Paper chromatography with new solvents applied to some commercial samples of thiazine stains, especially azures, resulted in improved resolution which gave more spots than had been obtained with previously reported solvents. To a Whatman No. 1 filter paper strip, 50 cm in length, 0.08 ml of a 0.1% solution in water was spotted 18 cm from the edge of the strip. The solvent used in most cases was a mixture of t-butylalcohol, methylcellosolve and a 1% NH4Cl solution in distilled water (60:20:20, v/v) with the ascending technique. Several batches of thionin were found to have a high degree of purity (concerning their content of coloured components). One sample of thionin (Chroma) contained only one component (violet). The thionin-SO2 reagent showed a chromatogram with more spots than the thionin sample from which the reagent was prepared. A paper electrophoresis method is described in which four serum lipoprotein components are separated without the use of albumin in the buffer. Tris-EDTA-boric acid buffer, Oil Red O staining solution prepared in 52.8% instead of the usual 60% alcohol, and solvent-extracted paper strips are the distinguishing features of this procedure. The system was effective not only in separating chylomicron, β- and α-lipoproteins into well-defined bands, but also in separating very low density or pre-β-lipoprotein distinctly in some samples and partially in others. The advantages of this procedure are low background staining and, in comparison to the procedure using albumin in the buffer, a sharper alpha band. This sharpened α-band gives a sharper peak in densitometric scanning. Electrophoresis could be performed for 16 hr or 4 hr. The 4-hr electrophoresis run produced electrophoregrams with even denser and sharper bands than the 16-hr run.

The problem of identification of botanical drug extracts is enormous in view of the large variety of botanical species as well as other factors affecting the biosynthesis of plant constituents. This is particularly true for those drugs which do not have a pharmacologically active ingredient or chemical component which can be assayed for in a routine manner. A rapid method is proposed which utilizes an ascending paper chromatographic technique with slight modification, ultraviolet light for observation, and the use of appropriate chromogenic spray reagents. This procedure produces a chromatogram consisting of base, stem, cone, and solvent front regions. The resulting characteristic picture, when viewed as a whole in comparison with a standard or control sample, can then be used for the identification and evaluation of the drug extract.

A simple paper chromatographic method was found to be effective for the study of phenylketonuria and tyrosyluria. It proved to be much more reliable than conventional tests for the detection of abnormal amounts of ketoacids in urine and was suitable and convenient for following the effects of dietary variations on the excretion of these compounds. Information obtained about ketones other than those of primary importance to the conditions studied included confirmation of the excretion of p-hydroxyphenylpyruvic acid in phenylketonuria. A modification of the method applicable to histidinaemia was devised.

Simple paper chromatography of urine obtained in the Figlu test has been found satisfactory for determining excess excretion of the imidazole metabolites of histidine. It has been confirmed that examination for these in addition to formiminoglutamic acid is necessary for a full assessment of this test. When excretion of the latter is not abnormal in folic-acid and/or vitamin B(12) deficiency, that of urocanic acid, a glycine conjugate of urocanic acid and/or imidazole propionic acid, is usually raised. An abnormally high excretion of hydantoin-5-propionic acid after histidine has also been found in these deficiencies, and its estimation may be of additional use in the evaluation of the Figlu test.

The possible risks of infection associated with the practice of licking envelopes, stamps and labels were investigated. Although pathogenic bacteria and viruses were not isolated from sample envelopes obtained from various sources, the gums used in manufacture were found to exert a protective effect against death from desiccation on the bacteria and viruses which had been introduced into them.Staph. aureus and Salm. paratyphi Bremained viable for several months in dried films of two out of four gums tested. An echovirus could be recovered from similar films for up to 30 days, and an adenovirus for up to 10 days.Bacterial multiplication occurred in the gum used for the manufacture of postage stamps. The comparison is drawn between the possible consequences of minimal contamination of this gum and of cans of corned beef in the light of recent studies.

