The purpose of the book is to advance in the understanding of brain function by defining a general framework for representation based on category theory. The idea is to bring this mathematical formalism into the domain of neural representation of physical spaces, setting the basis for a theory of mental representation, able to relate empirical findings, uniting them into a sound theoretical corpus.The innovative approach presented in the book provides a horizon of interdisciplinary collaboration that aims to set up a common agenda that synthesizes mathematical formalization and empirical procedures in a systemic way. Category theory has been successfully applied to qualitative analysis, mainly in theoretical computer science to deal with programming language semantics. Nevertheless, the potential of category theoretic tools for quantitative analysis of networks has not been tackled so far. Statistical methods to investigate graph structure typically rely on network parameters. Category theory can be seen as an abstraction of graph theory. Thus, new categorical properties can be added into network analysis and graph theoretic constructs can be accordingly extended in more fundamental basis. By generalizing networks using category theory we can address questions and elaborate answers in a more fundamental way without waiving graph theoretic tools. The vital issue is to establish a new framework for quantitative analysis of networks using the theory of categories, in which computational neuroscientists and network theorists may tackle in more efficient ways the dynamics of brain cognitive networks.The intended audience of the book is researchers who wish to explore the validity of mathematical principles in the understanding of cognitive systems. This book is a collection of up-to-date research reviews dealing with various aspects of the structure and function of excitable cells. Its overall objective is to further the search for a better understanding of the mechanism of excitation on a structural and physicochemical basis. The chapters are written by active investigators from a variety of disciplines, repre­ senting many different points of view. Their complementary fields of expertise give this book the rare feature of extraordinary breadth. Excitability is a fundamental property of many biological systems. The mechanisms by which nerve impulses are initiated and propagated, and by which rhythmical activities are produced in nerve, muscle, and cardiac cells, can be fully elucidated only when the process of excitation is derived from fundamental principles applied to known structural forms, at both the macroscopic and the molecular level. The problems of excitation are complex, requiring knowledge of many aspects of cells, including their morphology, elec­ trobiology, chemical physics, and biochemistry.

The dramatic advances in molecular genetics are becoming incorporated into neurobiologic studies at an ever increasing rate. In developmental neurobiology, the importance of cell­ cell interactions for neurogenesis and gene expression is be­ ginning to be understood in terms of the molecular bases for these interactions. This book seeks to emphasize the importance of molecular technology in the study of neurogenetic mechanisms and to explore the possible relationships between specific cell­ cell interactions and regulated gene expression in the develop­ ing nervous ~stem. This volume consists of nineteen chapters which address ques­ tions of gene expression and the importance of cell-cell interac­ tions as key factors in the developing nervous ~stem. Rather than viewing these two processes as separate mechanisms, as the organi­ zation of these chapters might suggest, we would like to emphasize the interplay of these genetic and epigenetic influences in all phases of neural ontogeny, a concept which is made clear by the subject matter of the contributions themselves. In neurosciences one may say,''All roads lead to Rome.'It seems as though wherever one starts, the course of investigation leads to the same major ques­ tions about nervous system function and dysfunction. In thinking about what to write in this preface, it occurred to me that it might be best to deal with that with which I am most familiar and to trace to some extent my own''road to Rome.''As I look over my work of the last 37 years, it becomes clear to me that it can be epitomized as a search for patterns. What usually began as a single­ minded devotion to in-depth analysis of one or a small number of variables always has led to questions of how the results might relate to the whole living unit, whether it is cell, tissue, or organism. For a number of years after my discovery in the vertebrate central nervous system of -y-aminobutyric acid (GABA) and the enzyme which forms it, L­ glutamate decarboxylase (GAD), and the identification of GABA as a major inhibitory neurotransmitter by others, I felt that my laboratory, largely bio­ chemical, was wandering in the wilderness of the complexities of the vertebrate CNS without definitively coming to terms with problems related to GABAergic transmitter functions and the roles of GABA neurons in information processing.

