Objective: Tissue electroporation is achieved by applying a series of electric pulses to destabilize cell membranes within the target tissue. The treatment volume is dictated by the electric field distribution, which depends on the pulse parameters and tissue type and can be readily predicted using numerical methods. These models require the relevant tissue properties to be known beforehand. This study aims to quantify electrical and thermal properties for three different tissue types relevant to current clinical electroporation. Methods: Pancreatic, brain, and liver tissue were harvested from pigs, then treated with IRE pulses in a parallel-plate configuration. Resulting current and temperature readings were used to calculate the conductivity and its temperature dependence for each tissue type. Finally, a computational model was constructed to examine the impact of differences between tissue types. Results: Baseline conductivity values (mean 0.11, 0.14, and 0.12 S/m) and temperature coefficients of conductivity (mean 2.0, 2.3, and 1.2 % per degree Celsius) were calculated for pancreas, brain, and liver, respectively. The accompanying computational models suggest field distribution and thermal damage volumes are dependent on tissue type. Conclusion: The three tissue types show similar electrical and thermal responses to IRE, though brain tissue exhibits the greatest differences. The results also show that tissue type plays a role in the expected ablation and thermal damage volumes. Significance: The conductivity and its changes due to heating are expected to have a marked impact on the ablation volume. Electrosurgical vessel joining is commonly performed in surgical procedures to maintain hemostasis. This process requires elevated temperature to denature the tissue and while compression is applied, the tissue can be joined together. The elevated temperature can cause thermal damages to the surrounding tissues. In order to minimize these damages, it is critical to understand how the tissue properties change and how that affects the thermal spread. The purpose of this study is to investigate the changes of tissue thermal conductivity and how the changes correlate to thermal dose during the joining process. We propose a hybrid method combining experimental measurement with inverse heat transfer analysis to determine thermal conductivity of thin tissue sample. Porcine aorta arterial tissues were used to investigate tissue thermal conductivity with variable thermal dose. Different joining times were used to create different amounts of thermal dose. A 36% decrease in tissue thermal conductivity was found when the thermal dose reaches the threshold for second-degree burn. When thermal dose is beyond the threshold of third-degree burn, the tissue thermal conductivity does not decrease significantly. A regression model was also developed and can be used to predict tissue thermal conductivity based on the thermal dose. [ABSTRACT FROM AUTHOR]

Laser-induced optical breakdown (LIOB), or photo- disruption, can generate individual microbubbles in tissues for biomedical applications. We have previously developed a co-localized high-frequency ultrasound system to detect and characterize these laser-induced microbubbles. Because ultrasound speed varies with temperature, this system can also be used to directly estimate thermal effects in the vicinity of photodisruption. In this study, individual bubbles (sizes 60-100 µm) were created at the bottom of a water tank using a 793-nm, 100-fs Ti:Sapphire laser pulsed at 250 kHz. During and after breakdown, pulse-echoes from the tank bottom in the region surrounding a bubble were recorded with a single-element 85-MHz ultrasonic transducer, and temperature-dependent pulse-echo displacements were calculated using phase-sensitive correlation tracking. These displacements were then fit to a finite-element heat transfer model to estimate the effective thermal distribution. Estimates were calculated for laser exposure times ranging from 6.25 to 312.5 ms (1600 to 78 000 laser pulses), at 1.5 and 4 J/cm² fluences. Results suggest a minimal temperature increase (<10 C) within 100 µm of a bubble created with <1600 laser pulses at 1.5 J/cm² fluence. In this paper, we present a radiotelemetry sensor, designed as a lab-in-a-pill, which incorporates a two-channel micro-fabricated sensor platform for real-time measurements of temperature and pH. These two parameters have potential application for use in remote biological sensing (for example they may be used as markers that reflect the physiological environment or as indicators for disease, within the gastrointestinal tract). We have investigated the effects of biofouling on these sensors, by exploring their response time and sensitivity in a model in vitro gastrointestinal system. The artificial gastric and intestinal solutions used represent a model both for fasting, as well as for the ingestion of food and subsequent digestion to gastrointestinal chyme. The results showed a decrease in pH sensitivity after exposure of the sensors for 3 h. The response time also increased from an initial measurement time of 10 s in pure GI juice, to Ca. 25 s following the ingestion of food and 80 s in simulated chyme. These in vitro results indicate that changes in viscosity in our model gastrointestinal system had a pronounced effect on the unmodified sensor. [ABSTRACT FROM AUTHOR]