Summary The electrophoretic pattern of serum proteins has been studied in 30 infants suffering from toxicosis. During the stage of acute dehydration the main changes were a moderate or marked increase of alpha globulin, particularly of the α2 fraction and a decrease of albumin. In some cases there was also an increase of the gamma globulin. During recovery the alpha globulin decreases while albumin increases. However, the return to normal of the electrophoretic pattern is not always achieved when clinical recovery occurs. The absolute amounts of total serum proteins have also been estimated and their significance has been discussed. Electrophorese sur papier des proteines du serum au cours de la toxicose. A PAPER Chromatographie study of urinary aminoacids has been carried out in 69 affected and 565 unaffected relatives from 64 families with various types of muscular dystrophy. Only a small proportion of subjects had either generalized or specific hyperaminoaciduria.

Birch pollen and dog epithelium allergen extracts were studied by ultracentrifugation in linear sucrose gradients. The various sucrose-gradient fractions were then tested for allergenic activity by the paper disc radioallergosorbent test, using sera from allergen sensitive patients.

Summary In 21 premature infants, the birth weights of whom ranged between 1500 and 2400 g and the ages between 1 and 56 days, the total serum protein values have been determined and the serum protein components have been electrophoretieally differentiated. All infants were fed on human milk but, in 10 infants additional protein was administered (Casein preparation Secoosan). Total protein values were found to be lower in premature infants than in full-term infants. During the first eight weeks of life total protein values decreased, from 5.63 g per cent to 4.03 g per cent. The decrease in total protein is caused by a decrease in the amount of albumin (from a little below 3 g per cent in the first week of life to a little above 2 g per cent in the eighth week) and of gamma-globulin (from about 1 g per cent to about 0.5 g per cent) while the alpha1-, alpha2-and beta-globulin values remain rather constant. If casein is added to the diet the total serum protein values and the serum albumin values will range at higher levels, but the decrease with increasing age will not be prevented. On the contrary, additional casein to the diet will cause a further reduction in the gamma-globulin concentrations (Fig. 1). Taux des proteines dans le serum des prematures. Dry wood specimens are sawed to 2mm thickness and impregnated with resin such as Lewisol 28 (Hercules Powder Co.). One side of the specimen is ground by hand on abrasive papers of grades #100, #180, #240, and #320. This side is then cemented to a petrographic glass slide with stick shellac and the other side similarly ground. Scratches can be eliminated by scraping the ground surface with the sharp edge of an ordinary glass microscopic slide. The section is removed by heating the slide, dissolving the shellac with alcohol and the resin of the embedding matrix with xylene. The sections can be stained in a hot saturated alcoholic solution of safranin O, counterstained with 0.01% fast green in an equal parts mixture of clove oil, methyl cellosolve, and absolute alcohol, and mounted in balsam.

A simple method of obtaining the ventilation rate in a thoron field and the relative non-equilibrium among the decay products of thoron is described. It is indicated how the ratio of the alpha activity after a delay of 150 min to that after a delay of 30 min can be used as a convenient parameter to do this. Curves are given showing the variation of the potential alpha energy values for thoron and radon daughters with the ventilation rate, which are useful when planning control by means of ventilation. SummaryA procedure has been developed whereby paper chromatograms of antiviral agents can be developed by applying them to virus infected, agar overlay, chick embryo cell tissue cultures. Plaque-free zones are produced, thus indicating the area on paper chromatogram where the active material can be located. Using antiviral substances antibiotic W-112 and isatin-monothiosemicarbazone, W. Nile and vaccinia viruses, and 4 chromato-graphic solvent systems, it was possible to localize the biological activity of these antiviral agents. As little as 1 μg of antibiotic W-112 produced large plaque-free zones when paper chromatogram was applied to the overlay culture 1 hr after virus infection. When 100 μg of same material was used, large plaque-free areas resulted even when the paper was applied 46 hr after virus infection. Isatin-monothiosemicarbazone at 50 μg level not only produced a plaque-free zone but also produced a yellow spot on paper chromatogram that coincided exactly with the zone of antiviral activity.

Paper tests for occult blood were assessed on faecal specimens from adults. An orthotolidine/sodium perborate and a modified orthotolidine/peroxide test were found to be reasonably satisfactory. The Hematest and Occultest tablets and a paper guaiac test were unsatisfactory.A large number of false positive results was obtained in children and infants. It is suggested that this might be due to meat derivatives reaching the faeces more easily in the young. Red cells labelled with either Cr(51) or Fe(59) were swallowed by human volunteers. Although between 45% and 73% of the red cell iron reappeared in the faeces, the peroxidase activity of the labelled material dropped six-fold on passage through the alimentary canal.

