The valuation and planning of complex projects are becoming increasingly challenging with rising market uncertainty and the deregulation of many industries, which have also raised the need for efficient risk management. We take the perspective of a private firm interested in sequential capacity expansion of a project and develop a framework for measuring the downside risk of the serial project and optimising the sequence of the stages. Under general distributional assumptions for the duration of each stage, we present an accurate representation of the project's net present value (NPV) distribution based on a Pearson curve fit, leading to closed-form expressions for the associated risk measures. We then assess the impact of duration variability on the value at risk and demonstrate its role in stochastic project scheduling. We also account for the trade-off between maximising the expected NPV and minimising the risk exposure, and obtain the optimal schedule for risk-averse decision-makers. It becomes obvious that both the duration variability of each stage and the decision-makers' risk preferences can significantly affect the optimal sequence of the stages and that high duration variability is not always undesirable, even for risk-averse decision-makers. • Introduction of flexible (or non-regular) work has a negative impact on productivity. • This negative impact is strongest under innovation regimes that rely on highly cumulative knowledge. • Supply-side deregulation of labor markets is likely to have contributed to a productivity growth slowdown in major OECD countries after 2004. Due to labor market reforms around 2003–4, Japan has a growing group of 'non-regular' workers who are easy to fire, and have poor carrier perspectives. This marks a break with the traditional Japanese model of life-time employment that allowed for intensive in-company training and commitment of personnel. Drawing from a national wage structure survey, we find indications that employment of non-regular workers has a negative impact on productivity (proxied by wages), this negative impact being largest under innovation regimes that require a high cumulativeness of knowledge. Our findings are consistent with neo-Schumpeterian research in Europe which concluded that certain labor market rigidities, while being un desirable from a neoclassical perspective, can be useful to innovation. Our paper confirms the impression from earlier research that structural reforms of labor markets along supply-side lines are likely to be one of the reasons for a substantial decline of productivity growth in major OECD countries since about 2004/05. Indonesian rural youth face challenges accessing farmland and sustaining an agricultural livelihood while their labour is not necessarily absorbed by other sectors. In that context, the Omnibus Law on Job Creation (Law 11/2020) promises to liberalise trade and investment across multiple sectors, including agriculture and food security. Combining legal research and political economy approaches to youth and agrarian challenges, we identify amendments to legislation that reduce safeguards for the environment, workers' and farmers' rights and their livelihoods. If fully implemented, the legislative amendments could further narrow youth's options both for secure formal work and futures in farming by accelerating the expansion of infrastructure, industrial plantations and extractive industries that utilise low‐wage labour and huge areas of land. This exposes inconsistencies in the government's approach to increase future food security by promoting intensification of agriculture and attracting youth to farming, while enabling agro‐ and resource extraction that absorbs land yet offers limited and precarious employment prospects. Background: To evaluate the ability of the estimated plasma expression levels of genes of microRNA (MiR-) 146a and 155 to differentiate between samples of pregnant women suspected to be infected by T. gondii. 50 newly pregnant women who had at least one of the criteria of high risk for toxoplasma infection and 50 newly primigravida women free of these criteria gave blood samples for qualitative determination of serum toxoplasma antibodies and estimation of plasma expression levels of MiR-146a and 155 using the qRT-PCR. During the pregnancy course, the incidence of pregnancy complications was recorded. Results: Twenty-six women were IgM−/IgG−, 17 women were IgM+/IgG− and 7 women were IgM+/IgG+. Thirty-two women had pregnancy complications with significantly lower incidence in IgM−/IgG− women. Plasma expression levels of MiR-146a and 155 were significantly higher in total patients compared to control levels and were significantly higher in samples of IgM+/IgG+ patients than in other samples. Statistical analyses defined a high plasma level of MiR-155 as the highly significant predictor for oncoming pregnancy complications and high levels of both microRNAs as predictors for the presence of toxoplasmosis despite seronegativity. Kaplan-Meier regression analysis defined increasing cumulative risk of having toxoplasmosis despite seronegativity