Plant root system architecture (RSA) has an important role in crop production, particularly for water and nutrient uptake under limiting conditions. In the last few years, several root phenotyping methods have been developed. Here we present a new technique which has been developed for non-destructive, inexpensive and high-throughput root growth studies and RSA analyses. To illustrate the potential applications, this method was tested in an experiment with nitrogen and phosphorous deficiencies in a nutrient solution, affecting RSA parameters of two spring barley varieties (Bojos and Barke). This technique is based on root growth on vertically positioned black filter paper (30 × 60 cm) placed between two black plastic (PVC-P) foils and micro-irrigation systems providing the recirculation of nutrient solution. The pre-germinated seeds were placed in the slit between two plastic bars which carry the filter paper and plastic sheets and fix the plant in the vertical position. This system allows easy repeated non-invasive access to roots for their measuring and sampling. Eighteen days after transplanting the root imaging was done using an RGB digital camera. To evaluate the root architecture parameters the "SmartRoot" software was used. The results revealed that the system is able to detect changes in RSA which are caused mainly by P deficiency (particularly changes in lateral root length and total root area). So far, a large number of transformation systems have been established for japonica rice, but only a few have been reported for cold-region varieties. In our study, we established highly efficient tissue culture systems for two cold-region rice cultivars, Dongnong 427 and Longdao 14. Plant growth regulator (PGR) levels were optimized by an orthogonal experimental design. The culture ability, constituted by induction and differentiation rate, served as the detection index of orthogonal experiments. The optimal combinations of PGRs for callus induction and regeneration of Dongnong 427 and Longdao 14 were 1 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) + 2 mg/l 6-benzyladenine (BA) + 4 mg/l kinetin (KIN) + 0.2 mg/l α-naphthaleneacetic acid (NAA) and 1 mg/l 2,4-D + 4 mg/l 6-BA + 4 mg/l KIN + 0.5 mg/l NAA, respectively. Agrobacterium strain EHA 105 containing the plasmid pCAMBIA1301 was used for transformation. The frequency of transient transformation was expressed as the ratio between the number of calli showing GUS expression and the total number of calli kept for staining. The highest transformation efficiency in Dongnong 427 was obtained when calli were immersed in 0.272 OD600 (optical density determined at 600 nm) for 10 min. While it was best for Longdao 14 calli to be infected with 0.592 OD600 for 20 min. Infected calli of the two varieties were co-cultivated on two pieces of sterile filter paper moistened with 1 ml liquid co-cultivation medium for three days. Sweet sorghum is a cereal crop that can potentially serve as a source for bioethanol production. This study aims to analyse the genetic diversity of promising genotypes of sweet sorghum at the molecular level. The genetic material consisted of 12 sweet sorghum genotypes. The genetic diversity estimated using 59 SSRs markers showed a polymorphism value of 0.48 and the coefficient of genetic diversity was classified as moderate. The unweighted pair group method arithmetic average (UPGMA) analysis assigned the tested genotypes into three major clusters with a similarity coefficient level of 0.596. This indicates that the genetic similarity of the tested genotypes is moderate to high. Low temperatures are one of the major abiotic stresses that affect alfalfa’s development and yield. Enhancing frost resistance through resistance-related genes is one of the most effective ways to address this issue in alfalfa. Therefore, exploring cold-resistant gene resources and the cultivation of cold-resistant alfalfa cultivars is inevitable in order to achieve high yield and quality. In this study, we conducted transcriptome profiling of roots obtained from two alfalfa genotypes, i.e., Qingda No.1 for freeze tolerance and Gannong No.9 for freeze sensitivity. We observed that Qingda No.1 had more lateral roots and a more developed root system after overwintering, while Gannong No.9 had fewer lateral roots and an underdeveloped root system. After overwintering, Qingda No.1 exhibited higher superoxide dismutase (SOD) activity compared to Gannong No.9, while Gannong No.9 showed higher perosuperoxide dismutasexidase (POD) activity than Qingda No.1. We identified 25,935 differentially expressed genes, with 12 979 and 12 956 differential genes found in the freeze-tolerant variety Qingda No.1 group and the freeze-sensitive Gannong No.9 group, respectively. The enrichment of the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways also differed between the two groups. We also discovered several gene family members, and the most frequent transcription factors were bHLH, B3, NAC, WRKY, and MYB_related. These findings provide comprehensive information to further understand the molecular mechanisms of adaptation to freezing stress in alfalfa and offer potential functional candidate genes for adaptation to abiotic stress. Several antimitotic agents were tested in three embryogenic doubled haploid (DH) lines of swede (Brassica napus ssp. napobrassica). No effect on embryogenesis was observed at the given concentrations of colchicine (5 μmol/L or 50 mg/L) and treatment time of 24 hours. Flow cytometric analysis of microspore embryos revealed a significant increase in the percentage of DH compared to the control for all substances: ethalfluralin, 62.2%; trifluralin, 58.3%; colchicine, 56.1%; amiprophos-methyl, 54.4%; pronamide, 35.0%; control, 13.3%. The occurrence of pure tetraploids was low (0–5%), mixoploids – especially n+2n – were high in all treatments, including the control (29.4–71.1%). Barley is one of the world’s most important crops. Barley is used for both food and feed and is important for the production of malt. Malt quality is a complex function of barley genetics, environmental conditions during barley growth, and the technological aspects of the malting process. Due to the high heritability of more than half of the malting parameters, barley can be bred for malting quality. Marker-assisted selection (MAS) is a good way to speed up the breeding process. In this study, nine molecular markers were used to screen 115 barley varieties and breeding lines over a four-year period. The results were compared with malting quality parameters. Multicomponent correlation analysis showed a good correlation (R = 0.63; P ≤ 0.01) between marker screening results and malting quality parameters. In 93 genotypes (80.9%), agreement was found between molecular marker prediction and malting quality determination. Differences between molecular marker screening and malt quality parameters and possible improvements are discussed. The use of molecular markers in MAS is highly appreciated by barley breeders. Powdery mildew (PM) is a major fungal disease in the Cucurbita species in the world, which can cause significant yield loss. The Mildew Locus O (MLO) family genes play important roles in the PM stress response. In this paper, twenty, twenty-one, and eighteen candidate MLO genes in Cucurbita moschata, Cucurbita maxima and Cucurbita pepo, respectively, were identified and designated as CmoMLO, CmaMLO and CpeMLO, respectively. The phylogenetic analysis indicated that these MLOs were divided into five clades and the number of MLOs belonging to clade V in the Cucurbita species was more than that in other crops. Furthermore, the expression analysis in the susceptibility (S) and resistance (R) lines showed that CpeMLO1, CpeMLO2 and CpeMLO5 might be involved in the