In Brazil, the use of Eucalyptus is focused on the production of wood or pulp for the paper industry but without any general recovery of waste, with leaves and branches being left on the ground. One possibility is to use these residues as raw materials in the production of industrially relevant and value-added compounds such as essential oil. The aim of the present study was to investigate the chemical composition, yield, anti-inflammatory/antinociceptive activities, and acute toxicity in mice, as well as the antimicrobial effects of essential oils from the leaves of 7 varieties of Eucalyptus and hybrids against Escherichia coli, Staphylococcus aureus, and Candida albicans. The extraction of oils was carried out using hydrodistillation, and they were analyzed by gas chromatography coupled to mass spectrometry. Urocam and Grancam were the plants that obtained the highest oil yield, with yields of 3.32 and 2.30%, respectively. The main chemical components identified in these plants were 1.8 cineole and α-pinene. The antinociceptive effect of the 7 oils (50 mg/kg, p.o.) was initially assessed in the acetic acid-induced writhing test. In this assay, a significant (p < 0.05) antinociceptive/anti-inflammatory effect was observed from 4 tested essential oils (E. benthamii, E. saligna, and the hybrids Urocam and Grancam) when compared to the vehicle-treated group. This effect was then confirmed in the formalin-induced paw licking test. No toxicological effects or alterations were observed in motor coordination after the administration of the studied oils to the animals. In the antimicrobial evaluation, the seven essential oils inhibited the growth of S. aureus, E. coli, and C. albicans at different concentrations. Collectively, these results demonstrate that the essential oil from the leaves and branches of Eucalyptus species and varieties present potential biomedical applications and represent a source of antimicrobial and/or anti-inflammatory compounds. Background: There is a characteristic Tibetan folk medicine in China named Corydalis hendersonii Hemsl. (CH) has been used for treatment of cardiovascular related diseases, called "plethora" in Tibetan medicine. Previous studies demonstrated that ethanol extract of CH showed anti-acute myocardial infarction (AMI) effect through inhibiting fibrosis and inflammation. Rich alkaloids fraction (RAF) is isolated from CH, but whether RAF possessing an equivalent effect with the CH ethanol extract and by which mechanism it protects against AMI has not yet reported. The paper aimed to study the potential role of RAF on myocardial injured mice and its underlying mechanism. Materials and methods: Liquid chromatography mass spectrometry-ion trap-time of flight (LCMS-IT-TOF) was used to analyze the chemical profile and isolate pure compounds. The ligation of left anterior descending (LAD) of coronary artery in mice was used to evaluate the in vivo anti-AMI effect, by dividing into eight groups: Sham, Model, Fosinopril (10 mg/kg, i.g.), total extract (TE, 400 mg/kg, i.g.), poor alkaloids fraction (PAF, 300 mg/kg, i.g.), and RAF (25, 50, and 100 mg/kg, respectively, i.g.) groups. Echocardiography was used to evaluate mice heart function through the index of left ventricular end-systolic diameter (LVEDs), left ventricular end-diastolic diameter (LVEDd), fractional shortening (FS) and ejection fraction (EF). We detected the lactate dehydrogenase (LDH) and creatine kinase-MB (CK-MB) in the serum and the plasma level of angiotensin II (AngII). The apoptosis of mice myocardial tissue was verified by TUNEL assay. The expression of p38 mitogen-activated protein kinases (p38 MAPK), Bcl-2 and Bcl-2-associated X protein (Bax) were detected through immunofluorescence staining, qRT-PCR and western blot in mice heart tissue and H9c2 cells. Results: Echocardiography data indicated that the values of LVEDd and LVEDs were reduced and the values of FS and EF were improved by TE and RAF significantly. RAF also decreased the levels of LDH, CK-MB and AngII and significantly inhibited inflammatory cells in the marginal zone of myocardial infarction. The TUNEL assay results showed that RAF significantly attenuated cell apoptosis. Immunofluorescence and qRT-PCR assay showed that RAF inhibited p38 MAPK, Bax, and Bcl-2 proteins in mice myocardium. Western blot results validated that the expressions of key proteins were inhibited by RAF. Also, the apoptotic cells and apoptosis-related proteins were dramatically reduced by RAF in vivo and in vitro. Besides, RAF and PAF were analyzed by LCMS-IT-TOF to identify the main compounds and to demonstrate the difference between them. The results showed that a total of 14 alkaloids were identified, which indicated that the isoquinoline alkaloids were the main ingredients