This comprehensive volume is a contribution to a new se ries initiated by the NATO Panel on'Gell to Gell Signals in Plants and Animals'. The book reflects the outcome of an NATO work­ shop and bri ngs to mi nd two im portant questions: consideri ng the mass of relevant I iteratu reavai- able, is there any necessity for a new series of books - and considering the flood of compa­ rable meetings - is there any point in workshops of this nature and their publication? In order to deal with such questions adequately, much more space would be needed than is available in a foreword. Thus, the answers must remain rather superficial and, of course, rather subjective. To simplify the issue, the question of publication can be narrowed down to two fac­ tors - the financial risk, undertaken by the publisher, and the scientific risk, borne by the editor. If the book is good (with respect to lay-out and content) it will be a success - nothing will be lost­ the people involved will enhance their reputation! We are left with the question of the usefulness of workshops. Without doubt, it is indeed a useful procedure for experts to come together, in an atmosphere of harmony, and freedom from external pressures and time limitations, to discuss a well-defined theme. Wether in agreement or disagreement, a fair and open forum can be expectet for a variety of contributions.

'I'll bet it will tum out that brains use both mechanisms, in different centers.'Much of my waking life and that of many of my friends is spent racking our brains over how brains work. This book claims that good science is often a form of betting on the outcome of research-the stakes being time and reputation and someone's money. Some scientists, to be sure, claim they avoid leaning this way or that, in the name of keeping an open mind. I recommend making expectations explicit in order to design controls against unconscious influence, formulate alternative outcomes more clearly-and to add zest. Both the immediately upcoming experiment and the expected result of many long years of work by many people after one is gone are proper subjects for betting or the most informed and serious guessing. The working title for this collection of new and old papers was for some time'Betting on how brains work'and then'Betting on brains.'It goes without saying that the book will not answer the title question but will speak to it, in particular making a series of propositions that I think are more likely to be confirmed by future research than the alternatives we can presently recognize. It follows that a significant message, implied in many chapters of the book is this.

Social systems are among the most complex known. This poses particular problems for those who wish to understand them. The complexity often makes analytic approaches infeasible and natural language approaches inadequate for relating intricate cause and effect. However, individual- and agent-based computational approaches hold out the possibility of new and deeper understanding of such systems. Simulating Social Complexity examines all aspects of using agent- or individual-based simulation. This approach represents systems as individual elements having each their own set of differing states and internal processes. The interactions between elements in the simulation represent interactions in the target systems. What makes these elements'social'is that they are usefully interpretable as interacting elements of an observed society. In this, the focus is on human society, but can be extended to include social animals or artificial agents where such work enhances our understanding of human society. The phenomena of interest then result (emerge) from the dynamics of the interaction of social actors in an essential way and are usually not easily simplifiable by, for example, considering only representative actors. The introduction of accessible agent-based modelling allows the representation of social complexity in a more natural and direct manner than previous techniques. In particular, it is no longer necessary to distort a model with the introduction of overly strong assumptions simply in order to obtain analytic tractability. This makes agent-based modelling relatively accessible to a range of scientists. The outcomes of such models can be displayed andanimated in ways that also make them more interpretable by experts and stakeholders. This handbook is intended to help in the process of maturation of this new field. It brings together, through the collaborative effort of many leading researchers, summaries of the best thinking and practice in this area and constitutes a reference point for standards against which future methodological advances are judged. This book will help those entering into the field to avoid'reinventing the wheel'each time, but it will also help those already in the field by providing accessible overviews of current thought. The material is divided into four sections: Introductory, Methodology, Mechanisms, and Applications. Each chapter starts with a very brief section called ‘Why read this chapter?'followed by an abstract, which summarizes the content of the chapter. This volume aims to document the most important worldwide accomplishments in converging knowledge and technology, including converging platforms, methods of convergence, societal implications, and governance in the last ten years. Convergence in knowledge, technology, and society is the accelerating, transformative interaction among seemingly distinct scientific disciplines, technologies, and communities to achieve mutual compatibility, synergism, and integration, and through this process to create added value for societal benefit. It is a movement that is recognized by scientists and thought leaders around the world as having the potential to provide far-reaching solutions to many of today's complex knowledge, technology, and human development challenges. Four essential and interdependent convergence platforms of human activity are defined in the first part of this report: nanotechnology-biotechnology-information technology and cognitive science (“NBIC”) foundational tools; Earth-scale environmental systems; human-scale activities; and convergence methods for societal-scale activities. The report then presents the main implications of convergence for human physical potential, cognition and communication, productivity and societal outcomes, education and physical infrastructure, sustainability, and innovative and responsible governance. As a whole, the report presents a new model for convergence. To effectively take advantage of this potential, a proactive governance approach is suggested. The study identifies an international opportunity to develop and apply convergence for technological, economic, environmental, and societal benefits. The panel also suggests an opportunity in the United States for implementing a program aimed at focusing disparate R and D energies into a coherent activity - a'Societal Convergence Initiative”. This study received input from leading academic, industry, government, and NGO experts from the United States, LatinAmerica, Europe, Asia, and Australia.

