Most car-following models have mainly focused on the effects of the lead vehicle on the target vehicle's behaviour or the driver's forward-looking effects, but not the effects of the vehicle behind the target vehicle (the following vehicle) or the driver's backward-looking effects. Therefore, this study proposes a data-driven car-following model that incorporates both backward- and forward-looking effects using a deep neural network (DNN). This model is called the "DNN with backward-looking effect (DNN-BE) model". The DNN-BE model produced higher prediction accuracy than the DNN model with forward-looking effects only and a conventional mathematical car-following model that considers both forward- and backward-looking effects. It was found that the target vehicle is more likely to accelerate when the spacing with the following vehicle is shorter and the spacing with the lead vehicle is longer. Introduction: The prediction of when the driver will react to a change in the lead vehicle motion is critical for assessing rear-end crash risk using car-following models. Past studies have assumed constant reaction time and driver's continuous reaction. However, these assumptions are not valid as the driver's reaction time can vary in different car-following situations and the driver does not continuously react to the lead vehicle motion. Thus, this study predicted the driver's reaction time using the Wiedemann car-following model and the Accumulator model. The Accumulator model assumes the driver's start of reaction based on the accumulation of looming and thereby reflects the driver's intermittent reaction. Method: Fifty drivers' behavior was observed using a driving simulator in two scenarios: (1) approach and follow a moving lead vehicle and (2) approach a stopped lead vehicle. The Accumulator model predicted the reaction times based on different looming variables (angular velocity and tau-inverse), lead vehicle type (car and truck), and lead vehicle brake lights (on or off). Results: The Accumulator model showed lower prediction errors of the reaction time than the Wiedemann model, which assumes reaction based on the fixed looming threshold. The Accumulator model predicted the reaction times more accurately when it was calibrated with the angular velocity due to width and height of lead vehicles. Moreover, the Accumulator model with tau-inverse produced the smallest prediction error of reaction times among different Accumulator models and the Wiedemann model when lead vehicle brake lights were on. Conclusions: This study demonstrates that the Accumulator model is a promising method of predicting the driver's reaction time in car-following situations, which affects rear-end crash risk. Practical Applications: The Accumulator model can be incorporated into a car-following model for the prediction of reaction times and can estimate the rear-end collision risk of vehicles more accurately. This study discusses the effect of semi-supervised learning in combination with pretrained language models for data-to-text generation. It is not known whether semi-supervised learning is still helpful when a large-scale language model is also supplemented. This study aims to answer this question by comparing a data-to-text system only supplemented with a language model, to two data-to-text systems that are additionally enriched by a data augmentation or a pseudo-labeling semi-supervised learning approach. Results show that semi-supervised learning results in higher scores on diversity metrics. In terms of output quality, extending the training set of a data-to-text system with a language model using the pseudo-labeling approach did increase text quality scores, but the data augmentation approach yielded similar scores to the system without training set extension. This study examines the impacts of cooperative adaptive cruise control (CACC) and cooperative lane changing (CLC) on the delay and the riding comfort in autonomous car–autonomous truck (AC–AT) mixed traffic at a freeway merging area. For this task, AC–AT mixed traffic on a 5.25 km section of freeway was analyzed using the Aimsun Next microscopic traffic simulation. The effects of different CACC parameters and CLC on the average speed and acceleration distributions as the measures of delay and riding comfort, respectively, were evaluated. It was found that (1) lower sensitivity to the lead vehicle reduced the merging time, (2) shorter time gaps between autonomous vehicles and between platoons decreased the delay, and (3) longer time gaps reduced the delay at higher percentage of ATs. These results demonstrate that the delay of AC–AT mixed traffic at a freeway merging area can be reduced and riding comfort can be increased by adjusting CACC parameters. [ABSTRACT FROM AUTHOR]

Bile acid metabolism is altered in neonates on parenteral nutrition (PN), predisposing them to parenteral nutrition-associated liver disease. Cholesterol 7α-hydroxylase (CYP7A1), the rate-limiting enzyme in the bile acid synthesis pathway, is repressed by fibroblast growth factor 19 (FGF19) and phytosterols (PS). We describe a case of a preterm infant who developed necrotizing enterocolitis (NEC) and received exclusive PN for over 2 months. Our objective was to serially assess CYP7A1 activity and plasma FGF19 and PS concentrations in this infant case compared to five healthy preterm infants. We found that CYP7A1 activity increased during the first 2 weeks of life in control infants but was undetectable in the infant case. FGF19 concentrations were high at birth in all infants and subsequently declined and did not differ between the case and control infants. As expected, PS concentrations were elevated in the infant case and continued to increase despite lipid minimization. In conclusion, CYP7A1 activity was gradually upregulated in healthy preterm infants but remained suppressed in the infant requiring prolonged PN. Introduction: Driver's evasive action is closely associated with collision risk in a critical traffic event. To quantify collision risk, surrogate safety measures (SSMs) have been estimated using vehicle trajectories. However, vehicle trajectories cannot clearly capture presence and time of driver's evasive action. Thus, this study determines the driver's evasive action based on his/her use of accelerator and brake pedals, and analyzes the effects of the driver's evasive action time (i.e., duration of evasive action) on rear-end collision risk. Method: Fifty drivers' car-following behavior on a freeway was observed using a driving simulator. An SSM called "Deceleration Rate to Avoid Crash (DRAC)" and the evasive action time were determined for each driver using the data from the driving simulator. Each driver tested two traffic scenarios – Cars and Trucks scenarios where conflicting vehicles were cars and trucks, respectively. The factors related to DRAC were identified and their effects on DRAC were analyzed using the Generalized Linear Models and random effects models. Results: DRAC decreased with the evasive action time and DRAC was closely related to drivers' gender and driving experience at the road sections where evasive action to avoid collision was required. DRAC was also significantly different between Cars and Trucks scenarios. The