Background Global estimates for cholera annually approximate 4 million cases worldwide with 95,000 deaths. Recent outbreaks, including Haiti and Yemen, are reminders that cholera is still a global health concern. Cholera outbreaks can rapidly induce high death tolls by overwhelming the capacity of health facilities, especially in remote areas or areas of civil unrest. Recent studies demonstrated that stool specimens preserved on filter paper facilitate molecular analysis of Vibrio cholerae in resource limited settings. Specimens preserved in a rapid, low-cost, safe and sustainable manner for sequencing provides previously unavailable data about circulating cholera strains. This may ultimately contribute new information to shape public policy response on cholera control and elimination. Methodology/Principal findings Whole genome sequencing (WGS) recovered close to a complete sequence of the V. cholerae O1 genome with satisfactory genome coverage from stool specimens enriched in alkaline peptone water (APW) and V. cholerae culture isolates, both spotted on filter paper. The minimum concentration of V. cholerae DNA sufficient to produce quality genomic information was 0.02 ng/μL. The genomic data confirmed the presence or absence of genes of epidemiological interest, including cholera toxin and pilus loci. WGS identified a variety of diarrheal pathogens from APW-enriched specimen spotted filter paper, highlighting the potential for this technique to explore the gut microbiome, potentially identifying co-infections, which may impact the severity of disease. WGS demonstrated that these specimens fit within the current global cholera phylogenetic tree, identifying the strains as the 7th pandemic El Tor. Conclusions WGS results allowed for mapping of short reads from APW-enriched specimen and culture isolate spotted filter papers. This provided valuable molecular epidemiological sequence information on V. cholerae strains from remote, low-resource settings. These results identified the presence of co-infecting pathogens while providing rare insight into the specific V. cholerae strains causing outbreaks in cholera-endemic areas., Author summary Cholera affects more than 4 million people globally every year; people predominantly living in poverty or in resource-constrained conditions including political crises or natural disasters. Cholera’s typical presentation is characterized by rapid onset of acute watery diarrhea and vomiting which can progress from watery stool to shock in as little as four hours. Laboratory conditions needed for culture confirmation and strain preservation are rarely to never present in these affected areas. In fact, many cholera endemic areas in Sub-Saharan African are so remote that even treatment response alone is often challenging. Here we present the genomic analysis of DNA extracted from dried filter paper, which is a low-cost, low-tech and sustainable method. Previously this method has facilitated cholera confirmation by PCR, but we demonstrate that this method is also suitable for whole genome sequencing and subsequent strain characterization by presenting the analysis of samples from an outbreak in a remote area of Cameroon. This method will facilitate the understanding of the molecular epidemiology in cholera-prone areas, which were previously too challenging to attempt. It also introduces a method that can be used on a broader scale for diarrheal disease surveillance, including providing a window into co-infection and microbiome analyses.

International audience; The incidence of arbovirus infections has increased dramatically in recent decades, affecting hundreds of millions of people each year. The Togaviridae family includes the chikungunya virus (CHIKV), which is typically transmitted by Aedes mosquitoes and causes a wide range of symptoms from flu-like fever to severe arthralgia. Although conventional diagnostic tests can provide early diagnosis of CHIKV infections, access to these tests is often limited in developing countries. Consequently, there is an urgent need to develop efficient, affordable, simple, rapid, and robust diagnostic tools that can be used in point-of-care settings. Early diagnosis is crucial to improve patient management and to reduce the risk of complications. A glass-fiber laser-cut microfluidic device (paper-based analytical device [PAD]) was designed and evaluated in a proof of principle context, for the analysis of 30 µL of patient serum. Biological raw materials used for the functionalization of the PAD were first screened by MAC-ELISA (IgM capture enzyme-linked immunosorbent assay) for CHIKV Immunoglobulin M (IgM) capture and then evaluated on the PAD using various human samples. Compared with viral lysate traditionally used for chikungunya (CHIK) serology, CHIKV pseudo-particles (PPs) have proven to be powerful antigens for specific IgM capture. The PAD was able to detect CHIKV IgM in human sera in less than 10 minutes. Results obtained in patient sera showed a sensitivity of 70.6% and a specificity of around 98%. The PAD showed few cross-reactions with other tropical viral diseases. The PAD could help health workers in the early diagnosis of tropical diseases such as CHIK, which require specific management protocols in at-risk populations.

International audience; The diagnosis of infectious diseases is entering a new and interesting phase. Technologies based on paper microfluidics, coupled to developments in isothermal amplification of Nucleic Acids (NAs) raise opportunities for bringing the methods of molecular biology in the field, in a low setting environment. A lot of work has been performed in the domain over the last few years and the landscape of contributions is rich and diverse. Most often, the level of sample preparation differs, along with the sample nature, the amplification and detection methods, and the design of the device, among other features. In this review, we attempt to offer a structured description of the state of the art. The domain is not mature and there exist bottlenecks that hamper the realization of Nucleic Acid Amplification Tests (NAATs) complying with the constraints of the field in low and middle income countries. In this domain however, the pace of progress is impressively fast. This review is written for a broad Lab on a Chip audience.

International audience; Scenario analysis constitutes a useful approach to synthesize knowledge and derive hypotheses in the case of complex systems that are documented with mainly qualitative or very diverse information. In this article, a framework for scenario analysis is designed and then, applied to global wheat health within a timeframe from today to 2050. Scenario analysis entails the choice of settings, the definition of scenarios of change, and the analysis of outcomes of these scenarios in the chosen settings. Three idealized agrosystems, representing a large fraction of the global diversity of wheat-based agrosystems, are considered, which represent the settings of the analysis. Several components of global changes are considered in their consequences on global wheat health: climate change and climate variability, nitrogen fertilizer use, tillage, crop rotation, pesticide use, and the deployment of host plant resistances. Each idealized agrosystem is associated with a scenario of change that considers first, a production situation and its dynamics, and second, the impacts of the evolving production situation on the evolution of crop health. Crop health is represented by six functional groups of wheat pathogens: the pathogens associated with Fusarium head blight; biotrophic fungi, Septoria-like fungi, necrotrophic fungi, soilborne pathogens, and insect-transmitted viruses. The analysis of scenario outcomes is conducted along a risk-analytical pattern, which involves risk probabilities represented by categorized probability levels of disease epidemics, and risk magnitudes represented by categorized levels of crop losses resulting from these levels of epidemics within each production situation. The results from this scenario analysis suggest an overall increase of risk probabilities and magnitudes in the three idealized agrosystems. Changes in risk probability or magnitude however vary with the agrosystem and the functional groups of pathogens. We discuss the effects of global changes on the six functional groups, in terms of their epidemiology and of the crop losses they cause. Scenario analysis enables qualitative analysis of complex systems, such as plant pathosystems that are evolving in response to global changes, including climate change and technology shifts. It also provides a useful framework for quantitative simulation modeling analysis for plant disease epidemiology.

