Integrins are heterodimeric cell surface receptors which anchor cells to different extracellular matrix proteins or act as cell-cell receptors. They play pivotal roles not only across a wide range of physiological processes including tissue morphogenesis, wound healing, and regulation of cell growth, but also in numerous pathological conditions such as autoimmunity, infectious disease, and carcinogenesis. The tumor environment is a dynamic network that includes cancer cells, immune cells, fibroblasts, endothelial cells, extracellular matrix, cytokines and receptors. The aim of this book is to summarize the role of these components, especially immune cells, in tumor suppression and/or progression and describe in detail why tumor cells can survive and spread in spite of the antitumor response of immune cells. Chemically and biochemically optimized preparation of adequate tools, their application for localization of receptor (tissue lectin) and ligand (cellular glycoconjugate) pairs in tumor cells and tumor sections and the usefulness of a lectin from a plant extract as potent immunomodulator indicate the prospect for a place of such techniques in pathology and oncology.

This volume of Current Topics in Microbiology and Immunology was planned in parallel with an EM BO workshop on cell-cell Interactions in Leukocyte Homing and Differentiation held at the Basel Institute for Immunology in November 1992, and many of the workshop speakers have contributed to it. Cell adhesion is one of the most dynamic fields of biological research and presented in this book is the current knowledge on the structure and function of the major families of cell adhesion molecules-the integrins, the selectins, the immunoglobulin superfamily, and CD44. Complex interactions between the members of these families mediate diverse adhesion functions, including leukocyte-leukocyte interactions, lymphocyte homing, inflammation, and lymphocyte-stromal cell interaction during hematopoiesis. A great deal of emphasis is placed on the regulatory elements that control the expression and function of adhesion molecules. Cytokines not only induce the expression of certain adhesion molecules, but may also modify their functional status. For instance, the integrins exist in either an inactive nonfunctional form or an active functional form, and a number of intracellular or extracellular stimuli modify integrin function. This is particularly important during leukocyte binding to endothelium and transendothelial migration, which proceeds through a cascade of adhesion events. Although cell adhesion molecules play an important role in many processes, this book concentrates on their role within the immune system. A number of chapters discuss the migration of lymphocytes between hematopoietic organs such as the thymus, lymph nodes, Peyer's patches, and spleen.

Chronic inflammation predisposes to some forms of cancer and the host response to malignant disease shows several parallels with inflammation and wound healing. The cells involved in inflammation are detected in a range of common cancers, together with the inflammatory cytokines and members of the chemokine ligand/receptor systems. Neutralization or deletion of the gene for some inflammatory cytokines confers resistance to tumour induction and experimental metastasis. Over-expression of such cytokines in tumour cells may enhance malignant potential. Certain chemokines are likely to subvert antitumour immunity by favouring development of ineffective Type 2 responses. Tumour cells may even utilize chemokine receptors in homing to lymph nodes and other organs. Thus, the cells, cytokines and chemokines found in tumours are more likely to contribute to tumour growth, progression and immunosuppression than they are to mount an effective host antitumour response. This book draws together contributions from an international group of scientists and clinicians from diverse disciplines, ranging from epidemiology to immunology, cell biology, molecular oncology, molecular medicine and pharmacology to debate these and related issues.