effect of the evasive action time on DRAC varied among different drivers, particularly in the Trucks scenario. Conclusions: Longer evasive action time can significantly reduce crash risk. Driver characteristics are more closely related to effective evasive action in complex driving conditions. Practical Applications: Based on the findings of this study, driver warning information can be developed to alert drivers to take specific evasive action that reduces collision risk in a critical traffic event. Chatbots have several features that may stimulate self-disclosure, such as accessibility, anonymity, convenience and their perceived non-judgmental nature. The aim of this study is to investigate if people disclose (more) intimate information to a chatbot, compared to a human, and to what extent this enhances their emotional well-being through feelings of relief. An experiment with a 2 (human vs. chatbot) by 2 (low empathetic vs. high empathetic) design was conducted (N  = 286). Results showed that there was no difference in the self-reported intimacy of self-disclosure between the human and chatbot conditions. Furthermore, people perceived less fear of judgment in the chatbot condition, but more trust in the human interactant compared to the chatbot interactant. Perceived anonymity was the only variable to directly impact self-disclosure intimacy. The finding that humans disclose equally intimate information to chatbots and humans is in line with the CASA paradigm, which states that people can react in a social manner to both computers and humans. Many ultra-diffuse galaxies (UDGs) have been discovered in the Coma cluster, and there is evidence that some, notably Dragonfly 44, have Milky Way-like dynamical masses despite dwarf-like stellar masses. We used X-ray, ultraviolet (UV), and optical data to investigate the star formation and nuclear activity in the Coma UDGs, and we obtained deep UV and X-ray data (Swift and XMM–Newton) for Dragonfly 44 to search for low-level star formation, hot circumgalactic gas, and the integrated emission from X-ray binaries. Among the Coma UDGs, we find UV luminosities consistent with quiescence but NUV − r colours indicating star formation in the past Gyr. This indicates that the UDGs were recently quenched. The r -band luminosity declines with projected distance from the Coma core. The Dragonfly 44 UV luminosity is also consistent with quiescence, with SFR |$\lt 6\times 10^{-4} \, \mathrm{M}_{\odot }$|  yr−1, and no X-rays are detected down to a sensitivity of 1038 erg s−1. This rules out a hot corona with a |$M \gt 10^8 \, \mathrm{M}_{\odot }$| within the virial radius, which would normally be expected for a dynamically massive galaxy. The absence of bright, low-mass X-ray binaries is consistent with the expectation from the Galaxy total stellar mass, but it is unlikely if most low-mass X-ray binaries form in globular clusters, as Dragonfly 44 has a very large population. Based on the UV and X-ray analysis, the Coma UDGs are consistent with quenched dwarf galaxies, although we cannot rule out a dynamically massive population. [ABSTRACT FROM AUTHOR]

This paper examines the tone-merging phenomenon in Hong Kong Cantonese. Both perception and production tasks were administered to 120 participants with ages ranging from 20 to 58 years. After considering the complicated interplay of perception and production confusion, the paper provides statistical evidence that three tonal contrasts have undergone merging in contemporary Hong Kong Cantonese. They are Full-merger T2 and T5, where contrast is collapsed in both perception and production; Partial-merger T3 and T6, where contrast is collapsed in production only; and Near-merger T4 and T6, where contrast is collapsed in perception but maintained in production. The kindergarten program, Tools of the Mind (Tools), has been shown to improve executive functions (as assessed by laboratory measures) and academic performance. The objective here was to see if Tools can improve executive functions in the real world (in the classroom), academic outcomes not previously investigated, reduce bullying and peer ostracism, and increase teachers’ and students’ joy in being in the classroom. This first randomized controlled trial of Tools in Canada included 351 kindergarten children (mean age 5.2 years at entry; 51% female) in 18 public schools. Stratified randomization resulted in teachers and students in both groups being closely matched. Teachers in both groups received the same number of training hours and same funds for new materials. Outcome measures were pre and post standardized academic skill assessments and teacher online survey responses. This study replicated that Tools improves reading and shows for the first time that it improves writing (far exceeding levels the school districts had seen before), self-control and attention-regulation in the real world (e.g., time on task without supervision), reduces teacher burnout and children being ostracized or excluded, and increases the joy students and teachers experience in school. By Spring, Tools teachers were still enthusiastic about teaching; control teachers were exhausted. These results were not only better than the control group but also better than Tools teachers experienced the year before Tools. Thus, children in a kindergarten curriculum that emphasized play, improving self-regulation, working together and helping one another, and hands-on learning performed better academically, showed less bullying and peer ostracism and more kindness and helping behavior than students in more traditional classes, and teacher enthusiasm for teaching soared. • Cyst nematodes deliver effector proteins into host cells to manipulate cellular processes and establish a metabolically hyperactive feeding site. The novel 30D08 effector protein is produced in the dorsal gland of parasitic juveniles, but its function has remained unknown. • We demonstrate that expression of 30D08 contributes to nematode parasitism, the protein is packaged into secretory granules and it is targeted to the plant nucleus where it interacts with SMU2 (homolog of suppressor of mec-8 and unc-52 2), an auxiliary spliceosomal protein. • We show that SMU2 is expressed in feeding sites and an smu2 mutant is less susceptible to nematode infection. In Arabidopsis expressing 30D08 under the SMU2 promoter, several genes were found to be alternatively spliced and the most abundant functional classes represented among differentially expressed genes were involved in RNA processing, transcription and binding, as well as in development, and hormone and secondary metabolism, representing key cellular processes known to be important for feeding site formation. • In conclusion, we demonstrated that the 30D08 effector is secreted from the nematode and targeted to the plant nucleus where its interaction with a host auxiliary spliceosomal protein may alter the pre-mRNA splicing and expression of a subset of genes important for feeding site formation. Haploinsufficiency of GRN, the gene encoding progranulin (PGRN), causes frontotemporal lobar degeneration (FTLD), the second most common cause of early-onset dementia. Receptor-mediated lysosomal targeting has been shown to regulate brain PGRN levels, and complete deficiency of PGRN is a direct cause of neuronal ceroid lipofuscinosis (NCL), a lysosomal storage disease. Here we show that the lysosomal cysteine protease cathepsin L (Cat L) can mediate the proteolytic cleavage of intracellular PGRN into poly-granulin and granulin fragments. Further, PGRN and Cat L co-localize in lysosomes of HEK293 cells, iPSC-derived neurons and human cortical neurons from human postmortem tissue. These data identify Cat L as a key intracellular lysosomal PGRN protease, and provides an intriguing new link between lysosomal dysfunction and FTLD. [ABSTRACT FROM AUTHOR]

