We developed and evaluated two-level, namely 2017011 and 2017012, serum-based reference materials (RMs) for 17 beta-estradiol (17 β-E2) by the reference method of isotope dilution liquid chromatography tandem mass spectrometry (ID-LC-MS/MS) from the remaining serum samples after routine clinical tests, to help improve clinical routine testing and provide the traceability of results. This paper describes the development process of these RMs. The National Metrology Institute of Japan (NMIJ) certified reference material (CRM) 6004-a was used as the primary RM for the measurement of 17 β-E2. These serum-based RMs showed satisfactory homogeneity and stability. They also assessed the commutability between the reference method and the three routine clinical immunoassay systems. Besides, a collaborative study was carried out in five reference laboratories, all of which had been accredited by the China National Accreditation Service for Conformity Assessment (CNAS) in accordance with ISO/WD 15725-1. Statistical analysis of raw results and uncertainty assessment obtained certified values: 2017011 was 445.2 ± 39.0 pmol/L, and 2017012 was 761.9 ± 35.5 pmol/L. [ABSTRACT FROM AUTHOR]

At present, the National Metrology Institute of Japan provides six national primary pH buffers under the Japan Calibration Service System. Each batch of these buffers is certified by the primary pH method using a Harned cell. On the basis of these primary buffers, the designated laboratories supply the secondary and working pH standards using a high-precision pH meter. This paper provides an estimate of the batch-to-batch reproducibility of the primary pH standard production based on the history of the certification of primary carbonate buffers in NMIJ. This buffer, which was chosen as the subject of the study because of the relative difficulty of its measurements (and thus a greater dispersion of results), is nominally the 0.025 mol kg−1 equimolal solution of disodium carbonate and sodium hydrogen carbonate. As its pH value is significantly affected by the purity of the reagents used, the evaluation of their source materials is made by both pH measurements and acidimetric gravimetric back titrations. Considering the experimentally determined pH reproducibility of ca. 0.010, potential risks to the pH accuracy are discussed when using recipe-based carbonate pH standards. [ABSTRACT FROM AUTHOR]

The article discusses reports published in the issue which focuses on papers on forensic toxicology presented at the 50th Annual Meeting of The International Association of Forensic Toxicologists held in Japan including five paper with analytical focus.

The article discusses the highlights of 11th International Symposium on Biological and Environmental Reference Materials (BERM11) which was held from October 29 to November 2, 2007 at the Tsukuba International Congresss Center EPOCHAL in Tsukuba, Japan. The conference reflected the increasing involvement of the Asia-Pacific region in the development of certified reference materials. It was hosted by the National Metrology Institute of Japan (NIMJ). It was the first event on metrology in chemistry since the establishment of NIMJ in 2001.

