Arbutus unedo L. is a resilient tree with a circum-Mediterranean distribution. Besides its ecological relevance, it is vital for local economies as a fruit tree. Several microorganisms are responsible for strawberry tree diseases, leading to production constrictions. Thus, the development of alternative plant protection strategies is necessary, such as bacterial endophytes, which may increase their host's overall fitness and productivity. As agricultural practices are a driving factor of microbiota, this paper aimed to isolate, identify and characterize endophytic bacteria from strawberry tree leaves from plants growing spontaneously in a natural environment as well as from plants growing in orchards. A total of 62 endophytes were isolated from leaves and identified as Bacillus, Paenibacillus, Pseudomonas, Sphingomonas and Staphylococcus. Although a slightly higher number of species was found in wild plants, no differences in terms of diversity indexes were found. Sixteen isolates were tested in vitro for their antagonistic effect against A. unedo mycopathogens. B. cereus was the most effective antagonist causing a growth reduction of 20% in Glomerella cingulata and 40% in Phytophthora cinnamomi and Mycosphaerella aurantia. Several endophytic isolates also exhibited plant growth-promoting potential. This study provides insights into the diversity of endophytic bacteria in A. unedo leaves and their potential role as growth promoters and pathogen antagonists. [ABSTRACT FROM AUTHOR]

Species mapping methods play a central role in biogeographical questions, as they may generate a domino effect on further works based on species distribution. In light of the massive recent increase in the availability of online occurrence data, we highlight the strengths and limitations of the mapping methods most widely used to display the geographic distribution of plants, namely geographic range maps and occurrence record maps. We use the modern distribution of the genus Arbutus in western Eurasia, North Africa, and Macaronesia, for which no occurrence record map has been published yet, to discuss critical issues in data collection and representation. The occurrence record map of A. unedo, A. andrachne, A. canariensis, and A. pavarii shows how well this mapping method captures the details of peripheral and isolated stands as well as the variability of population density. A number of biogeographical issues are addressed by this approach, including the determination of the chorological centre of gravity in relation to historical dynamics, genetic patterns in relation to range porosity, and the autochthony status of marginal stands. These issues constitute the necessary foundation for additional palaeobotanical research and ecological modelling to investigate the past-to-future dynamics of Arbutus and other species of the Mediterranean–Atlantic area. The use of alternative ingredients in the production of kombucha has seen a recent increase. Our research aimed to characterize the chemical, nutritional, microbial, and aromatic profiles of kombucha beverages prepared with strawberry tree (Arbutus unedo) fruits fermented with three different SCOBYs for 21 days. The analyses showed similar levels of microbiological groups (aerobic mesophilic microorganisms, lactic acid bacteria, acetic acid bacteria, and yeasts)among the SCOBYs used. The beverages studied displayed a decrease in pH value and carbohydrate content, and protein degradation was also observed as fermentation progressed. However, the increase in total phenolic compounds during the first week proved to be a point of interest. A total of 20 volatile organic compounds were detected, giving different sensory qualities to the beverages: higher ethanol, benzaldehyde-4-ethyl, or acetic acid depending on the SCOBY used. The results obtained indicated that strawberry tree kombucha might be an alternative beverage with notable nutritional and aromatic properties, with fermentation time and SCOBY composition being identified as crucial factors. In this study, original smoothies obtained with strawberry tree fruit puree and apple juice enriched with Diospyros kaki fruits, Myrtus communis purple berry extract, Acca sellowiana, and Crocus sativus petal juice were evaluated for their antioxidant activity and inhibition of targeted digestive enzymes. Values of CUPRAC, FRAP, ORAC, DPPH•, and ABTS•+ assays generally increased with plant enrichment, particularly for A. sellowiana addition (ABTS•+ 2.51 ± 0.01 mmol Trolox/100 g fw). The same trend was observed regarding the ability to scavenge reactive oxygen species (ROS) tested in Caco-2 cell cultures. Inhibitory activity on α-amylase and α-glucosidase