A close look at Günter Blobel's transformative contributions to molecular cell biology. The difficulty of reconciling chemical mechanisms with the functions of whole living systems has plagued biologists since the development of cell theory in the nineteenth century. As Karl S. Matlin argues in Crossing the Boundaries of Life, it is no coincidence that this longstanding knot of scientific inquiry was loosened most meaningfully by the work of a cell biologist, the Nobel laureate Günter Blobel. In 1975, using an experimental setup that did not contain any cells at all, Blobel was able to target newly made proteins to cell membrane vesicles, enabling him to theorize how proteins in the cell distribute spatially, an idea he called the signal hypothesis. Over the next twenty years, Blobel and other scientists were able to dissect this mechanism into its precise molecular details. For elaborating his signal concept into a process he termed membrane topogenesis—the idea that each protein in the cell is synthesized with an'address'that directs the protein to its correct destination within the cell—Blobel was awarded the Nobel Prize in Physiology or Medicine in 1999. Matlin argues that Blobel's investigative strategy and its subsequent application addressed a fundamental unresolved dilemma that had bedeviled biology from its very beginning—the relationship between structure and function—allowing biology to achieve mechanistic molecular explanations of biological phenomena. Since the 1970s, the origin of cancer is being explored from the point of view of the Somatic Mutation Theory (SMT), focusing on genetic mutations and clonal expansion of somatic cells. As cancer research expanded in several directions, the dominant focus on cells remained steady, but the classes of genes and the kinds of extra-genetic factors that were shown to have causal relevance in the onset of cancer multiplied. The wild heterogeneity of cancer-related mutations and phenotypes, along with the increasing complication of models, led to an oscillation between the hectic search of ‘the'few key factors that cause cancer and the discouragement in face of a seeming ‘endless complexity'. To tame this complexity, cancer research started to avail itself of the tools that were being developed by Systems Biology. At the same time, anti-reductionist voices began claiming that cancer research was stuck in a sterile research paradigm. This alternative discourse even gave birth toan alternative theory: the Tissue Organization Field Theory (TOFT). A deeper philosophical analysis shows limits and possibilities of reductionist and anti-reductionist positions and of their polarization. This book demonstrates that a radical philosophical reflection is necessary to drive cancer research out of its impasses. At the very least, this will be a reflection on the assumptions of different kinds of cancer research, on the implications of what cancer research has been discovering over 40 years and more, on a view of scientific practice that is most able to make sense of the cognitive and social conflicts that are seen in the scientific community (and in its results), and, finally, on the nature of living entities with which we entertain this fascinating epistemological dance that we call scientific research. This volume contains the Proceedings of the Serono Symposium on Pre­ implantation Embryo Development, held in Newton, Massachusetts, in 1991. The idea for the symposium grew out of the 1989 Serono Symposium on Fertilization in Mammals• at which preimplantation development was the predominant suggestion for a follow-up topic. This was indeed a timely subject in view of the recent resurgence of interest in this funda­ mental phase of embryogenesis and its relevance to basic research and applied fertility studies in humans, food-producing animals, and endangered species. The symposium brought together speakers from a broad range of disciplines in order to focus on key regulatory mechanisms in embryo development, using a wide variety of animal models, and on representative topics in human preimplantation embryogenesis. The culmination of preimplantation development is a blastocyst con­ taining the first differentiated embryonic tissues and capable of initiating and sustaining pregnancy. The central objective of the symposium was to throw light on the regulation of cellular and molecular events underlying blastocyst formation. It was particularly appropriate that the date of the symposium marked the 20th anniversary of the publication of the classic volume Biology of the Blastocyst, the proceedings of an international workshop held in 1970. This book, which summarized most of the information then available on this topic in mammals, was edited by the pioneer in blastocyst research, Dr. Richard B1andau, who was the guest speaker at the symposium.

Understanding the brain mechanisms which underlie behavior is one of the most challenging tasks of modern biology. The study of these mechanisms can be approached using a variety of biological systems as models, depending essentially on the type of question being asked and the technical approach which is considered. In vertebrates, the study of brain neurochemistry in relation to behavior e,xpression has made tremendous progress during the last two decades. In particular, much at­ tention has been devoted to the effect of steroid hormones on brain structure and activity in connection with social and mainly reproductive behavior. This book exemplifies some of the major trends in the field. I did not attempt to cover exhaustively all the work that has been done in this area but rather to present a series of selected reviews on the molec­ ular and cellular brain mechanisms most directly related to social behavior. This selection obviously reflects my own interests but also, I believe, highlights those areas of research in which important progress has been made in recent years. A number of brain biochemical or cellular mechanisms which could be related to behavior, but have not been formally demonstrated to be so, are consequently ignored. The focus here is on social behavior and thus steroid-dependent processes are covered in priority. This choice of course leads us to ignore major trends in brain and behavior research but this is, I hope, clearly reflected in the title of the volume.

In the mid-1980s the European Journal of Biochemistry set out to publish review articles. The enterprise proved successful, resulting in high-level reviews written by well-known scientists appearing in the Journal. The reviews represent emerging and rapidly growing fields of research in fundamental as well as applied areas of biochemistry, such as medicine, biotechnology, agriculture and nutrition.
