Motor dysfunction and cognitive impairment are major symptoms in both Huntington's Disease (HD) and Parkinson's Disease (PD). This book is the result of a convergence of scientific information regarding mechanisms that produce acute nerve cell death in the brain. Although seemingly disparate, stroke, brain and spinal cord trauma, coma from a low serum glucose concentration (hypoglycemia), and prolonged epileptic seizures have in common the inciting factor of excitotoxicity, the activation of a specific subtype of glutamate receptor by an elevated extracellular glutamate concentration that results in an excessive influx of calcium into nerve cells. The high calcium concentration in nerve cells activates several enzymes that are responsible for degradation of cytoplasmic proteins and cleavage of nuclear DNA, resulting in nerve cell death. The high calcium concentration also interferes with mitochondrial respiration, with the resultant production of free radicals that damage cellular membranes and nuclear DNA. Understanding the biochemical pathways that produce nerve cell death is the first step toward devising an effective neuroprotective strategy, the ultimate goal. As our understanding of apoptotic pathway expands, we are coming to realize the great potential of utilizing this pathway to treat diseases such as cancer. The book attempts to review, summarize, and speculate on the apoptotic pathways, how are they regulated and how targeted therapies are being used to treat a wide variety of diseases. Special emphasis is placed on cancer since new treatments either being developed or currently in the clinical setting are showing great promise to increase survival rates for cancer patients. Die vorliegende Arbeit sollte dazu beitragen, die molekularen Grundlagen der Apoptose im Gehirn aufzuklären. Für diesen Zweck wurde ein Primärkultursystem aus dem Hippocampus neonataler Ratten etabliert, welches sich nach acht Tagen aus 30% neuronalen und 70% nicht-neuronalen Zellen zusammensetzte. Bei den nicht-neuronalen Zellen handelte es sich in erster Linie um Gliazellen. Der Proteinkinaseinhibitor Staurosporin wurde verwendet, um in den hippocampalen Primärkulturen eine Apoptose auszulösen. 300 nM Staurosporin führten zu Vitalitätsverlusten und zu einer Fragmentierung der Zellkerne, wobei von der Kernfragmentierung als erstes die Neurone betroffen waren. Desweiteren konnte eine deutliche Fragmentierung der DNA in oligonukleosomale Bruchstücke detektiert werden. Eine Aktivität der Caspase-3 wurde ebenfalls festgestellt. Sie war nach 5 stündiger Behandlung maximal ausgeprägt und betrug ca. das 10 fache des Kontrollwertes. Der Caspaseinhibitor ZVAD-fmk war in der Lage, den durch Staurosporin hervorgerufenen Vitalitätsverlust um rund 30% zu vermindern. Eine Untersuchung der Zellkerne erbrachte jedoch keinen protektiven Effekt, vielmehr verursachte ZVAD-fmk einen Shift von fragmentierter zu kondensierter Kernmorphologie. Demnach ist eine Hemmung der Caspasen nicht hinreichend, um einen Zelltod in diesem Modell zu verhindern. Der Transkriptionsinhibitor Actinomycin D übte sowohl auf die Vitalität der hippocampalen Zellen als auch auf deren Kernmorphologie einen schädigenden Effekt aus. Folglich werden während der Apoptose in diesem Modell auf Ebene der mRNA nicht ausschließlich sogenannte'Killergene'aktiviert. Vielmehr ist denkbar, daß Staurosporin eine Erhöhung protektiver Gene induziert. Der zeitliche Expressionsverlauf verschiedener Gene, von denen man eine Beteiligung am apoptotischen Zellsterben annimmt, wurde nach Staurosporin-Behandlung mittels semiquanti­ta­tiver RT-PCR untersucht. Die Ergebnisse zeigten jedoch weder eine eindeutige Induktion noch eine Unterdrückung schädigender bzw. protektiver Gene. Die Erhöhung der Gen­expression für die SOD-2 und die HO-1 deutet auf eine Beteiligung von Sauerstoffradikalen bei der Staurosporin-induzierten Apoptose hin und kann als Schutzreaktion der Zellen interpretiert werden.
