Our knowledge of cerebellar functions and cerebellar disorders, called ataxias, is increasing considerably. Studies of the cerebellum are now a central focus in neuroscience. During the last four decades, many laboratories worldwide have dedicated their research activities to understanding the roles of the cerebellum in motor control, cognitive processes and biology of mental processes, behavioral symptoms, and emotion. It is now accepted that the cerebellum acts as a cognitive operator in learning, perception, and attention. Moreover, major improvements in our assessment of in vivo cerebellar architecture using imaging techniques have occurred. A typical example is the accurate description of cerebellar anatomy during fetal development with MRI, a progress which has direct impacts on patient care. These advances have been associated with discoveries of new clinical disorders, in particular in the field of genetic ataxias. More than 20 new genes have been identified these last 10 years. Only for dominant ataxias, more than 30 diseases have now been unravelled. The number of ataxic disorders will increase with aging, the cerebellum being the structure of the brain with the most important loss of neurons with age. More than 300 different cerebellar disorders are encountered during daily practice, but we are missing a single source of information explaining their pathogenesis. Most of the world's experts on prions met for a workshop in Erice in August 1996. The aim of the workshop was to discuss the fundamentals of the science ofprions. It was for­ tunate that so many could be present given the pressure that they were under because of the data presented in March 1996, indicating that Bovine Spongiform Encephalopathy, BSE or Mad Cow Disease, had penetrated the species barrier and was beginning to cause a new dis­ ease in humans--the new variant ofCreutzfeldt-Jakob Disease, nvCJD. This important and urgent subject became an additional major topic at the workshop. This is a book containing most of the talks plus the abstracts of those unable to find time to write up their talks. Almost all papers were written in the spring and summer of 1997 and contain material added after the workshop; thus Bob Will's paper on the new variant of cm contains data up to July 1997 and four contributions arrived in October 1997. In addition to the talks given at our workshop, there was a special joint session with the Planetary Emergencies Workshop where many distinguished scientists, including three Nobel laureates, discussed major issues affecting our planet. Six talks were given by us to this other workshop, five about prions, BSE, and nvCJD, and one on the broader issue of new epi­ demics by Luc Montagnier. Although most of the talks concerned research issues, there were a few special talks.

Prion diseases, also known as Transmissible Spongiform Encephalopathies (TSEs), exist in both humans (Creutzfeldt-Jakob disease (CJD)) and animals (scrapie, bovine spongiform encephalopathy (BSE), chronic wasting disease) and have the unique property of being infectious, sporadic or genetic in origin. Although the precise nature of the infectious agent responsible for TSEs is not definitely identified, it is now clearly demonstrated that a protein named PrP (for Prion Protein) plays a critical role in the transmission and pathogenesis of TSEs. This book provides the general description as well as the details of the techniques currently used for the study of prion diseases. Taking into account the pivotal role played by PrP it is not surprising that many Chapters of this book deal with the purification, the detection and the characterization of the different forms of this protein. In addition, in vitro, cellular and animal models specifically adapted to the study of TSEs, as well as bio-safety procedures are described. Each Chapter is written by scientists involved for many years in their respective domain of prion biology who give the best of their knowledge in this technical document. It has been recognized that viruses can induce neuronal damage by a variety of mechanisms. This volume summarizes, for the first time, the various ways that neurons can degenerate under the influence of viral infection, ranging from acute necrosis and virus-induced apoptosis to chronic damage in persistent infections. The mechanisms of neuronal latency are dealt with as well. The volume also includes chapters that review the chronic damage caused by viral proteins that interfere with differentiated functions of the neuron, including signaling by neurotransmitters and signal transduction by trophic factors. Other chapters deal with the controversial role of cellular immune reactions, which may be helpful in controlling infection, but may also be detrimental to the host nervous system. The specific mechanisms of damage involved in retroviral infections and in prion diseases of the nervous system are reviewed, and finally how autoimmune diseases can lead to neuronal damage.