International audience; The authors explored the risks and benefits of repeated irrigation of Populus alba saplings with aqueous paper sludge (APS). Saplings were cultivated in pots of forest soil (3 L) in a greenhouse for 7 weeks and watered twice a week with differing concentrations of APS (0, 10, 20, 30, 50, 75,and 100 % v/v with deionized water). Plant growth and ecophysiological variables along with zinc and aluminum transfer were monitored. A stimulation of plant growth was observed with sludge treatments of 30 or 50 %, significantly correlated to APS input (r = 0.81). This may be explained by the easily available nitrogen as is shown with the positive correlation of CO2 assimilation and leaf nitrogen (r = 0.70). However, a significant reduction in plant growth was observed when treatments of 75 and 100 % of APS were administered, despite a high nutritional level (nitrogen and phosphorus). The study suggests that APS concentrations from 30 to 50 % may positively affect the growth of poplar saplings; however, the higher concentrations indicated a risk for plant growth and the environment.

International audience; Water cleaning of paper leaves is a preliminary step in many customary deacidification treatments of ancient books in conservation workshops. Usually, the washing solutions are considered “waste waters” and they are thrown away. In this work, an analytical protocol for the analysis of wash waters for the assessment of the conservation state of ancient books is proposed. Wash waters of leaves under different conservation conditions of a 16th-century- rinted book were investigated using UV-Vis, IR, Raman, NMR, EPR, XRF, ESI-MS, ICP-MS, CE-PDA and elemental microanalysis. Analysis of the wash water extracts allowed one to identify a large number of degradation by-products, ranging from low-molecular mass organic acids to cellulose oligomers. Reasons for the very different conservation conditions of the leaves emerged, indicating that wash water analysis provides a deep insight into the conservation state and composition of leaves as well as degradation reactions occurred on them

Genetic polymorphism can be maintained over time by negative frequency-dependent (FD) selection induced by Rock-paper-scissors (RPS) social systems. RPS games produce cyclic dynamics, and have been suggested to exist in lizards, insects, isopods, plants, and bacteria. Sexual selection is predicted to accentuate the survival of the future progeny during negative FD survival selection. More specifically, females are predicted to select mates that produce progeny genotypes that exhibit highest survival during survival selection imposed by adult males. However, no empirical evidence demonstrates the existence of FD sexual selection with respect to fitness payoffs of genetic polymorphisms. Here we tested this prediction using the common lizard Zootoca vivipara, a species with three male color morphs (orange, white, yellow) that exhibit morph frequency cycles. In a first step we tested the congruence of the morph frequency change with the predicted change in three independent populations, differing in male color morph frequency and state of the FD morph cycle. Thereafter we ran standardized sexual selection assays in which we excluded alternative mechanisms that potentially induce negative FD selection, and we quantified inter-sexual behavior. The patterns of sexual selection and the observed behavior were in line with context-dependent female mate choice and male behavior played a minor role. Moreover, the strength of the sexual selection was within the magnitude of selection required to produce the observed 3-4-year and 6-8 year morph frequency cycles at low and high altitudes, respectively. In summary, the study provides the first experimental evidence that underpins the crucial assumption of the RPS games suggested to exist in lizards, insects, isopods, and plants; namely, that sexual selection produces negative-FD selection. This indicates that sexual selection, in our study exert by females, might be a crucial driver of the maintenance of genetic polymorphisms.

International audience; Microbial anodes were formed under polarization at −0.3 V/SCE on graphite plates in effluents from a pulp and paper mill. The bioanodes formed with the addition of acetate led to the highest current densities (up to 6 A/m2) but were then unable to oxidize the raw effluent efficiently (0.5 A/m2). In contrast, the bioanodes formed without acetate addition were fully able to oxidize the organic matter contained in the effluent, giving up to 4.5 A/m2 in continuous mode. Bacterial communities showed less bacterial diversity for the acetate-fed bioanodes compared to those formed in raw effluents. Deltaproteobacteria were the most abundant taxonomic group, with a high diversity for bioanodes formed without acetate addition but with almost 100% Desulfuromonas for the acetate-fed bioanodes. The addition of acetate to form the microbial anodes induced microbial selection, which was detrimental to the treatment of the raw effluent.

Waste copier paper is a potential substrate for the production of glucose relevant for manufacture of platform chemicals and intermediates, being composed of 51 % glucan. The yield and concentration of glucose arising from the enzymatic saccharification of solid ink-free copier paper as cellulosic substrate was studied using a range of commercial cellulase preparations. The results show that in all cellulase preparations examined, maximum hydrolysis was only achieved with the addition of beta-glucosidase, despite its presence in the enzyme mixtures. With the use of AccelleraseA (R) (cellulase), high substrate loading decreased conversion yield. However, this was overcome if the enzyme was added between 12.5 and 20 FPU g substrate(-1). Furthermore, this reaction condition facilitated continual stirring and enabled sequential additions (up to 50 % w/v) of paper to be made to the hydrolysis reaction, degrading nearly all (99 %) of the cellulose fibres and increasing the final concentration of glucose whilst simultaneously making high substrate concentrations achievable. Under optimal conditions (50 A degrees C, pH 5.0, 72 h), digestions facilitate the production of glucose to much improved concentrations of up to 1.33 mol l(-1).

Agar substrates forin vitroculture are well adapted to plant micropropagation, but not to plant rooting and acclimatization. Conversely, paper-pulp-based substrates appear as potentially well adapted forin vitroculture and functional root production. To reinforce this hypothesis, this study comparesin vitrodevelopment of nemesia on several substrates. Strong differences between nemesia roots growing in agar or in paper-pulp substrates were evidenced through scanning electron microscopy. Roots developed in agar have shorter hairs, larger rhizodermal cells, and less organized root caps than those growing on paper pulp. In conclusion, it should be noted that in this study,in vitromicroporous substrates such as paper pulp lead to the production of similar root hairs to those found in greenhouse peat substrates. Consequently, if agar could be used for micropropagation, rooting, and plant acclimatization, enhancement could be achieved if rooting stage was performed on micro-porous substrates such as paper pulp.

International audience; During the process of papermaking by pulp and paper plants, a thick and viscous deposits, termed slime, is quickly formed around the paper machines, which can affect the papermaking process. In this study, we explored the composition of the bacterial biota in slime that developed on shower pipes from 2 machines at a Canadian paper mill. Firstly, the composition was assessed for 12 months by DNA profiling with polymerase chain reaction coupled with denaturing gradient gel electrophoresis. Except for short periods (2-3 months), clustered analyses showed that the bacterial composition of the slime varied substantially over the year, with less than 50% similarity between the denaturing gradient gel electrophoresis profiles. Secondly, the screening of 16S rRNA gene libraries derived from 2 slime samples showed that the most abundant bacteria were related to 6 lineages, including Chloroflexi, candidate division OP10, Clostridiales, Bacillales, Burkholderiales, and the genus Deinococcus. Finally, the proportion of 8 bacterial lineages, such as Deinococcus sp., Meiothermus sp., and Chloroflexi, was determined by the Catalyzed Reporter Deposition-Fluorescence In Situ Hybridization in 2 slime samples. The results showed a high proportion of Chloroflexi, Tepidimonas spp., and Schlegelella spp. in the slime samples.

International audience; A new process using aminoalkylalkoxysilanes (AAAS) was developed recently for paper and book mass deacidification. This process is entirely novel in that, besides deacidifying, it also improves the mechanical properties of paper. In the present article, the properties of inhibition of fungal growth of the treated papers are investigated. It was found that the AAAS tested acted as surface-active antifungal agents when introduced in the paper, significantly reducing the growth of Aspergillus niger and Paecilomyces variotii, two fungal species commonly found in storage areas of libraries and archives. The AAAS which seemed the most efficient in preventing the fungal development was 3-aminopropylmethyldi-methoxysilane (AMDES), functionalised with a primary amine. The effect was significant in the different papers tested which demonstrated the protection against the proliferation of filamentous fungi in vitro for papers treated with AMDES. The higher concentration of AMDES inside the paper proved to be the most effective. The highest efficiency was reached for AAAS uptakes around 6%. At this concentration the sporulation of the two strains was inhibited and the development of A. niger mycelia was highly reduced.