susceptibility response. CpeMLO4 and CpeMLO6 showing opposite expression patterns in the R/S lines might be involved in the resistance response. Rf(fa), a new restoring fertility gene in rice, was previously located to a large region on Chromosome 10. The large number of genes within the region made cloning of Rf(fa) difficult. To perform the cloning and further elucidate the molecular mechanism, we reconstructed a mapping segregation population (BC1F1) of 12 000 plants. Using the population and polymorphism of simple sequence repeat (SSR) molecular markers, we finally mapped Rf(fa) between the two SSR molecular markers MM2000 and RM25658, within a 78.87 kb region. By de novo sequencing of a restoring line of CMS-FA hybrid rice, we obtained the genomic sequence of the mapping region, which provided the basis for the prediction of the candidate gene(s) of the target gene and for the comparison of genomic sequence differences between wild and cultivated rice. Within the mapping region, the genomic sequence of the wild rice was significantly different from that of cultivated rice. There were ten genes in the final mapping region. A pentatricopeptide repeat (PPR) protein gene was predicted as the candidate gene of Rf(fa). Our results laid a solid foundation for the final cloning and molecular mechanism analysis of the gene. Coloured rice has pigments deposited in the grain pericarp; red rice is the most common type of coloured rice. Red rice is rich in essential nutrients and has been grown and consumed in China for a long time. In this study, we report the genetic characterisation and preliminary molecular mapping of a mutant gene encoding red pericarp in rice (Oryza sativa L.). To analyse the genetic basis of the red pericarp mutant, a reciprocal cross between GER-3 (red pericarp, indica cv.) and 898 (white pericarp, indica cv.) was made. The genetic analysis results confirmed that there was only one dominant gene, temporarily designated Rp (Red pericarp) controlling the segregation of the red pericarp in the F2 population. For the molecular mapping of Rp, an F2 population derived from an inter-subspecific cross between Gene Engineering Rice-3 (GER-3) and C418 (japonica cv., white pericarp) was constructed. The genotype of the pericarp colour of the F2 individuals in the mapping population was validated by progeny testing of the F2:3 families. Simple sequence repeat (SSR) markers and the bulked segregation analysis (BSA) method were used; Rp was mapped to the short arm of chromosome 7 between the SSR markers RM21182 and RM21268, with a genetic distance of 3.5 and 12.0 cM, respectively. Fungal pathogens Plasmopara viticola (Berk. & M.A. Curtis) Berl. & De Toni (downy mildew) and Erysiphe necator Schwein. (powdery mildew) represent the biggest threats for grape growers worldwide. Under suitable conditions, these pathogens can spread very quickly through vineyards and cause significant damage. The most ecological way to reduce the possibility of infection in vineyards is growing interspecific genotypes which are able to suppress these pathogens and stop the spread of infection. With this in mind, 9 international and 11 Czech grapevine genotypes were analysed in order to genotype the resistance loci present in them. As a tool a set of SSR markers linked to known loci of resistance to downy and powdery mildew was used. Namely, presence of four loci responsible for resistance to Plasmopara viticola (Rpv3, Rpv4, Rpv7, and Rpv10) and two for Erysiphe necator (Ren3 and Ren9) were analysed with respective SSR markers. Simple sequence repeat (SSR) markers are valuable genetic and genomic research tools and are extensively used in major crops. However, a paucity of available molecular markers in lentils (Lens culinaris Medik.) has significantly hindered genomic studies in this vital legume crop. In this study, we developed 33 new SSR markers for lentils using an enriched genomic library and tested their polymorphism in 10 lentil cultivars. We found that 16 (48.5%) SSR markers were polymorphic. The expected heterozygosity values of the polymorphic SSR markers ranged from 0.095 to 0.820, while observed heterozygosity values varied between 0.100 and 1.00. In addition, we tested the transferability of 86 SSR markers, including the 33 newly developed ones, to other legume species. Transferability rates of lentil SSR markers to other species varied between 13.76% (Vicia sativa L.) and 2.58% (Phaseolus vulgaris L. and Trifolium pratense L.). Powdery mildew (PM) is a fungus that causes disease in both the field and the greenhouse. Utilizing resistant cultivars is the most effective approach of disease management. To develop insertion-deletion (InDel) markers associated to this trait, the whole genomes of the PM resistant line M17050 (P1) and the PM-susceptible line 28-1-1 (P2) were sequenced. A total of 1 200 InDels, with an average of 100 markers per chromosome, were arbitrarily chosen from the sequencing data for experimental validation. One hundred InDel markers were ultimately selected due to their informative genetic bands. Further, an F2 segregating population of melons generated from these two parents was inoculated by the PM pathogen. Based on bulk segregant analysis (BSA) using these 100 InDel markers, the powdery mildew resistance was associated with the genomic region LVpm12.1 on the melon chromosome 12. This region overlapped the previously described quantitative trait locus (QTL)-hotspot area carrying multiple PM-resistance QTLs. Moreover, conventional QTL mapping analysis was done, which located LVpm12.1 in the region between 22.72 and 23.34 Mb, where three highly polymorphic InDel markers MInDel89, MInDel92, and MInDel93 were detected. Genetic diversity is referred to as any variation at the phenotypic, DNA or genomic level of an individual, population or species. The appraisal of diversity is important to understand its pattern and evolutionary relationships between germplasms or genotypes, which will aid in sampling the genetic resources in a more systematic manner for conservation and crop improvement. The present study employed 50 simple sequence repeat (SSR) markers linked to the yield and fibre quality/colour traits for estimating the genetic diversity in 33 cotton genotypes of diploid and tetraploid species differing in fibre colour. The diversity analysis was performed in GenAlEx (Ver. 6.41) and Powermarker (Ver. 3.25) while DARwin (Ver. 6.0.21) software was used to establish the phylogenetic relationships following neighbour- -joining (NJ) and unweighted pair group method with arithmetic (UPGMA) mean method. Markers generated 186 polymorphic loci as genotypic data with an average of 3.72 alleles and an average polymorphic information content (PIC) value of 0.59 per SSR locus. The NJ and UPGMA grouped 33 genotypes into three major clusters I, II and III consisting of 21 tetraploid Gossypium hirsutum, 10 G. arboreum coloured and 2 white cotton genotypes, respectively. In the PCA, the first two components (PC1 and PC2) explained 74.69% of the variation and the biplot plotted the 33 genotypes in three groups. The study established the diverse nature of 33 cotton genotypes based on their fibre colour and ploidy level. With confirmation of the prevalent genetic diversity, we suggest that hybridisation can be planned among diverse genotypes to unleash greater variation in the fibre colour or to derive superior cross combinations. The present study was to determine the genotypic and environmental variability and stability in seed yield, oil content, oil yield, oleic and linoleic acid of 10 safflower lines derived from a cross of Dinçer 5-18-1 × Montola 2000 together with six cultivars under six environments at five locations. The effects of genotypes, environments and genotype × environment interactions were