Bovine spongiform encephalopathy (BSE) has become the most publicly recognizable example of a group of fatal neurodegenerative diseases caused by proteinaceous infectious particles called prions. The contributors to this volume, all internationally recognized experts in their fields, provide an introduction to prion biology, followed by reviews of the latest information on BSE, vCJD, and chronic wasting disease, an animal prion disease that has recently emerged in North America.

The misfolding and aggregation of specific proteins is an early and obligatory event in many of the age-related neurodegenerative diseases of humans. The initial cause of this pathogenic cascade and the means whereby disease spreads through the nervous system, remain uncertain. A recent surge of research, first instigated by pathologic similarities between prion disease and Alzheimer's disease, increasingly implicates the conversion of disease-specific proteins into an aggregate-prone b-sheet-rich state as the prime mover of the neurodegenerative process. This prion-like corruptive protein templating or seeding now characterizes such clinically and etiologically diverse neurological disorders as Alzheimer´s disease, Parkinson's disease, Huntington's disease, amyotrophic lateral sclerosis, and frontotemporal lobar degeneration. Understanding the misfolding, aggregation, trafficking and pathogenicity of the affected proteins could therefore reveal universal pathomechanistic principles for some of the most devastating and intractable human brain disorders. It is time to accept that the prion concept is no longer confined to prionoses but is a promising concept for the understanding and treatment of a remarkable variety of diseases that afflict primarily our aging society. ​

Recent work has implicated inflammatory processes in the pathophysiology of a variety of neurodegenerative diseases such as Alzheimer's disease, stroke and multiple sclerosis. Amyloid-forming proteins are implicated in over 30 human diseases. The proteins involved in each disease have unrelated sequences and dissimilar native structures, but they all undergo conformational alterations to form fibrillar polymers. The fibrillar assemblies accumulate progressively into disease-specific lesions in vivo. Substantial evidence suggests these lesions are the end state of aberrant protein folding whereas the actual disease-causing culprits likely are soluble, non-fibrillar assemblies preceding the aggregates. The non-fibrillar protein assemblies range from small, low-order oligomers to spherical, annular, and protofibrillar species. Oligomeric species are believed to mediate various pathogenic mechanisms that lead to cellular dysfunction, cytotoxicity, and cell loss, eventuating in disease-specific degeneration and systemic morbidity. The particular pathologies thus are determined by the afflicted cell types, organs, systems, and the proteins involved. Evidence suggests that the oligomeric species may share structural features and possibly common mechanisms of action. In many cases, the structure–function interrelationships amongst the various protein assemblies described in vitro are still elusive. Deciphering these intricate structure–function correlations will help understanding a complex array of pathogenic mechanisms, some of which may be common across different diseases albeit affecting different cell types and systems.

Metabolic Encephalopathy is meant to combine and correlate animal and human studies. This protein was called the prion protein. Earlier work had suggested that this diseases and others, loosely collected - gether as transmissible spongiform encephalopathies (TSEs), were not transmitted by conventional infectious agents. Prusiner suggested that 2 this new protein was the infectious agent in these diseases. Such a contentious suggestion lead to ferocious debate. Many researchers still maintained that there was no such thing as an infectious protein. - spite this, by 1990 most people accepted that the cause of the TSEs was the abnormal isoform of the prion protein his research group had id- ti?ed. The most convincing evidence for this had come from the work of Charles Weissmann, whose prion protein knockout mice could not be infected because they lacked expression of the protein that was now 3,4 forever linked to these disease. Since then it has become more widely accepted for these diseases to be termed prion diseases. In 1997 when 5 Stanley Prusiner won the Nobel Prize for his work on prion diseases. Even then, there was still an element of resistance in the scienti?c c- munity. It was considered that, in order the transmissible agent to truly be a protein only, the protein would have to be generated from a rec- binant source.
