[Objective] Scutellaria baicalensis Georgi (SBG) is a Chinese medicine. This paper studies the combined effect of initial soil water content and progressive drought on physiological traits and quality of SBG. [Method] The pot experiment was conducted in a mobile water shelter. The initial soil water content in the pots were controlled at 40%, 60% and 80% of the field capacity (FC) to represent severe, moderate, and mild drought, respectively. In each treatment, we measured the biomass, photosynthetic characteristics, osmotic substances, activities of antioxidant enzymes and active ingredients of the plants. [Result] ① No significant difference in biomass accumulation was found between the treatments. ② During the stress period, baicalin content was significantly higher in the 80%FC treatment, baicalein content was significantly higher in the 40%FC treatment and the baicalin content decreased with initial soil water treatment in the order of 40%FC>60%FC>80%FC. When the soil water content was 40%FC and 60%FC, the baicalin content was 17.3% and 11.3% higher than that when soil water content was 80%FC. Change in soil water contents did not have a significant impact on baicalein content. ③ Net photosynthetic rate, transpiration rate, stomatal conductance and intercellular carbon dioxide concentration all decreased as the plants grew, regardless of the soil water treatments. 8-10 d after the onset of water stress, the net photosynthetic rate did not show significant differences between the 60%FC and 80%FC treatments. ④ Under water stress, the content of malondialdehyde and proline in all treatments increased as the plants grew. The proline content was the highest in the 80%FC treatment, reaching 121.66 µg/g. ⑤ The activities of POD and CAT responded to water stress similarly, both increasing gradually as water stress persisted. The impact of soil water on activity of SOD was ranked in the order of 40%FC>60%FC>80%FC. ⑥ The active ingredients were higher in the 40%FC and 60%FC treatments. [Conclusion] Severe progressive drought with the soil water content kept at 40%FC reduced photosynthesis and increased baicalein content, osmolytes and activity of antioxidant enzymes. Mild and severe droughts by keeping the soil water content at 40%FC and 60%FC can improve accumulation of active ingredients in SBG. [ABSTRACT FROM AUTHOR]

Circular RNA (circRNA), an emerging non-coding RNA, is closely related to the processes of lipid deposition, inflammatory response, apoptosis, proliferation and repair in atherosclerosis (As), and affects the development of As. Traditional Chinese medicine (TCM) has an important academic value in the prevention and treatment of As, and its regulatory mechanism has not been comprehensively summarized. This paper reviewed the association of circRNA with lipid deposition, inflammatory response and macrophage in As, and summarized the research progress of the correlation between circRNA and atherosclerosis and the regulation of traditional Chinese medicine. Macrophage is a kind of immune cell which is closely related to atherosclerosis (As). With the change of internal environment, macrophage can polarize into different phenotype. Under the physiological environment, the polarization of macrophages is in dynamic balance. In the As state, the polarization balance is destroyed and the corresponding phenotypic macrophages are differentiated, which play different roles in each stage of As. The debate between Zheng and Xie exists in the whole process of As formation and development, while macrophage polarization runs through the whole stage of As syndrome differentiation and change. On account of this, this paper discusses the relationship between macrophage polarization and As based on the theory of Zheng and Xie, and propose that attention should be paid to the combination of macro and micro in the clinical prevention and treatment of As, and it can control the inflammatory response and stabilize plaque by regulating the macrophage polarization, thus providing an emerging target for the treatment of As with traditional Chinese medicine. The large language model (LLM), including ChatGPT, has shown outstanding performance in understanding and responding to human instructions, and has a profound impact on natural language question answering (Q&A). However, due to the lack of training in the vertical field, the performance of LLM in the vertical field is not ideal. In addition, due to its high hardware requirements, training and deploying LLM remains difficult. In order to address these challenges, this paper takes the application of traditional Chinese medicine formulas as an example, collects the domain related data and preprocesses the data. Based on LLM and knowledge graph, a vertical domain Q&A system is designed. The system has the following capabilities: (1) Information filtering. Filter out vertical domain related questions and input them into LLM to answer. (2) Professional Q&A. Generate answers with more professional knowledge based on LLM and self-built knowledge base. Compared with the fine-tuning method of introducing professional data, using this technology can deploy large vertical domain models without the need for retraining. 