highly significant (P < 0.01) for seed yield and oil content. Averaged across all environments, the seed yield was lowest in the cultivar Olas (2 352 kg/ha), and highest in the line Bay-Er 5 (2 869 kg/ha). According to mean (xi) and regression coefficient (bi) values, the Bay-Er 16 was better adapted to unfavourable environmental conditions, whereas the Bay-Er 1, Bay-Er 5 and Bay-Er 14 were better adapted to favourable environmental conditions. The highest oil content across environments, over 35%, was recorded in the line Bay-Er 15 and the cultivars Olas and Linas. The best adaptability to the environments was observed in the cultivar Olas. The oleic acid content of genotypes increased and the linoleic acid contents decreased from the north to the south latitudes. The oil content of genotypes grown in Southeastern Anatolia was higher than in the other regions. Cytochrome P450 proteins belong to one of the largest families of enzyme proteins in plants and play important roles in plant growth and development and the stress response. In our previous studies, a cytochrome P450 gene, GhCYP94C1 (cytochrome P450 94C1), was functionally characterized as a positive regulator of seed germination, main root elongation and early flowering. However, whether the gene has other potential functions remains to be further explored. In our study, expression analysis showed that GhCYP94C1 was highly expressed in roots and was suppressed by drought treatment. Endogenous silencing of GhCYP94C1 via virus-induced gene silencing (VIGS) increased drought resistance in cotton plants, which was accompanied by the upregulated expression of the abscisic acid (ABA) biosynthesis gene nine-cis-epoxycarotenoid dioxygenase 9 (GhNCED9) during drought stress. Our findings suggested that GhCYP94C1 may play an important role in drought resistance. Combined with previous research results, the present results provide a theoretical basis for future breeding of new cotton varieties with early maturation and drought resistance. Horsegram (Macrotyloma uniflorum) - an important, self-pollinated food legume, however due to limited genomic and genetic resources the genetic improvement could not be achieved as compare to other major legumes. Our work aims at finding novel microsatellite markers and their use for the construction of a linkage map from 157 individuals of F9 recombinant inbred lines (RILs) of horsegram. The determinate growth habit and plant height are important traits for its suitability for different cropping systems. The genotypic data were generated by screening 2 395 molecular markers, of which 600 (25.05 %) polymorphic markers were selected. Two-hundred eighty-seven markers were mapped on ten linkage groups (LGs) at a log of odds (LOD) of 3.5 straddling 796.76 cM with 2.78 cM of marker density. For the identification of the quantitative trait loci (QTLs), the phenotypic data recorded on the RILs for the plant height and growth habit were analysed using the statistical tools JoinMap® and Windows QTL cartographer, based on the composite interval mapping (CIM) technique. Across the ten linkage groups, we detected four QTLs (LOD ≥ 2.5) for four traits. All the traits were major QTLs as indicated by the percentage of phenotypic variance (PVE) (≥ 10%) that ranged from 13.5% to 40.3%, therefore, this is very important information which can be used in marker-assisted selection (MAS). To explore the role of WRKY transcription factors (TFs) in the resistance process of Qingke (Hordeum vulgare L. var. nudum Hook. f.), leaves of the leaf stripe disease-resistant variety Kunlun 14 and the susceptible variety Z1141 were sequenced by transcriptome sequencing (RNA-seq). A differentially expressed gene HvnWKRY1 was identified, and its disease-resistance function was preliminarily analysed. The result showed that the open reading frame (ORF) of the gene was 1 062 bp and encoded 354 amino acids. It contained the conserved WRKY domain (273-351) and belonged to the WRKY protein family. The phylogenetic tree results showed that HvWRKY1 was most closely related to Hordeum vulgare L. The WRKY family of Qingke, barley, maize and rice were divided into categories I, II, and III, among which HvWRKY1 was located in group III. Results of the quantitative real-time fluorescence PCR (qRT-PCR) showed that the expression of HvWRKY1 was significantly (P < 0.01) higher in leaf stripe infected leaves of Kunlun 14 than that of Z1141. In Arabidopsis thaliana transformed with HvWRKY1, resistance to Botrytis cinerea was enhanced. The RNA-seq analysis showed there were 824 differentially expressed genes (DEGs). Data of the Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment indicated, that a plant-pathogen interaction pathway was enriched. Orange-fleshed sweetpotato (OFSP) is a sustainable and inexpensive source of vitamin A that contributes to food and nutritional security in developing countries. Its adoption is low due to unconformity of the variety with community food culture, limited cultivar selection and breeding studies. The current study aimed at breeding of promising OFSP genotypes in Benin using three parental lines. Two hundred and seventy-four seeds and one hundred and six genotypes were obtained through biparental crosses of Tio-joe (imported orange-fleshed variety), Vobodouaho (local white-fleshed variety) and ACAB220 (local orange-fleshed variety). The phenotypic diversity of genotypes was assessed using 10 leaf characters and storage root flesh colour through multiple correspondence analysis and hierarchical cluster analysis. The results indicated cross-compatibility between Tio-joe and ACAB220 when ACAB220 was used as female. Reciprocal cross-compatibility was found between ACAB220 and Vobodouaho. The genotypes could be separated into two main populations and classified into five cluster groups. Promising intermediate and pale orange genotypes were obtained indicating possibilities to incorporate beta-carotene into the white background of the Vobodouaho variety through further backcrossing. Sorghum is an important crop, and starch and phenolic compounds are major and important components in the sorghum grain. However, the underlying critical genetic elements contributing to the rich portfolio of nutrients in sorghum grains are largely unknown. Transcriptomic methods were employed to characterize the expression patterns at five different grain developmental stages of Hongyingzi (an important brewing sorghum), and another two grain sorghums, Jinuoliang 1 and Hongliangfeng 1, for comparison. The uniquely expressed genes were identified at each developmental stage of Hongyingzi when compared with the other two sorghum cultivars. The co-regulated genes at different developmental stages and the regulatory network were determined; the determinant genes and single-nucleotide polymorphisms located at the promoters of these genes involved in starch and phenolic compounds biosynthetic pathways were also identified. These results will provide insights into the potential regulatory network and further contribute to the clarification of the key determinant genes involved in the biosyntheses of starch and phenolic compounds. Meanwhile, some new transcripts and genes were identified at five different developmental stages of grains of the three sorghum cultivars. Our work can provide impetus for further study of the genes responsible for the biosynthesis of starch and phenolic compounds in the sorghum grain, and pave a way for functional validation of a batch of potential genes and single-nucleotide polymorphisms proposed in current work. Wheat leaf rust is a devastating disease worldwide. Identification of leaf rust resistance genes in seedlings and of genes for slow rusting are important in resistance breeding and for gene deployment to control the