International audience; Soy protein isolates (SPI) and octenyl-succinate (OSA) modified starch were used as paper coating and inclusion matrices of two antimicrobial compounds: cinnamaldehyde and carvacrol. Antimicrobial compound losses from the coated papers were evaluated after the coating and drying process, and the two matrices demonstrated retention ability that depended on the compound nature and concentration. Whereas carvacrol losses ranged between 12 and 45%, cinnamaldehyde losses varied from 43 to 76%. The losses were always higher from OSA-starch-coated papers than from SPI-coated papers. During storage in accelerated conditions, at 30 °C and 60% relative humidity, carvacrol retention from coated papers was found to be similar whatever the coating matrices and the carvacrol rate. In contrast, the retention from SPI-coated papers was particularly high for the cinnamaldehyde concentration of 30% (w/w) compared to the lowest (10% w/w) or highest concentration (60% w/w). Compared to carvacrol, faster release was observed, particurlarly when OSA-starch was used. The antimicrobial properties of the coated papers were shown against Escherichia coli and Botrytis cinerea and explained by favorable conditions of total release of the antimicrobial agents.

International audience; The transport properties of two methyl ketones, 2-heptanone and 2-nonanone, through a paper coated with a wheat gluten (WG) solution were studied as a function of aroma compound concentration differential and relative humidity. Whatever the conditions, coating improved the aroma barrier properties of the paper. Whereas permeability of 2-nonanone through WG coated paper was not affected by the concentration differential, a high concentration of 2-heptanone induced a permeability increase, which can be explained by a slight plasticization effect of this compound. The moisture content of the films markedly affected the barrier properties depending on the aroma compound. For 2-heptanone, the most polar compound, the increase of permeability with relative humidity was related to the increase of diffusivity and solubility coefficient of this compound. With 2-nonanone, permeability and diffusivity coefficients strongly decreased with the increase of moisture content. This decrease could be linked to the hindrance in diffusivity of the less polar compound induced by the presence of water molecules. The different behavior in permeation properties seems to be related to the hydrophobic nature of the aroma compound. At intermediate and high humidity levels, coated papers are found to have better aroma barrier properties than LDPE films.

Wheat gluten (WG), calcium caseinate (CC) and corn gluten meal (CGM), were selected as protein raw material to prepare coating solutions. Different concentrations of protein (10–20%, w/w) and deposited solution layer (50–500 μm) were used to obtain coated papers with thicknesses varying between 143(± 3) and 227(± 8) μm. As expected, the thicknesses of coated paper increased with coating weight, but, for a given coating weight, different thicknesses were observed according to the raw material used and its concentration in the coating solution. A phenomenon involving a penetration of coating solution inside paper was suspected with WG-based solutions. The microstructure of coated papers was analyzed by SEM, OM and AFM and differences in structure and morphology were observed between the three raw materials. The percentages of coating agent impregnated inside paper were also calculated from the apparent density of coated paper and the density of self-supported films prepared in the same conditions but deposited on an inert and smooth Plexiglas® support. These percentages of impregnation ranged from 4.8 to 63.3% and increased as following: CC

Gutiérrez), Different model lipids—alkanes, fatty alcohols, fatty acids, resin acids, free sterols, sterol esters, and triglycerides—were treated with Pycnoporus cinnabarinus laccase in the presence of 1-hydroxybenzotriazole as mediator, and the products were analyzed by gas chromatography. The laccase alone decreased the concentration of some unsaturated lipids. However, the most extensive lipid modification was obtained with the laccase–mediator system. Unsaturated lipids were largely oxidized and the dominant products detected were epoxy and hydroxy fatty acids from fatty acids and free and esterified 7-ketosterols and steroid ketones from sterols and sterol esters. The former compounds suggested unsaturated lipid attack via the corresponding hydroperoxides. The enzymatic reaction on sterol esters largely depended on the nature of the fatty acyl moiety, i.e., oxidation of saturated fatty acid esters started at the sterol moiety, whereas the initial attack of unsaturated fatty acid esters was produced on the fatty acid double bonds. In contrast, saturated lipids were not modified, although some of them decreased when the laccase–mediator reactions were carried out in the presence of unsaturated lipids suggesting participation of lipid peroxidation radicals. These results are discussed in the context of enzymatic control of pitch to explain the removal of lipid mixtures during laccase–mediator treatment of different pulp types.

Nosocomial infections are contracted in a hospital or health care facility. Bacterial cause is the most common in 87% of cases. Studies have been conducted for the detection of these bacteria. The aim of this biosensor is to make an early detection tool, easy to use, reliable, not invasive and low cost. The goal is to detect in real time and simultaneously in a small amount of body fluids such as urine, sweat or saliva, the major bacterial pathogens causing these diseases: Escherichia Coli, Pseudomonas aeroginosa, Acinetobacter baumannii, Staphylococcus aureus and Clostridium difficile.

The paper wasp Polistes atrimandibularis is an obligatory social parasite of another Polistes species, P. biglumis bimaculatus. To control the host nest, the parasite sequentially changes the composition of its chemical signature, the cuticular hydrocarbons, during the colonial cycle. Gas chromatography-mass spectrometry of the cuticular hydrocarbons at every stage of the cycle showed that the parasite can switch on and off an entire chemical family, namely, the unsaturated hydrocarbons. In this way the parasite can match the host signature at a critical moment of the colonial cycle.

Adhesive properties of a recently isolated, mesophilic, cellulolytic Clostridium (strain A22) and ofClostridium cellulolyticum (ATCC 35319) were described. For study of these properties, a method with Whatman No. 1 filter paper was developed, allowing faster kinetic analysis measuring “unbound” cells. This method was confirmed with [methyl-3H] thymidine to measure directly “bound” cells. The results of kinetic analysis indicated for the two clostridia a two-step adhesion process. In the first step, cell-cellulose interaction was observed. In the second step, cell-cell interaction was determined with an aggregation of cells away from the surface. This result was confirmed by scanning electron microscopy. Competition between strain A22 andC. cellulolyticum for adhesion sites was investigated. The two clostridia have the same specific adhesion sites was investigated. The two clostridia have the same specific adhesion sites on cellulose (filter paper), and unspecific interactions between two strains occur.

We revisit an old Russian paper by V.V. Dynnik, R. Heinrich and E.E. Sel'kov (1980a,b) describing: "A mathematical model of carbohydrate energy metabolism. Interaction between glycolysis, the Krebs cycle and the H-transporting shuttles at varying ATPases load". We analyse the model mathematically and calculate the control coefficients as a function of ATPase loads. We also evaluate the structure of the metabolic network in terms of elementary flux modes. We show how this model can respond to an ATPase load as well as to the glucose supply. We also show how this simple model can help in understanding the articulation between the major blocks of energetic metabolism, i.e. glycolysis, the Krebs cycle and the H-transporting shuttles.

