Over the last few decades, various techniques have been developed to alter the properties of plants and animals. While the targeted transfer of recombinant DNA into crop plants remains a valuable tool to achieve a desirable breeding outcome, integration of transgenes into the host genome has been random, which in part, leads to reduced acceptance of GMOs by the general population in some parts of the world. Likewise, methods of induced mutagenesis, such as TILLING, have the disadvantage that many mutations are induced per plant, which has to be removed again by expensive backcrossing. Advances in genome sequencing have provided more and more information on differences between susceptible and resistant varieties, which can now be directly targeted and modified using CRISPR/Cas9 technology. By selecting specific gRNAs occurrence of off-target modifications are comparatively low. ZFNs and TALENs- based approaches required re-engineering a new set of assembled polypeptides for every new target site for each experiment. The difficulty in cloning and protein engineering prevented these tools from being broadly adopted by the scientific community. Compared to these technologies, designing the CRISPR toolbox is much simpler and more flexible. CRISPR/Cas9 is versatile, less expensive and highly efficient. It has become the most widely used technology for genome editing in many organisms.Since its inception as a powerful genome-editing tool in late 2012, this breakthrough technology has completely changed how science is performed. Cotton, the most important natural fiber crop, has been improved by conventional breeding—largely through planned hybridization of different cotton genotypes, since the discovery of Mendelian genetics. All these efforts resulted in the development of resilient high yielding cotton varieties. However, the progress through conventional breeding procedures is slow because of long lag periods for developing a variety, little control over the new genetic combinations, unwanted traits and lack of foolproof performance testing system. Genomic assays discovered over the last two decades have made it possible to understand the “language” of the genome by associating the genes with specific traits. Together with the more recently established gene-editing tools like CRISPR-Cas9, the cotton genome can be tailored much more precisely than ever before. In this regard, genetic information has been harnessed, through (i) sequencing of the progenitor and cultivated cotton species, (ii) ongoing mega pan-genome sequencing projects, (iii) genetic and physical mapping, and (iv) introgression of genes from alien sources, that resulted in the development of resilient cotton cultivars. These technologies have been deployed or are attempting to overcome the challenges of water shortage, excessive heat in most cotton growing regions, infectious diseases and infestation of insect pests, as well as rising production cost, for sustainable cotton production beyond 2030. In this book, new knowledge generated by the cotton research community and its application for developing resilient cotton are comprehensively summarized. The natural environment for plants is composed of a complex set of abiotic and biotic stresses; plant responses to these stresses are equally complex. Systems biology allows us to identify regulatory hubs in complex networks. It also examines the molecular “parts” (transcripts, proteins and metabolites) of an organism and attempts to combine them into functional networks or models that effectively describe and predict the dynamic activities of that organism in different environments. This book focuses on research advances regarding plant responses to abiotic stresses, from the physiological level to the molecular level. It highlights new insights gained from the integration of omics datasets and identifies remaining gaps in our knowledge, outlining additional focus areas for future crop improvement research. Plants have evolved a wide range of mechanisms for coping with various abiotic stresses. In many crop plants, the molecular mechanisms involved in a single type of stress tolerance have since been identified; however, in order to arrive at a holistic understanding of major and common events concerning abiotic stresses, the signaling pathways involved must also be elucidated. To date several molecules, like transcription factors and kinases, have been identified as promising candidates that are involved in crosstalk between stress signalling pathways. Mitochondria are the product of a long evolutionary history. It is now a well established fact that mitochondria did evolve from free living bacteria being the common ancestor of both, eukaryotic mitochondria and α-proteobacteria. Advances in genome sequencing, the establishment of in organello and in vitro assays to name only a few, contributed significantly to advances in plant mitochondrial research. Second generation sequencing and the ability to directly sequence and analyse the whole plant transcriptome certainly will help to develop the research on plant mitochondria to another level in the future. Plant biotechnology applies to three major areas of plants and their uses: (1) control of plant growth and development; (2) protection of plants against biotic and abiotic stresses; and (3) expansion of ways by which specialty foods, biochemicals, and pharmaceuticals are produced. The topic of recent advances in plant biotechnology is ripe for consideration because of the rapid developments in this?eld that have revolutionized our concepts of sustainable food production, cost-effective alt- native energy strategies, environmental bioremediation, and production of pla- derived medicines through plant cell biotechnology. Many of the more traditional approaches to plant biotechnology are woefully out of date and even obsolete. Fresh approaches are therefore required. To this end, we have brought together a group of contributors who address the most recent advances in plant biotechnology and what they mean for human progress, and hopefully, a more sustainable future. Achievements today in plant biotechnology have already surpassed all previous expectations. These are based on promising accomplishments in the last several decades and the fact that plant biotechnology has emerged as an exciting area of research by creating unprecedented opportunities for the manipulation of biological systems. In connection with its recent advances, plant biotechnology now allows for the transfer of a greater variety of genetic information in a more precise, controlled manner. The potential for improving plant productivity and its proper use in agric- ture relies largely on newly developed DNA biotechnology and molecular markers.

With regard to global climate changes, one of our future challenges will be to develop crop plants that cope better with changing environmental conditions. Abiotic stress is estimated to be the primary cause of crop loss worldwide, with the potential to cause a reduction of more than 50% in the average yield of the main crops. Climatic extremes are known to trigger senescence processes. Many different agriculturally important traits are affected by senescence, like number and quality of seeds, timing of seed set, fruit ripening, etc.. Despite the importance of the sen- cence processes, our knowledge on the regulatory mechanisms of senescence is still poor. However, senescence is not a chaotic breakdown, but an orderly loss of normal cell functions. In contrast to aging processes which have a passive and non-regulated degenerative character (for a review, see Krupinska et al. 2003), senescence is an active and highly regulated process. Senescence can be initiated by exogenous and endogenous triggers. The most important endogenous factors inducing senescence are the age of the leaves and the age and developmental stage of the plant. The leaves of annual plants show a continuous decrease in their photosynthesis rate after full expansion (Batt and Woolhause 1975; Hensel et al. 1993). In fast-aging plants like Arabidopsis, photosynthetic capacity of the leaves decreases by 50% within 4–6 days of full leaf expansion under continuous light conditions (Hensel et al. 1993).

The past two decades have witnessed an enormous progress in our understanding of plastid (chloroplast) biology.