disease. A total of 108 wheat cultivars from the Beijing and Shandong province and a set of 36 differentials, mostly near-isogenic lines in the background of Thatcher with known leaf rust resistance genes, were tested with 20 Puccinia triticina pathotypes (FHJS1, FGBQ, PGJQ, SHJT, FHGQ, PHTT1, FHGQ, FHGQ, PHJS, THSM, FHSQ, PHST, PRSQ, FNTQ, PHGM, KHGQ, PHTT2, TGTT, FHJS2, NHHT) at the seedling stage in the greenhouse. The cultivars and differentials were also planted in the field to test their slow rusting resistance using a mixture of races at Baoding, Hebei province and Zhoukou, Henan province, for two consecutive years. Ten leaf rust resistance genes, Lr1, 9, 10, 19, 20, 24, 26, 34, 37 and 46 were identified in the 57 commercial wheat cultivars, either singly or in combination, using molecular markers. Combined, the results from gene postulation and marker detection showed that one or more of the genes Lr1, 3, 10, 14a, 14b, 26, 36, 39, 34 and 46, were present in 57 cultivars, and that no known resistance gene was present in the remaining 51 cultivars. The resistance gene Lr26 was present in 42 cultivars, and nine cultivars contained Lr1. Lr46 was present in 10 cultivars, as indicated by the presence of the closely linked marker csLV46G22. Seven genotypes were identified as possibly carrying the gene Lr39. Lr3 and 10 were found in six and four genotypes, respectively. The genes Lr14b and 34 were each present in three cultivars, while evidence for the presence of Lr14a and 36 was obtained in single genotypes. Finally, 12 cultivars showed slow rusting resistance at two locations in two crop seasons. By choosing appropriate candidate reference genes (CRGs) and standardizing qPCR data, more accurate experimental data can be obtained. Herein, the expression stability of alpha-tubulin1 (TUA1), beta-tubulin (TUB), beta-tubulin 1 (TUB1), beta-tubulin 5 (TUB5), actin 1 (ACT1), actin 97 (ACT97), molecular chaperone dnaj (DNAJ), adenine phosphoribosyl transferase (APT), and histone H4 (HIS4) genes from Paulownia fortunei (Seem.) Hemsl. under different experimental conditions (different tissues, drought, salinity, Cd, and Cr treatments) was assessed with four statistical tools: RefFinder, BestKeeper, NormFinder, and geNorm. Notably, TUA1 and TUB5 were identified as CRGs for different tissues, ACT97 and TUB1 for drought treatment, ACT97 and APT for salinity treatment, TUB1 and ACT97 for Cd treatment, and DNAJ, TUB1 and TUB5 for Cr treatment. Furthermore, the results of "total" group, V4/V5 > 0.15 and V5/V6 < 0.15 revealed that the CRGs or gene combinations, which could meet all the test conditions, were not easy to identify. To further verify the reliability of CRGs, the expression levels of paulownia fortunei cellulose synthase A catalytic subunit2 (PfCesA2) and paulownia fortunei glutathione reductase (GR) genes were analysed. Medicago sativa is the most widely cultivated forage legume and one of the most economically valuable crops throughout the world. Qingda no.1 (Medicago sativa L. cv. Qingda no.1) is an excellent alfalfa local variety with strong cold, drought and salt resistance in the three rivers source area of Qinghai. In this study, the whole chloroplast (cp) genome of Qingda no.1 was sequenced, assembled and its structure was analysed by the Illumina high-throughput sequencing technology. The results showed that the chloroplast genome of Qingda no.1 exhibits no obvious typical quadripartite structure; the total length of the chloroplast genome is 125 637 bp; the chloroplast genome contained 111 genes, including 77 protein-coding genes, 30 tRNA genes, and 4 rRNA genes, with an overall GC content of 38.33%. The relative synonymous codon usage showed that 68.67% of the codons RSCU > 1 in Qingda no.1, with the preference ending with A and T. The simple sequence repeat (SSR) analysis identified 62 SSR loci. The phylogenetic analysis of the cp genome, Qingda no.1 clustered closely with Medicago sativa KU321683 (Medicago sativa L. cv. The objective of this study was to investigate the diversity of 59 accessions of barley using inter simple sequence repeat (ISSR), conserved DNA-derived polymorphism (CDDP), and start codon targeted (SCoT) markers. A total of 391 amplified polymorphic bands were generated using 44 ISSR, 9 CDDP, and 12 SCoT primers that produced 255, 35, and 101 polymorphic bands, respectively. The average values of gene diversity were 0.77, 0.67, and 0.81 for ISSR, CDDP, and SCoT markers, respectively. The mean values of polymorphism information content for ISSR, CDDP and SCoT markers were 0.74, 0.63, and 0.80 respectively. The discrimination power of the three approaches for assessing allelic diversity in barley accessions ranked as follows: SCoT > ISSR > CDDP. The barley accessions were classified and clustered into two main groups. Molecular variance analysis revealed 15, 9, and 14% variability among populations with ISSR, CDDP, and SCoT markers, respectively. The Mantel test results revealed that the three molecular marker matrices had significant positive relationships. Kaliurang is one of the local taro cultivars in Indonesia, potentially due to its high productivity and delicacy. This study analysed morpho-genetic variation between one diploid and three tetraploid Kaliurang taro clones from in vitro polyploid induction after plantlet acclimatization in the greenhouse for 6 weeks. Plants’ morphological characters were analysed using the unweighted pair group method with arithmetic mean (UPGMA) method based on 50 characters according to the taro descriptor from the International Plant Genetic Resources Institute (IPGRI). Furthermore, a morphometric characterization based on the Euclidean distance of 17 digitations from mature leaves, was analysed with the Analysis Phylogenic and Evolution package in R software. DNA band pattern was also performed with ISSR to analyse the clone’s genetic variance. The results showed morpho-genetic character diversity in diploid and tetraploid plants. A plant morphological study revealed that 22 out of 50 characters were distinct, with similarity coefficients ranging from 0.35 to 0.78. Leaf geometric morphometric analysis showed changes in five digitations and formed three clusters. Twelve ISSR primers out of twenty amplified the sequence of the Kaliurang taro genome to yield two clusters with a similarity coefficient of 0.71–0.83. The lignin content of forage crops significantly affects the livestock’s forage intake and its dry matter digestibility. The brown midrib (bmr) trait is known for its association with a reduced lignin content and increased digestibility. Pearl millet inbreds having the bmr trait were derived from two populations ICMV bmr and WRajPop bmr via continued selfing up to 6–8 generations. The results from the crosses revealed the presence of the same single recessive gene that controlled the bmr in both the populations. The stable expression of the bmr gene in the new genetic background indicated that there was no effect of modifier genes. Four inbreds were identified with the stable expression of bmr (three in ICMV 155 and one in the WRajPop background) with a high biomass potential, which can serve as a bmr source in future pearl millet forage breeding programmes. Grafting is the most important means of breeding peach, plum, apricot, and other fruit trees, and the selection of the rootstock is crucial to the quality of the grafting and the yield of the products. The traditional commonly used peach rootstock is susceptible to root-knot nematode infections, resulting in a decreased yield, while a variety of cherry plum rootstocks, Mirabolano 29C, is resistant to root-knot nematode. In this study, root-knot nematode infection experiments on seedlings of traditional peach rootstocks and Mirabolano 29C confirmed that Mirabolano 29C was