was increased by D. kaki, M. communis, and A. sellowiana. Total polyphenols evaluated by UPLC-PDA analysis ranged between 535.75 ± 3.11 and 635.96 ± 5.21 mg/100 g fw, and A. sellowiana provided the higher amount. Flavan-3-ols accounted for more than 70% of phenolic compounds, and only smoothies enriched with C. sativus showed a high amount of anthocyanins (25.12 ± 0.18 mg/100 g fw). The outcome of this study indicates these original smoothies as a possible ally in counteracting oxidative stress, as established by their favourable antioxidant compound profile, thus suggesting an interesting future application as nutraceuticals. Strawberry tree (Arbutus unedo L.) honey (STH) has been used since ancient times as a folk medicine remedy, especially in certain Mediterranean countries. This honey, rich in phenolic content, is well recognized for its antioxidant, anti-inflammatory, and antimicrobial activities, and is used for the treatment of skin lesions as well as gastrointestinal and respiratory disorders. This study investigated whether STH alleviates genome damage in human peripheral blood lymphocytes produced by the cytotoxic drug irinotecan. The phenolic profile of STH was previously estimated by ultra-high-performance liquid chromatography coupled to a linear ion trap–Orbitrap hybrid mass spectrometer. The effects of STH were evaluated at three concentrations (1×, 5×, and 10×), based on the daily consumption of the honey by an adult person. After 2 h of in vitro exposure, standard lymphocyte cultures for the analysis of chromosome aberrations and the cytokinesis-block micronucleus cytome assay were established. Our results demonstrate that STH offered remarkable geno- and cytoprotection when administered with irinotecan. These findings are relevant for drawing preliminary conclusions regarding the in vitro safety of the tested honey. Phenolic compounds are valuable cosmetic ingredients. They display skin protective potential and play an important role in preserving cosmetic formulations due to their ability to neutralize free radicals. Considering this fact, the current study aims to obtain a phenolic-enriched fraction from Arbutus unedo for topical application in cosmeceutical products. The chemical composition and the antioxidant, anti-inflammatory, and anti-tyrosinase activities of different extracts from the plant were investigated and compared. Samples were obtained by maceration, reflux, and ultrasound using water and ethanol. The findings indicated that the extraction methods impacted the phytochemical composition of the extracts. The high-performance liquid chromatography with diode-array detection (HPLC–DAD) analysis showed a wide range of phenolic compounds, comprising phenolic acids and flavonoids. Among the extracts, the water reflux had significant levels of both total polyphenols, flavonoids, and tannins and possessed the most important content on hyperoside. It displayed the most significant antioxidant activities with high antiradical and reducing power, as well as strong total antioxidant activity. It possesses a promising whitening effect with high anti-tyrosinase activities. Furthermore, it shows no cytotoxicity and moderate anti-inflammatory activity. Finally, due to its high yield efficiency and activities, water reflux was selected to formulate a cosmeceutical oil-in-water nanoemulsion that displayed optimal pH and stability. Strawberry trees (Arbutus unedo L.) are naturally grown in particular Black Sea and Mediterranean regions of Turkey with great diversity due to continuous seed propagation for centuries. The trees differ in terms of most of the horticultural characteristics. We investigated the phenolic compounds and the biochemical and pomological characteristics of the fruits of eight strawberry tree selections naturally grown in the western part of Turkey. Significant differences were found among the genotypes in terms of their phenolic compounds and their biochemical and pomological characteristics. Among soluble sugars, fructose (11.63 g 100 g-1) was the dominant sugar, followed by glucose (6.10 g 100 g-1) and sucrose (1.44 g 100 g-1) for all the genotypes. Positive correlation was found between fruit weight and soluble sugar content. Malic acid was the major organic acid (0.67-2.33 g 100 g-1), and the second major organic acid in strawberry tree fruits was citric acid (0.25-0.87 g 100 g-1). Vitamin C content was an average of 56.22 g 100 g-1 for the eight genotypes. Among phenolic compounds, gallic acid was dominant (1.62-7.29 mg 100 g-1), followed by chlorogenic acid (1.23-3.14 mg 100 g-1), on an average basis. [ABSTRACT FROM AUTHOR]

