This book addresses the differentiation control of skeletal muscle in different locations of the vertebrate body Particular attention is paid to novel regulatory molecules and signals as well as the heterogeneity of origin that have revealed a developmental overlap between skeletal and cardiac muscle. Different functional muscle groups are the product of the evolution of the vertebrate classes, making a phylogenetic comparison worthwhile for understanding the role of muscle stem cells and precursors in myogenesis. New insights into the hierarchy of transcription factors, particularly in the context of these different muscle groups have been gained from detailed investigations of the spatio-temporal and regulatory relationships derived from mouse and zebrafish genetics and avian microsurgery. Importantly, epigenetic mechanisms that have surfaced recently, in particular the role of MyomiRs, are also surveyed. With an eye to the human patient, encouraging results have been generated that identify parallels between embryonic myogenesis and regenerating myofibers due to common regulatory molecules. On the other hand, both processes differ considerably in quality and complexity of the processes employed. Interestingly, the heterogeneity in embryonic sources from which skeletal muscle groups in the vertebrate including the human body take origin is paralleled by differences in their susceptibility to particular muscle dystrophies as well as by the characteristics of the satellite cells involved in regeneration. The progress that has been made in the field of muscle stem cell biology, with special focus on the satellite cells, is outlined in this book by experts in the field. The definitions of stem cells, stemness, and the niche concept continue to undergo revisions. In adult vertebrates, hematopoietic and some non-hematopoietic progenitors are synthesized within specialized niches of bone marrow. They migrate to designated tissues, and are either trans-differentiated or become quiescent and settle down. These form the stem cell niche reservoir in all tissues. Not only the primary hematopoietic tissue but all organs and tissues are also capable of generating progenitors which are either synthesized from these migrants or are direct recruits from other tissues. In the niches, the cells settle down and await their turn to either make more clones like themselves or differentiate and mobilize in an exigency. Thus progenitors are important reserves, to be multiplied and deployed or travel as important message bearing molecules via blood as circulating progenitors which can home to their allocated destinations. Upon reaching, specific integrins and selectins help them “dock” and “port” and they may be reprogrammed to induce further differentiation and help repopulate the denuded tissue. In pulmonary fibrosis and asthma, progenitors from both systemic circulation and local stem cell niches have been found to participate in the myriad ramifications of repair, replacement and regeneration of lost or diseased tissue. ​​​​​​​Prominin-1 or otherwise known as CD133 is a glycoprotein that is present in humans and mice. Since the first description of prominin in 1997, in mouse neuroepithelial cells and in human hematopoietic stem cells as AC133 antigen, this molecule has aroused a large interest especially, as a stem cell marker, that gave rise to an ever growing body of publications and more recently its expression in cancer stem cells. Controversies as to its role as a cancer stem and its detection in different models, as well as its use as a prognostic marker have emerged. Yet, beyond its use as a stem cell and cancer stem cell marker, prominin-1/CD133 displays unique biological features and appears of importance in other processes like for example in retinal biogenesis. Indeed, this five-transmembrane plasma membrane glycoprotein, which marks membrane protrusions is associated with several essential processes like cell polarity, asymmetric cell division and membrane remodeling.