indeed more resistant to root-knot nematodes. At the same time, we compared the roots of the root-knot nematode uninfected and infected Mirabolano 29C by transcriptome sequencing and found 3 176 differentially expressed genes. A further functional enrichment analysis of these genes found that the secondary metabolites, phenylpropane and flavonoids, may be responsible for the high resistance of Mirabolano 29C to root-knot nematodes. Apples are available all year round since they can be stored for long time. However, the expression of the major apple allergen Mal d 1 can increase in the fruit during storage and incease so the risk of allergies in sensitive consumers. Therefore, we studied the change in the expression of the Mal d 1 gene during storage in the modern cultivar UEB 32642 (known under the brand name Opal®) and the cultivar Gala Brookfield (cv. Gala) as a reference. The cultivars were stored under two different conditions, ultra-low oxygen (ULO) and standard cold conditions. The gene expression was evaluated both in fresh fruits and in fruits stored for four, six, and nine months. Nine isoforms of the gene Mal d 1 with the highest published expression were quantified using real-time PCR. The most expressed isoforms Mal d 1.01, Mal d 1.02 and Mal d 1.06A were found in the fresh and also in the stored fruits. The expression of the Mal d 1.03G and Mal d 1.06D isoforms was higher in the stored fruits. Our study confirmed that (i) Opal® had a lower overall expression of the Mal d 1 gene than cv. Gala, both in the fresh and stored fruits; (ii) standard cold storage is superior in preserving lower Mal d 1 levels in Opal®apples compared to the ULO conditions; and (iii) less expressed isoforms may be responsible for the general increase in the Mal d 1 gene expression during storage. The CBF (C-repeat binding factors) genes play important roles in response to abiotic stress and environmental changes. In the present study, a total of 18 CBF genes were identified from a grapevine. Their domains, phylogenetics, and collinearity were analysed. The results revealed, that 18 VviCBF genes were distributed on 10 chromosomes unevenly in the grape genome. Promoter data analysis showed that the CBF gene has many cis-acting elements related to plant growth and development, light response, hormone, and abiotic stress response. We found that six VviCBF genes including, VviCBF5, VviCBF13, VviCBF14, VviCBF15, VviCBF16, and VviCBF18 differentially expressed during fruit developmental stages. Furthermore, four VviCBF genes including, VviCBF1, VviCBF3, VviCBF6, and VviCBF11 were expressed at the early stage of bud dormancy, whereas, nine VviCBF genes were expressed at the bud dormancy-breaking stage. Additionally, various VviCBFs genes respond to different abiotic and biotic stress. The effective use of high-quality and high-yielding germplasm resources is of great importance for the development of hulless barley varieties. Therefore, the search for such resources has long been a goal of the breeding community. In this study, a genotyping-by-sequencing (GBS) analysis was performed on an F2 (Nierumuzha × Kunlun 10) population. A high-density genetic map of hulless barley was constructed, which contained 1 475 efficient single-nucleotide polymorphism markers with 7 052 bin markers. The total length of the seven chromosomes was 1 139.4 cM, with an average marker distance of 0.59 cM. Based on this high-density linkage map, a total of 54 quantitative trait loci (QTLs) related to the seed traits were detected, including seed colour (SC), thousand kernel weight (TKW), seed average area (SAA), seed perimeter (SP), seed length (SL), seed width (SW), seed length/width (SLW), seed diameter (SD), and seed circular degree (SCD). These QTLs explained 3.32-35.73% (mean = 11.45%) of the phenotypic interpretation, including 24 main QTLs and 30 epistatic QTLs. A total of 24 candidate genes were identified within the QTL region, including one SC-associated transcription factor (ANT1), two TSW-related genes, a transcription factor (NAC021), a gene associated with the non-homologous end joining (NHEJ) pathway (ku70), three SAA-associated genes (LOL2, NAC021, TSK), two SL-associated genes (MADS21, MADS4), six SW-associated genes (FIP1, NAC021, DREB 1A, HVA22A, CYP78A6, SAUR71), five LW-related genes (NAM-B2, CRY1, LHY, CYP710A1, WRKY72), two SP-related genes (SKIP11, TCP18), two SD-related genes (NAC021, SKIP8), and three SCD-related genes (MYB1R1, RAX3, NAC100). These genes are involved in the regulation of the cell development, material transport, signal transduction, and plant morphogenesis and play an important role in the regulation of agronomic traits in hulless barley. One of the main issues addressed by phytology in recent years has been plant tolerance mechanisms for abiotic stress. No combined analysis has been made to identify the genes involved in drought stress tolerance. The meta-analysis of microarray data related to drought stress was analysed by the R software packages and showed 3 029 upregulated genes and 3 017 downregulated genes. The upregulated genes were mostly related to the drought tolerance protein, abiotic stress response, and the Cys2His2 Zinc Finger Transcription Factor (C2H2 zinc finger TF). The downregulated genes were mainly related to the late embryogenesis abundant protein, abiotic stress response, and the basic leucine zipper (bZIP) TF. The common gene ontology (GO) terms in the upregulated and downregulated genes were mainly related to the metabolic process, response to stimulus, cellular metabolic process, and photorespiration. The up and down meta-differential expressed genes (meta-DEGs) mainly belonged to the those following Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways including: the biosynthesis of secondary metabolites, plant hormone signal transduction, mitogen-activated protein kinase (MAPK) signalling pathway, and RNA degradation. Moreover, in the upregulated and downregulated genes, the TFs with a high percentage mainly belonged to the Teosinte branched1/Cincinnata/proliferating cell factor (TCP), basic helix loop--helix (bHLH) and bZIP. Next, the hub upregulated genes were mainly related to the thiamine biosynthesis protein thiC, 4-hydroxyphenylpyruvate dioxygenase, ribose-5-phosphate isomerase precursor and heat shock protein. The hub downregulated genes were mainly associated with the elongation factor Ts, aldehyde dehydrogenase, and trigger factor. Finally, the data from the present meta-analysis were compared with previous studies on the qRT-PCR results and their up and down expressions were confirmed. Based on the findings of the current study, novel insights into the drought stress molecular response can be provided and various candidate genes can be introduced for barley drought stress tolerance breeding. The present study was conducted to explore the variability generated through recombination breeding for nine economically important traits in Indian mustard (Brassica juncea L. Czern & Coss.). Delineation of the inbred lines to different gene pools, based on genetic diversity, enables their utilisation in hybrid breeding. The unweighted pair group method with arithmetic mean (UPGMA) and a Euclidean distance matrix was used to delineate the inbred lines to clusters. The variability was studied using the range and coefficients of variation of the traits. Significant variability was observed for all studied traits except for oil content and days to maturity. 