Background/Aims: The small molecule, Tolfenamic acid (TA) has shown anti-cancer activity in pre-clinical models and is currently in Phase I clinical trials at MD Anderson Cancer Center Orlando. Since specificity and toxicity are major concerns for investigational agents, we tested the effect of TA on specific targets, and assessed the cellular and organismal toxicity representing pre-clinical studies in cancer. Methods: Panc1, L3.6pl, and MiaPaCa-2 (pancreatic cancer), hTERT-HPNE(normal), and differentiated/un-differentiated SH-SY5Y (neuroblastoma) cells were treated with increasing concentrations of TA. Cell viability and effect on specific molecular targets, Sp1 and survivin were determined. Athymic nude mice were treated with vehicle or TA (50mg/kg, 3times/week for 6 weeks) and alterations in the growth pattern, hematocrit, and histopathology of gut, liver, and stomach were monitored. Results: TA treatment decreased cell proliferation and inhibited the expression of Sp1 and survivin in cancer cells while only subtle response was observed in normal (hTERT-HPNE) and differentiated SH-SY5Y cells. Mice studies revealed no effect on body weight and hematocrit. Furthermore, TA regimen did not cause signs of internal-bleeding or damage to vital tissues in mice. Conclusion: These results demonstrate that TA selectively inhibits malignant cell growth acting on specific targets and its chronic treatment did not cause apparent toxicity in nude mice. © 2013 S. Foil and vibrating wire strain gauges have an optimum strain measurement accuracy of one microstrain. However, they can only provide discrete strain readings over a single fixed-gauge length. Digital image correlation (DIC) offers an alternative to conventional strain gauges because a two-dimensional (2D) surface strain field can be obtained from a single sensor (camera). However, the benefits of 2D strain measurements are only worthwhile if a similar level of measurement accuracy to conventional strain gauges can be achieved. This paper presents the results of an investigation into the optimum strain measurement accuracy that can be achieved by using the 2D technique on artificial images (which eliminate errors associated with cameras and lighting). The principle of the 2D DIC technique and its historical development will be introduced. Then, three potential techniques for taking strain measurements will be presented and compared: single readings, averaged linear readings, and an approach on the basis of Mohr's circle. The Mohr's circle approach was found to be the most accurate and was not susceptible to image misalignment. Strain measurement accuracy was also found to be affected by the bias error of the subpixel interpolation scheme, but the use of an 8 coefficient B-spline was found to produce satisfactory results within the error of conventional strain gauges. Gauge length was also found to have a significant effect on strain measurement accuracy, indicating that measuring strains in a material in which there are variations across the strain field could result in a loss of measurement accuracy. Plant-parasitic cyst nematodes penetrate plant roots and transform cells near the vasculature into specialized feeding sites called syncytia. Syncytia form by incorporating neighboring cells into a single fused cell by cell wall dissolution. This process is initiated via injection of esophageal gland cell effector proteins from the nematode stylet into the host cell. Once inside the cell, these proteins may interact with host proteins that regulate the phytohormone auxin, as cellular concentrations of auxin increase in developing syncytia. Soybean cyst nematode (Heterodera glycines) Hgl9C07 is a novel effector protein expressed specifically in the dorsal gland cell during nematode parasitism. Here, we describe its ortholog in the beet cyst nematode (Heterodera schachtii), Hsl9C07. We demonstrate that Hsl9C07 interacts with the Arabidopsis (Arabidopsis thaliana) auxin influx transporter LAX3. LAX3 is expressed in cells overlying lateral root primordia, providing auxin signaling that triggers the expression of cell wall-modifying enzymes, allowing lateral roots to emerge. We found that LAX3 and polygalacturonase, a LAX3-induced cell wall-modifying enzyme, are expressed in the developing syncytium and in cells to be incorporated into the syncytium. We observed no decrease in H. schachtii infectivity in auxl and Iax3 single mutants. However, a decrease was observed in both the auxllax3 double mutant and the auxllaxllax21ax3 quadruple mutant. In addition, ectopic expression of 19C07 was found to speed up lateral root emergence. We propose that Hsl9C07 most likely increases LAX3-mediated auxin influx and may provide a mechanism for cyst nematodes to modulate auxin flow into root cells, stimulating cell wall hydrolysis for syncytium development. •Soybean cyst nematodes ( Heterodera glycines) produce secreted effector proteins that function as peptide mimics of plant CLAVATA3/ESR (CLE)-like peptides probably involved in the developmental reprogramming of root cells to form specialized feeding cells called syncytia. •The site of action and mechanism of delivery of CLE effectors to host plant cells by the nematode, however, have not been established. In this study, immunologic, genetic and biochemical approaches were used to reveal the localization and site of action of H. glycines-secreted CLE proteins in planta. •We present evidence indicating that the nematode CLE propeptides are delivered to the cytoplasm of syncytial cells, but ultimately function in the apoplast, consistent with their proposed role as ligand mimics of plant CLE peptides. We determined that the nematode 12-amino-acid CLE motif peptide is not sufficient for biological activity in vivo, pointing to an important role for sequences upstream of the CLE motif in function. •Genetic and biochemical analysis confirmed the requirement of the variable domain in planta for host-specific recognition and revealed a novel role in trafficking cytoplasmically delivered CLEs to the apoplast in order to function as ligand mimics. To arrest rockfall boulders, baffles are constructed as steel members affixed on top of a concrete footing. Existing designs aim to prevent any footing rotation and steel member bending. As such, existing baffle designs require massive reinforced concrete footings and complex bolt and weld connections. Such designs have limited constructability and result in complex failure mechanisms (i.e., tensile failure of concrete foundation at connections). In this paper, we introduce a new baffle design concept that optimizes the balance between steel member bending and footing