with plasma levels of MiR-146a and MiR-155 of 1.2 and 3, respectively. Conclusion: The incidence of pregnancy complications is high, irrespective of the seronegativity of women at high risk of toxoplasmosis. Estimated plasma levels of MiR-155 might identify women liable to develop complications and differentiate seronegative women vulnerable to having T. gondii infection. Trial registration: The study protocol was approved preliminarily by the Local Ethical Committee at Benha Faculty of Medicine. MYC has been implicated in the pathogenesis of a wide range of human tumors and has been described for many years as a transcription factor that regulates genes with pleiotropic functions to promote tumorigenic growth. However, despite extensive efforts to identify specific target genes of MYC that alone could be responsible for promoting tumorigenesis, the field is yet to reach a consensus whether this is the crucial function of MYC. Recent work shifts the view on MYC's function from being a gene‐specific transcription factor to an essential stress resilience factor. In highly proliferating cells, MYC preserves cell integrity by promoting DNA repair at core promoters, protecting stalled replication forks, and/or preventing transcription‐replication conflicts. Furthermore, an increasing body of evidence demonstrates that MYC not only promotes tumorigenesis by driving cell‐autonomous growth, but also enables tumors to evade the host's immune system. In this review, we summarize our current understanding of how MYC impairs antitumor immunity and why this function is evolutionarily hard‐wired to the biology of the MYC protein family. Street theater is historically a form of political performance art used to foster social change and is characterized by unscheduled performances in public spaces, interactive audience participation, and short performances often repeated in the same spot for passersby. Long used as a creative tool within social movements, street skits not only entertain but are used to educate and build awareness on pressing issues. It is an art form for the people, by the people, making theater available for all ("on the streets") rather than solely for those who can afford it. Street theater also has its uses in public health education and organizing and has been used globally as an effective method of health promotion, especially in rural and low-literacy areas. This paper looks at a street theater project that university students at Clark University in Worcester, MA participated in as part of a "Health Rights: Advocacy and Action" course. Students were tasked with developing a street theater skit that educates on a public health issue and publicly perform the skit on campus. Three skits were produced: (1) "Deadly Derailment": raised awareness on industry deregulation and the 2023 toxic chemical spill in East Palestine, OH; (2) "Harm Reduction Saves Lives": addressed humane and non-victim blaming ways to address substance use; and (3) "End the STIgma": educated on sexual health on a college campus. Since the 1980s, most advanced economies have massively 'flexibilized' their labour markets by reducing the protection against layoffs and introducing temporary and flexible contracts. Concurrently, a well-established economic stylized fact – the stability of the wage share of income – has been challenged as empirical evidence reveals a substantial decline in labour share across numerous countries. In this study, we aim to empirically investigate the causal link between these two phenomena by examining the impact of changes in the regulation of fixed-term contracts, as measured by variation over time in the temporary contracts EPL index, on the functional distribution of income. Background: Myotonic Dystrophy type I (DM1) is the most common muscular dystrophy in adults. Previous reports have highlighted that neuromuscular junctions (NMJs) deteriorate in skeletal muscle from DM1 patients and mouse models thereof. However, the underlying pathomechanisms and their contribution to muscle dysfunction remain unknown. Methods: We compared changes in NMJs and activity-dependent signalling pathways in HSALR and Mbnl1ΔE3/ΔE3 mice, two established mouse models of DM1. Results: Muscle from DM1 mouse models showed major deregulation of calcium/calmodulin-dependent protein kinases II (CaMKIIs), which are key activity sensors regulating synaptic gene expression and acetylcholine receptor (AChR) recycling at the NMJ. Both mouse models exhibited increased fragmentation of the endplate, which preceded muscle degeneration. Endplate fragmentation was not accompanied by changes in AChR turnover at the NMJ. However, the expression of synaptic genes was up-regulated in mutant innervated muscle, together with an abnormal accumulation of histone deacetylase 4 (HDAC4), a known target of CaMKII. Interestingly, denervation-induced increase in synaptic gene expression and AChR turnover was hampered in DM1 muscle. Importantly, CaMKIIβ/βM overexpression normalized endplate fragmentation and synaptic gene expression in innervated Mbnl1ΔE3/ΔE3 muscle, but it did not restore