Pentamidine, an antiprotozoal drug, was shown to have various cellular and molecular targets depending on the organism. In Leishmania, ultrastructural modifications of kinetoplast and mitochondria have been observed but no data is available on cellular and molecular events involved in development of pentamidine-resistance. The absence of modification of minicircle DNA in pentamidine treated L. donovani and L. amazonensis promastigotes suggested that topoisomerase II activity is not a target. This result was confirmed by quantitation of the enzyme by immunodetection. Southern blot experiments indicated that the kDNA network was altered in resistant clones. Molecular cloning and sequence analysis of kDNA minicircles showed transkinetoplastidy hitherto reported only for arsenite- and tunicamycin-resistant Leishmania. Comparison of wild-type and resistant sequences showed only 32-51% homology. The AT-rich regions, known as binding sites, of the drug occurred less frequently in the resistant clones and their locations were different. These minicircle sequence modifications leading to decreased binding sites for the drug might contribute to pentamidine-resistance in Leishmania.

International audience; Plasmodium falciparum secretes several proteins that cause changes in the erythrocyte membrane enabling it to survive within red blood cells. Little is known of the mechanisms involved in the secretion and targeting of parasite polypeptides to the various cell compartments. The P. falciparum gene homologous to the mammalian Sec61alpha, gene, which encodes a component of the translocation pore in the endoplasmic reticulum of eukaryotic cells, was characterised to investigate the translocation process in the parasite. PfSec61 is present as a unique copy in the parasite genome and was mapped to chromosome 13. It encodes a 40 kDa polypeptide, as shown by immunoblotting and immunoprecipitation of [35S]methionine metabolically-labelled parasite extracts. The deduced amino acid sequence of PfSec61 is 87% similar to the mammalian polypeptide, and the two proteins give similar hydropathy plots. These results strongly suggest that PfSec61 has the same topological orientation and functional role as Sec61alpha. Anti-PfSec61 antibodies were used to investigate the cellular location and kinetics of expression of the polypeptide in the parasite. Immunofluorescence confocal microscopy showed that PfSec61 was located in the parasite cytoplasm, close to the nucleus, in a position consistent with its being in the endoplasmic reticulum.

Using oligonucleotides derived from Pf60.1, a member of the Plasmodium falciparum Pf60 multigene family, numerous fragments were amplified from genomic and cDNA from the 3D7 P. falciparum clone. DNA sequencing showed that the various fragments presented considerable diversity, indicating that the 3D7 repertoire contains at least 20 distinct versions of the region analysed. The various sequences aligned with either of two prototype sequences. Characteristic of the A-type was the presence of a 21 bp motif, present in variable copy number, as well as a sequence homologous to the Babesia sp. RAP-1 consensus. The B prototype sequence did not present such features and substantially differed from the A-type, due to accumulation of point mutations and numerous triplet deletions. Consistent with the marked differences between both sub-families, individual members from each sub-family did not cross-hybridise, produced distinct multiple band patterns on Southern blots and distinct chromosome profiles. Numerous hybrid sequences were observed. Interestingly, most var genes and var-related unspliced cDNAs described so far are of A/B hybrid type. These data suggest that the family has evolved by successive amplifications from two ancestral copies, with accumulation of mutations, as well as recombination and/or gene conversion events.

Summary Plant architecture is central in determining crop yield. In the short‐day species strawberry, a crop vegetatively propagated by daughter‐plants produced by stolons, fruit yield is further dependent on the trade‐off between sexual reproduction (fruits) and asexual reproduction (daughter‐plants). Both are largely dependent on meristem identity, which establishes the development of branches, stolons and inflorescences.Floral initiation and plant architecture are modulated by the balance between two related proteins, FLOWERING LOCUS T (FT) and TERMINAL FLOWER 1 (TFL1). We explored in woodland strawberry the role of the uncharacterised FveFT2 and FveFT3 genes and of the floral repressor FveTFL1 through gene expression analyses, grafting and genetic transformation (overexpression and gene editing).We demonstrate the unusual properties of these genes. FveFT2 is a nonphotoperiodic florigen permitting short‐day (SD) flowering and FveTFL1 is the long‐hypothesised long‐day systemic antiflorigen that contributes, together with FveFT2, to the photoperiodic regulation of flowering. We additionally show that FveFT3 is not a florigen but promotes plant branching when overexpressed, that is likely to be through changing axillary meristem fate, therefore resulting in a 3.5‐fold increase in fruit yield at the expense of stolons.We show that our findings can be translated into improvement of cultivated strawberry in which FveFT2 overexpression significantly accelerates flowering.

BACKGROUND: This dataset relates to the biodiversity census carried out during the Belgica 121 (B121) expedition to the Western Antarctic Peninsula from February to March 2019. One of the aims of the campaign was to explore the surroundings of the Gerlache Strait and to carry out a detailed biodiversity census focusing on inter- and subtidal shallow-water areas using both classic descriptive marine ecology methods, as well as state-of-the art techniques (habitat mapping, genetics, trophic ecology). The biodiversity census was carried out onboard a nimble research vessel, RV Australis. This dataset will offer access to the raw data on biodiversity occurrences, obtained using a range of methods described in this data paper. International audience; VHH stands for the variable regions of heavy chain only of camelid IgGs. The VHH family forms a set of interesting proteins derived from antibodies that maintain their capacity to recognize the antigen, despite their relatively small molecular weight (in the 12,000 Da range). Continuing our exploration of the possibilities of those molecules, we chose to design alternative molecules with maintained antigen recognition, but enhanced capacity, by fusing four VHH with one Fc, the fragment crystallizable region of antibodies. In doing so, we aimed at having a molecule with superior quantitative antigen recognition (×4) while maintaining its size below the 110 kDa. In the present paper, we described the building of those molecules that we coined VHH2-Fc-VHH2. The structure of VHH2-Fc-VHH2 in complex with HER2 antigen was determined using electronic microscopy and modeling. The molecule is shown to bind four HER2 proteins at the end of its flexible arms. VHH2-Fc-VHH2 also shows an internalization capacity via HER2 receptor superior to the reference anti-HER2 monoclonal antibody, Herceptin®, and to a simple fusion of two VHH with one Fc (VHH2-Fc). This new type of molecules, VHH2-Fc-VHH2, could be an interesting addition to the therapeutic arsenal with multiple applications, from diagnostic to therapy.

International audience; The identification of quantitative trait loci (QTL) through genome-wide association studies (GWAS) is a powerful method for unravelling the genetic background of selected traits and improving early-stage predictions. In honey bees (Apis mellifera), past genetic analyses have particularly focused on individual queens and workers. In this study, we used pooled wholegenome sequences to ascertain the genetic variation of the entire colony. In total, we sampled 216 Apis mellifera mellifera and 28 Apis mellifera carnica colonies. Different experts subjectively assessed the gentleness and calmness of the colonies using a standardised protocol. Conducting a GWAS for calmness on 211 purebred A. m. mellifera colonies, we identified three QTL, on chromosomes 8, 6, and 12. The two first QTL correspond to LOC409692 gene, coding for a disintegrin and metalloproteinase domain-containing protein 10, and to Abscam gene, coding for a Dscam family member Abscam protein, respectively. The last gene has been reported to be involved in the domestication of A. mellifera. The third QTL is located 13 kb upstream of LOC102655631, coding for a trehalose transporter. For gentleness, two QTL were identified on chromosomes 4 and 3. They are located within gene LOC413669, coding for a lap4 protein, and gene LOC413416, coding for a bicaudal C homolog 1-B protein, respectively. The identified positional candidate genes of both traits mainly affect the olfaction and nervous system of honey bees. Further research is needed to confirm the results and to better understand the genetic and phenotypic basis of calmness and gentleness.