rotation to enable a compact design. Full-scale pendulum experiments are conducted to evaluate the proposed easy-to-construct and design baffle under successive boulder impacts of 50 kJ. It is demonstrated that by considering the effects of steel member deflection and concrete foundation rotation, the maximum impact force on baffles is reduced by up to two times when compared with those on a rigid barrier and the failure modes are simplified to either overturning failure of the footing or buckling failure of the steel member. A ratio that defines the flexural capacity of the steel member to the overturning capacity of the shallow concrete foundation is proposed to control the failure mode. The optimized baffle configuration for an impact energy of 50 kJ, with minimum residual deflection, has a target capacity ratio of 2.0. • A capacity ratio (flexural capacity of steel member to overturning capacity of footing) is proposed for baffle design. • A new baffle design concept is proposed and evaluated using full-scale pendulum experiments. • A failure criterion based on baffle rotation is proposed considering the residual boulder velocity. Abstract: The deposition of carbon layers on the surfaces of optics exposed to extreme ultraviolet (EUV) radiation has been observed in EUV lithography. It has become of critical importance to detect the presence of the carbon layer in the order of nanometer thickness due to carbon''s extremely strong absorption of EUV radiation. Furthermore, the development of efficient cleaning strategies requires that the nature of these carbon layers is well understood. Here, we present experimental results on the detection and characterization of carbon layers, grown on Mo/Si EUV reflecting optics, by laser-generated surface acoustic waves (LG-SAW). It was found that SAW pulses with a frequency bandwidth of more than 220 MHz can be generated and detected for multilayer mirrors and LG-SAW is sensitive enough to detect EUV induced carbon layers less than 5 nm thick. It was inferred from the low Young''s modulus (<100 GPa) that the carbon layer induced by EUV illumination in these vacuum conditions is mechanically soft and polymeric in nature with a high percentage of hydrogen. Objectives: Proposals to make decisions about coverage of new technology by comparing the technology's incremental cost-effectiveness with the traditional benchmark of dialysis imply that the incremental cost-effectiveness ratio of dialysis is seen a proxy for the value of a statistical year of life. The frequently used ratio for dialysis has, however, not been updated to reflect more recently available data on dialysis. Methods: We developed a computer simulation model for the end-stage renal disease population and compared cost, life expectancy, and quality-adjusted life expectancy of current dialysis practice relative to three less costly alternatives and to no dialysis. We estimated incremental cost-effectiveness ratios for these alternatives relative to the next least costly alternative and no dialysis and analyzed the population distribution of the ratios. Model parameters and costs were estimated using data from the Medicare population and a large integrated health-care delivery system between 1996 and 2003. The sensitivity of results to model assumptions was tested using 38 scenarios of one-way sensitivity analysis, where parameters informing the cost, utility, mortality and morbidity, etc. components of the model were by perturbed +/−50%. Results: The incremental cost-effectiveness ratio of dialysis of current practice relative to the next least costly alternative is on average $129,090 per quality-adjusted life-year (QALY) ($61,294 per year), but its distribution within the population is wide; the interquartile range is $71,890 per QALY, while the 1st and 99th percentiles are $65,496 and $488,360 per QALY, respectively. Higher incremental cost-effectiveness ratios were associated with older age and more comorbid conditions. Sensitivity to model parameters was comparatively small, with most of the scenarios leading to a change of less than 10% in the ratio. Conclusions: The value of a statistical year of life implied by dialysis practice currently averages $129,090 per QALY ($61,294 per year), but is distributed widely within the dialysis population. The spread suggests that coverage decisions using dialysis as the benchmark may need to incorporate percentile values (which are higher than the average) to be consistent with the Rawlsian principles of justice of preserving the rights and interests of society's most vulnerable patient groups. This paper makes use of a probabilistic model that predicts the likelihood of crashes (crash potential) on freeways on the basis of traffic flow conditions, in real-time crash prevention. The model was developed using incident logs and loop detector data collected over a 13-month period on the Gardiner Expressway in Toronto. Previous work suggested that an increase in levels of traffic turbulence generally yields high crash potential. Traffic turbulence was defined in terms of a series of crash precursors that represent traffic conditions that were present prior to crash occurrence. To apply the model in crash prevention, the link needs to be established between crash potential and real-time safety intervention. The objective of this paper is to explore this link for different thresholds of crash potential. The paper discusses the guidelines for evaluating the safety benefit of one crash prevention strategy (variable speed limits) and suggests the risk-based evaluation framework for real-time traffic control. Currently, there is little agreement as to how Natural Language Generation (NLG) systems should be evaluated, with a particularly high degree of variation in the way that human evaluation is carried out. This paper provides an overview of how (mostly intrinsic) human evaluation is currently conducted and presents a set of best practices, grounded in the literature. These best practices are also linked to the stages that researchers go through when conducting an evaluation research (planning stage; execution and release stage), and the specific steps in these stages. In this paper, we present an algorithm developed to detect Earth-orbiting object streaks within astronomical images. The NASA Wide-field Infrared Survey Explorer (WISE) database of imagery was selected, in order to assess the viability of infrared wavelengths for the detection and analysis of satellite, rocket body and debris objects. The algorithm was designed to scan a high volume of images for the presence of streaks and then to correlate these to known Earth-orbiting objects without the use of a-priori knowledge of position or orbit parameters of the detected object. As part of the streak analysis process, the algorithm performed a photometric characterisation of the detected objects in all four infrared bands observed by the WISE satellite. Accumulation of amyloid-b peptide (Aβ) aggregates in synapses may contribute to the profound synaptic loss characteristic of Alzheimer's disease (AD). The origin of synaptic Aβ aggregates remains elusive, but loss of endosomal proteostasis may trigger their formation. In this study, we identified the synaptic compartments