Twenty-one samples of 10 dyes were studied by paper chromatography, using Whatman #1 paper and developing by the ascending technique with 80% ethanol. Fifteen other Whatman papers were tested. Solvents faster than 80% ethanol (10% pyridine, 50% actone, 50% and 70% ethanol) were not useful. Slower solvents (isoamyl alcohol, isoamyl alcohol saturated with NH4OH, 90% acetone, acetone, 95% ethanol) were generally unsatisfactory but sometimes useful for extreme sharpness of spots produced in descending chromatograms. The use of filter paper in selective adsorption experiments, probably first described by Schonbein (1), has become a powerful tool in research, especially since the studies of Consden, Gordon, and Martin (2). Although in most modifications of paper chromatography only very small amounts of material can be resolved, Mitchell and his colleagues must be credited with preparative applications; recently these authors carried out successful enzyme separations in their chromatopile (3, 4).

The IgG M-components of 70 sera were quantitated by a paper electrophoretic method and by an immunochemical method with the use of an antiserum raised against purified polyclonal IgG. The results obtained by the two procedures were concordant and thereby showed that IgG M-components of different antigenic compositions can probably be quantitated by simple immunochemieal methods. Immunochemical determination of the total IgG content of sera containing IgG M-components by the Mancini technique agreed, both when the sera were assessed in their native and in their carbamylated form, with the immunochemical quantitation of Weeke. This indicates that different IgG M-components are immunochemically similarly altered by carbamylation.

ConclusionBy use of 30% Amberlite IRC 50 (W.A. 1) resin-exchange paper, it is possible to separate both insulin and glucagon independently from their mechanical mixtures with non-specific serum or gamma globulin.

Kuusinen (1969) compared the factor structures of individual subjects' ratings of their peers with the structure of peer ratings made by a group of subjects. He concluded that the group structure adequately represented the individual structures and that inter-individual differences were unimportant. This conclusion is challenged in the present paper. It is argued that the between-subject differences in Kuusinen's results were too pronounced to be dismissed as negligible. Data are also presented from a study in which subjects made personality ratings of stereotypic role figures. The factor structure underlying each subject's ratings was determined and compared with that of each other subject. The degree of congruence between the factor structures of pairs of subjects varied and in general only moderate convergence was observed. Implications of these individual differences for the concept of implicit personality theories were discussed.

A review of literature on the ocular toxicity of the anti-tuberculosis drug Ethambutol Hydrochloride. Ethambutol Hydrochloride is currently extensively used in tuberculosis therapy, so this review should be of interest to those who are concerned with patients under chemotherapy for tuberculosis.

SummaryDuodenal fluids from patients with cystic fibrosis of the pancreas and from control subjects were studied by paper strip electrophoresis. Components with greater electrophoretic mobility than any in normal duodenal fluid occurred consistently in cystic fibrosis duodenal fluids. The most rapidly migrating of these fractions was identical in electrophoretic mobility to the albumin fraction of normal human serum.

Summary and ConclusionsA method is presented for measuring Cortisol and corticosterone simultaneously in a single specimen of plasma. The steroids are purified by paper chromatography, eluted and measured by fluorometry. Losses incurred during analytical procedure are compensated by application of an isotope dilution correction. The normal range of corticosterone in human beings is 0 to 5 μtg/100 ml, with a mean of 1.3 ± 2.5 μg/100 ml. The normal range for Cortisol is 4.0 to 17.7 μg/100 ml, with a mean of 10.2 ± 3.6 μg/100 ml.

SummaryA technic for the two dimensional chromatography of blood plasma and protein mixtures on filter paper is described. Separations of protein constituents are greatly facilitated by the addition of surface active substances such as the “Tweens” or “Spans,” etc. The technic at present adopted is to add “Tween 85” or “Tween 81” and, using hemin as protein marker, to employ M/10 sucrose solution in the first dimension and M/10 sodium potassium tartrate solution in the second dimension. Differences between the protein patterns of chromatograms of human, rat and guinea pig plasma are noted. With protein mixtures, associations of proteins may occur that may be detected by chromatography.