Progress made in the last 20 years clearly indicates that the cell surface is an extremely dynamic structure involved in fundamental processes such as cell motility, innervation, and cell adhesion. Of particular interest is the finding that, in several tissues, the cell surface is differen­ tiated at the intercellular region, thereby providing communicating chan­ nels between apposing cells. Although our actual knowledge of the precise structures and mecha­ in the complex process of intercellular communication is still nisms involved scanty, evidence has been presented that ions and molecules diffuse from cell to cell, establishing a physiological continuum. Embryonic differentia­ tion, cell growth, neoplasia, electrical synchronization in nerve and mus­ cles, as well as immune response seem to be related to ceIl communication. In organizing this volume, it has been our intention to provide the reader with an actual review of the processes involved in intercellular communication in normal tissues as well as in neoplasia. We sincerely believe that the opinions and experiences described herein will be of help in establishing new perspectives for the future of this exciting new field of cell biology. This conference was instigated by a combination of factors: The nature of the problem, the wide­ spread occupational epidemiology reported on eye symptoms and eye fatigue in the workplace, and the organizers'awareness of the complexity of the scientific and clinical bases of knowledge that might be usefully applied. The introduction of new methods into system neurobiology provides new insights into how we receive and process information from the external world, and act upon it. New, non-invasive methods have opened the way to direct observation of the human brain in action. Due particularly to the interaction between the visual and oculomotor requirements involved, several clinical and scientific fields intersect when these issues are considered. To provide clear vision the accommodative and pupillary mechanisms are used. To maintain binocularity, the ver­ gence oculomotor system, sensitive to fatigue, must attain congruence with accommodative levels. This accommodation-vergence linkage was a focus of our symposium.

This fifth volume of Research Methods in Neurochemistry represents a milestone in that it marks almost a decade since the inception of the series. Over these ten years there has been an almost exponential growth in neuro­ chemistry accompanied by numerous technical developments. This is the justification for our series; inevitably we have only been able to cover a frac­ tion of the methodological innovations of the last decade, but we have tried as much as possible to create a balance between the different approaches and philosophies in the study of the chemical basis of brain function. Thus our original format of grouping chapters under various headings-for instance, studies in intact tissues as distinct from studies describing constituents and isolated enzymes-appears to be justified. Studies on whole animals or tissues retaining cellular organization are vital in providing insights into the neurochemical mechanism underlying functional processes; at the same time, the eventual understanding of function can only be attained on the basis of knowledge of the molecular architecture of the tissue. In the present volume, Oldendorfs chapter on the transport of radiolabeled metabolites across the blood-brain barrier illustrates one side of this equation, whereas Poduslo's chapter on the separation of oligo­ dendroglia! cells provides new information on the role of these cells in myelogenesis and the distinctive chemical composition of glia as compared to neurons.

A central problem in neurobiology concerns mechanisms that generate the pro­ found diversity and specificity of the nervous system. How does disruption of these processes result in human disease? In Chapter 3, Weston and co-workers describe the use of monoclonal antibodies that recognize specific neuronal epitopes (including specific gangliosides) for the purpose of defining heterogeneity in the neural crest, an important model system. Immunocyto­ chemical analysis reveals the existence of distinct sUbpopulations within the crest at extremely early stages; cells express neuronal or glial binding patterns at the time of migration. Consequently, interactions with the environment may select for predetermined populations. As a follow up to Volume 7, contributors continue to explore the latest developments in developmental psychology. Here, researchers focus on the integration of theory and research and evaluates theoretical progress and advanced research. Continuing with the successful format of previous volumes in Annals of Theoretical Psychology, Volume 10 presents four major contributions--each accompanied by commentaries and replies to commentaries.