where Aβ accumulates, and performed a longitudinal analysis of synaptosomes isolated from brains of TgCRND8 APP transgenic mice of either sex. To evaluate the specific contribution of Aβ-degrading protease endothelin-converting enzyme (ECE-1) to synaptic/endosomal Aβ homeostasis, we analyzed the effect of partial Ece1 KO in brain and complete ECE1 KO in SH-SY5Y cells. Global inhibition of ECE family members was used to further assess their role in preventing synaptic Aβ accumulation. Results showed that, before extracellular amyloid deposition, synapses were burdened with detergent-soluble Aβ monomers, oligomers, and fibrils. Levels of all soluble Aβ species declined thereafter, as Aβ42 turned progressively insoluble and accumulated in Aβ-producing synaptic endosomal vesicles with characteristics of multivesicular bodies. Accordingly, fibrillar Aβ was detected in brain exosomes. ECE-1-deficient mice had significantly increased endogenous synaptosomal Aβ42 levels, and protease inhibitor experiments showed that, in TgCRND8 mice, synaptic Aβ42 became nearly resistant to degradation by ECE-related proteases. Our study supports that Aβ accumulating in synapses is produced locally, within endosomes, and does not require the presence of amyloid plaques. ECE-1 is a determinant factor controlling the accumulation and fibrillization of nascent Aβ in endosomes and, in TgCRND8 mice, Aβ overproduction causes rapid loss of Aβ42 solubility that curtails ECE-mediated degradation. Aims: Changes in serotonergic sensory modulation associated with overexpression of 5‐HT3 receptors in the central nervous system (CNS) have been implicated in the pathophysiology of neuropathic pain after peripheral nerve damage. 5‐HT3 receptor antagonists such as ondansetron can potentially alleviate neuropathic pain, but have limited effectiveness, due potentially to limited CNS access. However, there is currently limited information on CNS disposition of systemically‐administered 5‐HT3 receptor antagonists. This study evaluated the cerebrospinal fluid (CSF) disposition of ondansetron, as a surrogate of CNS penetration. Methods: Fifteen patients were given a single 16 mg intravenous 15 minute infusion of ondansetron, followed by serial blood and a single CSF sampling. Population pharmacokinetic (PK) modelling was implemented to describe the average and individual plasma and CSF profiles of ondansetron. A two‐compartmental model was used to capture ondansetron plasma PK with a single CSF compartment to describe distribution to the CNS. Results: The individual model‐estimated CSF to plasma partition coefficients of ondansetron were between 0.09 and 0.20. These values were mirrored in the calculated CSF penetration ratios, ranging from 0.08 to 0.26. Conclusions: After intravenous administration, CSF concentrations of ondansetron were approximately 7‐fold lower than those observed in the plasma. A model could be developed to describe individual CSF concentration–time profiles of ondansetron based on a single CSF data point. The low CSF penetration of ondansetron may explain its limited analgesic effectiveness, and affords an opportunity to explore enhancing its CNS penetration for targeting conditions such as neuropathic pain. Krabbe disease (KD) is a progressive and devasting neurological disorder that leads to the toxic accumulation of psychosine in the white matter of the central nervous system (CNS). The condition is inherited via biallelic, loss-of-function mutations in the galactosylceramidase (GALC) gene. To rescue GALC gene function in the CNS of the twitcher mouse model of KD, an adeno-associated virus serotype 1 vector expressing murine GALC under control of a chicken β-actin promoter (AAV1-GALC) was administered to newborn mice by unilateral intracerebroventricular injection. AAV1-GALC treatment significantly improved body weight gain and survival of the twitcher mice (n = 8) when compared with untreated controls (n = 5). The maximum weight gain after postnatal day 10 was significantly increased from 81% to 217%. The median lifespan was extended from 43 days to 78 days (range: 74–88 days) in the AAV1-GALC-treated group. Widespread expression of GALC protein and alleviation of KD neuropathology were detected in the CNS of the treated mice when examined at the moribund stage. Functionally, elevated levels of psychosine were completely normalized in the forebrain region of the treated mice. In the posterior region, which includes the mid- and the hindbrain, psychosine was reduced by an average of 77% (range: 53–93%) compared to the controls. Notably, psychosine levels in this region were inversely correlated with body weight and lifespan of AAV1-GALC-treated mice, suggesting that the degree of viral transduction of posterior brain regions following ventricular injection determined treatment efficacy on growth and survivability, respectively. Overall, our results suggest that viral vector delivery via the cerebroventricular system can partially correct psychosine accumulation in brain that leads to slower disease progression in KD. Polarization provides useful quantitative information about scattering surfaces. In hyperspectral remote sensing of natural surfaces composed of granular materials, there are relatively few studies of polarization. Most earlier remote sensing studies of polarization have been based on multi-spectral data, and the majority focused on the negative branch of polarization, which typically appears at phase angles less than 20 degrees, using models with limited accuracy. Models of the positive branch have also shown limitations, particularly at longer phase angles. We review these earlier studies by Hapke and Shkuratov and present the results of our laboratory study using hyperspectral polarization imagery of particulate surfaces. Although the linear polarization ratio is typically a nonlinear function of phase angle, our results show that in an approximately linear region of the polarization curve, there is a correlation between the slope of the linear polarization ratio and the average grain size. This study investigates the impact of advanced driver-assistance systems on drivers' mental workload. Using a combination of physiological signals including ECG, EMG, EDA, EEG (af4 and fc6 channels from the theta band), and eye diameter data, this study aims to predict and categorize drivers' mental workload into low, adequate, and high levels. Data were collected from five different driving situations with varying cognitive demands. A functional linear regression model was employed for prediction, and the accuracy rate was calculated. Among the 31 tested combinations of physiological variables, 9 combinations achieved the highest accuracy result of 90%. These results highlight the potential benefits of utilizing raw physiological signal data and employing functional data analysis methods to understand and assess driver mental workload. Background: A significant proportion of children and young people with juvenile idiopathic arthritis (JIA) do not achieve inactive disease during the first two years following diagnosis. Refinements to clinical care pathways have the potential to improve clinical outcomes but a lack of consistent and contemporaneous clinical data presently