denervation-induced synaptic gene up-regulation. Conclusions: Our results indicate that CaMKIIβ-dependent and -independent mechanisms perturb synaptic gene regulation and muscle response to denervation in DM1 mouse models. Since 2013, Permitted Development Rights (PDR) in England have allowed commercial-to-residential conversions in locations once deemed suitable only for non-residential land-use. This deregulation of planning control has been justified as a way of encouraging more home-building in areas experiencing 'housing crisis', but its overall consequences remain unclear. This paper hence compiles quantitative evidence on a city-wide scale on the price, size, build and location of these conversions in London 2013–2021. It finds that homes produced through this route are generally smaller than the London average and are over-concentrated in neighbourhoods with fewer accessible green spaces and higher-than-average levels of air pollution. Here, larger conversion schemes (of more than 10 units) appear particularly problematic, potentially subjecting residents to forms of 'slow violence' that could have long-term consequences for their physical and mental health. The paper also finds that, on average, PDR conversions are marginally more affordable than other new developments in the capital, but are also more expensive per square metre, suggesting deregulation is allowing developers to 'extract' maximum value from these schemes rather than providing affordable homes per se. The implications of this are discussed in relation to the politics of housing in London and the wider forms of planning deregulation allowing developers to accrue increased profits from housing in an era of intense financialisation. The Perception of Anomie Scale (PAS) is a measure used to evaluate the state of society and whether it is disintegrated and deregulated. Although widely used, the psychometric properties of reliability, validity, and measurement invariance according to sex have not been studied in the Chilean university population. To explore these properties, a cross-sectional study was carried out with 383 students from public (45.7%) and private (54.3%) universities, with a mean age of 21.5 years (SD = 3.3). A CFA and ESEM were performed, which confirmed the two-factor correlated structure, achieving better goodness-of-fit indices by eliminating five items (RMSEA = 0.09; CFI = 0.98; TLI = 0.95). This also provided evidence of reliability and measurement invariance according to sex. This study provides evidence of the psychometric quality of the PAS scale, thus allowing its use in the Chilean university context. [ABSTRACT FROM AUTHOR]

This research turns the spotlight to the deregulation of once publicly funded affordable housing. Through a microsimulation that follows the conversion from affordable to market-rate units on Roosevelt Island New York, we estimate the expected demographic changes each year between 1976 and 2070. The simulation combines information from the American Community Survey, the island's masterplan, the privatization agreements, and interviews with residents, to produce interactive graphs at three urban scales: the neighborhood, the project, and the building. We found that while the households of market-rate units are gradually becoming younger and more affluent, the households of affordable units are becoming older and more impoverished. Despite an individual agreement for each building, the demographic changes are similar, and that, those changes will affect low-income buildings first. Simple Summary: UGT enzymes metabolize and detoxify numerous small molecules that are important in cancer, including carcinogens, endogenous growth regulators, and anticancer drugs. Alternatively spliced UGT transcripts can encode truncated proteins that inhibit canonical UGTs, thus reducing detoxification activity. We assessed the expression of specific variant transcripts, designated as UGT1A_v2 and _v3, in six different cancers using RNA-seq datasets with large cohorts of paired normal and tumor tissues. Our results show high interindividual variation in v2 and v3 transcript abundance, as well as tissue- and tumor-specific expression patterns. These findings suggest that the variants have tissue-specific impacts on glucuronidation and may have a more significant role in tumors than in normal tissues. The high interindividual variability is likely relevant to differing personalized drug metabolisms through the UGT conjugation pathway. Finally, our discovery of novel UGT1A variant transcripts further highlights the diversity of the UGT1A transcriptome and proteome. The UGT1A locus generates over 60 different alternatively spliced transcripts and 30 circular RNAs. To date, v2 and v3 transcripts are the only variant UGT1A transcripts that have been functionally characterized. Both v2 and v3 transcripts encode the same inactive variant UGT1A proteins (i2s) that can negatively regulate glucuronidation activity and influence cancer cell metabolism. However, the abundance and interindividual variability in the expression of v2 and v3 transcripts in human tissues and