In Part I of the present review series, I presented the current state of the water environment by focusing on Japanese cases and discussed the need to further develop microbial biosensor technologies for the actual water environment. I comprehensively present trends after approximately 2010 in microbial biosensor development for the water environment. In the first section, after briefly summarizing historical studies, recent studies on microbial biosensor principles are introduced. In the second section, recent application studies for the water environment are also introduced. Finally, I conclude the present review series by describing the need to further develop microbial biosensor technologies.Current water pollution indirectly occurs by anthropogenic eutrophication (Part I). In my 2010 review, I addressed conventional water analysis and biosensing of organic pollutants in Japan between 1960s and 2000s. It is now timely to reexamine current analytical and biomonitoring approaches in view of the new challenges in assessing pollution, particularly in closed water bodies, as pollutants tend to accumulate in these endorheic basins. In the present review series, I presented current water environment and its microbial biosensors. In this part, I presented current data of the water quality of these water bodies in Japan and established the need to further develop microbial biosensor technologies to address and monitor water quality here.Current water pollution indirectly occurring by anthropogenic eutrophication (Part I). A certified reference material, NMIJ CRM 7405-a, for the determination of trace elements and As(V) in algae was developed from the edible marine hijiki ( Hizikia fusiforme) and certified by the National Metrology Institute of Japan (NMIJ), the National Institute of Advanced Industrial Science and Technology (AIST). Hijiki was collected from the Pacific coast in the Kanto area of Japan, and was washed, dried, powdered, and homogenized. The hijiki powder was placed in 400 bottles (ca. 20 g each). The concentrations of 18 trace elements and As(V) were determined by two to four independent analytical techniques, including (ID)ICP-(HR)MS, ICP-OES, GFAAS, and HPLC-ICP-MS using calibration solutions prepared from the elemental standard solution of Japan calibration service system (JCSS) and the NMIJ CRM As(V) solution, and whose concentrations are certified and SI traceable. The uncertainties of all the measurements and preparation procedures were evaluated. The values of 18 trace elements and As(V) in the CRM were certified with uncertainty ( k = 2). The production and refinement of steel has followed very different paths in different parts of the Eurasian continent. In aiming to characterize the similarities and differences between various smelting and smithing methods, we have analysed steel samples from four different areas and historic periods: the Kotō Age in Japan (twelfth-sixteenth century), the Moghul Empire in India (seventeenth-nineteenth century), the Ottoman Turkish Empire (seventeenth century) and the late Middle Ages (fifteenth century) in Italy. The best quality steel was employed for forging arms and armour of high quality, so that we have selected samples from Japan, India, the Middle East and Italy belonging to such a category. Traditional methods, such as metallography, used to characterize different steels in terms of their carbon contents, microconstituents and slag inclusions, entailed an invasive approach. Since many of the selected artefacts are in a very good state of conservation, a different and non-invasive approach was desirable. To this aim, we have used time of flight neutron diffraction on the Italian Neutron Experimental Station diffractometer, located at the pulsed neutron source ISIS in the United Kingdom. By this technique, we were able to quantify the phase distribution of the metal phases, the slag inclusion content, and the oxidation state of the samples, both as average concentration on the whole artefact and in selected gauge volumes. Young lignite from two locations in Sendai City, Japan, were analyzed for polynuclear aromatic compounds (PACs). Lignite samples were extracted with toluene and PACs were isolated by a compound-class-selective, reactive matrix clean-up. This clean-up separates all compounds of polynuclear aromatic structure, for example hydrocarbons and ketones and their primary metabolites, from interfering organic compounds. The result of this isolation procedure is, therefore, a group of a large number of polycyclic compounds with different functional groups. Further analysis is done by glass capillary gas chromatography. Perylene was identified as the only compound obtained by the reactive matrix clean-up of lignite samples from both locations. Concentrations were in the range of 1–10 mg kg−1. Conversely, PACs resulting from particulate emissions from fossil fuel combustion always contain isomeric PAHs (polynuclear aromatic hydrocarbons) with some polycyclic aromatic ketones and thousands of primary and secondary metabolites at very low concentrations. This was demonstrated for a sample of urban air particulate matter, which is the source of PAC contamination of surface soil. The absence of accompanying PAHs and polynuclear aromatic ketones in the lignite samples confirms that perylene did not originate from a combustion process. It is assumed that the high concentrations of perylene are the result of a reduction of perylene quinone. Thus, the high perylene content of the lignite samples investigated is of biogenic origin. A new cod fish tissue certified reference material, NMIJ CRM 7402-a, for methylmercury analysis was certified by the National Metrological Institute of Japan in the National Institute of Advanced Industrial Science and Technology (NMIJ/AIST). Cod fish was collected from the sea close to Japan. The cod muscle was powdered by freeze-pulverization and was placed into 600 glass bottles (10 g each), which were sterilized with γ-ray irradiation. The certification was carried out using species-specific isotope dilution gas chromatography inductively coupled plasma mass spectrometry (SSID–GC–ICPMS), where 202Hg-enriched methylmercury (MeHg) was used as the spike compound. In order to avoid any possible analytical biases caused by nonquantitative extraction, degradation and/or formation of MeHg in sample preparations, two different extraction methods (KOH/methanol and HCl/methanol extractions) were performed, and one of these extraction methods utilized two different derivatization methods (ethylation and phenylation). A double ID method was adopted to minimize the uncertainty arising from the analyses. In order to ensure not only the reliability of the analytical results but also traceability to SI units, the standard solution of MeHg used for the reverse-ID was prepared from high-purity MeHg chloride and was carefully assayed as follows: the total mercury was determined by ID–ICPMS following aqua regia digestion, and the ratio of Hg as MeHg to the total Hg content was estimated by GC–ICPMS.