precludes standard setting and implementation of meaningful quality improvement programmes. This study was the first to pilot clinical data collection and analysis using the CAPTURE-JIA dataset, and to explore patient and clinician-reported feasibility and acceptability data. Methods: A multiphase mixed-methods approach enabled prospective collection of quantitative data to examine the feasibility and efficacy of dataset collection and of qualitative data informing the context and processes of implementation. An initial paper pilot informed the design of a bespoke electronic data collection system (the Agileware system), with a subsequent electronic pilot informing the final CAPTURE-JIA data collection tool. Results: Paper collection of patient data was feasible but time-consuming in the clinical setting. Phase 1 paper pilot data (121 patients) identified three themes: problematic data items (14/62 data items received >40% missing data), formatting of data collection forms and a clinician-highlighted need for digital data collection, informing Phase 2 electronic data collection tool development. Patients and families were universally supportive of the collection and analysis of anonymised patient data to inform clinical care. No apparent preference for paper / electronic data collection was reported by families. Phase 3 electronic pilot data (38 patients) appeared complete and the system reported to be easy to use. Analysis of the study dataset and a dummy longitudinal dataset confirmed that all eleven JIA national audit questions can be answered using the electronic system. Conclusions: Multicentre CAPTURE-JIA data collection is feasible and acceptable, with a bespoke data collection system highlighted as the most satisfactory solution. Abstract: The measurement of strain has always been central to developments in structural engineering. As structures around the world reach the end of their intended service life, there is a need for more robust field monitoring techniques to determine, in conjunction with numerical models, if these structures are still fit for purpose. Current strain gage technologies are only able to offer a limited number of discrete measurements, which often is not enough data to assess complex structures. This paper presents an alternative technique for measuring strain using digital images called Digital Image Correlation (DIC) that allows for the measurement of 2-D strain fields. The accuracy that can be achieved in physical experiments where sources of error such as lighting and camera quality are present is investigated. A series of tension tests on steel plates are conducted allowing measurements from the DIC technique to be compared to measurements from conventional foil strain gages. Out-of-plane motion is identified as one of the most significant potential sources of strain measurement error when using DIC. Five solutions for significantly reducing the effects of out-of-plane motion are presented and three of these solutions are applied to the steel plate experiment. Using these solutions it is possible to achieve mean strain errors of less than 5με when comparing DIC to foil strain gage measurements, which suggests that the DIC technique has the potential to replace conventional strain gages. The risk of developing cancer caused by environmental carcinogens lies in the balance between phase I carcinogen-activating enzymes and phase II detoxifying enzymes. Potentially genotoxic chemicals to which we are exposed to may require metabolic activation to exhibit their mutagenic and carcinogenic effects. Most of the enzyme systems, involved in carcinogen bioactivation, are concentrated in liver, but many extrahepatic organs and tissues have appreciable quantities of such enzymes. Most chemical carcinogens are activated in humans by specific cytochrome P450 (CYP) species, mainly CYP1A1, CYP1A2, CYP2E, and CYP3A. Activation of chemical carcinogens follows various metabolic pathways. The difference could range from only a single oxidation step or several sequential enzymatic steps. Polymorphic differences in carcinogen metabolism may result in difference in carcinogen risk since fast activators and/or slow detoxifiers may have higher steady state tissue concentration of the causative agent than slow activator and/or fast detoxifiers. Cancer prevention through better diet and nutrition has received considerable attention in the past and continues to show promise even today.” It has become increasingly apparent that adopting a diet may allow reduction in cancer incidence worldwide by functioning as inhibitors of tumor initiation, promotion, and progression, which may be mediated by either selective induction or inhibition of the hepatic xenobiotic metabolizing enzymes. Abstract: Problem: There is a growing concern with the safety of school-aged children. This study identifies the locations of pedestrian/bicyclist crashes involving school-aged children and examines the conditions when these crashes are more likely to occur. Method: The 5-year records of crashes in Orange County, Florida where school-aged children were involved were used. The spatial distribution of these crashes was investigated using the Geographic Information Systems (GIS) and the likelihoods of crash occurrence under different conditions were estimated using log-linear models. Results: A majority of school-aged children crashes occurred in the areas near schools. Although elementary school children were generally very involved, middle and high school children were more involved in crashes, particularly on high-speed multi-lane roadways. Driver''s age, gender, and alcohol use, pedestrian''s/bicyclist''s age, number of lanes, median type, speed limits, and speed ratio were also found to be correlated with the frequency of crashes. Discussion: The result confirms that school-aged children are exposed to high crash risk near schools. High crash involvement of middle and high school children reflects that middle and high schools tend to be located near multi-lane high-speed roads. Impact on Industry: The pedestrian''s/bicyclist''s demographic factors and geometric characteristics of the roads adjacent to schools associated with school children''s crash involvement are of interest to school districts. Our objective was to examine differences in cytokine/chemokine response in chronic hepatitis B(CHB) patients to understand the immune mechanism of HBsAg loss (functional cure) during antiviral therapy. We used an unbiased machine learning strategy to unravel the immune pathways in CHB nucleo(t)side analogue-treated patients who achieved HBsAg loss with peg-interferon-α(peg-IFN-α) add-on or switch treatment in a randomised clinical trial. Cytokines/chemokines from plasma were compared between those with/without HBsAg loss, at baseline, before and after HBsAg loss. Peg-IFN-α treatment resulted in higher levels of IL-27, IL-12p70, IL-18, IL-13, IL-4, IL-22 and GM-CSF prior to HBsAg loss. Probabilistic network analysis of cytokines, chemokines and soluble factors suggested a dynamic dendritic cell driven NK and T cell immune response associated with HBsAg loss. Bayesian network analysis showed a dominant myeloid-driven type 1 inflammatory response with a MIG and I-TAC central module contributing