their potential deregulation in cancers have not been comprehensively assessed. To address this knowledge gap, we quantified the expression levels of v1, v2, and v3 transcripts using RNA-seq datasets with large cohorts of normal tissues and paired normal and tumor tissues from patients with six different cancer types (liver, kidney, colon, stomach, esophagus, and bladder cancer). We found that v2 and v3 abundance varied significantly between different tissue types, and that interindividual variation was also high within the same tissue type. Moreover, the ratio of v2 to v3 variants varied between tissues, implying their differential regulation. Our results showed higher v2 abundance in gastrointestinal tissues than liver and kidney tissues, suggesting a more significant negative regulation of glucuronidation by i2 proteins in gastrointestinal tissues than in liver and kidney tissues. We further showed differential deregulation of wildtype (v1) and variant transcripts (v2, v3) in cancers that generally increased the v2/v1 and/or v3/v1 expression ratios in tumors compared to normal tissues, indicating a more significant role of the variants in tumors. Finally, we report ten novel UGT1A transcripts with novel 3′ terminal exons, most of which encode variant proteins with a similar structure to UGT1A_i2 proteins. Families in the United States suffer from a removal epidemic. The child welfare framework allows unnecessary and harmful intervention into family and parenting matters, traditionally left to the discretion of the parent. Many states allow Child Protective Services ("CPS") to investigate, intervene, and permanently separate a child from their parents for innocuous activities such as letting the child play outside unattended. This especially affects low-income and minority families. To prevent CPS from unnecessarily intervening in a family's decision to let their children engage in independent, unsupervised activities, Utah passed a "free-range" parenting act ("Act") in 2018. The Act explicitly excludes independent, age-appropriate activities from the definition of neglect. This Act has remained largely unexamined: whether the passage of the free-range parenting law has resulted in a decrease of non-supervision cases referred to and substantiated by CPS is unclear. It is also unclear whether free-range parenting laws are a viable solution to the issue of unnecessary and harmful state intervention, in general. This Note explores whether the free-range parenting law passed in Utah in 2018 has led to any discernible reduction in non-supervision cases and removals. Since the statistical analysis has significant limitations, the Note then takes a more general approach in examining whether these laws address the causes of unnecessary state intervention. The Note finds that many free-range parenting laws fail to address larger issues in the child welfare system and tend to mainly benefit middle-class and high-income families. A solution will likely require a concerted effort by all three branches of the government. With the advent of globalization, a scenario of high interdependence within the international community was established, highlighted by the affirmation of neoliberal premises such as the deregulation of the economy, monetarism and the intensive exploitation of environmental resources. Bearing in mind the tragic effects of neoliberalism on contemporary democracies, the objectives of this article, whose main theoretical reference is the Spaniard Gerardo Pisarello, are: a) to outline a panorama of neoliberal constitutionalism, highlighting its consequences; b) to present the New Latin American Constitutionalism as a possible alternative to traditional power structures, as recently experienced in Ecuador and Bolivia. It is concluded that, although subject to oppositions from the status quo and contradictions inherent to the societies of the countries that adopted it, the New Latin American Constitutionalism advanced significantly in the desiderata of strengthening popular representation in political instances and reducing the destructive power of the big capital. In 2016, the City of Toronto legalised the ridehail giant Uber under a particularly Uber‐friendly regulatory regime. Rather than understanding this interim outcome along the lines of now widespread narratives of corporate "disruption", in this article I take up Manuel B. Aalbers' notion of "regulated deregulation" in order to foreground the state's role as a manically prolific facilitator of early Uberisation. Based on ethnographic research in Toronto, I argue that the three longer‐standing state spatial strategies of (1) the common‐sense neoliberal state, (2) the labour‐averse competition state, and (3) the tech‐infatuated smart state were paramount in creating those "on‐the‐ground" conditions—social, legal, spatial, and other—on which Uber has been able to thrive in many cities across the North American continent. In order to solve the problem of power exchange between areas of multi-area power systems under the power market, this paper designs a power trading contract based on the generation