The division of the mature mammalian brain and spinal cord into regions of'white'matter and'gray'matter is observable upon the most cursory inspection. Microscopic examination indicates that the'white'matter regions are characterized by a multilamellar sheath (myelin) which sur­ rounds the axons. A morphologically similar myelin imparts the white color to tracts of the peripheral nervous system, although, as is empha­ sized throughout the book, there are very significant morphological and chemical differences between central and peripheral myelin. The rapid postnatal accumulation of myelin is temporally associated with increasing functional capacity and presumably indicates some important role related to neuronal activity. The ability of myelinated axons to conduct electrical impulses much more rapidly and with much lower use of energy, relative to unmyelinated axons, is now generally accepted. As is evident from the above capsule description, a considerable body of information concerning myelin is available. However, as in other areas of research the unknown expands before us more rapidly than we are able to add to our core of knowledge. This is a corollary of the axiom that a good experiment raises more questions than it answers. Among the areas currendy under investigation in many laboratories are the following: interaction of myelin-forming cells with neurons, assembly and metabo­ lism of myelin, molecular architecture of myelin, and the details of the functional role of myelin.

In evolutionary biology,'intelligence'must be defined in terms of traits that are subject to the major forces of organic evolution. Accordingly, this volume is concerned with the substantive questions that are relevant to the evolutionary problem. Comparisons of learning abilities are highlighted by a detailed report on similarities between honeybees and higher vertebrates. Several chapters are concerned with the evolution of cerebral lateralization and the control of language, and recent analyses of the evolution of encephalization and neocorticalization, including a review of effects of domestication on brain size are presented. The relationship between brain size and intelligence is debated vigorously. Most unusual, however, is the persistent concern with analytic and philosophical issues that arise in the study of this topic, from the applications of new developments on artificial intelligence as a source of cognitive theory, to the recognition of the evolutionary process itself as a theory of knowledge in'evolutionary epistemology'.

This series has addressed a constituency of scientists possessing biochemical background with the goal of providing them with specialized reviews of neu­ robiological interest. Since its initiation, neurochemistry and neuroscience have come of age, and the editors initiate with this volume the concept of a central theme. It is planned that each subsequent volume will also be topical. We note with sadness the passing of Dr. Henry Mahler who served as an advisory editor since the initiation of this series. Agranoff M. H. Aprison vii FOREWORD Oligodendroglia constitute one of the three principal cell types of the central nervous system. These cells, together with their elaborated membranes, account for at least 25 percent of the dry mass of an adult rat brain and an even greater percentage of the central nervous system of larger animals.

The NATO Advanced Study Institute held in the Hotel Metropole Llandrindod Wells, Powys, Wales from July 26th to August 4th 1981 provided an excellent platform for presentation and appraisal of our current knowledge of neurotransmitter systems. The proceed­ ings comprise the present volume and these were arranged to allow study of the pathways and the interactions of specific transmitter systems, one with another, in the various sub-regions of the central nervous system. In addition several of the key factors involved in neurotransmitter release were highlighted. These include the structural and molecular organisation of the nerve terminal, its organelles (such as vesicles, neurotubules and synaptic densities) and the mechanisms involved in neurotransmitter release including the participation of calcium. The organisation of transmitter synthesis, storage and transport were discussed together with their linkage to other streams of metabolism, and the cellular compartmentation of these processes. In addition to the principal lectures given at the Institute, many part1c1pants presented short reports for discussion and these are all included in this volume. The success of the Study Institute was due to the careful planning of the scientific programme to allow a smooth flow and integration of the topics included and this is reflected in the organisation of this book. After the completion of the first edition of this series, this editor thought that a new edition would not be warranted in less than IS, perhaps 20, years, but it seems that we live in a time in which rapid changes are the norm and findings in a field such as neurochemistry develop exponentially. The task of a future editor attempting to get a comprehensive neurochemical handbook for the year 2000 would be even less enviable, but by then information processing may be very different. The approach, the design, and the areas covered by each volume and each chapter are necessarily arbitrary, and it is likely that other editors or authors would have approached the coverage or the organization in a different manner. It is hoped, however, that readers will find the series helpful for beginning or for continuing work. There may be some overlap among the various chapters, but insisting on single coverage of an area would at times have restricted treatment to only one point of view and might have truncated and hurt the logical flow of some of the chapters.