in RAF may contributing to the cardioprotective effect in mice. Conclusions: RAF improves cardiac function by inhibiting apoptosis via p38 MAPK signaling pathway, and RAF contributes to the effect against myocardial ischemic injury of TE in mice, which provides a substantial reference for the clinical application against ischemia heart disease and quality control of CH. (CH) and the rich alkaloids fraction (RAF) of CH showed significant cardioprotective effect on mice following acute myocardial infarction. RAF improved cardiac function by inhibiting apoptosis via p38 mitogen-activated protein kinases (p38 MAPK) signaling pathway in vitro and in vivo. Background & Aims: Nanotechnology is one of the newest, most important and most efficient sciences in the world. It is an interdisciplinary technology that has caused significant changes and developments in the sciences by solving problems and shortcomings. Nanoscience and nanotechnology is the production of nanometer-sized material that has recently attracted the attention of many researchers in the fields of medicine, agriculture and industry. The great importance and application of this technology has attracted much attention. Nanoparticles are particles about 1 to 100 nm the properties of these materials depend on their nanometer size. Nano materials have unique properties including lightweight and small nanoparticles, High surface-to-mass ratio, using in small quantities, saving consumables, being multifunctional and safe for the environment. Chitosan is a linear amino polysaccharide that has been taken into consideration for its properties such as non-toxicity, antibacterial properties, adhesion properties, unique structure, multidimensional properties and high performance. Physical and chemical methods are mostly used to produce nanoparticles. Although these methods may be successful and well-known, they are generally expensive, time-consuming, and dangerous to the environment and humans. Therefore, alternative methods such as microorganisms, fungi, plants, plant extracts and plant biomass should be used instead, which have less environmental hazards. Among the methods of synthesis of chitosan nanoparticles, green synthesis using plant extracts is more beneficial than other methods. Many plant biomolecules such as polyphenols, flavonoids, amino acids, polysaccharides, alkaloids, alcohols, vitamins and proteins can play a role in the formation and stabilization of nanoparticles. The aim of this study was to develop a green method using the extract of the aerial parts of the Russian knapweed to prepare and produce chitosan nanoparticles. Methods: In this study, chitosan nanoparticles were synthesized in a process by the extract of Russian knapweed (Acroptilon repens L.) which containing phenolic compounds. In order to preparation the materials needed for the experiment, the aerial parts of the Russian knapweed in the pre-flowering stage were collected from a land in Ardabil with severe contamination. The collected plant was transferred to the weed laboratory and dried at 60 °C for 72 h in the oven. The dried samples were powdered by a grinding (mill) machine. The aqueous extract was prepared by maceration method. 100, 150 and 200 grams of Russian knapweed powder in to erlenmeyer flask spilled and then added a certain amount of twice distilled water until the final volume reaches of 1000 ml. Then they were placed on the shaker at 25 °C for 24 hours. The obtained extracts were smoothed using Whatman filter paper. At the final stage, the extracts were centrifuged at 5000 rpm for 15 minutes and supernatant were extracted as pure extract and were kept at 4 °C for testing. Chitosan nanoparticles were prepared using ionic gelation method. In the present study, commercial chitosan with medium molecular weight was used. At first, 3 g of chitosan was dissolved in 300 ml of distilled water and placed on a magnetic stirrer at 550 rpm to dissolve completely and evenly at room temperature (with one magnet). To adjust the pH =5, some acetic acid was added. Then a certain amount of Russian knapweed extract was added to it. In the next step, 0.5 g of sodium polyphosphate (TPP) in 50 ml of distilled water was completely dissolved and added drop by drop to the chitosan solution, that was stirring on a magnetic stirrer, and the stirring process continued for 60 minutes. This was repeated for all 3 concentrations of Russian knapweed extract separately. After the reaction time, the final solution was centrifuged for 5 min and after smoothing, the top solution of sediments deposited in the oven was dried for 24 h at 60 °C. Results: Basic techniques for investigation and determination of nanoparticle properties including: thermogravimetric analysis (TGA), Fourier transform infrared (FT-IR), X-ray diffraction (XRD) and Scanning Electron Microscope (SEM) tests. The samples obtained by these tests were analyzed. Infrared spectroscopy was used to investigate the vibrational frequencies of chemical compounds. Thermal weight measurement analysis method, which is based on weight changes during sample heating, was performed to investigate the difference in thermal stability of the samples in the temperature range of 0-700°C. X-ray diffraction pattern of the samples was performed using Cu-Ka radiation. Surface morphology and particle distribution were also obtained using scanning electron microscopy with accelerator voltage k 15. In Spectrophotometric analysis, the presence of a peak in the range of 700 nm indicates the biological synthesis of these nanoparticles with extract of Russian knapweed. FTIR spectroscopy showed the biomolecules present in the extract and involved in the synthesis, which confirms the process of green synthesis of chitosan nanoparticles. In the FTIR spectrum obtained from chitosan nanoparticles with aqueous extract of Russian knapweed and pure extract of Russian knapweed, the presence of factor groups confirming the existence and presence of effective antioxidant compounds such as phenols and flavonoids in the plant extract is used. Based on this, the participation and involvement of phenolic compounds in the Russian knapweed in the synthesis of chitosan nanoparticles is quite clearly observed and understood. The size and morphology of biologically synthesized nanoparticles were determined by scanning electron microscopy, and it was found that the shape of the nanoparticles produced is approximately spherical with an average size of about 33 to 76 nanometers. One of the main reasons for the difference in the shape and size of the synthesized nanoparticles is the different concentrations of the plant extract used. The results of X-ray diffraction analysis also showed the nanocrystals synthesized by the extract of Russian knapweed. Conclusion: According to the results, it can be said that the polyphenolic compounds in the Russian knapweed extract were acted reducing agents, and also as a complexing agent synthesize chitosan nanoparticles and make them sustainable. The process of ion reduction and synthesis of nanoparticles are done by primary and secondary metabolites such as antioxidants, flavonoids, flavonoids, isoflavones, anticyanidins, isothiocyanates, carotenoids and polyphenols present in the extract. According to the obtained results, plant extracts have a good compatibility with the environment, so that they can be used for rapid production of chitosan nanoparticles, which is a simple, green and efficient method for the synthesis of nanoparticles at room temperature. The present study was undertaken to investigate the effects of using different extracts of two types of trees (Jatropha curcas seeds and Taxodium distichium cones). The solvents used were distilled water and ethanol. The determination of bioactive components of each extract was carried out using gas chromatography/mass spectrometry (GC/MS) analysis. In general, ethanolic extract was more effective on both linear growth and sclerotial numbers and mycelium characteristics of Sclerotium cepivorum than either of the cold or hot water extracts compared to control. Under in vitro conditions, treated with 20% ethanol solvent showed a fungicidal effect. Meanwhile, fungistatic effects appeared at 5% for reduction of mycelium growth. In the greenhouse experiments, the results indicated that onion transplants treated with ethanolic extract of taxodium and jatropha at 20% resulted in a significant reduction in the disease with a significant increase in bulb yield compared with control treatments. This report is one of the few papers that uses taxodium cones and jatropha seeds as antifungal substances against plant pathogens, especially S. cepivorum under worldwide and Egypt conditions. Eucommia ulmoides Oliv. is a deciduous tree which contains various chemical ingredients. The main objective was to document the active chemical ingredients of Eucommia ulmoides Oliv. and their metabolic profiles in vivo, with a view to providing an experimental and theoretical basis for clarifying the mechanisms underlying the pharmacological activity of Eucommia ulmoides Oliv. against rheumatoid arthritis. Eight main active constituents of Eucommia ulmoides Oliv. bark (pinoresinol glucopyranoside, aucubin, geniposidic acid, geniposide, genipin, chlorogenic acid, quercetin and betulinic acid) were quantified using high-performance liquid chromatography (HPLC). This paper additionally identified and characterized prototype metabolites via ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) combined with the Human Metabolome Database (HMDB) and