and thus did not provide evidence to support the use of anti-proNGF/NGF or anti-TrkA therapies in advanced and metastatic forms of melanoma. Electrohydrodynamic (EHD) direct-writing has been widely used to fabricate micro/nanofibers that can serve as a building block in tissue engineering scaffolds. However, the application of EHD direct-writing in tissue engineering is limited by the lack of fundamental knowledge in the correlations among the process parameters, the fiber surface roughness, and the cell adhesion performance. Without a standardized experimental setting and the quantitative database, inconsistent results have been reported. Here, we quantitatively investigate the process–structure–property relationships as the first step towards a better understanding of the EHD direct-writing technology for tissue engineering. Polycaprolactone (PCL) solution is used as a model ink material, and human mesenchymal stem cells (hMSCs) are used to study cell adhesion on PCL fibers. We investigate the different jetting modes defined by the applied voltage, the feed rate, and the nozzle–collector distance. The quantitative effects of process parameters on the fiber surface roughness and the cell adhesion performance are experimentally determined. The quantitative process–structure–property relationships revealed in this study provide guidelines for controlling the surface roughness and the cell adhesion performance of EHD direct-written fibers. Lysozyme plays a crucial role in the innate immune response against bacterial phagocytosis by hydrolyzing the peptidoglycan layer of the bacterial cell wall. In this study, we characterized a goose-type lysozyme gene (TrLysG) in Japanese pufferfish. It is made up of an ORF of 573 bp that encodes a polypeptide of 190 amino acids. TrLysG includes a characteristic bacterial soluble lytic transglycosylase (SLT) domain, which contains three catalytic residues (Glu71, Asp84 and Asp95) and a highly conserved GLMQ motif (Gly90, Leu91, Met92 and Gln93). Phylogenetic analyses revealed that TrLysG is clustered together with its counterparts from other teleost fishes. Furthermore, mRNA expression analyses showed that TrLysG was highly expressed in healthy mucosal tissues (intestines and gills), and considerably up-regulated in response to Vibrio harveyi infection in the intestines, gills, and liver. At pH 6 and 55 °C, the pure recombinant TrLysG (rTrLysG) exhibits optimum activity. It also displayed antimicrobial activity against three Gram-positive bacteria (Streptococcus parauberis, Staphylococcus pasteuri and Staphylococcus epidermidis) as well as five Gram-negative bacteria (Shewanella, Aeromonas hydrophila, Escherichia coli, Vibrio parahaemolyticus and V. harveyi). Our results highlighted the significant role of TrLysG in immune defense against invading pathogens, thereby contributing to the prevention and alleviation of disease spread in aquaculture. Simple Summary: Glioblastoma (GBM) is the most lethal adult primary brain tumor, and has no cure. This study investigated the membrane protein sortilin as a prognosis biomarker for glioblastoma (GBM). We found that sortilin is overexpressed in GBM tumors and can be detected in the blood of GBM patients. In addition, in cell cultures, targeting sortilin resulted in the inhibition of GBM cell invasion. These data highlight the value of sortilin as a potential clinical biomarker and therapeutic target for GBM and warrant further translational investigation. Glioblastoma (GBM) is a devastating brain cancer with no effective treatment, and there is an urgent need for developing innovative biomarkers as well as therapeutic targets for better management of the disease. The membrane protein sortilin has recently been shown to participate in tumor cell invasiveness in several cancers, but its involvement and clinical relevance in GBM is unclear. In the present study, we explored the expression of sortilin and its potential as a clinical biomarker and therapeutic target for GBM. Sortilin expression was investigated by immunohistochemistry and digital quantification in a series of 71 clinical cases of invasive GBM vs. 20 non-invasive gliomas. Sortilin was overexpressed in GBM and, importantly, higher expression levels were associated with worse patient survival, pointing to sortilin tissue expression as a potential prognostic biomarker for GBM. Sortilin was also detectable in the plasma of GBM patients by enzyme-linked immunosorbent assay (ELISA), but no differences were observed between sortilin levels in the blood of GBM vs. glioma patients. In vitro, sortilin was detected in 11 brain-cancer-patient-derived cell lines at the anticipated molecular weight of 100 kDa. Interestingly, targeting sortilin with the orally bioavailable small molecule inhibitor AF38469 resulted in decreased GBM invasiveness, but cancer cell proliferation was not affected, showing that sortilin is targetable in GBM. A new detection device was designed by integrating fiber Bragg grating (FBG) and polyvinyl chloride (PVC) tube in order to monitor the slip surface of a landslide. Using this new FBG-based device, a corresponding slope model with a pre-set slip surface was designed, and seven tests with different soil properties were carried out in laboratory