Summary Ensuring faithful homologous recombination in allopolyploids is essential to maintain optimal fertility of the species. Variation in the ability to control aberrant pairing between homoeologous chromosomes in Brassica napus has been identified. The current study exploited the extremes of such variation to identify genetic factors that differentiate newly resynthesised B. napus, which is inherently unstable, and established B. napus, which has adapted to largely control homoeologous recombination.A segregating B. napus mapping population was analysed utilising both cytogenetic observations and high‐throughput genotyping to quantify the levels of homoeologous recombination.Three quantitative trait loci (QTL) were identified that contributed to the control of homoeologous recombination in the important oilseed crop B. napus. One major QTL on BnaA9 contributed between 32 and 58% of the observed variation. This study is the first to assess homoeologous recombination and map associated QTLs resulting from deviations in normal pairing in allotetraploid B. napus.The identified QTL regions suggest candidate meiotic genes that could be manipulated in order to control this important trait and further allow the development of molecular markers to utilise this trait to exploit homoeologous recombination in a crop., See also the Commentary on this article by Sourdille & Jenczewski, 229: 3041–3043.

International audience; Cerebral malaria is a neuroinflammatory disease induced by P. falciparum infection. In animal models, the neuro-pathophysiology of cerebral malaria results from the sequestration of infected red blood cells (iRBCs) in microvessels that promotes the activation of glial cells in the brain. This activation provokes an exacerbated inflammatory response characterized by the secretion of proinflammatory cytokines and chemokines, leading to brain infiltration by pathogenic CD8+ T lymphocytes. Astrocytes are a major subtype of brain glial cells that play an important role in maintaining the homeostasis of the central nervous system, the integrity of the brain-blood barrier and in mounting local innate immune responses. We have previously shown that parasitic microvesicles (PbA-MVs) are transferred from iRBCs to astrocytes. The present study shows that an unconventional LC3-mediated autophagy pathway independent of ULK1 is involved in the transfer and degradation of PbA-MVs inside the astrocytes. We further demonstrate that inhibition of the autophagy process by treatment with 3-methyladenine blocks the transfer of PbA-MVs, which remain localized in the astrocytic cell membrane and are not internalized. Moreover, bafilomycin A1, another drug against autophagy promotes the accumulation of PbA-MVs inside the astrocytes by inhibiting the fusion with lysosomes, and prevents ECM in mice infected with PbA. Finally, we establish that RUBCN/rubicon or ATG5 silencing impede astrocyte production in CCL2 and CXCL10 chemokines induced by PbA stimulation. Altogether, our data suggest that a non-canonical autophagy-lysosomal pathway may play a key role in cerebral malaria through regulation of brain neuro-inflammation by astrocytes.

International audience; Shellfish farming, a key sector of French aquaculture activity, allows the production of oyster spat in a controlled environment. Their production in commercial hatcheries requires control over the quality of the seawater used to sustain crossbreeding, breeding, and the production of fodder microalgae. Therefore, improving the filtration conditions of incoming water is crucial in ensuring the sustainability of production. An ultrafiltration pilot plant was therefore installed at Vendee Naissain. This ultrafiltration pilot plant allows filtration at 0.02 mu m; it is used upstream of hatcheries to eliminate pathogens and parasites that can influence the development of cultivated species and downstream to remove oyster gametes in hatchery effluents. The objectives of this work were: (i) to use ultrafiltered seawater for the culture of the microalga Isochrysis lutea (T-Iso) to determine whether better growth than that observed with borehole water, historically used by the producer, can be achieved; and (ii) to determine whether the use of ultrafiltered water results in better fertilization rates of the cupped oyster Crassostrea gigas compared to filtered and UV-treated seawater. Ultrafiltered water has shown definite efficiency for culturing T-Iso with rapid growth and significant reduction in contamination compared to cultivation in well water. The contribution of ultrafiltered water in hatching is more nuanced; ultrafiltered water does not stabilize hatch rates, and its quality is highly dependent on the quality of the seawater used.

International audience; Fine roots, and their functional traits, influence associated rhizosphere microorganisms via root exudation and root litter quality. However, little information is known about their relationship with rhizosphere microbial taxa and functional guilds. We investigated the relationships of 11 fine root traits of 20 sub-arctic tundra meadow plant species and soil microbial community composition, using phospholipid fatty acids (PLFAs) and high-throughput sequencing. We primarily focused on the root economics spectrum , as it provides a useful framework to examine plant strategies by integrating the co-ordination of belowground root traits along a resource acquisition-conservation trade-off axis. We found that the chemical axis of the fine root economics spectrum was positively related to fungal to bacterial ratios, but negatively to Gram-positive to Gram-negative bacterial ratios. However, this spectrum was unrelated to the relative abundance of functional guilds of soil fungi. Nevertheless, the relative abundance of arbuscular mycorrhizal fungi was positively correlated to root carbon content, but negatively to the numbers of root forks per root length. Our results suggest that the fine root economics spectrum is important for predicting broader groups of soil microorganisms (i.e. fungi and bacteria), while individual root traits may be more important for predicting soil microbial taxa and functional guilds.

The research done in this paper is focused on the properties of eight char samples that were produced by a pilot scale reactor: Spirajoule® located in France. This reactor converted different biomass and waste feedstock into three phase’s products: syngas, tars, and chars. In some cases, chars are considered as co-products without added value. However, they present numerous potential applications. Thus, this research is conducted precisely on the characterization of eight chars originating from biomass (waste wood, sawdust, miscanthus wood, and grape marc) and from waste (tires, municipal, and industrial RDF). Four property domains were studied in this research: physicochemical properties (moisture content, pH, skeletal density, and TGA), texture and morphology (SEM/EDX and BET specific surface area), surface chemistry (FTIR), and structure (XRD). Our results showed that the properties of these chars match the properties required in some applications such as soil amendment, catalyst, and water and gas treatment.

During the colonization process, the fungus secretes effector proteins that suppress immune responses and redirect the host metabolism in favor of the pathogen. As effectors play a critical role during plant colonization, their identification and functional characterization are essential to understanding biotrophy and disease.Using biochemical, molecular, and transcriptomic techniques, we performed a functional characterization of the U. maydis effector Jasmonate/Ethylene signaling inducer 1 (Jsi1).Jsi1 interacts with several members of the plant corepressor family Topless/Topless related (TPL/TPR). Jsi1 expression in Zea mays and Arabidopsis thaliana leads to transcriptional induction of the ethylene response factor (ERF) branch of the jasmonate/ethylene (JA/ET) signaling pathway. In A. thaliana, activation of the ERF branch leads to biotrophic susceptibility. Jsi1 likely activates the ERF branch via an EAR (ET-responsive element binding-factor-associated amphiphilic repression) motif, which resembles EAR motifs from plant ERF transcription factors, that interacts with TPL/TPR proteins.EAR-motif-containing effector candidates were identified from different fungal species, including Magnaporthe oryzae, Sporisorium scitamineum, and Sporisorium reilianum. Interaction between plant TPL proteins and these effector candidates from biotrophic and hemibiotrophic fungi indicates the convergent evolution of effectors modulating the TPL/TPR corepressor hub.