to HBsAg loss in the add-on arm. In the switch arm, HBsAg loss was associated with a T cell activation module exemplified by high levels of CD40L suggesting T cell activation. Our findings show that more than one immune pathway to HBsAg loss was found with peg-IFN-α therapy; by myeloid-driven Type 1 response in one instance, and T cell activation in the other. The aim of the present study is to compare the safety, efficacy and cost effectiveness of anesthetic regimens by compound, using etomidate and propofol in elderly patients undergoing gastroscopy. A total of 200 volunteers (65-79 years of age) scheduled for gastroscopy under anesthesia were randomly divided into the following groups: P, propofol (1.5-2.0 mg/kg); E, etomidate (0.15-0.2 mg/kg); P+E, propofol (0.75-1 mg/kg) followed by etomidate (0.075-0.1 mg/kg); and E+P, etomidate (0.075-0.01 mg/kg) followed by propofol (0.75-1 mg/kg). Vital signs and bispectral index were monitored at different time points. Complications, induction and examination time, anesthesia duration, and recovery and discharge time were recorded. At the end of the procedure, the satisfaction of patients, endoscopists and the anesthetist were evaluated. The recovery (6.1±1.2 h) and discharge times (24.8±2.8 h) in group E were significantly longer compared with groups P, P+E and E+P (P<0.05). The occurrence of injection pain in group P+E was significantly higher compared with the other three groups (P<0.05). In addition, the incidence of myoclonus and post-operative nausea and vomiting were significantly higher in group P+E compared with the other three groups (P<0.05). There was no statistical difference among the four groups with regards to the patients' immediate, post-procedure satisfaction (P>0.05). Furthermore, there was no difference in the satisfaction of anesthesia, as evaluated by the anesthetist and endoscopist, among the four groups (P>0.05). The present study demonstrates that anesthesia for gastroscopy in elderly patients can be safely and effectively accomplished using a drug regimen that combines propofol with etomidate. The combined use of propofol and etomidate has unique characteristics which improve hemodynamic stability, cause minimal respiratory depression and less side effects, provide rapid return to full activity and result in high levels of satisfaction. Krabbe Disease (KD) is an autosomal recessive disorder that results from loss-of-function mutations in the GALC gene, which encodes lysosomal enzyme galactosylceramidase (GALC). Functional deficiency of GALC is toxic to myelin-producing cells, which leads to progressive demyelination in both the central and peripheral nervous systems. It is hypothesized that accumulation of psychosine, which can only be degraded by GALC, is a primary initiator of pathologic cascades. Despite the central role of GALC in KD pathomechanism, investigations of GALC deficiency at a protein level are largely absent, due in part, to the lack of sensitive antibodies in the field. Leveraging two custom antibodies that can detect GALC at endogenous levels, we demonstrated that GALC protein is predominantly localized to oligodendrocytes in cerebral white matter of an infant brain, consistent with its functional role in myelination. Mature GALC could also be quantitatively detected as a 26 kDa band by western blotting and correlated to enzyme activity in brain tissues. The p.Ile562Thr polymorphic variant, which is over-represented in the KD population, was associated with reduced mature GALC protein and activity. In three infantile KD cases, homozygous null mutations in GALC lead to deficiency in total GALC protein and activity. Interestingly, although GALC activity was absent, normal levels of total GALC protein were detected by a sandwich ELISA using our custom antibodies in a later-onset KD brain, which suggests that the assay has the potential to differentiate infantile- and later-onset KD cases. Among the infantile KD cases, we quantified a 5-fold increase in psychosine levels, and observed increased levels of acid ceramidase, a key enzyme for psychosine production, and hyperglycosylated lysosomal-associated membrane protein 1, a marker for lysosomal activation, in periventricular white matter, a major pathological brain region, when compared with age-matched normal controls. While near complete demyelination was observed in these cases, we quantified that an early-infantile case (age of death at 10 months) had about 3-fold increases in both globoid cells, a pathological hallmark for KD, and CD8-positive T lymphocytes, a pathological marker for multiple sclerosis, in the white matter when compared with a slower progressing infantile case (age of death at 21 months), which suggests a positive correlation between clinical severity and neuropathology. Taken together, our findings have advanced the understanding of GALC protein biology in the context of normal and KD brain white matter. We also revealed new neuropathological changes that may provide insights to understand KD pathogenesis. Abstract: Dealloying is an important industrial technique for generating nanoporous metallic structures by selectively leaching out the more reactive metal component from an alloy material. A constant voltage is often applied to facilitate the dealloying process. Here we report the first study on dealloying with the application of a voltage waveform—specifically, pulsed voltage waveforms are applied for dealloying Ni–Cu alloys. It is found that pulsed dealloying voltage waveforms can exert a strong impact on the dealloying process by 1) significantly lowering the compositional threshold of the more reactive metal component for the dealloying reaction to take place, 2) more thoroughly removing the more reactive metal component and thus producing a porous metal of higher purity and higher porosity (volume fraction of voids), and 3) greatly affecting the morphology of the generated porous metal structure (e.g., leading to significantly thinner ligaments). The nanoporous metallic materials obtained by the pulsed voltage waveform enable supercapacitor electrodes of significantly better performance than the counterpart dealloyed with a constant voltage. HIV-1 group M viruses diverge 25%-35% in envelope, important for viral attachment during infection, and 10%-15% in the pol region, under selection pressure from common antiretrovirals. In Asia, subtypes B and CRF01_AE are common genotypes. Our objectives were to determine whether clinical, immunological, or virological treatment responses differed by genotype in treatment-naive patients initiating first-line therapy.Prospectively collected longitudinal data from patients in Thailand, Hong Kong, Malaysia, Japan, Taiwan, and South Korea were provided for analysis. Covariates included demographics, hepatitis B and C coinfections, baseline CD4 T lymphocyte count, and plasma HIV-1 RNA levels. Clinical deterioration (a new diagnosis of Centers for Disease Control and Prevention category B/AIDS-defining illness or death) was assessed by proportional hazards models. Surrogate endpoints were 12-month change in CD4 cell count and virologic suppression post therapy, evaluated by linear and logistic regression, respectively.Of 1105 patients, 1036 (93.8%) infected with