participation matrix, so as to simulate the specific process of power change in each area. With the integration of wind power into the large-scale power system, it is difficult to model the multi-area power system. For this situation, this paper designs a data-driven model-free adaptive load frequency control algorithm based on collecting input and output data, which gets rid of the dependence of the power system on the model. Along with the frequent transmission of input and output data in each area, the communication load of the power system also increases. Aiming at saving communication resources, this paper designs an event-triggered mechanism to reduce the communication bandwidth. The stability of the control algorithm is demonstrated theoretically. Several states have enacted specialized limited liability company legislation in an attempt to attract decentralized autonomous organizations. In this way, the regulatory competition debate surrounding states such as Wyoming, Tennessee, and Vermont, attempting to dethrone Delaware, has found a new battleground. According to Professor Lynn LoPucki, this will entail a regulatory race to the bottom, that is, a race to "laxity." I disagree. In fact, deregulation has already been achieved in the traditional limited liability company form. The decentralized autonomous organization limited liability company is no laxer or more attractive to investors, who willlikely prefer the traditional limited liability company or even other entity forms, given the diversity of investors' needs and aims. Moreover, some decentralized autonomous organization organizers may wish not to incorporate at all, hoping to avoid the law altogether. While increasingly risky, this strategy rests on the belief in the alegality liability shield, further diminishing the impact of the /ex specialis approach. After analyzing the statutory developments and the scholarly and industrial commentary, I conclude that in decentralized autonomous organizations, as elsewhere, the regulatory race to the bottom is a fad. We investigate the impact of US banking industry deregulation on intergenerational mobility. In order to do so, we use a quasi-natural experimental research design to investigate how a change in state level bank competition (following geographic deregulation) affects the intergenerational mobility of individuals born to (poorer) families at the lower end of the income distribution. We find that individuals born to poorer families who spend their first years as adults in an area with a more competitive banking system, experience higher mobility relative to counterparts located in areas characterised by lower levels of bank competition. With the liberalization and deregulation of the power industry, the power market presents increasingly invisible dynamics and uncertainty. Establishing an effective price forecasting model has always been an important issue for power market participants to avoid risks and maximize economic benefits. However, due to the inherent characteristics of high frequency and high volatility of electricity price data, it is still a difficult and challenging problem to achieve high-precision forecasting. To overcome these difficulties, this study constructed a novel multivariate electric price bi-forecasting system based on the proposed linear operator combination mechanism and deep learning model. Specifically, improved complete ensemble empirical mode decomposition with adaptive noise (ICEEMDAN) is used for data preprocessing, while the multiobjective golden eagle optimization algorithm (MOGEO) optimized deep belief network and bidirectional long short-term memory network are used as the main prediction models for realizing two forecasting modes of point forecast and interval forecast by a multi-input multi-output structure simultaneously. To verify the model forecasting ability and effectiveness, four datasets from Australia (New South Wales) from 2020.7 to 2021.1 are selected for experiments. According to the empirical results, the mean absolute percentage error results of the designed forecasting strategy in point forecasting are within 4% for the four datasets, and the average coverage width criterion values in interval forecasting are all approximately 0.3. The crucial role played by the oncogenic expression of TP53 , stemming from mutation or amyloid formation, in various human malignancies has been extensively studied over the past two decades. Interestingly, the potential role of TP53 as a crucial player in modulating immune responses has provided new insight into the field of cancer biology. The loss of p53's transcriptional functions and/or the acquisition of tumorigenic properties can efficiently modulate the recruitment and functions of myeloid and lymphoid cells, ultimately leading to the evasion of immune responses in human tumors. Consequently, the oncogenic nature of the tumor suppressor p53 can dynamically alter the function of immune cells, providing support for tumor progression and metastasis. This review comprehensively explores the dual role of p53 as both the guardian of the genome and