Low voltage electrostatic field (LVEF) and ultraviolet-C (UV–C) are non-thermal, safe, and environmentally friendly preservation technologies. This study explored the efficacy of LVEF, UV-C (0.1 kJ m−2), and their combination (LVEF + UV-C) in regulating the wound-healing response of injured potato tubers over a 21-day healing period at 20 °C. Results demonstrated that LVEF + UV-C treatment showed superior efficacy in accelerating wound healing of tubers compared to single-treatment and control groups, mitigating weight loss and disease index, promoting suberin and lignin accumulation, and enhancing firmness, browning index, and H 2 O 2 content. Biochemical changes observed during tuber healing exhibited a positive correlation with increased enzyme activities (PAL, 4CL, CAD, POD, and PPO) and metabolite levels (phenolic compounds, flavonoids, lignin) associated with phenylpropane metabolism. Our study findings suggest that the application of LVEF + UV-C stimulates phenylpropanoid metabolism, accelerates suberin and lignin accumulation at wounds, thus facilitates wound healing in potato tubers. • LVEF + UV-C achieve the best effect on accelerating wound healing process of tubers. • LVEF + UV-C accelerate the deposition of suberin (SPP, SPA) and lignin at wounds. • LVEF + UV-C boost enzyme activities and metabolite levels in phenylpropane metabolism. The use of highly efficient high-throughput screening (HTS) platform has recently gained more attention as a plausible approach to identify de novo therapeutic application potential of conventional anti-tumor drugs for cancer treatments. In this study, we used hepatocellular carcinoma (HCC) cells as models to identify cytotoxic compounds by HTS. To identify cytotoxic compounds for potential HCC treatments, 3271 compounds from three well established small molecule libraries were screened against HCC cell lines. Thirty-two small molecules were identified from the primary screen to induce cell death. Particularly, mitoxantrone (MTX), which is an established antineoplastic drug, significantly and specifically inhibited the growth and proliferation of HCC cells in vitro. Mechanistic studies of LC3-II, p62 and phosphorylation of p70S6K in HepG2 cells revealed that MTX treatment induced mTOR-dependent autophagy activation, which was further confirmed by the autophagic flux assay using lysosomal inhibitor chloroquine (CQ). In the combined treatment of MTX and CQ, where autophagy was inhibited by CQ, the elevations of cleaved Caspase-3 and PARP were observed, indicating the enhanced apoptosis in HepG2 cells. Taken together, we hypothesize that MTX-induced autophagy plays an pro-survival role in HCC treatment. Graphical abstract Abstract PRSS23 is a newly discovered serine protease that has been associated with tumor progression in various types of cancers. Our previous study showed PRSS23 is down-regulated obviously in Hedgehog pathway blocked gastric cancer cells. However, the correlation between PRSS23 and tumor progression of gastric cancer remains unclear. Here, the role and mechanism of PRSS23 in tumor progression of gastric cancer were determined. PRSS23 protein levels were significantly increased in gastric cancer tissues compared with the paired adjacent normal gastric mucosa tissues. The high expression of PRSS23 correlated strongly with both poor differentiated histology and cancer region of sinus ventriculi. Gastric cancer patients with low PRSS23 expression displayed a better prognosis. In gastric cancer cells, PRSS23 knockdown inhibited cell proliferation and induced apoptosis. In xenograft tumor model, PRSS23 knockdown led to dramatic decreases of the average tumor volume and the average tumor weight. In addition, PRSS23 knockdown suppressed gastric cancer growth through inhibiting EIF2 signaling using gene expression microarray analysis. Taken together, our results suggest PRSS23 is highly associated with human gastric tumorigenesis and progression. PRSS23 knockdown could suppress tumor growth of gastric cancer in vitro and in vivo through inhibiting EIF2 signaling, and EIF4E maybe a potential target of PRSS23.