CRF01_AE or subtype B were eligible for inclusion in clinical deterioration analyses and contributed 1546.7 person-years of follow-up (median: 413 days, interquartile range: 169-672 days). Patients >40 years demonstrated smaller immunological increases (P = 0.002) and higher risk of clinical deterioration (hazard ratio = 2.17; P = 0.008). Patients with baseline CD4 cell counts >200 cells per microliter had lower risk of clinical deterioration (hazard ratio = 0.373; P = 0.003). A total of 532 patients (48.1% of eligible) had CD4 counts available at baseline and 12 months post therapy for inclusion in immunolgic analyses. Patients infected with subtype B had larger increases in CD4 counts at 12 months (P = 0.024). A total of 530 patients (48.0% of eligible) were included in virological analyses with no differences in response found between genotypes.Results suggest that patients infected with CRF01_AE have reduced immunologic response to therapy at 12 months, compared with subtype B-infected counterparts. Clinical deterioration was associated with low baseline CD4 counts and older age. Asymmetry is a prominent feature of human brains with important functional consequences. Many asymmetric traits show population bias, but little is known about the genetic and environmental sources contributing to inter-individual variance. Anatomic asymmetry has been observed in Old World monkeys, but the evidence for the direction and extent of asymmetry is equivocal and only one study has estimated the genetic contributions to inter-individual variance. In this study we characterize a range of qualitative and quantitative asymmetry measures in structural brain MRIs acquired from an extended pedigree of Old World vervet monkeys (n = 357), and implement variance component methods to estimate the proportion of trait variance attributable to genetic and environmental sources. Four of six asymmetry measures show pedigree-level bias and one of the traits has a significant heritability estimate of about 30%. The area and volume of brain structural features, as assessed by high-resolution three-dimensional magnetic resonance imaging (MRI), are among the most heritable measures relating to the human CNS. We have conducted MRI scanning of all available monkeys>2 years of age (n=357) from the extended multigenerational pedigree of the Vervet Research Colony (VRC). Using a combination of automated and manual segmentation we have quantified several correlated but distinct brain structural phenotypes. The estimated heritabilities (h²) for these measures in theVRCare higher than those reported previously for such features in humans or in othernonhumanprimates: total brain volume (h²=0.99, SE=0.06), cerebral volume (h²=0.98, SE=0.06), cerebellar volume (h²=[0.86, SE=0.09), hippocampal volume (h² =0.95, SE = 0.07) and corpus callosum cross-sectional areas (h² =0.87, SE = 0.07). These findings indicate that, in the controlled environment and with the inbreeding structure of the VRC, additive genetic factors account for almost all of the observed variance in brain structure, and suggest the potential of the VRC for genetic mapping of quantitative trait loci underlying such variance. A position sensing glove called SmartScan, which creates a 3D virtual model of a real object, is presented. The data from the glove is processed by a volume minimization algorithm to validate the position sensor data. This allows only data from the object's surface to be retained. The data validation algorithm allows the user to progressively improve an image by repeatedly moving their hand over the object. In addition, the user can choose their own balance between feature resolution and invalid data rejection. The SmartScan glove is tested on a foot model and is shown to be robust against motion artifacts, having a mean accuracy of 2.9 mm (compared to a 3D model generated from optical imaging) without calibration. Staphylococcus aureus is an opportunistic pathogen and the major causative agent of numerous hospital- and community-acquired infections. Staphylococcus epidermidis has emerged as a causative agent of infections often associated with implanted medical devices. We have sequenced the ∼2.8-Mb genome of S. aureus COL, an early methicillin-resistant isolate, and the ∼2.6-Mb genome of S. epidermidis RP62a, a methicillin-resistant biofilm isolate. Comparative analysis of these and other staphylococcal genomes was used to explore the evolution of virulence and resistance between these two species. The S. aureus and S. epidermidis genomes are syntenic throughout their lengths and share a core set of 1,681 open reading frames. Genome islands in nonsyntenic regions are the primary source of variations in pathogenicity and resistance. Gene transfer between staphylococci and low-GC-content gram-positive bacteria appears to have shaped their virulence and resistance profiles. Integrated plasmids in S. epidermidis carry genes encoding resistance to cadmium and species.specific LPXTG surface proteins. A novel genome island encodes multiple phenol-soluble modulins, a potential S. epidermidis virulence factor. S. epidermidis contains the cap operon, encoding the polyglutamate capsule, a major virulence factor in Bacillus anthracis. Additional phenotypic differences are likely the result of single nucleotide polymorphisms, which are most numerous in cell envelope proteins. Overall differences in pathogenicity can be attributed to genome islands in S. aureus which encode enterotoxins, exotoxins, leukocidins, and leukotoxins not found in S. epidermidis. Objectives: This study evaluates the clinical course and identifies risk factors for sudden cardiac death (SCD) and clinical deterioration in hypertrophic cardiomyopathy (HCM) in a large community-based population. Comparison was made with data from six tertiary referral and six nonreferral institutions.Background: Hypertrophic cardiomyopathy is a disease with marked heterogeneity in clinical presentation and prognosis. Risk factors for SCD are not well defined in patients free of referral bias.Methods: Between 1970 and 1999, 225 consecutive patients (mean age [+/-SD] 41+/-16 years) were examined and followed at yearly intervals.Results: Forty-four deaths were recorded of which 27 cases were cardiovascular. Fourteen patients died suddenly, six were successfully resuscitated, and seven patients died of congestive heart failure. The annual mortality, annual cardiac mortality, and annual mortality due to sudden death were 1.3%, 0.8%, and 0.6%, respectively. At least one New York Heart Association (NYHA) functional class deterioration was reported in 33% of the patients with a significant (> or =50 mm Hg) left ventricular outflow tract (LVOT) gradient in contrast to 7% without obstruction. The presence of syncope was related to SCD (p < 0.05). Younger age and more severe functional limitation distinguishes patients from in hospital-based centers from the ones in community-based centers.Conclusions: Hypertrophic cardiomyopathy is a benign disease in an unselected population with a low incidence of cardiac death. Syncope was associated with a higher incidence of SCD and patients with a significant LVOT obstruction were more susceptible to clinical deterioration. [ABSTRACT FROM AUTHOR]