3) Extract conversion. By strengthening the information extraction ability of LLM and utilizing its generated natural language responses, structured knowledge is extracted and matched with a professional knowledge graph for professional verification. At the same time, structured knowledge can be transformed into readable natural language, achieving a deep integration of large models and knowledge graphs. Finally, the effect of the system is demonstrated and the performance of the system is verified from both subjective and objective perspectives through two experiments of subjective evaluation of experts and objective evaluation of multiple choice questions. Lung cancer is the leading cause of cancer-related deaths worldwide, with metastasis being the primary cause of mortality in lung cancer patients, and its prevention and control efficacy remain limited. In recent years, immunotherapy has emerged as a promising direction for overcoming the bottleneck of metastasis. Macrophages, as essential components of innate immunity, participate in the entire process of tumor initiation and progression. Tumor-associated macrophages (TAMs) represent the most abundant immune population in the tumor microenvironment (TME), displaying both anti-tumor M1- like and pro-tumor M2-like phenotypes. The latter promotes tumor invasion and metastasis, angiogenesis, lymphangiogenesis, immune suppression, and reactivation of dormant disseminated tumor cells (DTCs), thereby facilitating tumor metastasis. In recent years, traditional Chinese medicine (TCM) has shown significant efficacy in inhibiting tumor metastasis and has been extensively validated. It exerts anti-tumor effects by reducing the recruitment of TAMs, inhibiting M2-like polarization, and modulating cytokines and proteins in the TME. This paper reviews the relationship between TAMs and lung cancer metastasis, elucidates the targets and mechanisms of TCM in regulating TAMs to prevent and treat lung cancer metastasis, aiming to provide insights into lung cancer prevention and treatment. In the past decades, systematic isolation and structural identification of chemical components from plant material, especially traditional Chinese medicine, have provided a large number of chemical drugs and lead structures for clinic. However, there are still a large number of trace components in traditional Chinese medicine with novel chemical structures and specific pharmacological activities, which need to be further analyzed. With the development of modern analytical technology, the network technology based on tandem mass spectrometry and cluster analysis shows many advantages in trace analysis of complex components. In this paper, a method of high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF MS) was developed for the analysis of the main chemical constituents in the root bark of Lycium barbarum L. The aqueous extract sample was separated by methanol aqueous through C18-solid phase extraction, and then the 50% methanol aqueous elution was analyzed using HPLC-Q-TOF MS with C18 column. The ions were monitored in both positive mode and negative mode of ESI with the scanning ranges of m/z 100-300 and 300-2 000. The trace chemical components were analyzed by molecular network technology. The MS data files were converted to the mzXML format and the molecular network was generated on GNPS, then visualized with Cytoscape, and the structures of compounds were predicted based on literature data. Twenty one known compounds in Cortex Lycii were verified by manual analysis technique and eleven new components were firstly identified by molecular networking. It is worth reporting that compound 24 and 25 are different from cyclooctapeptides previously found in the root bark of Lycium chinense (another source of Cortex Lycii), indicating that they can be used as markers for the identification of two species of Cortex Lycii. The composition of traditional Chinese medicine (TCM) is complex, and analysis of those chemical compositions is vital for the research of TCM. Mass spectrometric-based technologies, especially ultrahigh performance liquid chromatography-high resolution mass spectrometry (UHPLC-HRMS), are considered to be powerful techniques with the advantages of high sensitivity, high selectivity, rapidity, and high resolution. In recent years, approaches of MS data acquisition and data post-processing strategies were continuously evolving, including MSE, SWATH, mass spectral trees similarity filter (MTSF), and molecular networking (MN), facilitating the exploration and identification of TCM components. In this paper, the applications of mass spectrometric-based technologies and strategies for the analysis of TCM components would be discussed as well as a brief outlook on the perspectives of such technologies and strategies. The technique of microdialysis combined with liquid chromatography-mass spectrometry not only can be used for real-time and continuous minimally invasive sampling in vivo, but also has the characteristics of high sensitivity, fast analysis speed and wide detection range, which has become a powerful means for the research of Chinese medicine. It has been widely used in many aspects such as the study of the in vivo process and action mechanism of traditional Chinese medicine. In this paper, the microdialysis technology was briefly introduced, then combined with relevant research examples, the research strategy and application of microdialysis-liquid chromatography-mass spectrometry technique in the screening of active components from Chinese medicine, in vivo and in vitro metabolism, pharmacokinetics, metabolomics were reviewed. Development of materials with both medicinal and edible values has become part of the national health strategy and a growth point of a new round of industrial development， which is of great significance for health management with Chinese characteristics. As a pioneer in reform， innovation and development， Shanghai has always been at the forefront of the the field of medicine and health in China and even globally. This paper reviewed relevant progress on the use and management of materials with both medicinal and edible values at home and abroad. Furthermore， it investigated and analyzed the foundation， institutional guarantees， and advantages of management， as well as the current status of management in Shanghai. This study provides reference for the government to further improve its management policy of the catalogue of materials with both medicinal and edible values.
