In her study Elisabeth Salzer describes three novel monogenic diseases. For CD27 deficiency Elisabeth Salzer describes a large cohort of patients. Although all patients shared the same causative missense mutation, they displayed diverse clinical presentations. In another patient she was able to identify a mutation in PRKCD resulting in a primary immunodeficiency with severe Lupus-like autoimmunity. The patient exhibited increased mRNA levels of IL6. Therefore, treatment with Tocilizumab, a humanized anti-IL-6 receptor monoclonal antibody was suggested. In a family with a history of deaths due to inflammatory bowel disease she identified a missense mutation in IL21. She produced wild type and mutated IL-21 protein and demonstrated a loss of function phenotype. As IL-21 is in clinical trials, she proposed a potentially curative treatment option. These discoveries contributed to the understanding of the multifaceted regulatory mechanisms of the immune systemand highlighted essential players in these complex signaling networks.

This book discusses the immunotherapeutic potential of Interleukin 12 in the context of clinical oncology, as well as antitumor effects confirmed in preclinical studies and clinical trials in cancer immunotherapy. This book provides an updated overview of agammaglobulinemia, a rare form of primary immunodeficiency which is considered the prototype of the congenital humoral defects, and which is characterized by the absence of peripheral B cells and very low serum immunoglobulin levels. The book opens by discussing the highly orchestrated early B cell development in the bone marrow and the genes involved based on both human and animal models. The pathogenesis and clinical presentation of X-linked agammaglobulinemia, caused by mutations in the BTK (Bruton's tyrosine kinase) gene, are then presented in detail, followed by descriptions of the clinical manifestations and molecular basis of the less frequent autosomal recessive and autosomal dominant forms of agammaglobulinemia. Primary immunodeficiency diseases (PIDs) are a heterogeneous group of inherited disorders characterized by different defects in the development and function of the immune system. A major process of rediscovery has taken place in the field of Cellular Immunology over the past 12 years—subsets of T lymphocytes exist that are specifically dedicated to regulation or as it should be more appropriately termed suppression of all aspects of immune responses. It was only after Prof. Sakaguchi identified the CD25 antigen in 1995 as a marker for a major population of T cells that had suppressor functions both in vitro and in vivo that the resurgence in the regulatory T cell area could begin. The regulatory T cells field has grown dramatically over the past decade. It is now impossible to read a journal that does not contain numerous papers whose titles deal with regulatory T cells. More importantly, it is also difficult to submit a new research grant proposal in any area of immunologic research that does not include a section on analysis on the contribution of regulatory T cells to the subject matter under study. Regulatory T-cells are essential components of the immune system, and several different subsets of regulatory T-cells have been described. Considerable regulatory function has been attributed to the CD4+CD25+ T-cell subset.