conditions. The FBG sensing fibers were fixed on the PVC tube to measure strain distributions of PVC tube at different elevation. Test results indicated that the PVC tube could keep deformation compatible with soil mass. The new device was able to monitor slip surface location before sliding occurrence, and the location of monitored slip surface was about 1-2 cm above the pre-set slip surface, which basically agreed with presupposition results. The monitoring results are expected to be used to pre-estimate landslide volume and provide a beneficial option for evaluating the potential impact of landslides on shipping safety in the Three Gorges area. With the development of urban underground space and increased infrastructure functions, both the scale of engineering construction and engineering difficulties have increased globally. In the construction of structures in soft strata, especially in coastal areas, the limited bearing capacity of the foundations poses a significant challenge. The composite pile technologies employing an organic combination of the rigid pile andthe flexible column can enable efficient soft ground treatment. In light of prominent global environmental issues, low-carbon energy-saving curing technologies have been rapidly developed for application in geotechnical engineering. This paper discusses progress in research on the mechanical properties of the efficient and low-carbon pile technologies, including the stiffened deep mixing (SDM) column, squeezed branch pile, pre-bored grouting plated nodular (PGPN) pile, precast cement pile reinforced by cemented soil with a variable section (PCCV), and carbonized composite pile (CCP). In addition, it reviews the technical characteristics and recent progress of feasible low-carbon energy-efficient curing technologies. The paper also proposes future directions for theoretical research and technological development of low-carbon pile technologies. The key contribution of this review is to provide insights into efficient and low-carbon pile technologies. In addition, the findings from the study of the pile technologies used in extra-thick soft strata also provide industry practitioners with a comprehensive guide regarding the specific applications and mechanical performance of the pile technologies, which can serve as a stepping stone to facilitate the technological development of the underground space industry. Background: Neuroendocrine neoplasms of the gallbladder (GB-NENs) are a rare group of histologically heterogeneous tumors, and surgical resection of the primary tumor is the mainstream treatment at the moment. The current study aimed to establish and validate novel nomograms for patients with GB-NENs undergoing primary tumor resection to predict the 6-, 12-, and 18-month overall survival (OS) and cancer-specific survival (CSS). Methods: Clinicopathological information of patients with GB-NENs undergoing primary tumor resection between 2004 and 2018 was derived from the Surveillance, Epidemiology, and End Results (SEER) database. Candidate prognostic factors were selected by Cox regression analyses, and the nomograms were constructed. Finally, concordance index (C-index), calibration plot, area under the curve from the receiver operating characteristic curve (AUC), and decision curve analysis (DCA) were utilized to assess the effective performance of the nomograms. Results: A total of 221 patients with GB-NENs undergoing resection were enrolled in this retrospective study. Using the Cox regression analyses, age, pathological classification, tumor size, and SEER stage were identified as the independent prognostic factors of patients with GB-NENs undergoing resection, and nomograms were constructed. The C-indexes of OS and CSS in training dataset were 0.802 (95% CI: 0.757–0.848) and 0.846 (95% CI: 0.798–0.895), while those of internal validation dataset were 0.862 (95% CI: 0.802–0.922) and 0.879 (95% CI: 0.824–0.934), respectively. Conclusions: Taken together, the nomograms are accurate enough to predict the prognostic factors of GB-NEN patients undergoing resection, allowing for treatment decision-making and clinical monitoring for future clinical work. Glioblastoma multiforme (GBM) is the most lethal adult brain cancer. Temozolomide (TMZ), the standard chemotherapeutic drug used in GBM, has limited benefit and alternate therapies are needed to improve GBM treatment. Nerve growth factor (NGF) and its precursor proNGF are increasingly recognized as stimulators of human tumor progression. The expression and stimulatory effect of NGF on GBM cell growth has previously been reported, but the status of proNGF in GBM is unreported. In this study, we have investigated proNGF expression and biological activity in GBM. A clinical cohort of GBM (n = 72) and low-grade glioma (n = 20) was analyzed by immunohistochemistry for proNGF and digital quantification. ProNGF expression was significantly increased in GBM compared to low grade gliomas and proNGF was also detected in patient plasma samples. ProNGF was also detected in most GBM cell lines by Western blotting. Although anti-proNGF blocking antibodies inhibited cell growth in GBM cells with methylated MGMT gene promoter, targeting proNGF could not potentiate the efficacy of TMZ. In