literature comparisons. Ultra pressure liquid chromatography-mass spectrometry/ mass spectrometry (UPLC-MS/MS) was subsequently employed to quantify these components in blood over time and evaluate their pharmacokinetic characteristics. The anti-rheumatoid arthritis effects of genipin, pinoresinol glucopyranoside and their combinations were assessed using in vitro cellular assays. We identified and characterized a total of 53 ingredients from Eucommia ulmoides Oliv. bark and plasma samples, among which 20 were confirmed as prototype metabolites. Meanwhile, this paper derived and analyzed the metabolic cleavage pathway of 8 index ingredients. Six of these compounds displayed rapid entry into blood, with high plasma exposure and fast elimination rates. Data from the in vitro cellular assay showed that aucubin, pinoresinol glucopyranoside, genipin, and combinations of these compounds effectively inhibit MH7A cell proliferation, reduce NO release, and decrease inflammatory factor levels. Connarus semidecandrus Jack (Family: connaraceae) is a medicinal plant known for its wide distribution throughout Southeast Asia. Renowned for its diverse therapeutic properties, it has been traditionally used for treating fever, skin irritation, and colic. Numerous individuals suffer from skin issues, including wrinkles, hyperpigmentation, and inflammation, due to environmental factors. Although many drugs are available to treat skin problems, chemical drugs have many shortcomings and side effects. Therefore, natural products are attractive potential medicines for alleviating skin troubles. We recently showed that Connarus semidecandrus Jack ethanol extract (Cs-EE) has anti-alopecia potential. This paper aims to explore the potential skin-protective effects and underlying molecular mechanisms of Connarus semidecandrus Jack in UVB-induced human keratinocytes (HaCaT). Before utilization, Cs-EE was dissolved in dimethyl sulfoxide (DMSO) and was preserved at a temperature of −20 °C. The phytochemical constituents of Cs-EE were detected by gas chromatography–mass spectrometry analysis (GC-MS). Sequentially, HaCaT cells were exposed to varying concentrations of Cs-EE prior to ultraviolet B (UVB) irradiation. Evaluations of cellular responses in HaCaT cells, including assessments of cell viability, deoxyribonucleic acid (DNA) damage, and gene and protein expressions, were carried out. To explore the specific signaling pathway involved, we conducted a luciferase assay in addition to validating these pathways using Western blot analysis. Nitric oxide (NO) and intracellular reactive oxygen species were decreased. Melanin production through the activation of melanocytes by α-melanocyte-stimulating hormone (MSH) was also inhibited by Cs-EE. Furthermore, the mRNA expression levels of key factors such as cyclooxygenase-2 (COX-2), tumor necrosis factor-α (TNF-α), interleukin 6 (IL-6), MMP-1, MMP-3, and MMP-9 exhibited a remarkable decrease. In addition, the phosphorylation of TAK1 within the signaling cascade exhibited a decline, and the activities of the transcription factor AP-1 were decreased according to a luciferase reporter assay. Taken together, these findings suggest that the anti-inflammatory, anti-aging, and anti-apoptotic effects of Cs-EE indicate the compound's potential usefulness as a natural component in pharmaceutical and cosmetic products. Ligusticum chuanxiong Hort. (Chuanxiong, LC), as an important traditional Chinese medicine (TCM), can not only be used as a monarch herb but also be used as a classic "Yin-Jing" (引经) medicine in compound prescriptions, e.g., Buyang Huanwu Decoction (BHD). Although LC has the effect of guiding components into the brain in BHD, there is still a lack of scientific evidence on this "Yin-Jing" effects. Herein, we used pharmacokinetics and tissue distributions to investigate "Yin-Jing" effects of LC. To simplify the study, four major constituents in BHD, i.e., Calycosin (CA), astragaloside IV (AI), paeoniflorin (PA), and amygdalin (AM) were combined to form a simple compound (abbreviated as CAPA here) to replace the original BHD in this paper. The Yin-Jing medical property of LC was confirmed by the compatibility of CAPA with LC or its different fractions (Fr. A ∼ Fr. F). To explore the "Yin-Jing" medical property of LC via pharmacokinetics and tissue distributions by ultra-performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-QQQ-MS). The contents of CA, AI, PA, and AM were simultaneously determined by the established and validated UPLC-QQQ-MS method in different rat tissues and plasma after administration of CAPA with the combination of LC or Fr. A ∼ Fr. F. The pharmacokinetic parameters, e.g., T max , C max , AUC 0–t and MRT 0–t , were calculated to evaluate the efficiency of "Yin-Jing". The