an oncogenic driver, especially in the context of regulation of autophagy, apoptosis, the tumor microenvironment, immune cells, innate immunity, and adaptive immune responses. In England, unaccompanied child migrants who seek asylum are the responsibility of the local state, who acts as their ‘corporate parent’. While these young people are ostensibly supported by children’s services in keeping with responsibilities under the Children’s Act 1989, in comparison to ‘local’ children unaccompanied children are disproportionately placed in unregulated, outsourced accommodation. Drawing on data from the Children Caring on the Move research project – including over 180 interviews with unaccompanied young people, frontline workers, accommodation owners and policy makers – I argue that this part of the care system can be understood as a double move of enclosure. First, I argue that claims about the unpredictability of arrivals of unaccompanied children are used as a rationale for the marketisation of children’s services, part of an enclosure of what was previously public. Second, I point to the enclosure, and commodification, of unaccompanied children themselves. They not only become a source of profit in this enclosed sector through their statuses as ‘child’ and ‘unaccompanied migrant’, but the limited forms of support provided in outsourced and unregulated provision make it more difficult for many to regularise their status before being subject to enforced destitution or deportability as non-citizen adults. The article contributes to efforts to theorise contemporary processes of enclosure in racial capitalism, adds to understandings of the UK’s ‘asylum market’ which to date has focused almost primarily on adult or family migrants, and sheds new light on causalities of care in the UK’s marketised and bordered children’s services. • We measure housing regulation using a novel dataset of unbuilt housing capacity. • This approach improves upon conventional indexes of regulation in several ways. • Our measure is likely better at capturing sentiment toward housing production. • It performs similarly or better than conventional measures in regressions of permits. • Interaction terms capture the added importance of capacity in high demand cities. We introduce a new way to measure the stringency of housing regulation. Rather than a standard regulatory index or a single aspect of regulation like Floor Area Ratio, we draw on cities' self-reported estimates of their total zoned capacity for new housing. This measure, available to us as a result of state legislation in California, offers a more accurate way to assess local antipathy toward new housing, and also offers a window into how zoning interacts with existing buildout. We show, in regressions analyzing new housing permitting, that our measure has associations with new supply that are as large or larger than conventional, survey-based indexes of land use regulation. Moreover, unbuilt zoning capacity interacts with rent to predict housing production in ways conventional measures do not. Specifically, interacting our measure with rent captures the interplay of regulation and demand: modest deregulation in high-demand cities is associated with substantially more housing production than substantial deregulation in low-demand cities. This research seeks to answer the following research question "What kind of regulation architecture can stimulate creative destruction and growth of two different types of new industries like telemedicine, and for-profit-hospital?" Online interview method was used to investigate the telemedicine industry and for-profit-hospitals at Daegu-Gyeongbuk province in South Korea and Cardiff-Wales in the UK. Findings provided following grounded theories. First, the way to create a converted new industry is not deregulation but regulation conversion. Though, deregulation helps to expand the modern market, it could disturb the emergence and growth of new markets. Second, the regulation can be an essential policy method to create emerging new markets when there are enough (potential) suppliers owing to technological developments or the accumulation of a new workforce equipped with new technologies. According to the UK for-profit-hospitals, regulations for consumer protection and decreasing transaction costs or complexity in the new market could trigger market creation. Third, digital transformation has motivated open innovation dynamics in all industries. The expanded open innovation dynamics with digital transformation can promote, most of all, the appearance of converted new industries like telemedicine. • First, the way to create a 'converted new industry' is not deregulation but regulation conversion. • Second, the regulation can be an essential planning method to create an' emerging new industry' when there are enough (potential) suppliers including technological developments or new workforce equipped with new technologies. • Third, the expanded open innovation dynamics with digital transformation can promote, most of all, the appearance of converted new industries' like telemedicine in Korea.