Triple‐negative breast cancer (TNBC) is a highly aggressive breast cancer subtype characterized by a remarkable molecular heterogeneity. Currently, there are no effective druggable targets and advanced preclinical models of the human disease. Here, a unique mouse model (MMTV‐R26Met mice) of mammary tumors driven by a subtle increase in the expression of the wild‐type MET receptor is generated. MMTV‐R26Met mice develop spontaneous, exclusive TNBC tumors, recapitulating primary resistance to treatment of patients. Proteomic profiling of MMTV‐R26Met tumors and machine learning approach show that the model faithfully recapitulates intertumoral heterogeneity of human TNBC. Further signaling network analysis highlights potential druggable targets, of which cotargeting of WEE1 and BCL‐XL synergistically kills TNBC cells and efficiently induces tumor regression. Mechanistically, BCL‐XL inhibition exacerbates the dependency of TNBC cells on WEE1 function, leading to Histone H3 and phosphoS33RPA32 upregulation, RRM2 downregulation, cell cycle perturbation, mitotic catastrophe, and apoptosis. This study introduces a unique, powerful mouse model for studying TNBC formation and evolution, its heterogeneity, and for identifying efficient therapeutic targets., Triple‐negative breast cancer (TNBC) is highly aggressive and currently lacks effective treatment. This study reports the generation of a unique mouse model developing spontaneous, exclusive TNBC, recapitulating heterogeneity and primary resistance to treatments. SummaryChloroplasts retain elements of a bacterial stress response pathway that is mediated by the signalling nucleotides guanosine penta- and tetraphosphate, or (p)ppGpp. In the model flowering plant Arabidopsis, ppGpp acts as a potent regulator of plastid gene expression and influences photosynthesis, plant growth and development. However, little is known about ppGpp metabolism or its evolution in other photosynthetic eukaryotes.Here, we studied the function of ppGpp in the diatom P. tricornutum using transgenic lines containing an inducible system for ppGpp accumulation. We used these lines to investigate the effects of ppGpp on growth, photosynthesis, lipid metabolism and protein expression.We demonstrate that ppGpp accumulation reduces photosynthetic capacity and promotes a quiescent-like state with reduced proliferation and ageing. Strikingly, using non-targeted proteomics, we discovered that ppGpp accumulation also leads to the coordinated upregulation of a protein protection response in multiple cellular compartments.Our findings highlight the importance of ppGpp as a fundamental regulator of chloroplast function across different domains of life, and lead to new questions about the molecular mechanisms and roles of (p)ppGpp signalling in photosynthetic eukaryotes.

Forum Paper Insect chemical ecology (ICE) evolved as a discipline concerned with plant–insect interactions, and also with a strong focus on intraspecific pheromone-mediated communication. Progress in this field has rendered a more complete picture of how insects exploit chemical information in their surroundings in order to survive and navigate their world successfully. Simultaneously, this progress has prompted new research questions about the evolution of insect chemosensation and related ecological adaptations, molecular mechanisms that mediate commonly observed behaviors, and the consequences of chemically mediated interactions in different ecosystems. Themed meetings, workshops, and summer schools are ideal platforms for discussing scientific advancements as well as identifying gaps and challenges within the discipline. From the 11th to the 22nd of June 2018, the 11th annual PhD course in ICE was held at the Swedish University of Agricultural Sciences (SLU) Alnarp, Sweden. The course was made up of 35 student participants from 22 nationalities (Fig. 1a) as well as 32 lecturers. Lectures and laboratory demonstrations were supported by literature seminars, and four broad research areas were covered: (1) multitrophic interactions and plant defenses, (2) chemical communication focusing on odor sensing, processing, and behavior, (3) disease vectors, and (4) applied aspects of basic ICE research in agriculture. This particular article contains a summary and brief synthesis of these main emergent themes and discussions from the ICE 2018 course. Abnormal numerical and structural chromosome content is frequently found in human cancer. To test the role of aneuploidy in tumor initiation and progression, we generated mice with random aneuploidies by transient induction of polo-like kinase 4 (Plk4), a master regulator of centrosome number. Short-term chromosome instability (CIN) from transient Plk4 induction resulted in formation of aggressive T-cell lymphomas in mice with heterozygous inactivation of one p53 allele and accelerated tumor development in the absence of p53. Transient CIN increased the frequency of lymphoma-initiating cells with a specific karyotype profile, including trisomy of chromosomes 4, 5, 14, and 15 occurring early in tumorigenesis. Tumor development in mice with chronic CIN induced by an independent mechanism (through inactivation of the spindle assembly checkpoint) gradually trended toward a similar karyotypic profile, as determined by single-cell whole-genome DNA sequencing. Overall, we show how transient CIN generates cells with random aneuploidies from which ones that acquire a karyotype with specific chromosome gains are sufficient to drive cancer formation, and that distinct CIN mechanisms can lead to similar karyotypic cancer-causing outcomes.

Susceptibility to fungal pathogens and plant immunity in rice and wheat can be modulated by the presence of intraspecific neighbours and does not require the neighbours to be infected., As part of a trend towards diversifying cultivated areas, varietal mixtures are subject to renewed interest as a means to manage diseases. Besides the epidemiological effects of varietal mixtures on pathogen propagation, little is known about the effect of intraspecific plant–plant interactions and their impact on responses to disease. In this study, genotypes of rice (Oryza sativa) or durum wheat (Triticum turgidum) were grown with different conspecific neighbours and manually inoculated under conditions preventing pathogen propagation. Disease susceptibility was measured together with the expression of basal immunity genes as part of the response to intra-specific neighbours. The results showed that in many cases for both rice and wheat susceptibility to pathogens and immunity was modified by the presence of intraspecific neighbours. This phenomenon, which we term ‘neighbour-modulated susceptibility’ (NMS), could be caused by the production of below-ground signals and does not require the neighbours to be infected. Our results suggest that the mechanisms responsible for reducing disease in varietal mixtures in the field need to be re-examined.

The main protease (3CLp) of the SARS‐CoV‐2, the causative agent for the COVID‐19 pandemic, is one of the main targets for drug development. To be active, 3CLp relies on a complex interplay between dimerization, active site flexibility, and allosteric regulation. The deciphering of these mechanisms is a crucial step to enable the search for inhibitors. In this context, using NMR spectroscopy, we studied the conformation of dimeric 3CLp from the SARS‐CoV‐2 and monitored ligand binding, based on NMR signal assignments. We performed a fragment‐based screening that led to the identification of 38 fragment hits. Their binding sites showed three hotspots on 3CLp, two in the substrate binding pocket and one at the dimer interface. F01 is a non‐covalent inhibitor of the 3CLp and has antiviral activity in SARS‐CoV‐2 infected cells. This study sheds light on the complex structure‐function relationships of 3CLp and constitutes a strong basis to assist in developing potent 3CLp inhibitors., We report the liquid‐sate NMR spectroscopy analysis of the dimeric SARS‐CoV‐2 main protease (3CLp), including its backbone assignments, to study its complex conformational regulation. Background Atezolizumab is an inhibitor of programmed death-ligand 1 (PD-L1), used to treat advanced or metastatic bladder cancer, and in trials for non-invasive disease. In order to be eligible for treatment, patients require a PD-L1 immune cell score ≥ 5%, using the Ventana SP142 PD-L1 assay. Many laboratories do not have access to the required Ventana Benchmark Ultra stainer, and it is unclear if the assay performs similarly on other stainers. In this study, we compare SP142 assay results between Ventana Benchmark Ultra and Leica Bond-III stainers. Methods Serial sections of 90 samples of transurethral bladder resections (comprising 51 pTaHG, 8 pTis, 18 pT1, 10 pT2 tumors) were stained using the SP142 PD-L1 antibody on Ventana Benchmark Ultra and Leica Bond-III stainers, manually scored, and compared using accuracy and Cohen’s kappa measures. Results Both devices yielded highly concordant PD-L1 immune cell scores (accuracy 0.84, Cohen’s κ 0.732). Moreover, we found similar tumor cell (TC) PD-L1 scores using both stainers, and a trend towards greater TC scores in pT2 stage samples (p = 0.05). Conclusion This study is the first to compare the SP142 antibody in bladder cancer on two different stainers. Our results indicate that both Benchmark Ultra and Bond-III stainers yield highly concordant results using the SP142 PD-L1 antibody.