Motivation addresses a central problem in psychology: Why does an animal's behavior fluctuate in the face of an unaltered environment? In a sense this is the opposite of the question from which work on motivation began, and for which Claude Bernard invented the concept of the fixity of the internal milieu: How does an animal maintain constancy in the face of a fluctuating environment? Dealing with motivation has become extremely complex as new experiments, phenomena, and theories have extended the concept. This book embodies some of the ways in which work on motivation is currently proceeding. One of the major changes has been the recognition that motivation cannot be explained without an understanding of the biological rhythms and activational systems that underlie behavior. Another is that ecological and evolutionary perspectives add enormously to answering the central problem of why an animal does what it does when it does. The book suffers from several omissions. There is no chapter on the devel­ opment of motivated behavior. There is none on reward systems in the brain, owing to the untimely death of James Olds, whose contribution would have enriched this book appreciably, and to whom we dedicate it. Over the past several years, the pace of research on the control of axonal growth has increased at a remarkable rate, and this activity is reflected in a growing literature dealing with various aspects of axonal growth and regener­ ation. It appears timely to review the role played by axonal transport in the intrinsic responses of neurons in the growth and regrowth processes. Through the cooperation of the senior editors of this series, we have been given the opportunity to bring such a focus to the current volume. We wish to acknowledge that the contributing authors attended a confer­ ence on'The Role of Axonal Transport in Neuronal Growth and Regenera­ tion'held in Tallahassee, Florida in March, 1983, sponsored by the Psycho­ biology Research Center of the Florida State University. It is our hope that many of the perceptions and insights expressed in these chapters resulted from our interactions.

Developmental Neurobiology provides an up-to-date survey of the cellular events and the molecular contributors that contribute to the assembly of the vertebrate nervous system. The text will serve as a readily tractable source for advanced undergraduate neuroscience majors and beginning graduate students who will benefit from a single source to begin their study of a more detailed understanding of neural development. Each chapter is peppered with a sound mixture of historical context and descriptions from both the vertebrate and invertebrate literature that best illustrate specific aspects of development. The liberal use of simple diagrams and tables, which readily illustrate complex issues, is a welcome addition for instructor and student alike. While classic topics of neural development, including axial patterning, cell proliferation, migration, cell death and synapse formation are covered, of particular interest are subjects that oftentimes received superficial coverage in texts, including separate, detailed chapters on oligodendrocyte and astrocyte development, and developmental mechanisms that relate to the process of aging. Multi-authored texts are often tricky to assemble for consistency, but Developmental Neurobiology succeeds in providing a sound introduction to the most exciting questions that neuroscientists will address experimentally for years to come. Detwiler's book Neuroembryology in 1936. Although progress had been made in the intervening 34 years, the author, Marcus Jacobson, was correct in noting that in 1970'most aspects of neural ontogeny couldbe surveyed at a glance'. In contrast, by the time the 3rd edition appeared in 1991, the size of the book had increased by 40% and the number of references cited went from around 2,000 to over 8,000. Since 1991, however, the field has grown at an even more rapid rate and has now reached a point that makes it virtually impossible for a single individual to comprehensively and authoritatively assess the entire gamut of neural ontogeny. Before his death in 2001, Jacobson together with the co-editor of the present edition, M. Rao, conceived the plan for a 4th edition that would be multiauthored with each chapter being written by experts in a sub-field. Although one of the joys of reading the previous three editions was the consistency of Jacobson's inimitable prose style, in the present book there remains a smoothness and consistency of style that is unusual and refreshing in a multiauthored text. In 14 chapters that begins with neural induction and ends with developmental mechanisms of aging, virtually all of the major topics of neural development are discussed in a clear and coherent fashion and with the aid of ample illustrations. The inclusion of historical antecedents, past and present controversies, technical and conceptual advances together with a comprehensive discussion of each topic all add up to an excellent assessment of the field as it enters the 21st century. Although one misses the Jacobsonian idiosyncrasies of previous editions, the 4th edition is a fitting legacy of Marcus Jacobson's four decades of empirical and pedagogical contributions to developmental neurobiology. In our attempts to interrogate Nature about the development of the nervous system, we ask such questions as'How do the nerve cells originate and how do the correct types of cells differentiate at their correct positions; how do the neurons link together to form circuits whose functions are properly coordinated; and how are the functions of nerve cells related to behavior, to thought, and to conscious­ ness?'Those problems are intellectually challenging, not only because solving them would give us practical advantages but also because while they remain unsolved they stimulate the imagination and challenge the intelligence. It is precisely because they are difficult and controversial and have defied complete solution that such problems continue to attract subtle minds. The understanding that we now have of neural ontogeny seems to me to be farther from complete knowledge than from total ignorance. Nonetheless, it gives us a slightly elevated position from which to survey the vicissitudes of the past, to appraise our present understanding, and to consider ways in which our knowl­ edge might develop in the future. The history of this subject affords a particularly piquant illustration of Arthur Lovejoy's comment that the'adequate record of even the confusions of our forebears may help, not only to clarify those confu­ sions, but to engender a salutary doubt whether we are wholly immune from different but equally great confusions.

