Background: The high disability rate of osteoarthritis (OA), a joint disease with an insidious onset and widespread effects, places a heavy financial burden on patients, families, and society. Traditional diagnostic approaches, including radiology and physical examination, cannot achieve early-stage screening of OA and thus, miss early intervention for patients. Therefore, the need of biomarkers for the early diagnosis of OA is crucial. Results: A total of 390 differentially expressed genes (DEGs) were identified from the training set, and 1077 key module genes were found by constructing a weighted gene co-expression network, and 161 key genes were obtained as a result. Four diagnostic marker genes highly associated with OA were screened for key genes using machine learning algorithms, and the resulting nomogram model showed excellent predictive power and clinical value. After further background studies, immune infiltration and functional enrichment analysis, we found that FKBP5 may play an important role in the prognosis and immune infiltration of multiple cancers, and this hypothesis was verified by pan-cancer analysis. Conclusions: We screened four diagnostic marker genes (FKBP5, EPYC, KLF9 and PDZRN4) that are highly associated with OA. And this led to a diagnostic model, which was assessed to have good predictive power and clinical value. Anthocyanins play crucial roles in pollen protection and pollinator attraction in flowering plants. However, the mechanisms underlying flower color determination and whether floral anthocyanin regulators participate in other processes remain largely unresolved in soybeans (Glycine max). In this study, we investigated the genetic components and mechanisms governing anthocyanin biosynthesis in soybean flowers. Molecular and genetic studies have characterized two antagonistic regulators, the positive activator GmMYBA3 and the negative repressor GmMYBR1, that modulate the gene expression of anthocyanin biosynthesis in soybean flowers. Further findings revealed a regulatory interplay between GmMYBA3 and GmMYBR1 bridged by GmTT8a, highlighting the complexity of anthocyanin regulation in different soybean organs. Exploration of additional soybean cultivars demonstrated the universality of GmMYBA3 and GmMYBR1 in regulating floral anthocyanin biosynthesis-related genes, with GmF3050H identified as a crucial determinant of white flower color. This study provides a molecular mechanism underlying soybean flower color determination, paving the way for the molecular modification of soybean flowers to probably enhance their resistance to abiotic stresses and attractiveness to pollinators. Gram-negative, non-motile, short rod-shaped or spherical bacterial strain that accumulates poly-β-hydroxybutyrate (PHB) granules was isolated from the Beibu Gulf in the South China Sea. Cells have no polar or subpolar flagella, dividing by binary fission. Phylogenetic analyses based on 16S rRNA gene sequences showed that the strain formed a monophyletic branch at the periphery of the evolutionary radiation occupied by the genus Paracoccus, family Rhodobacteraceae, order Rhodobacterales, class Alphaproteobacteria. The closest neighbours were Paracoccus aestuarii strain B7 (97.2% similarity), Paracoccus zeaxanthinifaciens ATCC 21588 (97.1% similarity) and Paracoccus homiensis DD-R11 (96.8%). The predominant fatty acids were C ω7c (82.1%), and significant amounts of C (5.6%), C 3-OH (2.3%) and C (1.5%) were present. The predominant respiratory ubiquinone of strain JLT1284 was Q-10 and the DNA G+C content of strain JLT1284 was 67.0 mol%. The isolate was also distinguishable from members of the genus Paracoccus on the basis of phenotypic and biochemical characteristics. It is evident from the genotypic, chemotaxonomic and phenotypic data, therefore, that strain JLT1284 represents a novel species of the genus Paracoccus, for which the name Paracoccus beibuensis sp. nov. is proposed. The Rac1-WAVE-Arp2/3 pathway pushes the plasma membrane by polymerizing branched actin, thereby powering membrane protrusions that mediate cell migration. Here, using knockdown (KD) or knockout (KO), we combine the inactivation of the Arp2/3 inhibitory protein arpin, the Arp2/3 subunit ARPC1A and the WAVE complex subunit CYFIP2, all of which enhance the polymerization of cortical branched actin. Inactivation of the three negative regulators of cortical branched actin increases migration persistence of human breast MCF10A cells and of endodermal cells in the zebrafish embryo, significantly more than any single or double inactivation. In the triple KO cells, but not in triple KD cells, the 'super-migrator' phenotype was associated with a heterogenous downregulation of vimentin (VIM) expression and a lack of coordination in collective behaviors, such as wound healing and acinus morphogenesis. Re-expression of vimentin in triple KO cells largely restored normal persistence of single cell migration, suggesting that vimentin downregulation contributes to the maintenance of the super-migrator phenotype in triple KO cells. Heat stress (HS) causes significant economic losses and has become a continual challenge in the dairy industry worldwide. The objective of this study was to evaluate the effects of a dietary supplement on milk performance and immune function in late-lactation cows under HS conditions. The supplement was a fermented Chinese herbal medicines (CHMs) mixture consisting of 18 herbs. Forty lactating Holstein cows (560 ± 51.0 kg of initial BW, 230 ± 10.0 DIM, 16 ± 3.0 kg of milk per day) were randomly assigned into 4 treatment groups (10 cows per group). Each group was fed a dietary supplemented with 0, 25, 50, or 100 g CHMs per cow per day. Cows were housed at high ambient temperature-humidity index (average 74.5) for an experimental period of 42 d during the summer months. Milk yield, composition, immune responses involving blood lymphocyte apoptosis rate, serum biochemical parameters, and genes expression in lymphocytes were evaluated on days 14, 28, and 42, respectively. Results