128 Indian mustard genotypes were grouped into four distinct gene pools based on genetic diversity. A set of 20 most diverse genotype combinations was produced. Promising inbred lines were identified and recommended as donors for the respective trait. The pedigree analysis of the inbred line groups revealed, that recombination breeding caused a large diversity as confirmed by the assignment of inbred lines with the same parentage to specific clusters. Knowledge of gene actions governing begomovirus resistance and plant architectural traits is a prerequisite for a successful hybrid breeding programme. Therefore, the gene actions associated with these traits were studied in two intervarietal crosses of Cucurbita moschata (C1: Punjab Nawab × MVSR-6711 and C2: Punjab Nawab × P-135). We used the generation mean analysis of six generations for this purpose. Significant differences between the generation means were observed for all the traits in both crosses. The parental lines differed significantly in most of the studied traits. The nature and magnitude of the gene effects of seventeen traits varied by trait and cross. A simple additive dominance model was adequate for the internode number, leaf length and width, petiole length, fruit weight and cavity diameter in C1 and the number of fruits/plant in C2. The non-allelic interaction was found to be significant for a majority of the traits including the per cent disease index of the squash leaf curl China virus, tomato leaf curl New Delhi virus and their mixed infections, which indicated, that recurrent selection in biparental progeny might be useful for the accumulation of genes with additive effects. Duplicate epistasis was observed for the vine, internodal and peduncle length in C1 and the internode number, petiole and peduncle length, peduncle and fruit polar diameter in C2. We investigated the phenotypic and genetic diversity among 25 grapevine accessions of white grape maintained in a germplasm collection ex situ. The selected varieties were characterised using standardised ampelographic descriptors and microsatellite markers to obtaintheir complete description. The ampelographic analysis based on 24 OIV descriptors and the obtained first dendrogram based on common features, revealed the extent of the phenotypic diversity ranging from 0.29 (between Coarnă albă and Chardonnay) to 0.92 (between Crâmpoşie and Cioinic), without any connection with their supposed origin or the current geographic area of distribution. The SSR allelic analysis with nine standard markers proved and confirmed its high discrimination potential for genotype identification and diversity. The second cluster analysis aimed to evaluate the genetic relationships among analysed accessions, based on Dice's similarity coefficient, revealed two groups of varieties: the first group (A) included twelve accessions with a degree of similarity between 0.37 and 0.5. The second (B) included eleven accessions, with a similarity degree between 0.33 and 0.67. The genotypic cluster analysis showed a distinct lower similarity between the Majarcă albă variety and all the varieties from the first group, and between Busuioacă de Bohotin and all the analysed varieties. The phenotypic and genetic matrices and dendrograms, although not correlated, were complimentary and provided valuable information about the diversity of the grapevine genotypes and the identification of the synonyms. This paper studies QTL (quantitative trait locus) resistance to Sharp Eyespot in wheat using a genetic population of the recombinant inbred line (RIL) hybridized from Niavt 14 and Xuzhou 25. Based on three-year phenotypic data and a resultant RIL genetic map, three QTLs in total associated with Sharp Eyespot resistance were detected. Two out of the three loci were detected on chromosome 2B, namely Qses.jaas-2b1 and Qses.jaas-2b2, of which Qses.jaas-2b1 accounted for 5.46 and 8.56% of the phenotypic variation in the field after inoculation in two successive years and Qses.jaas-2b2 accounted for 6.04, 8.10 and 12.92% after field inoculation for three successive years. A further QTL for resistance gene named Qses.jaas-7d was detected on chromosome 7D in addition to the two on 2B and this exceeding 11.25% of the phenotypic variation. These results indicate that QTLs associated with Sharp Eyespot possibly exist on the linkage groups of chromosomes 2B and 7D in wheat and lay the foundation for further research of QTL resistances to Sharp Eyespot in wheat. The external fruit colour is an important parameter of the fig fruit quality. Fig anthocyanin content is critical for the peel colour. The peel of mature fruits of the fig cultivar Orphan and its red peel bud mutant Hongyan were separated for a transcriptomic and proteomic analysis. A total of 162 different abundance proteins (DAPs) and 5 015 differentially expressed genes (DEGs) were identified. The correlation analysis revealed that only two and 15 genes were downregulated and upregulated, respectively, at both the transcriptome and proteome levels. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that the enrichment pathways including Tropane, piperidine and pyridine alkaloid biosynthesis, phenylalanine metabolism and isoquinoline alkaloid biosynthesis for DEGs, and protein processing in the endoplasmic reticulum and flavonoid biosynthesis may contribute to the mutant color phenotype. Sustainable global wheat production requires wheat varieties, that are sufficiently resistant to the main wheat diseases. The economically important fungal pathogens worldwide include powdery mildew (PM), yellow rust (YR), leaf rust (LR) and blotch causing pathogens including Septoria nodorum blotch (SNB) and Septoria tritici blotch (STB). Here, we present the evaluation of winter wheat varieties of diverse origin against the prevalent local populations of PM, YR, LR, STB and SNB under natural infection conditions through image-based phenotyping in two consecutive years (2019 and 2020). We found several varieties to be resistant against multiple diseases. Following the association mapping, we obtained a total of 206 marker trait associations for all the parameters scored which were condensed to 79 quantitative trait loci (QTLs) (eight QTLs for PM, 25 QTLs for LR, 11 QTLs for YR, 19 QTLs for SNB and eight QTLs for STB) based on the linkage disequilibrium among the molecular markers. The known genes present at these QTLs are discussed in detail. Knowledge of the molecular mechanisms of response to environmental stress is fundamental for the development of genetically stress-tolerant crops. This study aims to find vegetable soybean accessions tolerant to cultivation in stressful tropical environments. Fourteen accessions of the vegetable soybean (Glycine max (L.) Merrill) were grown in mineral and beach ridges interspersed with swale (BRIS) soils. The genetic diversity, estimated using inter-simple sequence repeat (ISSR) markers, revealed 42.50% polymorphism and was regarded as moderate. The unweighted pairgroup method arithmetic average (UPGMA) analysis allocated the tested accessions into five major clusters at a similarity coefficient level of 0.43. The lowest values of the genetic distance were between IIUMSOY11 and IIUMSOY13 & IIUMSOY13 and IIUMSOY14, indicating that these accessions were more genetically distant from the other accessions. Ten differentially expressed proteins were identified in the three selected accessions IIUMSOY1, IIUMSOY11 and IIUMSOY14 using mass spectrometry, revealing a unique expression of the proteins involved in the storage, flavonoid metabolism, protein modification, oxidative stress defence, carbohydrate metabolism and respiratory chain. The findings may be valuable for the selection of genetically diverse accessions, to enhance the breeding of vegetable soybean genotypes suitable for stressful tropical environments. Chilling is an important abiotic stress in plants. Sweet potato is sensitive to cold damage due to its tropical origin. In this study, we identified a basic helix-loop-helix (bHLH) gene, IbbHLH33, from our cold-tolerance-related transcriptomic data. Further