My colleagues and I have been gratified by how rapidly the first edition of Myelin has been aeeepted as a standard referenee work by myelin researehers. This is undoubtedly the primary faetor aecounting for the high rate of reeidivism among the authors with respeet to preparation of a seeond edition; eleven of the original twelve contributors were again involved. Four new authors (Wendy Cammer, Marjorie Lees, Ute Traugott, and Seymour Greenfield) have also eontributed to the present volume. This new edition retains many aspects of the format of the original, even including use of the same chapter headings. Thus, comments in the preface to the first edition concerning the level at which material is presented and the relationships between chapters covering the'basic'and'c1inical'material are still applicable. Despite certain similarities in overall organization, comparison of the present edition with the original demonstrates that our eHorts to restriet expansion in the length of the text were not notably successful; the new edition is mueh lengthier than the original. Nevertheless, the increase in text length is modest relative to the increase in information in the seven years separating the preparation of the two editions. Most of the material in the individual chapters has had to be completely redone in order to include this new information. For example, our view of the metabolism of myelin increasingly must take cognizance of a very rapidly turning over pool of certain of its components.

Neurochemistry, having the objective of elucidating biochemical processes subserving nervous activity, emerged as an application of chemistry to the of neurobiological problems as a post-World War II phenomenon. investigation However, only in the last 40 years has the chemical community recognized neurochemistry as a distinct, if hybrid, discipline. During this period great strides have been made. However, recently neurochemistry, along with neu­ rophysiology, neuropharmacology, neuroanatomy, and the behavioral sci­ ences, has emerged to form neuroscience, a new community of scientists with its own national society, journals, and meetings. Actually, this recently formed hybrid, neuroscience, is in the process of merging with another well-established discipline, molecular genetics (frequently called molecular biology, and itself a hybrid), which appears to have sufficient hybrid vigor to form yet a new community of scientists, which, for want of a more imaginative term, has been called molecular genetic neuroscience. Clearly, advantages resulting from such mergers or hybridizations accrue not only from the merging discipline (neurochemistry in this case) to the new community (molecular genetic neuroscience), but also in the reverse direction. This Foreword will be concerned primarily with examples of this latter process.