The Mediterranean region is a hot spot of climate change vulnerable to increased droughts and heat waves. Scaling carbon fluxes from leaf to landscape levels is particularly challenging under drought conditions. We aimed to improve the mechanistic understanding of the seasonal acclimation of photosynthesis and morphology in sunlit and shaded leaves of four Mediterranean trees (Quercus ilex L., Pinus halepensis Mill., Arbutus unedo L. and Quercus pubescens Willd.) under natural conditions. Vc,max and Jmax were not constant, and mesophyll conductance was not infinite, as assumed in most terrestrial biosphere models, but varied significantly between seasons, tree species and leaf position. Favourable conditions in winter led to photosynthetic recovery and growth in the evergreens. Under moderate drought, adjustments in the photo/biochemistry and stomatal/mesophyllic diffusion behaviour effectively protected the photosynthetic machineries. Severe drought, however, induced early leaf senescence mostly in A. unedo and Q. pubescens, and significantly increased leaf mass per area in Q. ilex and P. halepensis. Shaded leaves had lower photosynthetic potentials but cushioned negative effects during stress periods. Species-specificity, seasonal variations and leaf position are key factors to explain vegetation responses to abiotic stress and hold great potential to reduce uncertainties in terrestrial biosphere models especially under drought conditions. Introduction. Several studies reveal the important role played by ‘lesser-known’ wild fruits since they contain nutritional and functional compounds which have biological properties. Materials and methods. Our work studied the presence of bioactive compounds such as vitamin C (ascorbic acid and dehydroascorbic acid), total phenolic content, phenolic acids, flavonols, anthocyanins and the antioxidant capacity (FRAP, ABTS•+ and DPPH• in vitro tests) in wild fruits of Arbutus unedo L. and Rubus ulmifolius Schott of Spanish origin, including samples from different seasons and locations. Results and discussion. A wide variability was found in the composition of fruits of the same species, which substantiates the importance of analyzing several batches of wild fruits, to achieve representative results, taking into account the natural variability. Arbustus unedo fruits showed higher vitamin C, mainly in the ascorbic acid form, and phenolic content than R. ulmifolius [(172–419) mg ascorbic acid×100 g–1 fw vs. (5.99–26.83) mg ascorbic acid×100 g–1 fw, and (773–1621) mg total phenolic compounds×100 g–1 fw vs. (376–1326) mg total phenolic compounds×100 g–1 fw, respectively]; phenolic acids and anthocyanins are the major groups of phenolic compounds found in both species, with gallic acid and cyanidin 3-glucoside as the main compounds. Arbustus unedo fruits showed significantly higher Folin-Ciocalteu values than those of R. ulmifolius. These values are higher than those reported for the majority of berries. The significant correlations found among different antioxidant compounds (r > 0.6300, P < 0.001) may reveal a protective effect between ascorbic acid and phenolic acids or anthocyanins in the fruits. Woody species in Mediterranean ecosystems form intra-annual density fluctuations (IADFs) in tree rings in response to changes in environmental conditions, especially water availability. Dendrochronology, quantitative wood anatomy and high-resolution isotopic analysis (using a laser ablation technique) were used to characterize IADFs in Arbutus unedo shrubs grown on two sites with different water availability on the island of Elba (Italy). Our findings show that IADF characterization can provide information about the relationship between environmental factors and tree growth at the seasonal level. At the more xeric site, IADFs mainly located in the early and middle parts of the annual ring, showed a decrease in vessel size and an increase in δ13C as a result of drought deficit. Opposite trends were found at the more mesic site, with IADFs located at the end of the ring and associated with a lower δ13C. Moreover, at the first site, IADFs are induced by drought deficit, while at the second site IADFs are linked with the regrowth in the last part of the growing season triggered by favourable wet conditions. This combined approach is a promising way for dating problematic wood samples and interpreting the phenomena that trigger the formation of IADFs in the Mediterranean environment. Pressure infiltration of water into a leaf via the stomatal pores can be used to quickly determine whether all stomata are open, or as recently described for several mesophytic and xerophytic species, whether there is a non-homogeneous distribution of stomatal opening (stomatal