showed that milk yield, milk fat, and protein content were greater (all P < 0.05) for 50 or 100 g/d CHMs compared with the group without CHMs supplements throughout the experimental period. On the other hand, increasing CHMs dose demonstrated a greater lymphocyte or leukocyte count (P < 0.01). By flow cytometry analysis, early or late apoptosis rate of the lymphocytes was decreased (P < 0.05) by CHMs supplements. The immunity-related biochemistry and genes transcript responses involving cytokines (IL-1, IL-2, IL-6, and IL-12), apoptosis (Bak, Mcl-1, Bax, Bcl-2, Bcl-xl, and P53), and immunoglobulins (IgA, IgG, and IgM) were investigated. Compared with the unsupplemented group, the serum IL-2 and IL-6 levels, as well as IL-2 mRNA expression, increased (P < 0.05) for 100 g/d. However, the serum IL-1 level tended to decrease (P = 0.08) with increasing CHMs dose, and IL-1 mRNA expression was down-regulated (P = 0.02) by up to 24% for 100 g/d. Additionally, the serum Bax level decreased (P < 0.01) and Bcl-2 level increased (P = 0.01) for 100 g/d. Bax and Bak mRNA expressions were down-regulated (P < 0.05), and Bcl-2 and Bcl-xl expression were up-regulated (P < 0.05) for 50 or 100 g/d. The mRNA expressions of P53 and Mcl-1 were not affected by CHMs (P > 0.10). Besides, serum IgG levels were greater (P < 0.01) for 50 or 100 g/d, compared with unsupplemented group. Background/Aims: Slug protein, a transcription factor for the induction of epithelialmesenchymal transition (EMT) and cancer cell invasion and metastasis, is frequently upregulated in human epithelial cancers. However, mutation of this gene in cancer is rare, and the mechanism of its dysregulation remains unknown, especially in head and neck squamous cell carcinoma (HNSCC). Methods: We examined the role of TNF-α in the stabilization of Slug by immunoprecipitation-westernblot analysis. Migration of HNSCC cells with or without knockdown of Slug gene expression was assayed by a wound healing assay. Immunohistochemical staining analysis was used to measurement Slug levels in both normal and HNSCC tumor tissues. Results: The inflammatory cytokine TNF-α stabilized Slug protein by inhibiting its ubiquitination through the NF-κB pathway. Inhibition of NF-κB or knockdown of p65 abrogated the TNF-α-induced stabilization of Slug. Knockdown of Slug expression inhibited cancer cell migration and EMT characteristics induced by TNF-α. Moreover, increased levels of Slug were found to correlate with lymph node metastasis and predict poor prognosis in patients with HNSCC. Conclusions: NF-κB-mediated stabilization of Slug underlies the inflammation-induced EMT and metastasis in HNSCC, which may serve as a therapeutic target for metastatic HNSCC. [ABSTRACT FROM AUTHOR]

Background: Sperm-associated antigen 9 (SPAG9) is upregulated in several malignancies and its overexpression is positively correlated with cancer cell malignancies. However, the specific biological roles of SPAG9 in hepatocellular carcinoma (HCC) are less understood. Methods: We analyzed SPAG9 and ETS-like gene 1, tyrosine kinase (ELK1) expression in 50 paired HCC specimens and adjacent noncancerous liver specimens using immunohistochemistry. SPAG9 small interfering RNA (siRNA) was used to knockdown SPAG9 expression in HCCLM3 and HuH7 cell lines. We used plasmids to upregulate ELK1 expression and siRNA to downregulate ELK1 expression in HuH7 cells. Quantitative real-time polymerase chain reaction and Western blot were used to evaluate the expression of SPAG9 and ELK1 at the mRNA and protein level, respectively. Wound healing, matrigel migration, and invasion analyses were performed to determine the effect of SPAG9 and ELK1 on HCC metastasis. Results: SPAG9 and ELK1 were overexpressed in HCC tissue specimens and their expressions were higher in HCCLM3 and HuH7 cells compared to the low-metastatic HepG2 cells. Overexpression of SPAG9 was positively associated with tumor-node-metastasis staging (P=0.032), metastasis parameters (P=0.018) of HCC patients, and ELK1 expression (r=0.422, P<0.001) in HCC tissue specimens. In addition, knockdown of SPAG9 in HCCLM3 and HuH7 cells using siRNA significantly suppressed cell migration and invasion. Furthermore, we observed inhibition of ELK1 expression and p38 signaling. However, ELK1 overexpression reversed the inhibitory effects of SPAG9 siRNA on HCC cell metastasis and ELK1 depletion inhibited HuH7 cell migration and invasion. Conclusion: SPAG9 overexpression was positively correlated with HCC metastasis and SPAG9-induced migration and invasion were partially dependent on ELK1 expression in HCC cell lines. Background: During early HIV-1 infection (EHI), the interaction between the immune response and the virus determines disease progression. Although CD1c + myeloid dendritic cells (mDCs) can trigger the immune response, the relationship between CD1c + mDC alteration and disease progression has not yet been defined. Methods: EHI changes in CD1c + mDC counts, surface marker (CD40, CD86, CD83) expression, and IL-12 secretion were assessed by flow cytometry in 29 patients. Results: When compared with the normal controls, patients with EHI displayed significantly lower CD1c + mDC counts and IL-12 secretion and increased surface markers. CD1c + mDC counts were positively correlated with CD4+ T cell counts and inversely associated with viral loads. IL-12 secretion was only positively associated with CD4+ T cell counts. Rapid progressors had lower counts, CD86 expression, and IL-12 secretion of CD1c + mDCs comparing with typical progressors. Kaplan-Meier analysis and Cox regression models suggested patients with low CD1c + mDC counts (<10 cells/μL) had a 4-fold higher risk of rapid disease progression than those with high CD1c +mDC counts. However, no relationship was found between surface markers or IL-12 secretion and disease progression. Conclusions: During EHI, patients with low CD1c + mDC counts were more likely to experience rapid disease progression than those with high CD1c + mDC counts. [ABSTRACT FROM AUTHOR]