Volume V deals with the problems of turnover in the nervous system.'Turnover'is defined in different ways, and the term is used in different contexts. It is used rather broadly in the present volume, and intentionally so. The turnover of macromolecules is only one aspect; here'turnover'in­ dicates the simultaneous and coordinated formation and breakdown of macromolecular species. The complexities of cerebral protein turnover are shown in a separate chapter dealing with the synthesis of proteins, in another on breakdown, and in still another on the relationship of these two (showing how the two halves of turnover are controlled). The fact that most likely the two halves of protein turnover, synthesis and breakdown, are separated spatially and the mechanisms involved are different further emphasizes the complexity of macromolecular turnover.'Turnover'is used in a different context when the turnover of a cycle is discussed; but here again a number of complex metabolic reactions have to be interrelated and controlled; some such cycles are discussed briefly in this volume, additional cycles have been discussed with metabolism, and some cycles still await elucidation or discovery.

Two new chapters have been added to reflect the increased interest in the development and differentiation of the nervous system and in the reconstruction of its circuitry in tissue culture. One chapter deals with slice cul­ tures in which the organization of the nervous system is preserved. When slice cultures are combined with explant cultures, afferent and efferent projections can be reconstructed. The other chapter deals with aggregating neural cell cul­ tures, in which'minibrains'can form. Theses are small, uniformly sized spheres of nervous tissue, usually having nerve cells in the center and astrocytes, oligo­ dendrocytes, and microglia in the periphery. The emergence of a new scientific book series requires some explanation regarding how it hopes to compensate the reader for the discomforts it undoubtedly produces both in the realms of informational input-overload and in the financial strain on personal and institutional budgets. This series recognizes that investigators who have entered neurochemistry from the biochemical tradition have a rather specialized view of the brain. Too often, interdisciplinary offerings are initially attractive but turn out to recite basic biochemical considerations. We have come to believe that there are now sufficiently large numbers of neurochemists to support a specialized venture such as the present one. We have begun with consideration of traditional areas of neurochemistry which show considerable scientific activity. We hope they will serve the neurochemist both for general reading and for specialized information. The reader will also have the opportunity to reftect on the unbridled speculation that results from the disinhibiting effects on the author who has been invited to write a chapter. A fundamental problem in neuroscience is the elucidation of the cellular and molecular mechanisms underlying the development and function of the nervous system. The complexity of organization, the heteroge­ neity of cell types and their interactions, and the difficulty of controlling experimental variables in intact organisms make this a formidable task. Because of the ability that it affords to analyze smaller components of the nervous system (even single cells in some cases) and to better control experimental variables, cell culture has become an increasingly valuable tool for neuroscientists. Many aspects of neural development, such as proliferation, differentiation, synaptogenesis, and myelination, occur in culture with time courses remarkably similar to those in vivo. Thus, in vitro methods often provide excellent model systems for investigating neurobiological questions. Ross Harrison described the first culture of neural tissue in 1907 and used morphological methods to analyze the cultures. Since that time the technique has been progressively modified and used to address an ever widening range of developmental questions. In recent years a con­ vergence of new or improved cell culture, biochemical, electrophysiol­ ogical, and immunological methods has occurred and been brought to bear on neurobiological questions. This volume is intended not to be comprehensive but rather to highlight some of the latest findings, with a review of previous important work as well, in which combinations of these methods are used.

An excellent survey of the most recent advances in the area of cellular and molecular biology of glial cells and their involvement in remyelination and functional repair in the CNS. A major part of this book deals with signaling paths within and among neural cells. Glial cells (astrocytes, oligodentrocytes, microglial cells) themselves release substantial amounts of cytokines, growth factors and other signaling molecules, which play an important role during injury and regeneration. Another signaling route between neurons and glial cells follows ion channels and neurotransmitter receptors. In addition to animal models, human oligodentrocytes in cell culture and in the human MS brain are characterized with respect to their immunocytochemistry and function.