International audience; Climate change and energy demand are calling for more sustainable fuels such as biomethane produced by anaerobic digestion of organic waste. Biochar addition to waste is presumed to enhance the efficiency of methane production, yet individual reports disclose contradictory results. Therefore, we performed a meta-analysis of 27 selected publications containing 156 paired measurements of control and biochar-amended treatments to assess the impact of biochar on the methanogenic performance. Results show that biochar promotes biomethane production substantially with a high Hedge's d value of 5.7 +/- 1.04, yet sporadic publications report a methane decline. Methanogenic performance is statistically controlled by feedstock type, pyrolysis temperature and biochar concentration, but not controlled by pH, size, surface area and methanogen species. These findings should help to tune the parameters of anaerobic digestion with biochar to optimize biomethane productions. Moreover, our results cast some doubt on the efficiency of adding biochar to soil to sequester carbon in soils because biochar promotes methane generation and, in turn, emissions of methane, a greenhouse gas, to the atmosphere.

International audience; Background: Wastewater surveillance was proposed as an epidemiological tool to define the prevalence and evolution of the SARS-CoV-2 epidemics. However, most implemented SARS-CoV-2 wastewater surveillance projects were based on qPCR measurement of virus titers and did not address the mutational spectrum of SARS-CoV-2 circulating in the population.Methods: We have implemented a nanopore RNA sequencing monitoring system in the city of Nice (France, 550,000 inhabitants). Between October 2020 and March 2021, we monthly analyzed the SARS-CoV-2 variants in 113 wastewater samples collected in the main wastewater treatment plant and 20 neighborhoods.Findings: We initially detected the lineages predominant in Europe at the end of 2020 (B.1.160, B.1.177, B.1.367, B.1.474, and B.1.221). In January, a localized emergence of a variant (Spike:A522S) of the B.1.1.7 lineage occurred in one neighborhood. It rapidly spread and became dominant all over the city. Other variants of concern (B.1.351, P.1) were also detected in some neighborhoods, but at low frequency. Comparison with individual clinical samples collected during the same week showed that wastewater sequencing correctly identified the same lineages as those found in COVID-19 patients.Interpretation: Wastewater sequencing allowed to document the diversity of SARS-CoV-2 sequences within the different neighborhoods of the city of Nice. Our results illustrate how sequencing of sewage samples can be used to track pathogen sequence diversity in the current pandemics and in future infectious disease outbreaks.; Contexte : La surveillance des eaux usées apparait comme un outil épidémiologique prometteur pour définir la prévalence et suivre l'évolution des épidémies de SRAS-CoV-2 sur un territoire. Jusqu’à présent, la plupart des projets de surveillance du SRAS-CoV-2 dans les eaux usées reposaient sur une mesure des titres de virus par PCR quantitative et ne fournissaient pas une vision exhaustive de toutes les mutations du SRAS-CoV-2 pouvant circuler dans la population. Méthodes : Nous avons mis en place un système de surveillance par séquençage de l’ARN viral sur séquenceur Oxford Nanopore à l’échelle de toute la ville de Nice (France, 550 000 habitants). Entre octobre 2020 et mars 2021, nous avons analysé chaque mois les différents variants du SRAS-CoV-2 à partir de 113 échantillonnages d'eaux usées collectés au niveau de la station centrale d'épuration et de 20 quartiers différents de la ville.Résultats : Nous avons initialement détecté les lignées prédominant en Europe à la fin de 2020 (B.1.160, B.1.177, B.1.367, B.1.474 et B.1.221). En janvier, l’émergence d'un variant de la lignée B.1.1.7 s'est produite dans un des quartiers de la ville, caractérisé par une mutation A522S sur la spicule. Ce variant s'est rapidement répandu dans toute la ville où il est devenu dominant à partir du mois de février. D'autres variants préoccupants (B.1.351, P.1) ont également été détectés dans certains quartiers, mais toujours à une faible fréquence. La comparaison avec les échantillons cliniques individuels collectés au cours de la même semaine a montré que le séquençage des eaux usées identifiait correctement les mêmes lignées que celles trouvées chez des patients COVID-19.Interprétation : Le séquençage des eaux usées a permis de documenter de façon très précise la diversité des séquences du SRAS-CoV-2 présentes dans les différents quartiers de la ville de Nice. Nos résultats illustrent comment le séquençage des échantillons d'eaux usées peut être utilisé comme outil « micro-épidémiologique » pour suivre la dissémination d’agents pathogènes lors d’épisodes pandémiques et/ou épidémiques, présents ou futurs.

International audience; Ten-Eleven Translocation (TET) proteins convert 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC) leading to a dynamic epigenetic state of DNA that can influence transcription and chromatin organization. While TET proteins interact with complexes involved in transcriptional repression and activation, the overall understanding of the molecular mechanisms involved in TET-mediated regulation of gene expression still remains limited. Here, we show that TET proteins interact with the chromatin remodelling protein lymphoid-specific helicase (LSH/HELLS) in vivo and in vitro. In mouse embryonic fibroblasts (MEFs) and embryonic stem cells (ESCs) knock out of Lsh leads to a significant reduction of 5-hydroxymethylation amount in the DNA. Whole genome sequencing of 5hmC in wild-type versus Lsh knock-out MEFs and ESCs showed that in absence of Lsh, some regions of the genome gain 5hmC while others lose it, with mild correlation with gene expression changes. We further show that differentially hydroxymethylated regions did not completely overlap with differentially methylated regions indicating that changes in 5hmC distribution upon Lsh knock-out are not a direct consequence of 5mC decrease. Altogether, our results suggest that LSH, which interacts with TET proteins, contributes to the regulation of 5hmC levels and distribution in MEFs and ESCs.