analyses revealed that IbbHLH33 encoded a nuclear protein and was most closely related to AtbHLH33. RT-qPCR analysis showed that IbbHLH33 was expressed at the highest level in the roots, and its expression was strongly induced by low temperature (4 °C), H2O2 and abscisic acid (ABA) treatments. Transgenic tobacco plants overexpressing IbbHLH33 were obtained by Agrobacterium-mediated transformation, which enhanced the chilling resistance of tobacco. At low temperatures, the proline content, superoxide dismutase (SOD) activity and malondialdehyde (MDA) content increased significantly, while the relative conductivity decreased significantly. At the same time, the expression of proline synthesis related genes and antioxidant activity related genes increased, while the expression of ABA synthesis related genes decreased. The results showed that IbbHLH33 is a transcription factor encoding a gene of the bHLH family that regulates chilling tolerance. The wheat leaf rust resistance gene Lr1 encodes a typical coiled-coil nucleotide-binding site leucine-rich repeat (CC-NBS-LRR) of resistance protein containing 1 344 amino acids. WR003, a cleaved amplified polymorphic sequence (CAPS) marker is derived from the LRR regions of Lr1. In this study, a worldwide collection of 120 Aegilops tauschii accessions and 282 hexaploid wheat varieties was screened for Lr1 alleles using WR003, and the specificity of WR003 for Lr1 was confirmed by pathogenicity tests and genotype analysis. The sequence alignment and phylogenetic tree analysis of 38 Lr1 haplotypes provided a further view of the molecular evolution of Lr1. The results showed that there were very few polymorphisms between the Lr1 alleles from Ae. tauschii and hexaploid wheat with the same resistance phenotype. The polymorphisms of the Lr1 haplotypes were mainly between the different resistance lines, rather than between the different ploidy levels. These results indicate that Lr1 originated from Ae. tauschii and differentiated into resistant and susceptible genotypes before its introgression into hexaploid wheat. Therefore, it is likely that wheat Lr1 has at least two major variants for disease resistance and susceptibility, and except for certain point mutations, few gene conversions and genetic re-combinations occurred during the hexaploid wheat domestication. The relationship between the early ground cover and the grain yield in winter wheat is not yet fully understood. In a winter wheat breeding programme, selection for early ground cover is traditionally made using visual scoring. Although visual scoring is preferred as a phenotypic screening tool by wheat breeders, its output may not be reliable, as it requires experience. A smartphone camera-based digital image technique can be recommended as a feasible, reliable, repeatable, affordable, and fast selection tool for early ground cover in wheat as an alternative to visual scoring. For this purpose, two wheat trials were conducted in the 2017--2018 and 2019--2020 seasons. In both seasons, 215 wheat genotypes in total, together with three checks from spring wheat, were tested under rain-fed conditions in the spring wheat zone in Turkey. All the tested wheat genotypes were grouped into spring, facultative, and winter growth habit using visual scoring. Simultaneously, photos were taken from each plot with a smartphone camera, and the early ground cover (%) was estimated using the smartphone camera-based digital image technique. The relationships between grain yield, visual scoring, and early ground cover could so be estimated. In both seasons, significant negative correlation between grain yield and visual scoring (r = -0.679** and r = -0.704**, respectively) and significant positive correlation between the grain yield and the early ground cover (r = 0.745** and r = 0.747**, respectively) were observed. The correlation between visual scoring and early ground cover were negative (r = -0.862** and r = -0.926**, respectively). The broad sense heritability estimates in both seasons were 0.51 and 0.85, respectively, for early ground cover, 0.91 and 0.94 for visual scoring, and 0.86 and 0.69 for grain yield. In this study, we revealed that testing winter wheat genotypes in the spring wheat zone rather than in the winter wheat zone could be a more effective way to unveil the positive relationship between the early ground cover and the grain yield. Breeding for aluminium (Al) tolerance is a vital approach for enhancing the productivity of field crops in acidic soil regions where Al toxicity seems to be the most restraining factor for crop performance. Sunflower is generally considered extremely sensitive to Al toxicity; although no comprehensive information on the evaluation of sunflower genotypes for Al tolerance is available. In this study, 50 sunflower genotypes (set-I and set-II) were evaluated for Al tolerance at the seedling stage under hydroponic conditions. Substantial genetic variability in Al tolerance was observed among the studied genotypes. High estimates of heritability were obtained for both the total root length (TRL) and root regrowth (RRG), together with high estimates of genetic advance. A cluster analysis separated the genotypes into five different groups among the studied germplasm, the genotypes; NDLR-06 and EC-601861 were observed to be highly Al tolerant in terms of root regrowth under Al stress. In conclusion, the findings lreveal the complex mechanisms of Al tolerance in sunflower and may help to find new genetic resource for the improvement of Al tolerance in sunflower breeding. The inheritance of anther orientation of 154 individuals from two B1 populations of Brassica rapa (syn. Brassica campestris) was evaluated under controlled conditions in a greenhouse. The anther orientation was evaluated visually at the time of fully open flowers. The observed extrorse:introrse ratios were 1: 1 in R-o-18 × (R-o-18 × RM29) population and 3: 1 in R-o-18 × (R-o-18 × R c-50) population. It was concluded that this trait is controlled by two duplicated pairs of genes (A1, A2) for extrorse anthers, either of which can produce extrorse anthers when a single dominant allele is present. In breeding plants for resistance to disease pathogens major genes are often used. They mostly confer a resistance, which can be overcome by a corresponding pathogen virulence specific to the resistance conferred by that gene. Therefore, monitoring of new virulences in the pathogen population is an integral part of both the study of resistance and practical breeding. The aim of this paper is to present the detection of rare virulences that have been found in aerial populations of Blumeria graminis f.sp. hordei collected in the Czech Republic during the last six years. In total, 822 random isolates originating from cultivated spring and winter barley fields were evaluated on 37 differential varieties and 12 isolates carrying rare virulences were found. At least four virulences to resistances in Laverda, Psaknon, Venezia and SJ048311 were novel. In contrast, four rare isolates avirulent to the resistance Ha were also found. No isolate avirulent to resistances Ra, HH (a8) and Lo was detected. The present paper summarized some knowledge of modern technologies, applied in apple (Malus domestica) genome studies. New generation sequencing allowed single nucleotide polymorphism (SNP) chip technologies for genotyping, description of functional apple genes, characterization of the evolutionary results in apple genome fragment transition, as well as phylogenetic reconstruction of the genus Malus, being the confirmed progeny of M. sieviersii. Based on these technologies, newly developed putative markers may give the most important biological data such as age, geographical origins, tissue type determination, and external visible characters. The