This volume of Cerebral Cortex is dedicated to Sir John Eccles, who was an active member of the advisory board for the series until his death in May 1997. His input as to what topics should be covered in future volumes of this series will be sorely missed. The present volume is concerned with neurodegenerative disorders and age­ related changes in the structure and function of the cerebral cortex, a topic that has attracted increasing interest as longevity and the number of aged individuals in the population increase. Although much of the research on the neurodegenerative effects of aging has been centered on Alzheimer's disease, most of the aging popu­ lation will not be afflicted by this disease. The problem of the relation between our bodies and our minds, and espe­ cially of the link between brain structures and processes on the one hand and mental dispositions and events on the other is an exceedingly difficult one. Without pretending to be able to foresee future developments, both authors of this book think it improbable that the problem will ever be solved, in the sense that we shall really understand this relation. We think that no more can be expected than to make a little progress here or there. We have written this book in the hope that we have been able to do so. We are conscious of the fact that what we have done is very conjectur­ al and very modest. We are aware of our fallibility; yet we believe in the intrinsic value of every human effort to deepen our understanding of our­ selves and of the world we live in. We believe in humanism: in human rationality, in human science, and in other human achievements, however fallible they are. We are unimpressed by the recurrent intellectual fashions that belittle science and the other great human achievements. An additional motive for writing this book is that we both feel that the debunking of man has gone far enough - even too far. It is said that we had to learn from Copernicus and Darwin that man's place in the universe is not so exalted or so exclusive as man once thought. That may well be.

Presented with a choice of evils, most would prefer to be blinded rather than to be unable to move, immobilized in the late stages of Parkinson's disease. Yet in everyday life, as in Neuroscience, vision holds the centre of the stage. The conscious psyche watches a private TV show all day long, while the motor system is left to get on with it'out of sight and out of mind.'Motor skills are worshipped at all levels of society, whether in golf, tennis, soccer, athletics or in musical performance; meanwhile the subconscious machinery is ignored. But scientifically there is steady advance on a wide front, as we are reminded here, from the reversal of the reflexes of the stick insects to the site of motor learning in the human cerebral cortex. As in the rest of Physiology, evolution has preserved that which has already worked well; thus general principles can often be best discerned in lower animals. No one scientist can be personally involved at all levels of analysis, but especially for the motor system a narrow view is doomed from the outset. Interaction is all; the spinal cord has surrendered its autonomy to the brain, but the brain can only control the limbs by talking to the spinal cord in a language that it can understand, determined by its pre-existing circuitry; and both receive a continuous stream of feedback from the periphery.

A great deal of the success of science has rested on its specific methods. One of which has been to start with the study of simple phenomena such as that of falling bodies, or to decompose systems into parts with well-defined properties simpler than those of the total system. In our time there is a growing awareness that in many cases of great practical or scientific interest, such as economics or the hu­ man brain, we have to deal with truly complex systems which cannot be decomposed into their parts without losing crucial properties of the total system. In addi­ tion, complex systems have many facets and can be looked at from many points of view. Whenever a complicated problem arises, some scientists or other people are ready to invent lots of beautiful words, or to quote Goethe'denn immer wo Begriffe feh­ len, dort stellt ein Wort zur rechten Zeit sich ein'('whenever concepts are lack­ king, a word appears at the right time'). Quite often such a procedure gives not only the layman but also scientists working in fields different from that of the in­ ventor of these new words the impression that this problem has been solved, and I am occasionally shocked to see how influential this kind of'linguistics'has become.

The author of this remarkably comprehensive review, PIERRE DUSTIN, has performed an invaluable service in bringing to­ gether in one volume the observations and theory on micro­ tubules that have accumulated over the last fifteen years. He has understood the magnitude of the task from the beginning and has met it thoroughly and, I must say, courageously. From here on, and for many years to come, young investiga­ tors, and some not so young, will have a ready reference as they seek to discover what has been done and remains to be done in achieving a better understanding of these important cell components. Since the early 1960's when it became clear that the fila­ ments (micro tubules) making up the 9 + 2 complex of cilia and flagella were to be found very widely in cells, microtub­ ules have attracted an ever-increasing amount of attention. Now it is known, as reviewed in this volume, that they influ­ ence the morphogenesis of anisometry in the shapes of cells and cell extensions; that they function as frames for the intra­ cellular movement of granules and chromosomes; that they are subject to control in their assembly and disassembly by externally applied substances such as colchicine and cyclic AMP; and that they display abnormalities in their numbers and orientations in transformed (malignant) cells, to mention only a few of their several functions.