International audience; We developed a chemical method to covalently functionalize cellulose nanofibers and cellulose paper with mannoside ligands displaying a strong affinity for the FimH adhesin from pathogenic E. coli strains. Mannose-grafted cellulose proved efficient to selectively bind FimH lectin and discriminate pathogenic E. coli strains from non-pathogenic ones. These modified papers are valuable tools for diagnosing infections promoted by E.coli, such as cystitis or inflammatory bowel diseases, and the concept may be applicable to other life-threatening pathogens. The worldwide spread of antibiotic resistances raises serious health problems, and has driven the identification of new virulence factors and development of alternative antibacterial therapeutics. Mannose-binding FimH adhesin, expressed by Escherichia coli strains has been extensively studied as a target for disrupting bacterial attachment to the host cells. 1 Impressive results were obtained in the context of urinary tract infections (UTI), a prevalent infection type generally mediated by the attachment of uropathogenic E. coli strains (UPEC) to the highly mannosylated uroplakin transmembrane protein of urothelial lining cells. FimH antagonists orally administered to in vivo UTI mouse models, were shown to decrease the E.coli load in the bladder by several orders of magnitude, 2-4 making them competitive with conventional antibiotic treatment. 5,6 This concept was more recently extended to Crohn's disease (CD), an inflammatory disorder of the intestine where an altered gut microbiota, particularly the presence of adherent-invasive E. coli strains (AIEC), is suspected to play a key role in the pathogenesis. 7 Synthetic derivatives of heptylmannoside (HMan), a nanomolar FimH antagonist, 8 were shown to lower the AIEC bacterial level, signs of colitis and gut inflammation when administered per os (10 mg/kg) in CD mouse models. 9,10 Sensitive and rapid diagnostic systems are essential to evaluate the presence of E. coli expressing FimH adhesin in gut microenvironments in order to properly stratify patients before treatment. While the high prevalence of UPEC in the normally sterile urinary tract environment facilitates diagnosis, the AIEC niche is more complex, located at the ileal mucosa in 21 to 63% of CD patients, 11,12 within an ecological community of hundreds of symbiotic microorganisms. Furthermore, no specific biomarkers are currently effective at distinguishing AIEC from other commensal E. coli of the gut microbiota. Previously, it has been shown that AIEC pathobiont possesses specific allelic variants in the fimH gene, conferring them a high ability to adhere in vitro and to colonize the gut of CEABAC10 mice. 13 Establishing an approach to discriminate the strong mannose-binders from other bacteria would therefore be of tremendous importance for efficient diagnosis. In this work, we developed a heterogeneous support to specifically trap and accumulate pathogenic E.coli from biological fluids. Heptyl-α-D-mannoside (HMan) was grafted by click chemistry techniques onto cellulose nanofibers (CN) and cellulose paper (CP). HMan was previously identified as a potent binder of the isolated FimH lectin domain. 8 It should be noted that the lectin domain represents the high-affinity state of FimH under mechanical force and that full-length FimH display a lower affinity for mannosides. 14,15 Covalently functionalized CN or CP were characterized by Fourier transform spectroscopy (FTIR), elemental analysis, X-ray photo-electron spectrometry (XPS), and scanning electron microscopy (SEM). HMan-CN was first compared in vitro against CN grafted with lower FimH affinity ligands i.e. Man-CN lacking the hydrophobic heptyl chain, and HGlc-CN an analog with a glucose sugar that is not recognized by FimH lectin (Scheme 1). In addition, we switched the anomeric oxygen atom to a sulfur and synthesized HSMan-CN to prevent potential sugar hydrolysis from the surface by mannosidases. The modified CN were first assessed for their faculty to bind FimH and to prevent AIEC adhesion to intestinal cells. HMan-CN was then orally administered to the CEABAC10 mouse model of CD to assess its capacity to decrease AIEC in the feces of AIEC-infected mice and to act as a potential CD therapeutic. Endocasts (i.e., replicas of the inner surface of the bony braincase) constitute a critical proxy for qualifying and quantifying variations in brain shape and organization in extinct taxa. In the absence of brain tissues preserved in the fossil record, endocasts provide the only direct evidence of brain evolution. However, debates on whether or not information inferred from the study of endocasts reflects brain shape and organization have polarized discussions in paleoneurology since the earliest descriptions of cerebral imprints in fossil hominin crania. By means of imaging techniques (i.e., MRIs and CT scans) and 3D modelling methods (i.e., surface‐based comparisons), we collected consistent morphological (i.e., shape) and structural (i.e., sulci) information on the variation patterns between the brain and the endocast based on a sample of extant human individuals (N = 5) from the 3D clinical image database of the Steve Biko Academic Hospital in Pretoria (South Africa) and the Hôpitaux Universitaires Pitié Salpêtrière in Paris (France). Surfaces of the brain and endocast of the same individual were segmented from the 3D MRIs and CT images, respectively. Sulcal imprints were automatically detected. We performed a deformation‐based shape analysis to compare both the shape and the sulcal pattern of the brain and the endocast. We demonstrated that there is close correspondence in terms of morphology and organization between the brain and the corresponding endocast with the exception of the superior region. By comparatively quantifying the shape and organization of the brain and endocast, this work represents an important reference for paleoneurological studies., Is the endocast a reliable proxy for reconstructing the brain of fossil specimens? By quantitatively comparing extant human brains and endocasts using MRIs and CT‐scans, here we address this fundamental question and support the significance of fossil endocasts in paleoneurological studies.

Aroma compounds are known to be efficient active agents for a broad range of applications (antimicrobial, anti-oxidant, insect repellent…) that are highly sought when aiming at extending shelf life of food or biological products. However, they are intrinsically odorant and volatile at ambient temperature, which restricts the processing routes used to introduce them in a polymeric matrix and can affect their mode of action and limit efficiency. Indeed, due to their high sensitivity toward temperature they can be lost or transformed during processing. Acting after being released in the headspace, their concentration has to be controlled to avoid any odorant contamination of the targeted products. Hence, the ability for an aroma compound to be retained in a polymeric matrix, and then released when submitted to a triggering effect, are the two main requirements that should be satisfied. The volatile nature of the aroma compound offer the possibility when introduce in the packaging to act by direct or indirect contact with the product and thus to be used in different ways; as a coating layer directly applied on the product surface, as a self-supported film or as coated paper when associated with a paper sheet, as well as an object that could be inserted in the package. As biopolymers such as proteins and polysaccharides are able to retain aroma compounds but also to favor their release by modification of their structure when the relative humidity (RH) and temperature change, they are relevant carriers of these specific aroma compounds. Examples of how active packaging systems with limonene, eugenol and carvacrol as active agents were designed and elaborated. These examples will be presented with a special focus on the processing conditions and the way to improve their aroma compound retention and the release control (biopolymer nature, cyclodextrin clay addition…). Avrami's equation has been used to model the transfer of aroma compound and to advantageously compare it taking into account the mechanism in relation to the biopolymer structural changes.

International audience; Hydrogen (H2) production by dark fermentation can be performed from a wide variety of microbial inoculum sources, which are generally pre-treated to eliminate the activity of H2-consuming species and/or enrich the microbial community with H2-producing bacteria. This paper aims to study the impact of the microbial inoculum source on pre-treatment behavior, with a special focus on microbial community changes. Two inocula (aerobic and anaerobic sludge) and two pre-treatments (aeration and heat shock) were investigated using glycerol as substrate during a continuous operation. Our results show that the inoculum source significantly affected the pre-treatment efficiency. In aerobic sludge no pre-treatment is necessary, while in anaerobic sludge the heat pre-treatment increased H2 production but aeration caused unstable H2 production. In addition, biokinetic control was key in Clostridium selection as dominant species in all microbial communities. Lower and unstable H2 production were associated with a higher relative abundance of Enterobacteriaceae family members. Our results allow a better understanding of H2 production in continuous systems and how the microbial community is affected. This provides key information for efficient selection of operating conditions for future applications.
