Cockayne syndrome (CS) is a rare autosomal genetic disorder that was first identified almost 62 years ago by Alfred Cockayne and was named after him. The earliest publication record (PubMed) available is a paper by Marie et al in 1958. Since then 815 research papers including excellent reviews have been published (PubMed, December 2008), yet we are a long way from fully understanding the exact molecular mechanisms of this disease. Ironically, like many other inborn genetic defects, CS is still incurable; the mean life expectancy of the patients is 12.5 years. Major milestones in the study of CS were the discovery that the patients have a defect in DNA repair, the identification of the two complementation groups CSA and CSB, and the finding that CS cells were defective in the specialized pathway of nucleotide excision repair, transcriptional-coupled repair (TCR), that removes certain lesions from actively transcribed DNA. The editor of this book (SIA) has considerable interest in this field; recent studies have revealed a number of new enzymes (unpublished data) that may be responsible for the scavenge of ROS. Our future studies might show if deficiency of any of these newly discovered enzymes (as a result of genetic mutations) may lead to the neurodegeneration and other ROS-induced diseases. We hope that this book will stimulate both experts and novice researchers in the field with excellent overview of the current status of research and pointers to future research goals. The insights gained may also be valuable for the development of new therapeutic regimens for dealing with the clinical problems raised by this rare but devastating human condition.

While every cell of an organism has an identical genomic content, extremely complex networks exist to tailor the genomic output to the needs of that cell. This program of gene expression is different for every cell type and stage of development. In addition, the cell can respond to its environment by modulating its gene expression program in a fairly dramatic manner. For many decades gene transcription has been investigated in systems from bacteria to mammalian cells and along the way many landmark findings have set new paradigms that often apply across wide evolutionary distances. Studying individual genes, however, especially in mammalian systems has been a painstaking business and although we know the transcription activators and other complexes that control specific genes in minute detail, generalizing these findings has often proven to be difficult. It has become clear that transcription factors do not operate alone but form complex networks in the cell. If one component of this complexity is disturbed then there are repercussions across the entire network, but it has been impossible to study these networks until very recently. The advent of microarray technology within the last decade has revolutionized how we study gene transcription. There are several types of array technology that essentially screen for relative mRNA levels for many thousands of genes at once. We do not focus here on the technology as this has become routine and is available to many researchers. Microarray technology has given us the ability to measure the entire gene expression program of a cell in a single experiment and compare it to other cells thus allowing a global view of cell behaviour at the level of gene transcription. Expression profiling, as this endeavour has become known, is now a relatively simple undertaking and hundreds, probably thousands of papers have been published demonstrating the power of this technology. Expression profiling has been applied to many diverse biological problems and is also being developed as a method for disease diagnosis especially in the cancer classification field. There are constant improvements or modified uses of the technology that are allowing more and more high throughput experiments to be carried out.

The germfree animal is reared in the laboratory to be bacteria free; its counterpart, the gnotobiotic animal, is exposed to select microorganisms. The need for such an animal model for use in biomedical studies was first expressed by Pasteur in the late 1800s. Women are contributing to disciplines once the sole domain of men. Field biology has been no different. The history of women field biologists, embedded in a history largely made and recorded by men, has never been written. Compilations of biographies have been assembled, but the narrative—their story—has never been told. In part, this is because many expressed their passion for nature as writers, artists, collectors, and educators during eras when women were excluded from the male-centric world of natural history and science. The history of women field biologists is intertwined with men's changing views of female intellect and with increasing educational opportunities available to women. Given the preponderance of today's professional female ecologists, animal behaviorists, systematists, conservation biologists, wildlife biologists, restoration ecologists, and natural historians, it is time to tell this story—the challenges and hardships they faced and still face, and the prominent role they have played and increasingly play in understanding our natural world.For a broader perspective, we profile selected European women field biologists, but our primary focus is the journey of women field biologists in North America.