C max and AUC 0–t of CA, AI, PA, and AM were remarkably increased in rat brain tissues compared with those of the control group after compatibility of LC. This demonstrated that LC has the Yin-Jing effects on brain tissues. Additionally, Fr. B or Fr. C might be the material basis by specifically studying the distributions of CA, AI, PA, and AM in brain tissue based on mutual compatibility. The effects of Fr. B and Fr. C on distributions of these constituents in other tissues or plasma was also studied to verify the effects of Yin-Jing of LC. The results showed that the same upward trend is found in heart, liver and plasma, but the intensity is insignificant as that in brain tissue. Furthermore, the C max and AUC 0–t of some analytes in the rat spleen, lung, and kidney were significantly decreased compared with the control group (P < 0.05 or 0.01). LC has the function of Yin-Jing, especially guiding the components into the brain tissue. Moreover, Fr. B and Fr. C is suggested to be the pharmacodynamic material basis for the effect of Yin-Jing of LC. These finding explained that it was recommended to add LC into some prescriptions for treating cardiovascular and cerebrovascular diseases caused by Qi deficiency and blood stasis. This has laid a certain foundation for the research on the Yin-Jing efficacy of LC to better clarify the theory of TCM and guide the clinical application of Yin-Jing drugs. Different Physalis plants have been widely employed in traditional medicine for management of diabetes mellitus. Previous studies with respect to the in vivo antidiabetic activity of Physalis plants illustrated that they improved glucose and lipid metabolism in streptozotocin (STZ) -induced diabetic rats yet the mechanism of action of bioactive constituents of the different organs of Physalis plants on diabetes remains obscure. Our objective is to study the effects of the different organs of ground cherry (P. pruinosa) on diabetes in rat models and elucidate their mechanism of actions through serum pharmacochemistry combined to network pharmacology analyses and in-vivo testing. Characterization of the constituents in the drug-dosed serum samples relative to the blank serum after treatment with different extracts was performed by UPLC -MS/MS technique. The absorbed metabolites where then subjected to network pharmacology analysis to construct an interaction network linking "compound-target-pathway". In vivo verification was implemented to determine a hypothesized mechanism of action on a STZ and high fat diet induced type II diabetes mellitus (T2DM) model based on functional and enrichment analyses of the Kyoto Encyclopedia of Genes and Genome and Gene Ontology. Identification of a total of 73 compounds (22 prototypes and 51 metabolites) derived from P. pruinosa extracts was achieved through comparison of the serum samples collected from diabetic control group and extracts treated groups. The identified compounds were found to belong to different classes according to their structural type including withanolides, physalins and flavonoids. The absorbed compounds in the analyzed serum samples were considered as the potential bioactive components. The component-target network was found to have 23 nodes with 17 target genes including MAPK8, CYP1A1 and CYP1B1. Quercetin and withaferin A were found to possess the highest combined score in the C-T network. Integrated serum pharmacochemistry and network pharmacology analyses revealed the enrichment of leaves extract with the active constituents, which can be utilized in T2DM treatment. In the top KEGG pathways, lipid and atherosclerosis metabolic pathways in addition to T2DM pathways were found to be highly prioritized. The diabetic rats, which received leaves extract exhibited a substantial increment in GLUT2, INSR, IRS-1 , PI 3 K-p85 and AKT-ser473 proteins by 105%, 142%, 109%, 81% and 73%, respectively relative to the untreated diabetic group. The immunoblotting performed for MAPK and ERK1/2 part of the inflammatory pathway studied in STZ induced diabetic rats revealed that leaves, calyces and stems extracts resulted in a substantial diminish in p38-MAPK, ERK 1/2 , NF-κB, and TNF-α. Histopathological examination revealed that the hepatic histoarchitecture was substantially improved in the leaves, stems, and clayces-treated rats in comparison with untreated diabetic rats. Further, pancreatic injuries, which induced by STZ were dramatically altered by the treatment with P. pruinosa leaves, calyces and stems extracts. β-cells in diabetic rats received leaves extract disclosed moderate insulin immunostaining with a notable increase in the mean insulin area%. The study in hand offers a comprehensive study to clarify the bioactive metabolites of the different organs of P. pruinosa.