subcutaneous xenograft of human GBM cells, anti-proNGF antibodies slightly reduced tumor volume but had no impact on TMZ efficacy. In conclusion, this data reveals that proNGF is overexpressed in GBM and can stimulate cancer cell growth. Bullacta exarata has been consumed in Asia, not only as a part of the normal diet, but also as a traditional Chinese medicine with liver- and kidney-benefitting functions. Several scientific investigations involving extraction of biomolecules from this mollusk and pharmacological studies on their biological activities have been carried out. However, little is known regarding the antitumor properties of polysaccharides from B. exarata, hence the polysaccharides from B. exarata have been investigated here. One polysaccharide conjugate BEPS-IA was isolated and purified from B. exarata. It mainly consisted of mannose and glucose in a molar ratio of 1:2, with an average molecular weight of 127 kDa. Thirteen general amino acids were identified to be components of the protein-bound polysaccharide. Methylation and NMR studies revealed that BEPS-IA is a heteropolysaccharide consisting of 1,4-linked-α-D-Glc, 1,6-linked-α-D-Man, 1,3,6-linked-α-D-Man, and 1-linked-α-D-Man residue, in a molar ratio of 6:1:1:1. In order to test the antitumor activity of BEPS-IA, we investigated its effect against the growth of human hepatocellular carcinoma cells HepG2 in vitro. The result showed that BEPS-IA dose-dependently exhibited an effective HepG2 cells growth inhibition with an IC50 of 112.4 μg/mL. Flow cytometry analysis showed that BEPS-IA increased the populations of both apoptotic sub-G1 and G1 phase. The result obtained from TUNEL assay corroborated apoptosis which was shown in flow cytometry. Western blot analysis suggested that BEPS-IA induced apoptosis and growth inhibition were associated with up-regulation of p53, p21 and Bax, down-regulation of Bcl-2. Heterodera schachtii is a well-known cyst nematode that causes serious economic losses in sugar beet production every year. Rapid and visual detection of H. schachtii is essential for more effective prevention and control. In this study, a species-specific recombinase polymerase amplification (RPA) primer was designed from a specific H. schachtii sequence-characterized amplified region (SCAR) marker. A band was obtained in reactions with DNA from H. schachtii, but absent from nontarget cyst nematodes. The RPA results could be observed by the naked eye, using a lateral flow dipstick (LFD). Moreover, we combined CRISPR technology with RPA to identify positive samples by fluorescence detection. Sensitivity analysis indicated that 10−4 single cysts and single females, 4−3 single second-stage juveniles, and a 0.001 ng genomic DNA template could be detected. The sensitivity of the RPA method for H. schachtii detection is not only higher than that of PCR and qPCR, but can also provide results in <1 h. Consequently, the RPA assay is a practical and useful diagnostic tool for early diagnosis of plant tissues infested by H. schachtii. Sugar beet nematodes were successfully detected in seven of 15 field sugar beet root samples using the RPA assay. These results were consistent with those achieved by conventional PCR, indicating 100% accuracy of the RPA assay in field samples. We developed safe and stable mixed polymeric micelles with low lipids and free propofol for intravenous administration, to overcome the biological barrier of the reticuloendothelial system (RES), reduce pain upon injection, and complications of marketed propofol formulation. The propofol-mixed micelles were composed of distearoyl-phosphatidylethanolamine-methoxy-poly (ethylene glycol 2000) (DSPE mPEG2k) and Solutol HS 15 and were optimized using Box Behnken design (BBD). The optimized formulation was evaluated for globule size, zeta potential, loading content, encapsulation efficiency, pain on injection, histological evaluation, hemolysis test, in vivo anesthetic action, and pharmacokinetics, in comparison to the commercialized emulsion Diprivan. The optimized micelle formulation displayed homogenous particle sizes, and the free drug concentration in the micelles was 60.9% lower than that of Diprivan. The paw-lick study demonstrated that propofol-mixed micelles significantly reduced pain symptoms. The anesthetic action of the mixed micelles were similar with the Diprivan. Therefore, we conclude that the novel propofol-mixed micelle reduces injection-site pain and the risk of hyperlipidemia due to the low content of free propofol and low-lipid constituent. We demonstrate a simple and highly sensitive optical fiber relative humidity (RH) sensor based on a long-period fiber grating (LPFG) coated with polyethylene glycol (PEG)/polyvinyl alcohol (PVA) composite films. The resonance wavelength of the LPFG is sensitive to environmental humidity due to the change in effective refractive index caused by the strong surface absorption and desorption of the porous PEG/PVA coatings. The sensor is sensitive in a wide range from 50% to 95% RH, with a highest sensitivity of 2.485 nm/%RH in the range 50–75% RH. The proposed RH sensor has the advantages of compact size, good reversibility, and stability, which makes it attractive for high-humidity environments. [ABSTRACT FROM AUTHOR]