patchiness) on the leaf surface. Information about this phenomenon is important since the commonly used algorithms for calculation of leaf conductance from water vapor exchange measurements imply homogeneously open stomata, which in the occurrence of stomatal patchiness will lead to erroneous results. Infiltration experiments in a growth chamber with leaves of the Mediterranean evergreen shrub Arbutus unedo, carried out under simulated Mediterranean summer day conditions, where the species typically exhibits a strong midday stomatal closure, revealed a temporary occurrence of stomatal patchiness during the phase of stomatal closure in the late morning and during the stomatal reopening in the afternoon. Leaves were, however, found to be fully (i.e. homogeneously) infiltratable in the morning and in the evening. At midday during maximum stomatal closure, leaves were almost non-infiltratable. During the day, the infiltrated amount of water was found to be linearly correlated with porometer measurements of leaf conductance of the same leaves, carried out with the attached leaves immediately before infiltration. The quantification of climate–growth relationships is a fundamental step in tree-ring sciences. This allows the assessment of functional responses to climate warming, particularly in biodiversity and climate-change hotspots including the Mediterranean Basin. In this region, broadleaf tree and shrub species of pre-Mediterranean, subtropical origin, have to withstand increased aridification trends. However, they have not been widely studied to assess their long-term growth responses to climate and drought. Since these species evolved under less seasonal and wetter conditions than strictly Mediterranean species, we hypothesized that their growth would mainly respond to higher precipitation and water availability from spring to early summer. Here, we quantified climate–growth relationships in five of these broadleaf species showing different leaf phenology and wood type (Pistacia terebinthus L., Pistacia lentiscus L., Arbutus unedo L., Celtis australis L., and Laurus nobilis L.) by using dendrochronology. We calculated Pearson correlations between crossdated, indexed, mean ring width series of each species (chronologies) and monthly climate variables (mean temperature, total precipitation). We also calculated correlations between the species' chronologies and a drought index on 7-day scales. Lastly, we compared the correlation analyses with "climwin" analyses based on an information-theoretic approach and subjected to cross-validation and randomization tests. As expected, the growth of all species was enhanced in response to wet and cool conditions during spring and early summer. In some species (P. lentiscus, A. unedo, C. australis,) high prior-winter precipitation also enhanced growth. Growth of most species strongly responded to 9-month droughts and the correlations peaked from May to July, except in L. nobilis which showed moderate responses. The "climwin" analyses refined the correlation analyses by (i) showing the higher explanatory power of precipitation (30%) vs. temperature (7%) models, (ii) selecting the most influential climate windows with June as the median month, and (iii) providing significant support to the precipitation model in the case of P. terebinthus confirming that the radial growth of this species is a robust proxy of hydroclimate variability. We argue that "climwin" and similar frameworks based on information-theoretic approaches should be applied by dendroecologists to critically assess and quantify climate–growth relationships in woody plants with dendrochronological potential. A comparison between High-Performance Thin-Layer Chromatography (HPTLC) analysis and Liquid Chromatography High Resolution Mass Spectrometry (LC–HRMS), coupled with Principal Component Analysis (PCA) was carried out by performing a combined metabolomics study to discriminate Arbutus unedo (A. unedo) plants. For a rapid digital record of A. unedo extracts (leaves, yellow fruit, and red fruit collected in La Maddalena and Sassari, Sardinia), HPTLC was used. Data were then analysed by PCA with the results of the ability of this technique to discriminate samples. Similarly, extracts were acquired by non-targeted LC–HRMS followed by unsupervised PCA, and then by LC–HRMS (MS) to identify secondary metabolites involved in the differentiation of the samples. As a result, we demonstrated that HPTLC may be applied as a simple and reliable untargeted approach to rapidly discriminate extracts based on tissues and/or geographical origins, while LC–HRMS could be used to identify which metabolites are able to discriminate samples.