new generation genotyping platforms, representing very high efficiency, are now successfully applied for random apple genome-wide association (GWA) studies as well pedigree-based analysis and marker-assisted selection (MAS). Fusarium oxysporum Schlecht. f.sp. melongenae (FOM), Verticillium dahliae (Ve) and Ralstonia solanacearum are major limiting pathogens affecting eggplant cultivation and their yield in the world. Natural resistance genes are the most environmentally friendly method to control the disease. Thus, marker-assisted selection (MAS) is preferred as a tool for screening resistance genes (R-genes) in eggplant resistance breeding. In this study, markers that are specifically linked to major disease resistance genes conferring resistance to F. melongenae, V. dahliae and R. solanacearum were tested in a population containing breeding materials to validate the resistance. Resistant Solanum melongena accessions LS1934 and LS2436 and their reciprocal crosses were used as the resistance resource for this validation study. Moreover, classical resistance tests to FOM and Ve were performed with the root-dip inoculation method for classification of all the accessions based on their resistance/susceptibility responses. The SCAR426, CAPs_903 and SIVR844 markers were highly informative for the determination of resistance genes (Fomg, Ve and ERs1). Therefore, in areas with high susceptibility to diseases, a highly efficient combination of the relevant R-genes and their pyramiding into commercial eggplant varieties are proposed to be implemented as a pragmatic approach. Rice is one of the frontline cereals in the world and the major cultivated crop in Bangladesh. A total of eleven simple sequence repeats (SSRs) and thirteen sequence-tagged site (STS) markers were used to characterize twenty-four rice cultivars in Bangladesh. Twenty-four markers generated 60 alleles with 2.5 alleles per locus. The average polymorphism information content (PIC) value was 0.40, while the mean value of heterozygosity, gene diversity, and major allele frequency were recorded as 0.10, 0.48 and 0.62, respectively. However, the SSR markers showed more specificity and a higher discrimination power than the STS markers. The cluster analysis displayed four major clusters with a genetic similarity coefficient value of 0.73. The morphological analyses of the grain identified that Binadhan-20 and BRRI dhan34 had the longest and the shortest seed size, respectively, with a variable correlation between the seed length, width and length/width ratio. The phenol reaction test distinguished seven cultivars as japonica and seventeen cultivars as indica or an intermediate type. The stem juice yield is a key factor that influences both the biological and economic production of sweet sorghum [Sorghum dochna (Forssk.) Snowden]. To elucidate upon the genetic basis of the stem juice yield, an F5 population developed from a cross between the low juice yielding Xinliang52 (XL52) and high juice yielding W455 lines, were used in a quantitative trait locus (QTL) analysis. A main effect of the QTL controlling stem juice yield was separated with an SSR marker called Xtxp97, which explained 46.7% of the phenotypic variance. In addition, F5 and F6 populations were constructed with XL52 and W452 as the parents to further verify the QTLs, and a significant correlation was found between the juice yield trait and the Xtxp97 marker. Based on the progeny tests of 29 recombinants, QJy-sbi06 was located in a region of about 21.2 kb on chromosome 6, where a candidate gene encoding an NAC transcription factor (sobic.006G147400) was identified. Combining the different population association analysis and sequencing technology showed that XL52 inserted a 1.8 kb transposon in the NAC to directly interrupt and inactivate the juice yield gene. This study also demonstrated that the colour of the leaf midribs was controlled by a single gene and was significantly positive correlated with juiciness (r = 0.784, P < 0.01). These results could lay the foundation for map-based cloning of QJy-sbi06 and provide genes or QTLs for breeding sorghum lines with a high juice yield and quality. [ABSTRACT FROM AUTHOR]

Hedera helix L. is an invasive, but medicinally important plant. In Iran, there is no available study on the H. helix population to reveal the genetic diversity and population structure. Fifty-six individual plants belonging to nine geographical populations were collected in four provinces of Iran. High genetic diversity, polymorphisms, and a Shannon diversity index of 0.269 were detected in Mazandaran, Kandovan (Population 3). Analysis of the molecular variance indicated 40% of total genetic variation of the whole population was present in the subpopulation. A high genetic similarity (0.922) between plant Populations 5 (Kermanshah; Islamabad) and 6 (Kermanshah; Paveh) was noted. On the other hand, a low genetic similarity was observed between plant Populations 1 (Tehran; Darband) and 8 (Ardabil; Hur). The Mantel test revealed a correlation between the genetic and geographical distances. Furthermore, it demonstrated the isolation mechanism responsible for the population structure in the H. helix plant populations. The principal component analysis explained the majority of the variation in the morphological characteristics. Three components explained 87% of the variation, and the first component explained 60% of the variation. For instance, the leaf morphology showed a correlation of > 0.7 between leaf morphological and floral characters. The plant leaves and quantitative flower characteristics separated the plant populations according to the differences in length. Many plant genetic engineering taskss require the spatial expression of genes which in turn depends upon the availability of specific promoters. The present paper analyses the green-tissue characteristics of a new L. gibba rbcS promoter driving the expression of the gus gene in transgenic tobacco. A 1491 bp rbcS (small subunit of ribulose bisphosphate carboxylase) promoter was isolated from Lemna gibba. The sequence analysis revealed that this promoter is different from the previously reported rbcS promoter and is named SSU5C. A 1438 bp fragment of the SSU5C promoter was fused with the gus gene and transgenic tobacco plants were generated. The analysis of T1 tobacco p1438-gus revealed that GUS expression driven by the SSU5C promoter was detected in the green part of vegetative organs. The promoter deletion analysis confirmed a region from position -152 to -49 relative to the start of transcription containing boxes X, Y and Z, while a positive regulatory region conferred green tissue-specific expression. Further functional analysis of constructs of box-X, Y, Z, which was fused with the basal SSU5C promoter, confirmed that the boxes X, Y and Z represent the new minimized functional promoter, respectively, and are able to direct green tissue-specific expression. Twenty hop genotypes were selected for the evaluation of resistance to primary and secondary Pseudoperonospora humuli infection and of alpha acid and hop oil content in the hops. From the wild hop genotypes, two from Canada and one from Belgium showed resistance. Among the registered hop varieties, the Czech varieties Kazbek and Boomerang were the most resistant. Both wild hop genotypes from Canada showed the highest content of alpha acids among the wild hop entries, namely 4% w/w. The lowest variability of the alpha acid content in the wild hop category was found in two wild hop varieties from the Caucasus, one from Austria and one from Lithuania. The highest content of hop oils was determined in two hop genotypes from Canada and two from Belgium. Wild hop genotypes from the Caucasus have the lowest variability of hop oils among the wild hop entries.
