We have surpassed the omics era and are truly in the Age of Molecular Therapeutics. The fast-paced development of SARS-CoV-2 vaccines, such as the mRNA vaccines encoding the viral spike protein, demonstrated the need for and capability of molecular therapy and nanotechnology-based solutions for drug delivery. In record speed, the SARS-CoV-2 viral RNA genome was sequenced and shared with the scientific community, allowing the rapid design of molecular therapeutics. The mRNA vaccines exploit the host cell endoplasmic reticulum to produce viral spike proteins for antigen presentation and recognition by the innate and adaptive immune system. Lipid nanoparticles enable the delivery of the fragile, degradation-sensitive nucleic acid payloads. Molecular-based therapeutics and nanotechnology solutions continue to drive the scientific and medical response to the COVID-19 pandemic as new mRNA, DNA, and protein-based vaccines are developed and approved and the emergency use approved vaccines are rapidly manufactured and distributed throughout the globe. The need for molecular therapies and drug delivery solutions is clear, and as these therapies progress and become more specialized there will be important advancements in organelle targeting. Stark indications of the multi-faceted dangers we face can be seen in the chemical weapons attacks against civilians and combatants in Iraq and Syria, and also in more targeted chemical assassination operations in Malaysia and the UK. Using a multi-disciplinary approach, and drawing upon an international group of experts, this book analyses current and likely near-future advances in relevant science and technology, assessing the risks of their misuse. In this collection, the authors review the currently available data on interactions of ribosomal components with canonical mRNAs and with hepatitis C virus (HCV)-like IRESes during translation and on conformational rearrangements in the ribosomes accompanying these interactions. This review considers data obtained by biochemical approaches, primarily by site-directed cross-linking with the application of mRNA and IRES analogues bearing reactive groups in definite locations, and models for translational complexes of higher eukaryotic ribosomes derived from cryo-electron microscopy. Afterwards, the authors discuss how the addition of the poly(A) tail of 30-100 adenosines to the 3´-end of mRNA has been shown to be important for the stability, nuclear export, export of mRNA to the cytoplasm and translation of a transcript. The molecular mechanism responsible for the definition of a poly(A) site includes several recognition, cleavage and polyadenylation factors that identify the poly(A) signal, usually AAUAAA hexamer and similar variants, located 10-35 nucleotides upstream of the cleavage site. The process known as SUMO (small ubiquitin-related modifier) conjugation to a lysine residue in the target protein or sumoylation is a rapid, reversible post-transcriptional modification is also examined. Similar to ubiquitin, SUMO isoforms are conjugated to their targets by a isopeptide bond between their C-terminal glycine and the ɛ-NH2 group of the target lysine residue. Next, the authors discuss splicing of pre-mRNAs coding for proteins. Since the discovery of frequent spliceosomal gene mutations which occur in about 65% of patients with myelodysplastic syndrome (MDS), we know that splicing factor gene mutations are the most frequent mutations found in MDS patients. Predominantly, four splicing factor genes (SF3B1, SRSF2, U2AF1, ZRSR2) are mutated in MDS and related malignancies. In closing, the authors present their findings that splicing is not only necessary for obtaining correct open reading frames, but also leads to deposition of the exon junction complex core. Essentially, all pre-mRNAs are cleaved and polyadenylated with exception of histone mRNAs.

Artemisinin, a sesquiterpene lactone originally extracted from the medicinal plant Artemisia annua L., is an effective antimalarial agent, particularly for multi-drug resistant and cerebral malaria. However, the concentration of artemisinin in the plant is very low. Because the chemical synthesis of artemisinin is complicated and not economically feasible in view of the poor yield of the drug, the intact plant remains the only viable source of artemisinin production. Therefore, it is necessary to increase the concentration of artemisinin in A. annua to reduce the cost of artemisinin based antimalarial drugs. Transboundary animal diseases (TADs) are a major threat to livestock. They are highly contagious or transmissible, and they have the potential to cause high morbidity and mortality in both susceptible animal populations and humans. In addition, not only are TADs detrimental to national economies, they are also a serious threat to world food security.
